Ductile iron bars (DIBs) with a diameter of 145 mm, used for plunger pump production, were made by the horizontal continuous casting (HCC). The microstructure of the samples cut at three locations with different distances away from the surface (~20 mm from the surface, half of the radius and the center of the HCCDIBs) were investigated. The mechanical properties were measured by tensile and torsion tests. Results show that after the spheroidization of graphite, the iron matrix incorporates the nodules of Format I, Size 8 close to the surface, Format I, Size 7 at the half of the radius from the surface, and Format II, Size 6 in the centre of the bar, according to the ASTM A247 standard. The content of pearlite in the matrix changes from 55% (~20 mm from the surface) to 70% (half of the radius) and 80% (the center of the HCCDIBs). The strengths in tension are 552, 607 and 486 MPa with the elongations of 12.5%, 10.5% and 5.8% in samples cut at these three locations from the surface to the centre, respectively. The strength in torsion is equal to 558, 551 and 471 MPa at corresponding torsion angles of 418°, 384° and 144° respectively to the same distance from the bar surface. Fracture in tension is manifested via crack propagation through the interface between graphite nodules and matrix (Mode I), while in torsion the fracture is caused by the shear of graphite nodules (Mode II). It is shown that the transition between two fracture modes is also dependent on the size of graphite nodule. Typically, fracture Mode I was observed for nodules of smaller diameter (less than 22.7 µm) and fracture Mode II was seen for nodules of greater diameter (more than 24.8 µm). [ABSTRACT FROM AUTHOR]

Suaeda salsa calluses cultured in darkness for 28 d were transferred to Murashige and Skoog (MS) media containing various growth regulators under white light conditions for 10 d to investigate cell growth, betacyanin accumulation, and expression of dopa-4,5-dioxygenase ( DODA). Callus growth was markedly promoted when 0.2 mg·L 2,4-D and 0.5 mg·L 6-BA were added to the MS medium. Surprisingly, of the auxins tested, IAA had no effect on betacyanin content, but 2,4-D strongly decreased betacyanin content. Betacyanin content was positively correlated with 6-BA concentrations in the range of 0.1-2.0 mg·L. DODA mRNA levels were consistent with the response of betacyanin content to exogenous growth regulators. These results suggest that betacyanin metabolism in S. salsa calluses is regulated under white light conditions by growth regulators through the regulation of genes such as DODA that are involved in betacyanin synthesis. Abstract Purpose  To evaluate single photon emission computed tomography (SPECT)/spiral computed tomography (CT) fusion imaging for the diagnosis of bone metastasis in patients with known cancer and to compare the diagnostic efficacy of SPECT/CT fusion imaging with that of SPECT alone and with SPECT  CT. Materials and methods  One hundred forty-one bone lesions of 125 cancer patients (with nonspecific bone findings on bone scintigraphy) were investigated in the study. SPECT, CT, and SPECT/CT fusion images were acquired simultaneously. All images were interpreted independently by two experienced nuclear medicine physicians. In cases of discrepancy, consensus was obtained by a joint reading. The final diagnosis was based on biopsy proof and radiologic follow-up over at least 1 year. Results  The final diagnosis revealed 63 malignant bone lesions and 78 benign lesions. The diagnostic sensitivity of SPECT, SPECT  CT, and SPECT/CT fusion imaging for malignant lesions was 82.5%, 93.7%, and 98.4%, respectively. Specificity was 66.7%, 80.8%, and 93.6%, respectively. Accuracy was 73.8%, 86.5%, and 95.7%, respectively. The specificity and accuracy of SPECT/CT fusion imaging for the diagnosis malignant bone lesions were significantly higher than those of SPECT alone and of SPECT  CT (P 2 = 9.855, P = 0.002). The numbers of equivocal lesions were 37, 18, and 5 for SPECT, SPECT  CT, and SPECT/CT fusion imaging, respectively, and 29.7% (11/37), 27.8% (5/18), and 20.0% (1/5) of lesions were confirmed to be malignant by radiologic follow-up over at least 1 year. Conclusions  SPECT/spiral CT is particularly valuable for the diagnosis of bone metastasis in patients with known cancer by providing precise anatomic localization and detailed morphologic characteristics. The nanometric La1− x K x CoO3 ( x = 0–0.30) perovskite-type oxides were prepared by a citric acid-ligated method. The catalysts were characterized by means of XRD, IR, BET, XPS and SEM. The catalytic activity for the simultaneous removal of soot and nitrogen oxides was evaluated by a technique of the temperature-programmed oxidation reaction. In the LaCoO3 catalyst, the partial substitution of La3+ at A-site by alkali metal K+ enhanced the catalytic activity for the oxidation of soot particle and reduction of NO x . The La0.70K0.30CoO3 oxides are good candidate catalysts for the simultaneous removal of soot particle and NO x . The combustion temperatures for soot particles over the La0.70K0.30CoO3 catalyst are in the range from 289 to 461 °C, the selectivity of CO2 is 98.4% and the conversion of NO to N2 is 34.6% under loose contact conditions. The possible reasons that can lead to the activity enhancement for the K-substitution samples compared to the unsubstituted sample (LaCoO3) were given. The running status of hydraulic tube tester is reflected by the boosting pressure curve in Hydrostatic testing process. The authors present the extreme learning machine (ELM), a novel good learning scheme much faster than traditional gradient-based learning algorithms, as a mechanism for clustering the pressure curves. However, it caused low accuracy for clustering pressure curves for hydraulic tube tester. In this paper, a multi-stage ELM is proposed to improve the accuracy of clustering. During the process of this new ELM, the input data were divided into several stages, then, every stage was analyzed independently. At last, this method has been used in hydraulic tube tester data. The reaction mechanisms for diesel soot oxidation over V4/ZrO2 or V4/TiO2 oxide catalysts were studied by the means of in-situ UV-Raman spectroscopy. The results indicate that the formation of surface oxygen complexes (SOC) is a key step and the SOC species mainly exist as carboxyl groups. The presence of NO in the reaction gas stream can promote the formation of SOC species. For soot oxidation over V4/TiO2 catalyst, NO2 can be produced and remarkably accelerate soot oxidation. Based on the UV-Raman experimental results, two different reaction mechanisms are proposed for soot oxidation over V4/ZrO2 or V4/TiO2 samples, respectively. [ABSTRACT FROM AUTHOR]

The CrHZSM-5 catalysts with trace amount of Cr were firstly used for catalytic cracking of isobutane, and the effect of Cr-loading on the catalytic performances of CrHZSM-5 catalysts for the cracking of isobutane was also studied. The results suggested that when the loading of Cr in the CrHZSM-5 catalysts was less than 0.038 mmol/g Cr, especially at Cr loading of 0.004 mmol/g, both the reactivity of isobutane cracking and the selectivity to light olefins of CrHZSM-5 samples were greatly enhanced compared with the unpromoted HZSM-5, and very high yields of olefins(C2+C3) and ethylene were obtained. For instance, the yield of olefins(C2+C3) and ethylene reached 56.1% and 30.8%, respectively, at 625 °C when 0.004 mmol/g Cr was loaded on HZSM-5 sample. [ABSTRACT FROM AUTHOR]

The La1− xK xMnO3 perovskite-type oxides whose sizes were in nanometric range were prepared by the citric acid-ligated method. The structures of these perovskite-type oxides were examined by XRD and FT-IR. The catalytic activity for the combustion of soot particulate was evaluated by a technique of the temperature-programmed reaction. In the LaMnO3 catalyst, the partial substitution of K for La at A-site enhanced the catalytic activity for the combustion of soot particle. In the La1− xK xMnO3 catalysts, the combustion temperature of soot particle decreases with increasing x values. The La1− xK xMnO3 oxides with the substitution quantity between x=0.20 and x=0.25 are good candidate catalysts for the soot particle removal reaction, and the combustion temperature of soot particle is between 285 and 430 °C when the contact of catalysts and soot is loose, and their catalytic activities for the combustion of soot particle are as good as supported Pt catalysts, which is the best catalyst system so far reported for soot combustion under loose contact conditions. The optical properties of structures containing InGaAs quantum dots in GaAs and AlGaAs matrices grown by molecular-beam epitaxy are investigated. It is shown that increasing the In content in the quantum dots has the effect of raising the energy of carrier localization and increasing the energy distance between the ground state and the excited states of carriers in the quantum dots. An investigation of the influence of postgrowth annealing on the optical properties of the structures shows that the formation of vertically coupled quantum dots and the use of a wide-gap AlGaAs matrix enhances the thermal stability of the structures. Moreover, high-temperature (830°C) thermal annealing can improve the quality of the AlGaAs layers in structures with vertically coupled InGaAs quantum dots in an AlGaAs matrix. It is important to improve the comprehensive performance of the ductile iron bars (DIBs) for the cylinder block of the extra high pressure hydraulic plunger pump and accelerate the industrial application. In this work, the LZQT600-3 DIBs with the diameter of 145 mm were prepared by the horizontal continuous casting (HCC) process, that is, LZQT600-3 HCCDIBs. The microstructure and room temperature tensile properties of different sections [left-edge (surface layer), left-1/2R (left half of the radius), and the center of the HCCDIBs] were studied. The results show that the spheroidization of LZQT600-3 HCCDIBs matrix from the left-edge, left1/2R to the center is at nodulizing grade II and above. As the cooling rate gradually decreases from surface to the center of the HCCIBs, the number of spheroidized graphite is gradually reduced, the size is gradually increased, the shape factor is decreased, and the pearlite content and lamellate spacing are increased. Along the horizontal direction of the section, the hardness of the material is distributed symmetrically around the center of the HCCDIBs. In the vertical direction, the hardness distribution in the center of the HCCDIBs is asymmetrical due to the gravity during the solidification process. Therefore, the microstructure in the lower part of the section solidifies relatively quickly. The left-edge has the best tensile mechanical properties, and the ultimate tensile strength, yield tensile strength and elongation are 597.3 MPa, 418.5 MPa and 9.6%, respectively. The tensile fracture belongs to the ductile-brittle hybrid fracture. Seed development is a complex process governed by highly coordinated changes in the expression of a large protein set. DIGE (Difference Gel Electrophoresis)-based proteomics was used to study developing Chinese fir seeds. 153 spots were obtained by using the analysis of DeCyder software (v. 6.5). Cluster analysis showed that they could be joined into three main groups. Eleven spots, more actively expressed at early cotyledonary stage of developing seeds, were identified by LC/MS/MS (tandem MS). Ten spots were identified by searching NCBInr or EST databases. They included two legumin-like storage proteins, LEA protein, small heat-shock protein, PR10-1.13, a protein similar to eukaryotic translation initiation factor, a protein similar to maternal effect embryo arrest 51, ORF115, a protein similar to monodehydroascorbate reductase, and unknown proteins. Adsorption and desorption of Cu(II) and Cd(II) in the soils derived from basalts aged 10, 1,330, and 2,290 kiloyears (ky) in the tropical regions of Hainan, China were investigated. X-ray diffraction analysis indicated that the clay minerals were dominated by smectite, halloysite, and kaolinite mixed with montmorillonite in the soil derived from 10 ky-basalt and by kaolinite, halloysite, and gibbsite in the soils derived from 1,330 and 2,290 ky-basalt. Soil cation exchange capacity (CEC) decreased with the increase in basalt age, while the content of soil-free iron oxides decreased for such increase in basalt age. The amount of Cu(II) and Cd(II) adsorbed by the soil derived from 10 ky-basalt was much higher than that adsorbed by the other two soils. A similar trend was observed for the desorption of Cu(II) and Cd(II) pre-adsorbed to these soils in 1 M KNO3. The desorption rate of Cu(II) decreased with the increase in age of basalts. This indicated that the electrostatic adsorption of Cu(II) decreased with the increase in degree of soil development. Similarly, the specific adsorption of Cu(II) increased with increase in degree of soil development. The desorption rate of Cd(II) showed much higher than the Cu(II), which ranged from 87.5% to 92.6%. This indicated that Cd(II) was adsorbed mainly through the electrostatic attraction. Removal of soil organic matter leads to the decrease in Cu(II) adsorption and the increase in Cu(II) desorption. The overall effect of organic matter removal on the adsorption and desorption of Cd(II) and desorption rate of Cd(II) was smaller compared to Cu(II). [ABSTRACT FROM AUTHOR]

There is an increasing recognition of the potential role of variants in mitochondrial DNA and nuclear-encoded mitochondrial proteins in modifying disease risk. This has led to a rise in the number of mitochondrial association studies being undertaken. The unique inheritance pattern of mitochondria makes mitochondrial DNA variation susceptible to having geographical structure. Such a structure may have a dramatic impact on mitochondrial association studies, particularly in heterogeneous populations. By combining self-reported ancestry data and mitochondrial genotype data for a sample of 3839 individuals from 1037 Australian families, population substructure is tested by looking for evidence of differences in mitochondrial haplogroup and single nucleotide polymorphism (SNP) frequencies between different ancestral groups in Australia. In addition, the substructure within ancestral groups is tested by comparing the similarity of mates to randomly drawn pairs of individuals from the same ancestral group. It is shown that there are significant differences in the frequency of variants both between European and non-European groups, and within Europe. This agrees with previous studies of European mitochondrial variation. No evidence was found for structure within ancestral groups. Spectrofluorometric titration, electrospray ionization time-of-flight mass spectrometric and UV melting methods were employed to study the binding of chelerythrine and sanguinarine to bulged DNA. The results showed that both alkaloids bind specifically to single pyrimidine (C, T) bulge sites. The ability of sanguinarine to bind to both regular and bulged hairpins was found to be stronger than that of chelerythrine, but the binding selectivity of chelerythrine toward single-base bulges was much larger than that of sanguinarine. In a sheet forming process, drawbead plays an important role on the control of the material flow. In this paper, a numerical procedure for the design of forming processes is described. It is based on the coupling of an optimization technique and the simplified one step finite element method (also called inverse approach). The optimization technique allows adjustment of the process parameters so that specified criteria are fulfilled. Response surface methodology (RSM) is a global approximation method, which is ideally suited for solving highly nonlinear optimization problems. The finite element method, in addition to predicting the response of the process to certain parameters, allows assessment of the effect of a variation in these parameters on this response. The authors utilize the one step method at the preliminary design stage to supply stress or strain information for the following optimization using RSM. The procedure for this optimization process is fully described. The front fender for Numisheet 2002 is presented and the real defect free workpiece is produced to demonstrate the usefulness of the proposed optimization procedure. A comparison between the two forming limit curves (FLC) before and after optimization and results obtained using the precise incremental commercial software DYNAFORM based on the explicit dynamic approach verify that the optimization design method of drawbead could be successfully applied in designing actual tools of auto body cover panels. Zeolite fibers have attracted growing interest for a range of new applications because of their structural particularity while maintaining the intrinsic performances of the building blocks of zeolites. The fabrication of uniform zeolite fibers with tunable hierarchical porosity and further exploration of their catalytic potential are of great importance. Here, we present a versatile and facile method for the fabrication of hierarchical ZSM-5 zeolite fibers with macro-meso-microporosity by coaxial electrospinning. Due to the synergistic integration of the suitable acidity and the hierarchical porosity, high yield of propylene and excellent anti-coking stability were demonstrated on the as-prepared ZSM-5 hollow fibers in the catalytic cracking reaction of iso-butane. Convincing epidemiological data suggest an inverse association between cancer and neurodegeneration, including Alzheimer's disease (AD). Since both AD and cancer are characterized by abnormal, but opposing cellular behavior, i.e., increased cell death in AD while excessive cell growth occurs in cancer, this motivates us to initiate the study into unraveling the shared genes and cell signaling pathways linking AD and glioblastoma multiform (GBM). In this study, a comprehensive bioinformatics analysis on clinical microarray datasets of 1,091GBM and 524AD cohorts was performed. Significant genes and pathways were identified from the bioinformatics analyses - in particular ERK/MAPK signaling, up-regulated inGBM and Angiopoietin Signaling pathway, reciprocally up-regulated in AD - connecting GBM and AD (P , 0.001), were investigated in details for their roles in GBM growth in an AD environment. OBJECTIVES:Crohn's disease (CD; MIM 266600) is one of the most common forms of inflammatory bowel disease (IBD), and represents a significant burden to health care in developed countries. Our aim was to determine whether a gene in the IBD linkage region on chromosome 19q13, with a role in Paneth cell secretion and T-cell activation, conferred genetic susceptibility to the development of CD.METHODS:In total, 792 CD cases and 1,244 controls of Australian origin (Caucasian) were genotyped for seven single-nucleotide polymorphisms (SNPs) in the gene encoding the intermediate conductance calcium-activated potassium channel protein (KCNN4) at 19q13.2. CD cases were phenotyped using the Montreal classification. The replication set comprised an additional 326 CD cases and 951 population-based Caucasian controls. Analysis of the KCNN4 mRNA transcript was carried out using quantitative reverse transcriptase-PCR.RESULTS:KCNN4 SNP rs2306801 was associated with CD (primary P=0.0008, odds ratio (OR) (95% confidence interval (CI)): 0.76 (0.65-0.89); replication P=0.01, OR (95% CI): 0.77 (0.61-0.97). Stratification by disease location identified the association between SNP rs2306801 and ileal CD (P=0.01). Non-inflamed ileal mucosa from CD patients carrying any of the common disease-predisposing NOD2 variants (R702W, G908R, 1007fs) had significantly reduced levels of KCNN4 mRNA expression (P=0.001). KCNN4 protein expression was detected in Paneth cells, and in T cells in inflamed lamina propria.CONCLUSIONS:Our data implicate the role of KCNN4 in ileal CD. By investigating the effects of recombinant human IGF-2 (rhIGF-2) on the diopter, axial eye length and the expression of IGF-2 in non-form-deprivation (FD) and FD eyes of guinea pig, we tried to elucidate the relationship between the effects of rhIGF-2 on eye growth and FD in guinea pig. Eighty 3-week-old guinea pigs were included in the study, which were divided into two groups randomly. Group A ( n = 40) was the non-FD group, Thirty-two guinea pigs received intravitreal injections of either 1 ng, 10 ng, 100 ng rhIGF-2 or bovine serum albumin (BSA) to the right eye; eight guinea pigs who received no intravitreal injections served as control. Group B ( n = 40) was the FD group; the right eyes were form-deprived, and received the same disposals as group A. The diopter and the axial eye length of all guinea pigs were measured at day 14. The expression of IGF-2 in the retina was measured by Western blot. After 14 days, FD eyes were high myopia. The axial length of FD eyes was significant for longer than that of non-FD eyes. The expression of IGF-2 in the retina of FD eyes was up-regulated. In non-FD group, there was no significant difference in the diopter and axial eye length among rhIGF-2 injection eyes, BSA injection eyes and control eyes, but the expression of IGF-2 in the retina of 10 ng and 100 ng rhIGF-2 injection eyes was obviously down-regulated. In the FDM (form-deprivation myopia) group, the myopic diopter and axial eye length increased in 10 ng and 100 ng rhIGF-2 injection eyes compared with those in BSA injection eyes and control eyes. The level of IGF-2 expression in the retina of 10 ng and 100 ng rhIGF-2 injection eyes was obviously down-regulated. The diopter and expression of IGF-2 in the retina of rhIGF-2 injection eyes was negatively correlated with the dose of each injection, which was positively correlated with the axial length. RhIGF-2 intravitreous injection does not affect the diopter and axial length in non-FD eyes of guinea pig, while it can induce a significant increase in myopic diopter and axial length of FD eyes. OBJECTIVES: Crohn's disease (CD) and ulcerative colitis (UC) are the two most common forms of inflammatory bowel disease (IBD), representing a significant health-care burden. A variant in the autophagy gene ATG16L1 (T300A) has been newly identified as a CD susceptibility locus by genome-wide association. Our aim was to assess the contribution of T300A in determining disease susceptibility and phenotype in two independent Australian IBD cohorts and explore the relationship between T300A and known CD risk factors (NOD2[nucleotide-binding oligomerization domain containing 2] status and smoking). METHODS: In total, 669 CD and 543 UC cases, and 1,244 controls (study 1), 154 CD cases and 420 controls (study 2), and 702 unaffected parents from both groups were genotyped. We conducted case–control and family association analyses, and investigated relationships between T300A and disease subgroups and between NOD2 status and cigarette smoking (CD only). RESULTS: The strong association between CD and T300A was confirmed ( P < 0.001), with a two-fold increase in disease risk associated with the GG genotype (odds ratio [OR] 1.96, 95% confidence interval [CI] 1.49–2.58), while ileal CD risk was almost three-fold (OR 2.73, CI 1.87–4.0). ATG16L1 and NOD2 were found to contribute independently to CD risk. A greater than seven-fold increased CD risk was observed for current smokers with a GG genotype ( vs nonsmoking AA genotype; P < 0.001, OR 7.65, CI 4.21–13.91). A significant inverse association was found between T300A and UC ( P= 0.002). This was strongest for patients with extensive, severe disease. A reversed-phase HPLC method has been developed for determination of twelve intact glucosinolates—glucoiberin, glucocheirolin, progoitrin, sinigrin, epiprogoitrin, glucoraphenin, sinalbin, gluconapin, glucosibarin, glucotropaeolin, glucoerucin, and gluconasturtiin—in ten traditional Chinese plants. The samples were extracted with methanol and the extracts were cleaned on an activated Florisil column. A mobile phase gradient prepared from methanol and 30 mmol L−1 ammonium acetate at pH 5.0 enabled baseline separation of the glucosinolates. Glucosinolate detection was confirmed by quadrupole time-of-flight tandem mass spectrometric analysis in negative-ionization mode. Detection limits ranged from 0.06 to 0.36 μg g−1 when 5 g of dried plant was analyzed. Recoveries of the glucosinolates were better than 85% and precision (relative standard derivation, n = 3) ranged from 5.3 to 14.6%. We conducted a genome-wide association pooling study for cutaneous melanoma and performed validation in samples totaling 2,019 cases and 2,105 controls. Using pooling, we identified a new melanoma risk locus on chromosome 20 (rs910873 and rs1885120), with replication in two further samples (combined P < 1 × 10−15).
