Intensive economic and human activities present challenges to the carbon storage capacity of terrestrial ecosystems, particularly in arid regions that are sensitive to climate change and ecologically fragile. Therefore, accurately estimating and simulating future changes in carbon stocks on the northern slope economic belt of Tianshan Mountains (NSEBTM) holds great significance for maintaining ecosystem stability, achieving high-quality development of the economic belt, and realizing the goal of "carbon neutrality" by 2050. This study examines the spatiotemporal evolution characteristics of the NSEBTM carbon stocks in arid regions from 1990 to 2050, utilizing a combination of multi-source data and integrating the Patch-generating Land use Simulation (PLUS) and Integrated Valuation of Ecosystem Services and Trade-offs (InVEST) models. Additionally, an attribution analysis of carbon stock changes is conducted by leveraging land use data. The findings demonstrate that (1) the NSEBTM predominantly consists of underutilized land, accounting for more than 60% of the total land area in the NSEBTM. Unused land, grassland, and water bodies exhibit a declining trend over time, while other forms of land use demonstrate an increasing trend. (2) Grassland serves as the primary reservoir for carbon storage in the NSEBTM, with grassland degradation being the leading cause of carbon loss amounting to 102.35 t over the past three decades. (3) Under the ecological conservation scenario for 2050 compared to the natural development scenario, there was a net increase in carbon storage by 12.34 t; however, under the economic development scenario compared to the natural development scenario, there was a decrease in carbon storage by 25.88 t. By quantitatively evaluating the land use change in the NSEBTM and its impact on carbon storage in the past and projected for the next 30 years, this paper provides scientific references and precise data support for the territorial and spatial decision making of the NSEBTM, thereby facilitating the achievement of "carbon neutrality" goals. Members of the Flaviviridae family, encompassing the Flavivirus and Hepacivirus genera, are implicated in a spectrum of severe human pathologies. These diseases span a diverse spectrum, including hepatitis, vascular shock syndrome, encephalitis, acute flaccid paralysis, and adverse fetal outcomes, such as congenital heart defects and increased mortality rates. Notably, infections by Flaviviridae viruses have been associated with substantial cardiovascular compromise, yet the exploration into the attendant cardiovascular sequelae and underlying mechanisms remains relatively underexplored. This review aims to explore the epidemiology of Flaviviridae virus infections and synthesize their cardiovascular morbidities. Leveraging current research trajectories and our investigative contributions, we aspire to construct a cogent theoretical framework elucidating the pathogenesis of Flaviviridae-induced cardiovascular injury and illuminate prospective therapeutic avenues. Based on data from the Gurbantunggut Desert, the largest fixed/semi-fixed desert in China, and ERA5-Land reanalysis data, the long-term variations and spatial surface heat source (SHS) differences in the Gurbantunggut Desert are discussed herein. The results show the following: (1) The hourly SHS at the Kelameili station during the 2013–2021 period was a weak heat source at night; contrastingly, it was a strong heat source during the day. The duration of the hourly SHS increased gradually from January to July, but it decreased gradually from July to December. The daily SHS showed obvious seasonal variation, reaching the maximum in summer and the minimum in winter. The ERA5-Land reanalysis can reproduce all the variation characteristics of the SHS well. (2) The climatology (i.e., multi-year mean) of the monthly SHS intensity was lower than 50 W/m2 during the January–March and September–December periods in the Gurbantunggut Desert, indicating a weak heat source. On the other hand, the climatology recorded in April–August was higher than 50 W/m2, with a strong heat source. From the perspective of spatial distribution, the eastern and western regions of the Gurbantunggut Desert show strong heat sources, while the central region shows weak heat sources. The spatial distribution of the first and second modes of the empirical orthogonal function (EOF) decomposition reflected the consistent spatial variability and a north–south (or east–west) polarity variation of the monthly SHS in the Gurbantunggut Desert, respectively. (3) The yearly SHS showed negative anomalies during the 1950–1954, 1964–1982 and 2004–2015 periods, and positive anomalies during the 1955–1963, 1983–2003 and 2016–2021 periods in the Gurbantunggut Desert. Additionally, the time series of the SHS anomalies was positively correlated with the Interdecadal Pacific Oscillation (IPO) index. During the negative IPO phase, the yearly SHS showed a negative anomaly in the Gurbantunggut Desert, while the yearly SHS showed a positive anomaly during the positive IPO phase in most regions of the Gurbantunggut Desert. Urban heat islands have garnered significant attention due to their potential impact on human life. Previous studies on urban heat islands have focused on characterizing temporal and spatial variations over longer periods of time. In this study, we investigated the urban heat island (UHI) in Birmingham from September 2013 to August 2014 using higher temporal resolution SEVIRI satellite surface temperature data along with data from the Birmingham Urban Climate Laboratory (BUCL) meteorological station and the UK Meteorological Office meteorological station. Our aim was to characterize the diurnal variations in the surface urban heat island intensity (SUHII) and canopy urban heat island intensity (CUHII) and to explore their relationship under the influence of three factors (day/nighttime, season, and wind speed) using regression analysis. Our findings reveal that SUHII and CUHII exhibit relatively stable patterns at night but vary significantly during the day with opposite diurnal trends. In addition, SUHII and CUHII were more variable in spring and summer but less variable in winter. During the nighttime, SUHII represents CUHII with high confidence, especially during spring and summer, but less so during the cold season. In addition, SUHII represents CUHII with greater confidence under low-wind conditions. This study deepens our understanding of the diurnal dynamics of urban heat islands and the influence of atmospheric conditions on the relationship between surface and canopy heat islands in urban areas. Microcystin-LR (MC-LR) contamination is a worldwide environmental problem that poses a grave threat to the water ecosystem and public health. Exposure to MC-LR has been associated with the development of intestinal injury, but there are no effective treatments for MC-LR-induced intestinal disease. Probiotics are "live microorganisms that are beneficial to the health of the host when administered in sufficient quantities". It has been demonstrated that probiotics can prevent or treat a variety of human diseases; however, their ability to mitigate MC-LR-induced intestinal harm has not yet been investigated. The objective of this study was to determine whether probiotics can mitigate MC-LR-induced intestinal toxicity and its underlying mechanisms. We first evaluated the pathological changes in colorectal tissues using an animal model with sub-chronic exposure to low-dose MC-LR, HE staining to assess colorectal histopathologic changes, qPCR to detect the expression levels of inflammatory factors in colorectal tissues, and WB to detect the alterations on CSF1R signaling pathway proteins in colorectal tissues. Microbial sequencing analysis and screening of fecal microorganisms differential to MC-LR treatment in mice. To investigate the role of microorganisms in MC-LR-induced colorectal injury, an in vitro model of MC-LR co-treatment with microorganisms was developed. Our findings demonstrated that MC-LR treatment induced an inflammatory response in mouse colorectal tissues, promoted the expression of inflammatory factors, activated the CSF1R signaling pathway, and significantly decreased the abundance of Lactobacillus. In a model of co-treatment with MC-LR and Lactobacillus fermentum (L. fermentum), it was discovered that L. fermentum substantially reduced the incidence of the colorectal inflammatory response induced by MC-LR and inhibited the protein expression of the CSF1R signaling pathway. This is the first study to suggest that L. fermentum inhibits the CSF1R signaling pathway to reduce the incidence of MC-LR-induced colorectal inflammation. Grassland ecosystems are an important component of global terrestrial ecosystems and play a crucial role in the global carbon cycle. Therefore, it is important to study the carbon dioxide (CO2) process in the Middle Tien Shan grassland ecosystem, which can be regarded as a typical representative of the mountain grasslands in Xinjiang. Eddy covariance (EC) and the global carbon fluxes dataset (GCFD) were utilized to continuously monitor the Middle Tien Shan grassland ecosystem in Xinjiang throughout the 2018 growing season. The findings revealed notable daily and monthly fluctuations in net ecosystem exchange (NEE), gross primary productivity (GPP), and ecosystem respiration (Reco). On a daily basis, there was net absorption of CO2 during the day and net emission during the night. The grassland acted as a carbon sink from 6:00 to 18:00 and as a carbon source for the remaining hours of the day. On a monthly scale, June and July served as carbon sinks, whereas the other months acted as carbon sources. The accumulated NEE, GPP, and Reco during the growing season were −329.49 g C m−2, 779.04 g C m−2, and 449.55 g C m−2, respectively. On the half-hourly and daily scales, soil temperature (Ts) was the main contributor to CO2 fluxes and had the greatest influence on the variations in CO2 fluxes. Additionally, air temperature (Ta) showed a strong correlation with CO2 fluxes. The grassland ecosystems exhibited the strongest CO2 uptake, reaching its peak at soil temperatures of 25 °C. Moreover, as the air temperatures rose above 15 °C, there was a gradual decrease in NEE, while CO2 uptake increased. The applicability of GCFD data is good in the grassland ecosystem of the Middle Tien Shan Mountains, with correlations of 0.59, 0.81, and 0.73 for NEE, GPP, and Reco, respectively, compared to field observations. The exchange of energy between the land surface and atmosphere is dependent upon crucial parameters, including surface roughness, emissivity, bulk transfer coefficients for momentum (CD) and heat (CH). These parameters are calculated through site observation data and remote sensing data. The following conclusions are drawn: (1) the aerodynamic roughness of the Gurbantunggut Desert measures 1.1 × 10−2 m, which is influenced by the varying conditions of the underlying surface. The roughness decreases as wind speed increases and is seen to be directly proportional to the growth of vegetation. From April to June, the aerodynamic roughness increases with increasing vegetation cover, but begins to gradually decrease after July. Spatially, the middle regions show higher roughness values than the eastern and western areas. In the central part of the desert, the roughness is between 2.37 × 10−2 m and 2.46 × 10−2 m from April to November. The northwest and northeast regions measure 1.41 × 10−2 m–2.04 × 10−2 m and 1.53 × 10−2 m–2.39 × 10−2 m, respectively. (2) The surface emissivity is 0.93, and it varies depending on the snow and vegetation present in the underlying area. (3) CD and CH exhibit an inverse relationship with wind speed. When wind speed falls below 6 m/s, the CD declines rapidly as wind speed increases. It has been found that the topology effect and the possible emergent hidden scale and hidden local flavor symmetries at high density reveal a novel structure of compact star matter. When N f ≥ 2 , baryons can be described by skyrmions when the number of color N c is regarded as a large parameter and there is a robust topology change—the transition from skyrmion to half-skyrmion—in the skyrmion matter approach to dense nuclear matter. The hidden scale and local flavor symmetries, which are sources introducing the scalar meson and vector mesons, are significant elements for understanding the nuclear force in nonlinear chiral effective theories. We review in this paper how the robust conclusions from the topology approach to dense matter and emergent hidden scale and hidden local flavor symmetries figure in generalized nuclear effective field theory (GnEFT), which is applicable to nuclear matter from low density to compact star density. The topology change encoded in the parameters of the effective field theory is interpreted as the hadron-quark continuity in the sense of the Cheshire Cat Principle. A novel feature predicted in this theory that has not been found before is the precocious appearance of the conformal sound velocity in the cores of massive stars, although the trace of the energy-momentum tensor of the system is not zero. That is, there is a pseudoconformal structure in the compact star matter and, in contrast to the usual picture, the matter is made of colorless quasiparticles of fractional baryon charges. A possible resolution of the longstanding g A quench problem in nuclei transition and the compatibility of the predictions of the GnEFT with the global properties of neutron star and the data from gravitational wave detections are also discussed. Metal-organic frameworks (MOFs)-based optical nanoprobes for luminescence and surface-enhanced Raman spectroscopy (SERS) applications have been receiving tremendous attention. Every element in the MOF structure, including the metal nodes, the organic linkers, and the guest molecules, can be used as a source to build single/multi-emission signals for the intended analytical purposes. For SERS applications, the MOF can not only be used directly as a SERS substrate, but can also improve the stability and reproducibility of the metal-based substrates. Additionally, the porosity and large specific surface area give MOF a sieving effect and target molecule enrichment ability, both of which are helpful for improving detection selectivity and sensitivity. Wind profile light detection and ranging (LiDAR) is an important tool for observing features within the atmospheric boundary layer. Observations of the wind field and boundary layer height from coherent Doppler wind LiDARs (CDWLs) under sandy and dusty weather conditions were evaluated using observations from two CDWLs and one GTS radio sounding located at the northern edge of the Tibetan plateau from 1 May to 30 August 2021. The results showed that CDWL has good applicability in reproducing wind fields in dust, precipitation, and in clear-sky conditions, and that it is superior to the v wind field for real measurements of the u wind fields. In terms of the planetary boundary layer height (PBLH), the validity of the inversion of PBLH in dusty weather was higher than that under clear-sky conditions. It was found that the PBLH retrieved by the CDWL at 20:00 (BJT) was better than that at 08:00 (BJT). The diurnal variation amplitude of the PBLH before the occurrence of a sandstorm was larger than the diurnal variation amplitude of the PBLH occurring during a sandstorm. [ABSTRACT FROM AUTHOR]

Currently, no consensus exists regarding Sotos syndrome in the Chinese population. Here, we present a case of neonatal Sotos syndrome, followed by a retrospective analysis of five cases of neonatal Sotos syndrome, reported in China. The study subject was a twin premature infant, heavier than gestational age, with characteristic facial features, limb shaking, and hypertonia. Transient hypoglycemia, abnormal cranial magnetic resonance imaging, multiple nodules in polycystic kidneys and liver, abnormal hearing, patent ductus arteriosus, and an atrial septal defect were also noted. The subject showed overgrowth and developmental retardation at 3 months of age. Sequencing revealed a novel missense mutation, c.5000C>A, in the nuclear receptor binding the SET domain protein 1 gene, resulting in an alanine-to-glutamate substitution. The bioinformatics analysis suggested high pathogenicity at this site. This study provides insights into diagnosis of neonatal Sotos syndrome based on specific phenotypes. A new depsidone derivative (1), aspergillusidone G, was isolated from a marine fungus Aspergillus unguis, together with eight known depsidones (2-9) and a cyclic peptide (10): agonodepside A (2), nornidulin (3), nidulin (4), aspergillusidone F (5), unguinol (6), aspergillusidone C (7), 2-chlorounguinol (8), aspergillusidone A (9), and unguisin A (10). Compounds 1-4 and 7-9 were obtained from the plasma induced mutant of this fungus, while 5, 6, and 10 were isolated from the original strain under chemical induction. Their structures were identified using spectroscopic analysis, as well as by comparison with literature data. The HPLC fingerprint analysis indicates that chemical induction and plasma mutagenesis effectively influenced the secondary metabolism, which may be due to their regulation in the key steps in depsidone biosynthesis. In bioassays, compound 9 inhibited acetylcholinesterase (AChE) with IC50 in 56.75 µM. Compounds 1, 5, 7, 8, and 9 showed moderate to strong activity towards different microbes. Compounds 3, 4, and 5 exhibited potent larvicidality against brine shrimp. In docking studies, higher negative CDOCKER interaction energy and richer strong interactions between AChE and 9 explained the greater activity of 9 compared to 1. The Taklimakan Desert is a typical arid area. Due to the needs of production and life, a total of 2 km2 of constructed green land (hereinafter referred to as CGL) has been formed in the sand dune, resulting in the uniform underlying surface of the desert having been changed, which has led to the change in the near-surface energy distribution pattern and the formation of a local climate of the CGL that is obviously different from that of the desert climate. Therefore, it is necessary to study the varied interval of the threshold of meteorological factors and the regional climate characteristics of the CGL under the background of desert. The main results are as follows. Firstly, from sunrise to noon, the increasing rate of temperature in natural sandy land (hereafter, NSL) was higher than that in CGL, and the opposite results occurred between noon and sunset. The peak temperature of CGL was 2 h later than that of NSL. At night, the temperature at the boundary of the CGL was generally higher than that of the NSL and the central area of the CGL. In addition, the results show that under the combined influence of underlying conditions, local circulation and small terrain, the CGL (middle) daily range of temperature > NSL (west) > CGL (east) > CGL (west). The positive temperature change period of CGL was significantly shorter than that of NSL in all seasons. However, temperature inversion occurred at night in all seasons. The intensity of the temperature inversion was strongest in winter, with a maximum temperature difference of 12.8 °C, followed by autumn, spring, and summer, with a maximum difference of 6.4 °C. Secondly, the wind speed in the daytime was higher than that at night, and the wind speed in NSL was higher than that in CGL. In summer, if the average wind speed of the NSL was quantified as 1.0 m/s, the wind speed lapse rate reached 30% at the boundary of the CGL. In the present study, we developed a liquid chromatography-diode array detector-electrospray ionization/mass spectrometric (LC-DAD-ESI/MS) method for analysis of saikosaponins in Bupleurum falcatum. The LC method employed a ZORBAX SB-Aq analytical column (150 × 4.6 mm i.d., 5 μm) at a flow rate of 0.8 mL/min coupled with a diode array detector at 204 nm. A step gradient of acetonitrile-water (v/v) containing 0.5% formic acid from 30 to 70% was applied, leading to a sample analysis time of 30 min. The ESI-MS was carried out in positive and negative modes from 500 to 1,500 m/z. Saikosaponins c, a, and d gave strong sodium adducts at m/z 949.6, 803.5 and 803.6, respectively, in positive mode. The data indicate that the present LC-DAD-ESI/MS assay is an effective method for the determination of saikosaponins c, a and d from the roots of Bupleurum falcatum. [ABSTRACT FROM AUTHOR]

The increasing cyanobacterial blooms have recently been considered a severe environmental problem. Microcystin-leucine arginine (MC-LR) is one of the secondary products of cyanobacteria metabolism and most harmful cyanotoxins found in water bodies. Studies show MC-LR negatively affects various human organs when exposed to it. The phenotype of the jejunal chronic toxicity induced by MC-LR has not been well described. The aim of this paper was to investigate the effects of MC-LR on the jejunal microstructure and expression level of inflammatory-related factors in jejunum. Mice were treated with different doses (1, 30, 60, 90 and 120 μg/L) of MC-LR for six months. The microstructure and mRNA expression levels of inflammation-related factors in jejunum were analyzed. Results showed that the microstructure of the jejunum was destroyed and expression levels of inflammation-related factors interleukin (IL)-1β, interleukin (IL)-8, tumor necrosis factor alpha, transforming growth factor-β1 and interleukin (IL)-10 were altered at different MC-LR concentrations. To the best of our knowledge, this is the first study that mice were exposed to a high dose of MC-LR for six months. Our data demonstrated MC-LR had the potential to cause intestinal toxicity by destroying the microstructure of the jejunum and inducing an inflammatory response in mice, which provided new insight into understanding the prevention and diagnosis of the intestinal diseases caused by MC-LR. Microcystin-LR (MC-LR) is the most toxic and frequently detected monocyclic heptapeptide hepatotoxin produced by cyanobacteria, which poses a great threat to the natural ecosystem and public health. It is very important to seek environment-friendly and cost-efficient methods to remove MC-LR in water. In this study, the MC-degrading capacities of a novel indigenous bacterial community designated as YFMCD4 and the influence of environmental factors including various temperatures, MC concentrations and pH on the MC-degrading activities were investigated utilizing high-performance liquid chromatography (HPLC). In addition, the MC-degrading mechanism of YFMCD4 was also studied using HPLC coupled with a mass spectrometry equipped with electrospray ionization interface (HPLC-ESI-MS). The data showed MC-LR was completely removed at the maximum rate of 0.5 µg/(mL·h) under the optimal condition by YFMCD4. Two pure bacterial strains Alcaligenes faecalis and Stenotrophomonas acidaminiohila were isolated from YFMCD4 degraded MC-LR at a slower rate. The MC-degrading rates of YFMCD4 were significantly affected by different temperatures, pH and MC-LR concentrations. Two intermediates of a tetrapeptide and Adda appeared in the degradation process. These results illustrate that the novel YFMCD4 is one of the highest effective MC-degrading bacterial community, which can completely remove MC-LR and possesses a significant potential to treat water bodies contaminated by MC-LR. Acetaminophen overdose-induced hepatotoxicity is the most common cause of acute liver failure in many countries. Previously, alpha-mangostin (α-MG) has been confirmed to exert protective effects on a variety of liver injuries, but the protective effect on acetaminophen-induced acute liver injury (ALI) remains largely unknown. This work investigated the regulatory effect and underlying cellular mechanisms of α-MG action to attenuate acetaminophen-induced hepatotoxicity in mice. The increased serum aminotransferase levels and glutathione (GSH) content and reduced malondialdehyde (MDA) demonstrated the protective effect of α-MG against acetaminophen-induced hepatotoxicity. In addition, α-MG pretreatment inhibited increases in tumor necrosis factor (TNF-α) and interleukin-1β (IL-1β) caused by exposure of mice to acetaminophen. In liver tissues, α-MG inhibited the protein expression of autophagy-related microtubule-associated protein light chain 3 (LC3) and BCL2/adenovirus E1B protein-interacting protein 3 (BNIP3). Western blotting analysis of liver tissues also proved evidence that α-MG partially inhibited the activation of apoptotic signaling pathways via increasing the expression of Bcl-2 and decreasing Bax and cleaved caspase 3 proteins. In addition, α-MG could in part downregulate the increase in p62 level and upregulate the decrease in p-mTOR, p-AKT and LC3 II /LC3 I ratio in autophagy signaling pathways in the mouse liver. Taken together, our findings proved novel perspectives that detoxification effect of α-MG on acetaminophen-induced ALI might be due to the alterations in Akt/mTOR pathway in the liver. Microcystin-LR is a cyclic heptapeptide hepatotoxin produced by harmful cyanobacteria. A panel of microRNAs containing miR-451a were found to be significantly changed in normal human liver cells HL7702 after exposure to microcystin-LR (MC-LR) in our previous study. However, the functions of miR-451a in hepatotoxicity induced by MC-LR remained unclear. The study aimed to investigate the impacts of miR-451a in HL7702 cells following treatment with 5 or 10 μM MC-LR. The comet assay indicated that MC-LR can influence Olive tail moment (OTM) in HL7702 cells. Furthermore, increase of miR-451a significantly repressed DNA damage and the protein expression level of γ-H2AX induced by MC-LR. Moreover, over-expression of miR-451a inhibited the expression level of p-AKT1 protein in cells following treatment by MC-LR. These results showed that miR-451a may protect from MC-LR-induced DNA damage by down-regulating the expression of p-AKT1, which provides new clues for the diagnosis and therapy policies for liver damage induced by MC-LR.

In bioassays, compound 9 inhibited acetylcholinesterase (AChE) with IC 50 in 56.75 μM. Compounds 1 , 5 , 7 , 8 , and 9 showed moderate to strong activity towards different microbes. Compounds 3 , 4 , and 5 exhibited potent larvicidality against brine shrimp. In docking studies, higher negative CDOCKER interaction energy and richer strong interactions between AChE and 9 explained the greater activity of 9 compared to 1 . Chemical induction and plasma mutagenesis can be used as tools to expand the chemodiversity of fungi and obtain useful natural products.

In the present study, we developed a liquid chromatography-diode array detector-electrospray ionization/mass spectrometric (LC-DAD-ESI/MS) method for analysis of saikosaponins in Bupleurum falcatum. The LC method employed a ZORBAX SB-Aq analytical column (150 x 4.6 mm i.d., 5 μm) at a flow rate of 0.8 mL/min coupled with a diode array detector at 204 nm. A step gradient of acetonitrile-water (v/v) containing 0.5% formic acid from 30 to 70% was applied, leading to a sample analysis time of 30 min. The ESI-MS was carried out in positive and negative modes from 500 to 1,500 m/z. Saikosaponins c, a, and d gave strong sodium adducts at m/z 949.6, 803.5 and 803.6, respectively, in positive mode. The data indicate that the present LC-DAD-ESI/MS assay is an effective method for the determination of saikosaponins c, a and d from the roots of Bupleurum falcatum.
