Two previously undescribed monoterpenoid indole alkaloids, N -formylyunnanensine (1) and scholaphylline (2), along with eight known alkaloids, namely, 19,20- E -vallesamine (3), 19,20- Z -vallesamine (4), 19,20- E -vallesamine N -oxide (5), 6,7- seco -19,20α-epoxyangustilobine B (6), 16 R -19,20- E -isositsirikine (7), N -demethylalstogustine N -oxide (8), E -vallesiachotamine (9), and Z -vallesiachotamine (10), were isolated from the bark of Alstonia scholaris. Alkaloid 1 was determined to be the N -formyl derivative of the known alkaloid, yunnanensine, comprising a pair of inseparable E / Z -formamide rotamers, while scholaphylline (2) was identified as the first member of the seco stemmadenine-yunnanensine type bisindole alkaloid. The structures of these alkaloids and their absolute configurations were established based on detailed analysis of the spectroscopic data in conjunction with the TDDFT-ECD method. A possible biogenetic pathway to 1 and 2 , involving stemmadenine as the primary precursor, was proposed. Scholaphylline (2) showed modest cytotoxicity against Panc1 (IC 50 27.7 ± 5.1 µM), MDA-MB-231 (IC 50 32.0 ± 2.2 µM), and MDA-MB-468 (IC 50 34.1 ± 3.3 µM) cancer cell lines, while N -formylyunnanensine (1) did not show appreciable cytotoxicity against any of the tested cell lines. Abstract The utilization of plants for the production of metallic nanoparticles is gaining significant attention in research. In this study, we conducted phytochemical screening of Alstonia scholaris (A. scholaris) leaves extracts using various solvents, including chloroform, ethyl acetate, n-hexane, methanol, and water. Our findings revealed higher proportions of flavonoids and alkaloids in both solvents compared to other phytochemical species. In the methanol, extract proteins, anthraquinone and reducing sugar were not detected. On the other hand, the aqueous extract demonstrated the presence of amino acids, reducing sugar, phenolic compounds, anthraquinone, and saponins. Notably, ethyl acetate and chloroform extracts displayed the highest levels of bioactive compounds among all solvents. Intrigued by these results, we proceeded to investigate the antibacterial properties of the leaf extracts against two major bacterial strains, Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). All extracts exhibited significant zones of inhibition against both bacterial isolates, with S. aureus showing higher susceptibility compared to E. coli. Notably, the methanol extract displayed the most potent I hibitory effect against all organisms. Inspired by the bioactivity of the methanol extract, we employed it as a plant-based material for the green synthesis of copper nanoparticles (Cu-NPs). The synthesized Cu-NPs were characterized using Fourier infrared spectroscopy (FT-IR), UV–visible spectroscopic analysis, and scanning electron microscopy (SEM). The observed color changes confirmed the successful formation of Cu-NPs, while the FTIR analysis matched previously reported peaks, further verifying the synthesis. The SEM micrographs indicated the irregular shapes of the surface particles. From the result obtained by energy dispersive X-ray spectroscopic analysis, Cu has the highest relative abundance of 67.41 wt%. Confirming the purity of the Cu-NPs colloid. These findings contribute to the growing field of eco-friendly nanotechnology and emphasize the significance of plant-mediated approaches in nanomaterial synthesis and biomedical applications.

Rheumatoid arthritis (RA) is a chronic autoimmune inflammatory disease leading to bone and joint damage. The current study reports the development of an herbal anti-arthritic gel formulation incorporating the latex of Alstonia scholaris (AS). Six batches of topical herbal gels (G1-G6) were formulated by dispersion method using different concentrations of AS latex (0.5 % to 3.0 % w/w) in Carbopol 934 (1.5% w/w) gel base. The gels were evaluated for various physicochemical properties using standard methods and G4 was selected as an optimized formulation based on the desirable properties of a topical gel. The in vivo anti-arthritic potential of G4 was evaluated using a complete Freund's adjuvant (CFA)-induced arthritic rat (Sprague Dawley) model for 28 days. Assessment of various arthritic parameters including hematological and biochemical parameters and radiography of hind legs in arthritic rats was carried out. The developed herbal gels were found to be uniform and stable. The G4 gel formulation showed a maximum drug release of 95.22% following Higuchi kinetic model. The topical application of G4 gel exhibited significant (p<0.05) anti-arthritic activity compared to diseased rats. The herbal gel G4 effectively restored the arthritic, hematological and biochemical parameters in arthritic rats. Radiographic analysis demonstrated its ability to reduce soft tissue swelling and thus prevented bone degeneration and the narrowing of joint spaces. These results supported that the developed herbal gel may offer a novel therapeutic approach in the treatment of RA with fewer side effects, good patient compliance, high efficacy, and low cost. The biosorption of lead (Pb) ions on Alstonia scholaris fruit biomass was investigated in batch and column experiments in an aqueous solution, emphasising the possible binding mechanism of Pb ions on the fruit biomass. The adsorption process was found to be dependent on parameters like adsorbent dose, initial Pb ion concentration, contact time, and pH. No impact was found on Pb adsorption during competitive adsorption. The results suggested that the fruit biomass could remove more than 97% of Pb ions from the aqueous solution. The Freundlich isotherm model best explained the obtained results. The monolayer adsorption capacity of the A. scholaris fruit biomass was 50 mg g-1. The reaction rate was fast initially with most Pb adsorption taking place during the first 80 minutes, while the equilibrium was attained at 420 minutes. Scanning electron microscopy revealed changes in the surface texture of A. scholaris fruit biomass after adsorption of Pb. The biomass’s surface area was determined using Brunauer-Emmett-Teller technique. Desorption, Fourier transform infrared spectroscopy, and functional group modification revealed that the bonding of Pb ions on Alstonia scholaris fruit biomass involved complex mechanisms.

Serum ALT and triglyceride were significantly decreased in all treatment groups. Serum glucose was also affected by the extract. The results revealed that A. scholaris flower extract exhibits a beneficial effect on C. striata growth and health. The phytochemical study was carried out on the ethanolic extract of the leaves of Alstonia scholaris. This plant has been used traditionally for medicinal purposes in the treatment of several diseases such as cancer, jaundice, malaria, gastrointestinal troubles, dysentery, snake-bite and diarrhea. Chromatographic purification of this extract led to the isolation and characterization of nine compounds. Their structures were elucidated as betulin (1), α-amyrin acetate (2), mixture of β-sitosterol (3) and stigmasterol (4), tetratriacontyl-trans-p-coumarate (5), ursolic acid (6), picralstonine (7), β-sitosterol glucoside (8) and scholaricine (9). Scholaricine and β-sitosterol-glucoside exhibited potent lipoxygenase inhibitory activity with IC50 of 15.2 ​± ​0.41 and 18.5 ​± ​0.14 respectively compared to Baicalein (22.6 ​± ​0.08) used as positive control. Molecular docking studies were conducted to support lipoxygenase inhibitory activities of the test compounds.

In the current study, we evaluated the efficacy of Ayush-64 (A64), a polyherbal formulation containing Alstonia scholaris (L.) R. Br. (A. scholaris), Caesalpinia crista L. (C. crista), Picrorhiza kurroa Royle ex Benth (P. kurroa), and Swertia chirata (Roxb.) H. Karst. (S. chirata) against COVID-19 in a Syrian hamster infection model. Preventative use of A64 resulted in the late-phase recovery of body weight loss in severe acquired respiratory syndrome coronavirus-2 (SARS-CoV-2)-infected hamsters, suppression of pro-inflammatory cytokines, and blunted pulmonary pathology. In addition, we also investigated the efficacy of individual ingredients of A64, viz., A. scholaris, C. crista, P. kurroa, and S. chirata, in the hamster model. The hamster challenge data showed robust anti-viral and immunomodulatory potential in A. scholaris, followed by P. kurroa. However, C. crista and S. chirata of A64 showed prominent immunomodulatory potential without limiting the lung viral load. In order to better understand the immunomodulatory potential of these herbal extracts, we used an in vitro assay of helper T cell differentiation and found that A. scholaris mediated a more profound suppression of Th1, Th2, and Th17 cell differentiation as compared to A64 and other ingredients. Taken together, our animal study data identifies the ameliorative potential of A64 in mitigating coronavirus disease-19 (COVID-19) pulmonary pathology. A. scholaris, a constituent extract of A64, showed relatively higher anti-viral and immunomodulatory potential against COVID-19. Background: The different parts of Alstonia scholaris (AS) viz. leaf, bark root, and latex are used traditionally to treat pain, swelling, stiffness, immune deficiency, malaria, ulcer, and arthritis. The traditional healers in Dhemaji district of Assam prescribe raw AS latex for topical treatment of inflammatory diseases. The unique feature of this study is that the AS latex has been evaluated scientifically for the first time for its anti-inflammatory properties. Objectives: The present investigation evaluates the anti-inflammatory potential of the AS latex in arthritis using in vitro assays. Materials and Methods: Anti-arthritic potential of AS latex was evaluated using in vitro assays of bovine serum albumin (BSA) denaturation, egg albumin denaturation, and human red blood cell (HRBC) membrane stabilization. Results: It was observed that the standard diclofenac sodium and the AS latex showed good anti-arthritic potential. In BSA denaturation method, the latex, and diclofenac sodium showed 71.59% and 85.23% inhibition of proteinase enzyme at 250 μg/mL, respectively. In the egg albumin denaturation method, the latex, and standard showed the maximum percentage of inhibition of protein denaturation of 70.42% and 81.32%, respectively, at 250 μg/mL. In the HRBC membrane stabilization model, the latex produced considerable anti-arthritic potential in a concentration–dependent manner. At 250 μg/ mL in hypotonic solution, latex, and diclofenac sodium showed 68.77% and 72.12% membrane stabilization, respectively. The habitation and environment are affected by the stable isotopes of caesium (Cs) and strontium (Sr), as well as by their radioactive isotopes. The current work gives insight on Alstonia scholaris' capacity to phytoextract stable caesium (Cs) and strontium (Sr), as well as the plant's ability to protect against the toxicity of both elements. Experiments with Cs [0–5 mM (CsCl)] and Sr [0–3 mM (SrCl2. 6H2O)] dosing in controlled light, temperature, and humidity condition in greenhouse for 21 days were undertaken. Cs and Sr accumulation in different plant parts was quantified with atomic absorption spectroscopy (AAS) and inductively coupled plasma-optical emission spectrometry (ICP-OES) respectively. Hyper-accumulation capacity for Cs and Sr was estimated with indices like transfer factor (TF) and translocation factors (TrF). The uptake pattern of caesium in Alstonia scholaris is 5452.8–24,771.4 mg/kg DW (TF = 85.2–57.6) and in the case of Sr is 1307.4–8705.7 mg/kg DW (TF = 85.3–1.46). The findings demonstrated the plant's ability to transfer Cs and Sr to aboveground biomass on the basis of dry weight, with the majority of the metals being deposited in the shoot rather than the root portion of the plant. For Cs and Sr, with increasing concentration, the plants exhibited the enzymatic expression for defence against metal toxicity by free radicals compared to control. Field emission electron microscopy with energy-dispersive spectroscopy (FESEM with EDS) was employed to assess the spatial distribution of Cs and Sr in plant leaf, indicating the accumulation of Cs, Sr, and their homologous components. Cement production, quarrying and stone crushing are major emission sources of alkaline dust that can affect human health and vegetation. The main objectives of this study were to evaluate the potential of using bark pH, soil pH and lichen community for indicating alkaline dust pollution. Twelve polluted sites were in a limestone industrial area. Bark pH and the lichen community were observed on Alstonia scholaris trees, and soil pH was obtained from topsoil samples. The bark pH at all polluted sites was significantly higher (5.5 to 7.3) than that at the unpolluted site (4.3). Among the polluted sites, the highest bark pH value was observed at the nearest site to the center of the industrial area, while the lowest value was discovered at the farthest site. Bark pH showed a strongly negative correlation with the distance from the center. Soil pH at the unpolluted site (6.3) was also significantly lower than that at the polluted sites (7.6 to 8.1), except at the farthest site (6.5). The soil pH also tended to increase closer to the center. Seven lichen species were observed on the trunks of investigated trees in all polluted sites and were observed only at sites more than 4.7 km away from the center, where bark pH ranged from 5.5 to 6.3. The extent of dust impact on vegetation seemed to be within 6–7 km from the center. The results of this study confirm the potential of the bark pH of A. scholaris, soil pH and lichen community as long-term indicators of alkaline dust pollution. [ABSTRACT FROM AUTHOR]

Alstonia scholaris is a perennial evergreen tree of family Apocynaceae and one of the graceful ornamental and medicinal plants. In general, this tree suffers the problem, that is, gall formation on a leaf. Most probably, it is caused by leaf gall insects (Pauropsylla tuberculata) belonging to order Hemiptera and is commonly known as jumping lice. The insect has five nymphal stages and forms the gall by sucking the plant sap. It lacks the pupal stage. Such a heavy attack by this insect affects the rate of photosynthesis and overall growth of plants. Psoriasis, a widespread skin condition impacting over 100 million individuals globally, is characterised by uncontrolled hyperproliferation of keratinocytes, abnormal apoptosis, and excessive secretion of inflammatory cytokines and angiogenic factors. Traditional use of Alstonia scholaris (L.) R.Br. , Wrightia tinctoria (Roxb.) R.Br. and Solanum xanthocarpum Schrad. & Wendl. in Ayurveda and Siddha medicinal systems have shown promising anti-inflammatory and wound-healing properties. However, underlying mechanisms of their phytoactivity in addressing psoriasis-like skin inflammation on human keratinocytes remain largely unexplored. The study was aimed to investigate anti-psoriatic potential of ethyl acetate and ethanolic extracts of A. scholaris, W. tinctoria and S. xanthocarpum in human keratinocyte cell line (HaCaT). Ethyl acetate and ethanolic extracts of A. scholaris (ASEA and ASE), W. tinctoria (WTEA and WTE) and S. xanthocarpum (SXEA and SXE) were first subjected to phytochemical screening through high-performance liquid chromatography (HPLC) using their marker compound loganin, kaempferol and chlorogenic acid, respectively. The proliferation inhibition efficiency of these extracts was measured using MTT assay on HaCaT cell line. Subsequently, the apoptotic effect of these extracts on HaCaT cell line was determined by JC-1 and Annexin V assays. Furthermore, IL-8 and RANTES levels were measured in TNF-alpha-induced HaCaT cell line post-treatment with these extracts to determine their anti-inflammatory properties. ASEA, ASE, WTEA, WTE, SXEA and SXE significantly inhibited proliferation of keratinocytes (HaCaT cells) and resulted in the induction of apoptotic markers (mitochondrial membrane potential and phosphatidyl serine externalization). Additionally, pro-inflammatory markers (IL-8 and RANTES levels) were downregulated in HaCaT cells. The anti-proliferative effects were particularly distinct at higher concentrations (200 μg/mL), with inhibition rates reaching over 85% for W. tinctoria and S. xanthocarpum extracts. In apoptotic assays, notable increases in late apoptotic or necrotic cell populations and significant losses in mitochondrial membrane potential were observed. All extracts markedly reduced the secretion of inflammatory mediators IL-8 and RANTES. All three plants exerted an anti-psoriatic effect at the cellular level via multiple parameters (anti-proliferative, pro-apoptotic, anti-inflammatory effect). This study provides insight into the mechanism of action of ASEA, ASE, WTEA, WTE, SXEA and SXE and highlights their promising potential for development as herbal therapeutic agents for psoriasis. Monoterpene indole alkaloids (MIAs) are valuable metabolites produced in numerous medicinal plants from the Apocynaceae family such as Alstonia scholaris, which synthesizes strictamine, a MIA displaying neuropharmacological properties of a potential importance. To get insights into the MIA metabolism in A. scholaris, we studied here both the spatial and transcriptional regulations of MIA genes by performing a robust transcriptomics analysis of the main plant organs, leaf epidermis but also by sequencing RNA from leaves transiently overexpressing the master transcriptional regulator MYC2. These transcriptomic studies notably demonstrated that the first steps of the MIA pathway are successively distributed in the internal phloem associated parenchyma and epidermis, and that MYC2 exerts a remarkable transcriptional effect by modulating the expression of around 1000 genes. By combining these distinct datasets, we initiated the search for MIA-related genes encoding CYP71, based on the similarity of expression compared to already known MIA genes. Transient expression of these candidates in Nicotiana benthamiana leaves and yeast notably led to the identification of a related isoform of rhazimal synthase (RHS) capable of converting the MIA precursor geissoschizine into akuammicine, strictamine and 16-epi-pleiocarpamine. Investigating its catalytic mechanism revealed that strictamine results from rhazimal deformylation and that a similar mechanism may also explain 16-epi-pleiocarpamine synthesis. This prompted us to rename these enzymes geissoschizine cyclase due to their capacity of cyclizing geissoschizine into three different MIA scaffolds and to form both C-C and C-N bonds. Plant-insect interactions where the insect changes the course of host-plant growth by building a new structure called as a "gall" to protect and nourish the growing insect continue to fascinate scientists. Alstonia scholaris, a medicinal plant extensively used in Indian Traditional Medicine has shown to have such interactions with insect Pauropsylla tuberculata Crawford producing numerous galls on the plant leaves. This intricate and poorly understood natural relationship demands further investigation to unravel the distinctions between gall tissue and unaffected leaves. We examined the tissue ultrastructure by scanning electron microscope (SEM) and phytochemistry by thin layer chromatography and liquid chromatography - high resolution mass spectrometry to understand variations between the gall tissues and ungalled leaves of A. scholaris. The data showed dense population of sclereids in the gall tissues in addition to parenchyma cells. The concentration of aluminium, phosphorus and carbon were found to be reduced in the galls while cobalt and oxygen were increased. The SEM images gives insight into the ecological relationship between these species. The present study details the synthesis and characterisation of iron (Fe) doped activated carbon from Alstonia Scholaris (AS-Fe) natural wood waste. Investigation addresses the utilization of natural wood waste for useful and potential high temperature Alstonia Scholaris activated carbon (HT-AS). Iron doped activated carbon (AS-Fe) is used for the treatment of industrial waste water. Activated carbon and utilization performances are well attributed to the preparation methods and hence a range of characteristic interpretation like Fourier transform spectroscopy (FTIR), X-Ray powder diffraction, UV-Vis spectra, Field emission Scanning Electron Microscopes (FE-SEM) and EDAX analysis are evaluated. The result of AS-Fe is well distinguished by comparing the features with the porous high temperature Alstonia Scholaris activated carbon (HT-AS). This work includes green synthesis of magnesium oxide nanoparticles (MgO NPs) by using Alstoniascholaris, which is indigenous to many countries such as China, Australia, Sri Lanka, Pakistan, and India. Its pharmacological activities include antidiabetic, antioxidant, anticancer, analgesic, antitussive, and anti-diarrheal activities. In this study, the antioxidant and anti-inflammatory activities of bio-inspired magnesium oxide nanoparticles, MgO NPs, were investigated. MgO NPs were prepared by using the leaf extract of Alstonia scholaris, followed by characterization using EDX, XRD, and SEM techniques. The crystallite size of magnesium oxide nanoparticles was 19.57 nm. XRD analysis confirmed the crystallinity and the purity of MgO NPs. Anti-inflammatory activity was carried out to observe inhibition of protein denaturation. Since the IC50 of MgO nanoparticles was lower than the standard, it was found to be more effective. IC50 values were compared, and results reveal that bioinspired MgO NPs undergo more scavenging of free radicals than standard (ascorbic acid) MgO NPs. These MgO nanoparticles are useful in cosmetics such as scrubs, moisturizers, and an active ingredient in microdermabrasion and in formulating effective drugs for maintaining the protein structure of the body, which will reduce inflammation.

5,6-Benzoflavone (1), a new natural product, has been isolated from the EtOH extract of the flowers of Alstonia scholaris (L.) R. Br. (Apocynaceae), along with the known compound dicoumarol (2). Phytochemical studies on the fruits also provided bergenin (3), 9,19-cycloart-23-ene-3,25-diol (4), 11-oxo-α-amyrin acetate (5), and marsformoxide B (6). Compounds 2–6 have been isolated for the first time from the genus Alstonia. Their structures were elucidated by spectroscopic techniques, including two-dimensional NMR. Biological screening of the isolates revealed the significant urease inhibitory potential of compounds 1–6 while 4–6 also showed significant antibacterial activity against S. typhi and P. mirabilis. A polyphenolic flavone Luteolin (3',4',5,7-tetrahydroxyflavone) is found in various plants and is traditionally used in Chinese medicine. It is obtained from Alstonia scholaris (L.) R.Br Flower belonging to the family Apocynaceae while investigation. Various studies have been demonstrated the antioxidant or antiulcer potential of luteolin from different plant sources. In the present investigation the antioxidant or antiulcer effect of the Luteolin has been carried out using molecular docking simulations. The objective of this study was to analyze the antioxidant and antiulcer potential of luteolin obtained during isolation. The in vitro biological evaluation has been supported by the in silico studies using Autodock vina 4 shows the ligand-protein interaction of lute olin with 1HD2, 4GY7 and 3O1Q. Luteolin showed significant DPPH scavenging and urease inhibition activity i.e., 23.4 ± 0.87, 6.21±0.45 IC50 (uM) respectively as compared to the standard BHA and thiourea 44.2±0.45, 22.4±0.29 IC50 (uM) respectively. The docking simulations showed significant binding pocket sites with the respective proteins1HD2, 4GY7 and 3O1Q with the least binding energy -6.8, -8.0 and -8.2 kcal/mol respectively. Thus, Strong evidence has been presented with their confirmation structural interaction via molecular docking with proteins that serve as binding sites for available Luteolin molecule. Abstract Alstonia scholaris (L.) R. Br., an evergreen tropical plant rich in indole alkaloids with significant physiological activity, is traditionally used to treat respiratory diseases in China. This study was conducted to establish the toxicity profile of the alkaloid extract (TA) of A. scholaris leaves in non-rodents. After oral administration of a single dose (4 g/kg.bw), a number of transient symptoms, such as unsteady gait, drooling, emesis, and reddening of peri-oral mucosa, were observed, but no treatment-related mortality. A sub-chronic toxicity study with a range of doses of TA (20, 60 and 120 mg/kg.bw) was conducted for a 13-week treatment period, followed by 4-week recovery observation. Except for emesis and drooling in majority of animals in 120 mg/kg.bw treatment group, no clinical changes were observed in TA-treated animals. Data from electrocardiography, bone marrow, urine, fecal, hematology and clinical chemistry analyses were comparable between TA-treated and control animals. No significant differences in the relative organ weights and histopathological characteristics were evident between the TA-treated and control groups. Accordingly, the non-observed-adverse-effect-level (NOAEL) of TA was established as 120 mg/kg.bw. Our results add further knowledge to the safety database for indole alkaloid extracts from A. scholaris with potential utility as novel drug candidates. Graphic Abstract

Abstract Indole alkaloids extract (IAAS) was prepared from leaves of Alstonia scholaris (L.) R. Br., an evergreen tropical plant widely distributed throughout the world. This plant has been used historically by the Dai ethnic people of China to treat respiratory diseases. This study evaluated the genotoxicity and safety pharmacology of IAAS to support clinical use. The bacterial reverse mutation (Ames) test, in vitro mammalian chromosomal aberration test, and in vivo mammalian erythrocyte micronucleus (MN) test were performed to evaluate genotoxicity. Mice were administered IAAS (240, 480, or 960 mg/kg bw) once orally to observe adverse central nervous system effects. Furthermore, beagle dogs were administered IAAS (10, 30, 60 mg/kg bw) once via the duodenum to evaluate its effects on the cardiovascular and respiratory systems. IAAS with or without S9-induced metabolic activation showed no genotoxicity in the Ames test up to 500 μg/plate, in the mammalian chromosomal aberration test up to 710 μg/mL, or in the MN test up to 800 mg/kg bw. No abnormal neurobehavioral effects were observed in mice following treatment with up to 960 mg/kg bw of IAAS. Moreover, blood pressure, heart rate, electrocardiogram parameters, and depth and rate of breathing in anesthetized beagle dogs did not differ among the IAAS doses or from the vehicle group. These data indicated that IAAS did not induce mutagenicity, clastogenicity, or genotoxicity, and no pharmaco-toxicological effects were observed in the respiratory, cardiovascular, or central nervous systems. Abstract Alstonia scholaris (L.) R. Br. (Apocynaceae) is an evergreen tree that has been used to treat lung diseases. In this study, the toxicity profile of indole alkaloids from leaves of A. scholaris was investigated. In acute toxicity tests, mice were administered total alkaloids (TA) and five indole alkaloids. In a chronic toxicity test, rats were continuously administered TA (50, 100, and 300 mg/kg bw) for 13 weeks, followed by a 4-week recovery. A single administration of TA affected the behavior of mice, and at 12.8 g/kg bw, prone position, shortness of breath, wheezing, and convulsion were observed. The half-lethal dose (LD50) in mice was 5.48 g/kg bw, almost 2740 times the clinical dose in humans. Among the five indole alkaloids, the maximum tolerance dose in mice ranged from 0.75 to 4 g/kg bw. The TA-treated rats did not die and showed no adverse effects or dose-dependent changes in weight or food and water consumption, despite fluctuations in hematological and biochemical parameters compared with historical data. Furthermore, both gross and histopathological observations revealed no abnormalities in any organ. With daily oral administration to rats, the non-observed-adverse-effect-level of TA was 100 mg/kg bw. The results indicate that TA is safe for clinical use. Graphic Abstract

The incredible benefits of Alstonia scholaris are piquing researchers' attention in extracting its cellulose and utilizing it in further therapeutic applications. This study is based on cellulose extraction from its stalks and processed through chemical pre-treatments to manifest its cellulose content by using different bleaching reagents. A comparison was made on efficiencies of three reagents and it is found that the hydrogen peroxide exposed maximum cellulose than sodium hypochlorite and sodium chlorite. The experimental results revealed that A. scholaris possess 68–70% cellulose content. FTIR spectrum shows that OH- and CH- vibrations of cellulose appeared at 3320 cm−1 & 2892 cm−1 respectively whereas SEM images show fibrillation, rough surface, and lumens in bleached fiber that attributes to the removal of lignin and hemicelluloses and confirms cellulose extraction. The XRD pattern certifies the crystalline nature and compactness of cellulose whereas tensile properties and TGA help in understanding its flexibility, mechanical strength, and thermal stability at 370 °C respectively. Alstonia scholaris is an important indole alkaloid rich medicinal plant with diverse pharmacological activity. To understand the effect of extraction techniques, the stem bark sample of A. scholaris was subjected to continuous hot percolation, ultrasonic extraction, and cold maceration techniques. Continuous hot percolation technique extractive exhibited a potential pancreatic lipase (PL) inhibitory activity and further bio-assay guided fractionation resulted in the isolation of echitamine with a PL inhibitory activity (IC50 = 10.92 µM). A new validated HPTLC-HRMS method was developed for the quantification of echitamine by using a mobile phase of chloroform: methanol (80:20, v/v) with 0.04% formic acid. Echitamine content in the individual extractives were in direct correlation with the PL inhibitory activity (Pearson's r = −0.9409). The molecular docking studies further confirmed the PL inhibitory potential of echitamine. Tuberculosis is the second most common cause of death from infectious disease. Current antituberculosis treatments consist of long course of combination of antibiotics and toxic side effects and lead to poor patient compliance. So, the present research work has been undertaken for the development and standardization of polyherbal formulation for tuberculosis. In the present study polyherbal formulation was developed using powder of plants of Ocimum sanctum Linn, Tinospora cordifolia (Willd.) Miers, Alstonia scholaris (L.) R.Br and Trikatu churna. The phytochemical screening was performed for presence of various phytoconstituents. Thin Layer Chromatography densitometric methods were developed using High-Performance Thin-Layer Chromatography for the quantifications of gallic acid and piperine in formulation and found to be 0.049 % and 0.051 % respectively in formulation. In vitro antituberculosis activity was performed using Lowenstein Jensen Medium and percentage inhibition at 50 µg/ml, 500 µg/ml and 1000 µg/ml was found to be 11.71 %, 74.15 % and 94.11 % respectively. Results showed dose dependant in vitro antituberculosis activity. These activities may be due to the presence of various phytoconstituents like flavonoids, tannins and phenoilc in formulations. Nine previously unreported various types of monoterpenoid indole alkaloids, together with seven known analogues were isolated from the stem barks of Alstonia scholaris through a silica gel free methodology. The structures of 1 – 9 were elucidated by spectroscopic data analysis, electronic circular dichroism calculations, and single-crystal X-ray diffraction. Compound 1 is a modified echitamine-type alkaloid with a novel 6/5/5/7/6/6 hetero hexacyclic bridged ring system, and 8 and 9 exist as a zwitterion and trifluoroacetate salt, respectively. The anti- Toxoplasma activity of all isolates on infected Vero cells were evaluated, which revealed that compound 14 at 0.24 μM displayed potent activity. This study expanded the structural diversity of alkaloids of A. scholaris , and presented their potential application in anti- Toxoplasma drug development. The production of environmentally friendly and long-lasting supercapacitor electrodes employing activated carbons obtained from industrial wood waste from Alstonia Scholaris by traditional chemical activation using H 2 SO 4 at High temperature activation is shown in this paper. The new traits of metal oxide (Fe 2 O 3) doped Alstonia Scholaris wood derived activated carbon employing a slow precipitation technique. The produced activated carbon and the Fe 2 O 3 doped activated carbon samples were examined using a variety of analytical methods, including XRD, FESEM, TEM-HRTEM, elemental mapping, N 2 adsorption-desorption, Raman, XPS, and VSM. The BET result of prepared Fe 2 O 3 doped activated carbon exhibits high surface area of 443 m2g−1. However, the assembled ASC cell exhibits high energy density of 38 Wh/kg and power density of 3181 W/kg. Two new analogues of alstoscholarisine A, containing a phenyl or butyl substituent at the C-19 position, have been prepared in racemic form from the known skatole derivative. The syntheses of these compounds were accomplished in 13 steps, with a late-stage formation of the C-19 stereocenter. These derivatives are expected to have significantly changed biological activity, compared to alstoscholarisine A – a potent neuroactive natural product. Development of acaricidal resistance and environmental pollution has driven need for eco-friendly pesticides and herbal acaricides. Leaf extracts of Alstonia scholaris and Sida cordifolia in four different solvents were used in the range of 0.25-8.0% for larval immersion test against unfed larvae of Hyalomma anatolicum. Mortality rates of larval ticks were in dose-dependent manner ranged from 0.5 to 100% for A. scholaris and from 0.8 to 100% for S. cordifolia. Acetone extract of A. scholaris and methanolic extract of S. cordifolia were most effective against larval ticks with LC50 values of 0.71% and 0.42%, respectively. Significant larvicidal activity of A. scholaris and S. cordifolia leaf extracts against unfed larvae of H. anatolicum qualify them as green pesticides which could be combined with other tools for integrated pest management.

Alstonia scholaris could be used as a traditional medicinal plant in China for the treatment of acute respiratory, which might be caused by respiratory tract infections. The investigation tested the anti-infective effects of total alkaloids extract (TA) from leaves of A. scholaris, and as a result, TA inhibited herpes simplex virus type 1 (HSV-1), respiratory syncytial virus (RSV) and influenza A virus (H1N1) in vitro respectively. In addition, the survival days of mice were prolonged, and the lung weights and mortality of mice were decreased significantly, after oral administrated TA in H1N1 and beta-hemolytic streptococcus infectious models in vivo respectively. Five natural extract sources were acacia bark (Acacia spp.), pulai bark (Alstonia scholaris), kumis kucing leaves (Orthosiphon spp.), mimba leaves (Azardirachta indica), and gadung tubers (Dioscorea spp.). Two non-polar solvents, i.e., n-hexane and petroleum ether were used for five natural source extractions following ASTM soxhlet extraction. The research showed that triterpene and fatty acid derivatives were the major compounds present in five natural extracts. Introduction: Urolithiasis has afflicted human race since the days of yore. Conventional treatment acts only on associated ailments whereas pharmacologically active phytoconstituents derived from plants are reported to possess lithotriptic as well as anti-urolithiatic effect, thus herbal sources need to be scientifically explored. Present study was aimed to evaluate anti-urolithiatic potential of leaves of Alstonia scholaris and its isolated pentacyclic triterpenoids: lupeol and ursolic acid in ethylene glycolinduced renal calculi rat model. Materials and Methods: Leaves of Alstonia scholaris were extracted with various solvents, amount of triterpenoids in each extract was quantified using HPTLC. Extract which had higher amount of triterpenoid was evaluated for its anti-urolithiatic effect. Albino wistar rats of either sex were divided into 6 groups. All groups except vehicle were fed with 0.75%v/v of ethylene glycol for 28 days. Treatment with standard cystone, low and high dose of extract and pentacyclic triterpenoids were given to respective groups from 4th-28th day of study. On 28th day, serum creatinine, uric acid, BUN, TNF-α, IL-6 and urinary electrolyte levels were estimated. Kidneys were isolated, one was used for histopathological study and other for antioxidant assays. Results and Discussion: Histopathological examination of kidneys revealed destructed morphology due to deposition of large polymorphic crystals and cystic dilatation of tubules. Whereas treatment with extract and its pentacyclic triterpenoids prevented renal damage and deposition of crystals. Perturbations in levels of nitrogenous wastes, inflammatory mediators and urinary electrolytes were also prevented. Alstonia scholaris rich in triterpenoid significantly arrested associated oxidative stress and inflammation thus preventing formation of renal calculi. Endophytes are a group of bacteria live in plant tissues, and have been numerously reported to promote plant growth. The aim of this study was to observe bacterial endophytes from blackboard tree (Alstonia scholaris) in inhibiting pathogenic fungal growth on chili plants, promoting plant growth and producing indole acetic acid, as well as in solubilizing phosphate. Sixteen endophytic bacteria were isolated from blackboard tree root and leaf samples, in which two, A1 and A3, out of sixteen showed to highly inhibit pathogenic fungi Fusarium sp. and Colletrotichum sp., to produce indol acetic acid, and to solubilize phosphate. 16S rRNA gene sequenceshowed that A1 was relativey closed to Carnobacterium gallinarum strain DSM4847 and A3was relativey closed to Carnobacterium maltaromaticum strain JCM 1154 with similarity of 84 and 78%, respectively.

Infestation of leaf galls can be seen on many plants. One such incidence of infestation has been observed from the various localities of Udalguri district of Assam, where the galls were prominently seen on the leaves of Alstonia scholaris tree. The whole study is based on the infestation of gall fly, Pauropsylla tuberculata on the Alstonia tree. Alstonia scholaris in Udalguri districts are found mostly on road sides. They are basically used in the manufacturing of pencils because of their rapid growth. The observations and identification of the gall fly was based on the survey of various places of Udalguri district to get a better idea about its prominence and population density. The study includes the observation and identification of the infestation done by the fly, its feeding habitats, various life stages and various morphological characters as well. Aim: Immunomodulatory Effect of Isolated Triterpenes from Alstonia scholaris Linn. in Wistar Albino Rats. Material & Methods: The flower part of Alstonia scholaris were extracted by using n-hexane, dichloromethane, methanol, and water. All the extracts were screened for the presence of various active compounds. DCM extract showed the occurrence of terpenoids and flavonoids. The methanolic extract showed the presence of flavonoids and other active components. DCM extract was then allowed to isolate the active compound by the means of column chromatography. N-hexane and dichloromethane solvents in various ratios were used for the isolation of active compounds. Then isolated compound was identified by the various spectral analysis techniques. The isolated compound was evaluated by cyclophosphamide and E. Coli model. Results: In the case of cyclophosphamide induced myelosuppression, there was a decrease in the WBC reckoning in the control group. In treatment groups, the WBC count was found to be augmented with p<0 .01and p< 0.001 at 10 and 50 mg/kg dose respectively on the 11th day. In E. coli induced abdominal sepsis, mortality due to peritonitis was evaluated in control and treatment groups. There was 100% mortality in the control group. In the groups treated with a-amyrin 10 mg/kg body, wt 36 % mortality was observed respectively. Conclusion: The current discoveries are huge for the improvement of options, cheap and maybe safer strategies for the treatment of diseases. A detailed study is also required on structure determination of the compounds from bioactive fractions in order to find the structure- activity relationship. [ABSTRACT FROM AUTHOR]

Alstonia scholaris (L.) R. Br. (Apocynaceae) is an evergreen tree that has been used to treat lung diseases. In this study, the toxicity profile of indole alkaloids from leaves of A. scholaris was investigated. In acute toxicity tests, mice were administered total alkaloids (TA) and five indole alkaloids. In a chronic toxicity test, rats were continuously administered TA (50, 100, and 300 mg/kg bw) for 13 weeks, followed by a 4-week recovery. A single administration of TA affected the behavior of mice, and at 12.8 g/kg bw, prone position, shortness of breath, wheezing, and convulsion were observed. The half-lethal dose (LD50) in mice was 5.48 g/kg bw, almost 2740 times the clinical dose in humans. Among the five indole alkaloids, the maximum tolerance dose in mice ranged from 0.75 to 4 g/kg bw. The TA-treated rats did not die and showed no adverse effects or dose-dependent changes in weight or food and water consumption, despite fluctuations in hematological and biochemical parameters compared with historical data. Furthermore, both gross and histopathological observations revealed no abnormalities in any organ. With daily oral administration to rats, the non-observed-adverse-effect-level of TA was 100 mg/kg bw. The results indicate that TA is safe for clinical use. [ABSTRACT FROM AUTHOR]
