Lipids play crucial roles in regulating aging and longevity. In the past few decades, a series of genetic pathways have been discovered to regulate lifespan in model organisms. Interestingly, many of these regulatory pathways are linked to lipid metabolism and lipid signaling. Lipid metabolic enzymes undergo significant changes during aging and are regulated by different longevity pathways. Lipids also actively modulate lifespan and health span as signaling molecules. In this review, we summarize recent insights into the roles of lipid metabolism and lipid signaling in aging and discuss lipid-related interventions in promoting longevity.

Lysosomes are sites of active metabolism in a cell. They contain various hydrolases that degrade extracellular and intracellular materials during endocytosis and autophagy, respectively. In addition to their long-recognized roles in degradation and recycling, emerging studies have revealed that lysosomes are organizing centers for signal transduction. Lysosome-derived signaling plays crucial roles in regulating nutrient sensing, metabolic adaptation, organelle crosstalk, and aging. In particular, how the degradative role of the lysosome cooperates with its signaling functions to actively modulate lifespan is beginning to be unraveled. This review describes recent advances in the role of the lysosome as a 'signaling hub' that uses three different lysosome-derived signaling pathways to integrate metabolic inputs, organelle interactions, and the control of longevity.

To determine whether localized hyperosmotic spikes on the pre-lens tear film (PrLTF) due to tear break up results in hyperosmotic spikes on the ocular surface during soft-contact-lens (SCL) wear and whether wear of SCLs can protect the cornea against PrLTF osmotic spikes.Two-dimensional transient diffusion of salt was incorporated into a computationally designed SCL, post-lens tear film (PoLTF), and ocular surface and solved numerically. Time-dependent localized hyperosmolarity spikes were introduced at the anterior surface of the SCL corresponding to those generated in the PrLTF. PurposeTo investigate the effect of various oxygen transmissibilities (Dk/t) of scleral lenses and corneal thickness recovery time from overnight eye closure with patching on corneal edema during 5 h lens wear.MethodsScleral lenses (hofocon A, 15.6 mm diameter) were worn bilaterally with three different Dks (100, 140, and 160 Barrer). Central and peripheral corneal thickness (CCT and PCT) were measured using optical coherence tomography. Four subjects were randomly selected for one additional visit and asked to patch one eye before night sleeping. The patch was not removed until lens insertion to avoid corneal deswelling. Then CCT of both eyes was measured.ResultsTen neophytes with healthy eyes participated in the study. Mean [95% CI] Dk/t of the study lenses was 32.0 [29.2, 34.7] hBarrer/cm. This study implemented real-world tests in Nanjing, China for measuring emission factors (EFs) of air pollutants, including Carbon Monoxide (CO), Hydrocarbon (HC), Nitrogen Oxides (NOx), and Particulate Matter (PM) from ten construction machines in three operational modes (idling, moving, and working) with a Portable Emission Measurement System. The idling mode shows the least variation of EFs, and its average CO EFs can be higher than the moving and working modes by 43% and 34%, respectively. The working mode generates the highest emission for all other pollutants with the highest variation. The EFs suggested by the Guide (an official guidebook for developing emission inventory in China) are in general lower than the measured EFs, and the gap becomes larger for older machines. The EFs of CO, NOx, and PM of China Stage II machines are 24%, 120%, and 66% higher than those of the Guide, respectively. The differences go up as high as 126%, 1066%, and 559% for China Stage I machines, indicating the upgrade of engine technology from Stage I to Stage II, as well as the effect of machine deterioration. The result of this study reveals the effectiveness of stringent emission standards in controlling emissions from construction machines. High emissions from older machines emphasize the importance of a more rigorous machine replacement policy and a regulated maintenance strategy. The result also stresses the need to update the Guide with differentiated activity modes, region variations, and machine deterioration effects.

Purpose With active investigation underway for embedded-circuit contact lenses, safe oxygen supply of these novel lenses remains a question. Central-to-peripheral corneal edema for healthy eyes during wear of soft contact (SCL) and scleral lenses (SL) with embedding components is assessed. Methods Various 2-dimensional (2D) designs of SL and SCL with embedded components are constructed on Comsol Multiphysics 5.5. Local corneal swelling associated with the designed lenses is determined by a recently developed 2D metabolic-swelling model. Settled central post-lens tear-film thicknesses (PoLTFs) are set at 400 μm and 3 μm for SL and SCL designs, respectively. Each lens design has an axisymmetric central and an axisymmetric peripheral embedment. Oxygen permeability (Dk) of the lens and the embedments ranges from 0 to 200 Barrer. Dimensions and location of the embedments are varied to assess optimal-design configurations to minimize central-to-peripheral corneal edema. Results By adjusting oxygen Dk of the central embedment, the peripheral embedment, or the lens matrix polymer, corneal swelling is reduced by up to 2.5 %, 1.5 %, or 1.4 % of the baseline corneal thickness, respectively, while keeping all other parameters constant. A decrease in PoLTF thickness from 400 μm to 3 μm decreases corneal edema by up to 1.8 % of the baseline corneal thickness. Shifting the peripheral embedment farther out towards the periphery and towards the anterior lens surface reduces peak edema by up to 1.3 % and 0.6 % of the baseline corneal thickness, respectively. Conclusions To minimize central-to-peripheral corneal edema, embedments should be placed anteriorly and far into the periphery to allow maximal limbal metabolic support and oxygen transport in the polar direction (i.e., the θ-direction in spherical coordinates). High-oxygen transmissibility for all components and thinner PoLTF thickness are recommended to minimize corneal edema. Depending on design specifications, less than 1 % swelling over the entire cornea is achievable even with oxygen-impermeable embedments.

Background: Chronic stimulant use has been associated with disruptions in fronto-striatal systems implicated in as- sociative learning [1]. Experimental studies have also shown that individuals with stimulant drug addiction experience difficulties in selecting the appropriate action following feedback [2]. However, the precise impairments in feedback learning in stimulant-addicted individuals are still unclear. A possible explanation might lie in an abnormal prediction error mechanism, as stimulant drugs directly target striatal dopaminergic neurons [3].

© 2016 Elsevier B.V. A Schirmer tear test (STT) is commonly used to gauge human tear production, especially when dry-eye symptoms present. In an STT, the rounded tip of a standardized paper strip is inserted into the lower fornix of the eye, and the wetted length extending out from the lower lid is recorded after 5 min of eye closure. Longer wetted lengths suggest higher tear production rates. To date, however, there is no methodology to transform STT transient wetting lengths into basal tear- production rates. We develop a physical model to elucidate wetting kinetics in a Schirmer strip. Tear evaporation from the exposed portion of the strip and gravity are accounted for. Careful consideration of the initial depletion of tear in the closed-eye tear prism reveals an initial fast increase in wetted length followed by slower growth. Excellent agreement of the proposed model is achieved with experimental observation. When evaporation is negligible, the slow-growth regime exhibits a linear increase of wetted length in time. The linear-length-growth time regime permits simple calculation of quantitative tear-production rates. We suggest measuring several dynamic wetting lengths along a sheathed Schirmer strip and near the 5-min insertion duration followed by fitting to a straight line. The slope of the length-versus-time data gives the basal lacrimal-supply rate.

Summary A new study has found that, in the nematode worm Caenorhabditis elegans, biomass conversion from the intestine to yolk, mediated by autophagy and insulin/IGF-1 signaling, promotes reproduction in early life but aging in late life.

A novel methodology for assessing evaporation (up to 48 h) through lipid-nanofilms in vitro was developed. The influence of lipid-mixture compositions on evaporation rates was studied. The evaporative fluxes and rheology of lipid-nanofilms were compared with those of human tear-lipid nanofilms in vitro.A sessile-drop technique with precise drop-volume control was adapted to measure evaporation rates at constant temperature of 36 °C and humidity of 75 %. Model lipid solutions were deposited on the surface of aqueous drops to create nanofilms of 10-100 nm. The measurements of dynamic surface pressure vs. nanofilm-thickness were performed under the same conditions. The lipid-mixtures compositions were chosen to mimic that of human tear lipids. Evaporation through lipid nanofilms decreased with film thickness and aging. Evaporation through 70-nm films was 2.5-3 time slower than through 10-nm-thick films. Nonpolar-lipid mixtures reduced evaporation by approximately 35 %. The optimized model-lipid mixtures containing polar phospholipids reduced evaporation by 70-75 %, matching the evaporation-reduction by human-lipid nanofilms in vitro. These model mixtures exhibited interfacial rheology similar to human tear lipids in vitro.This methodology substantiated that aged lipid-nanofilms significantly reduced evaporation in vitro. These findings contradict to the previous reports suggesting that model lipid and meibum films do not retard evaporation in vitro. Polar phospholipids enhance evaporative resistance close to the level observed for human tear-lipid films in vivo. We hypothesize that unique rheological properties of tear-lipid nanofilms are germane to the specific mono- and bi-layered structures formed by phospholipids at the lipid-air and lipid-aqueous interfaces.

Zhou et al. challenge the well-known beneficial effect of autophagy in promoting longevity. Evidence presented demonstrate that autophagy induction coupled with increased mitochondrial permeability is detrimental to organismal health in both the nematode Caenorhabditis elegans and mammals.
