Summary: Background: Equine anoplocephalid cestodes are tapeworms that occur worldwide in horses and are known to cause pathological manifestations and clinical disease within the digestive system of horses. Objective: While these clinical manifestations and the efficacy of currently available anthelmintics against equine tapeworms have been well documented, a recent study has demonstrated treatment failure of praziquantel and pyrantel pamoate, suggesting resistance; however, this has not been heavily investigated. Study design: This case study examined 24 Thoroughbred yearlings on one Kentucky Thoroughbred horse farm to investigate the elimination of tapeworm eggs after administration of routine anticestodal anthelmintics after confirmation of positive faecal egg counts. Methods: The Mini‐FLOTAC faecal egg counting method was used to count anoplocephalid eggs in yearlings treated with pyrantel pamoate paste (13.2 mg base/kg), moxidectin‐praziquantel (0.4 and 2.5 mg/kg) and ivermectin‐praziquantel (0.2 and 1 mg/kg) over a 6‐month period in 2022. Results: Data demonstrated that tapeworm egg shedding persisted post‐deworming with all three anthelmintic products. Out of the 24 Thoroughbred enrolled, 11/24 (45.8%) had persistent infections in the month of March and 3/24 (12.5%) had persistent infections in April after deworming with ivermectin‐praziquantel. With moxidectin‐praziquantel, 2/24 (8.3%) remained egg count positive. For pyrantel pamoate, 3/24 (12.5%) were detected with tapeworm eggs post‐treatment. During July and August, selected yearlings had faecal sampling done before and 2 weeks post‐deworming with moxidectin‐praziquantel. For July 1/6 (16.7%) horses had tapeworm eggs both before and after deworming, and one additional horse was positive in the post‐treatment sample only. For the August treatment, 4/14 (28.6%) were egg count positive both before and after deworming, with one additional horse being positive in the post‐treatment sample only. Conclusions: This study demonstrated evidence of anthelmintic treatment failure against equine tapeworms by means of regular faecal egg count monitoring. Bertiella studeri, a typical intestinal cestode of nonhuman primates, accidentally infects human beings. However, B. studeri infection in monkeys has been rarely reported in recent years. A case of B. studeri infection was identified in one captive rhesus macaque with natural infection of Mycobacterium tuberculosis. Appropriate diagnoses of parasites of apex marine predators are crucial to understand their biodiversity, host specificity, biogeography, and life cycles. Such diagnoses are also informative of ecological and biological characteristics of both host and environment in which the hosts and their parasites live. We here (i) investigate the parasite fauna of a bluntnose sixgill shark Hexanchus griseus (Bonnaterre, 1788) obtained from the Gulf of Naples (Tyrrhenian Sea), (ii) characterize molecularly all its metazoan parasites, and (iii) resurrect and report the main morphological features and phylogenetic position of Grillotia acanthoscolex, a cestode species previously synonymized with Grillotia adenoplusia. A rich parasite fauna represented by eight different taxa was found, including two monogeneans (Protocotyle grisea and Protocotyle taschenbergi), one digenean (Otodistomum veliporum), four cestodes (Crossobothrium dohrnii, Clistobothrium sp., G. acanthoscolex, and G. adenoplusia), and one copepod (Protodactylina pamelae). Sequencing of these samples accounts for an important molecular baseline to widen the knowledge on the parasitic fauna of bluntnose sixgill sharks worldwide and to reconstruct their correct food chains. The bluntnose sixgill shark was found to be a definitive host for all endoparasites found here, confirming that it occupies an apex trophic level in the Mediterranean Sea. The taxa composition of the trophic parasite fauna confirms that the bluntnose sixgill shark mostly feeds on teleost fish species. However, the occurrence of two phillobothrid cestodes (C. dohrnii and Clistobothrium sp.) suggests that it also feeds on squids. Finally, we emphasize the importance of using integrative taxonomic approaches in the study of parasites from definitive and intermediate hosts to elucidate biology and ecology of taxa generally understudied in the Mediterranean Sea. Detection of intestinal parasites in fish typically requires autopsy, resulting in the sacrifice of the fish. Here, we describe a non‐lethal method for detecting the tapeworm Eubothrium crassum in fish using anal swabs and real‐time PCR detection. Two assays were developed to detect cytochrome oxidase I (COI) mitochondrial DNA and 18S ribosomal DNA sequences of E. crassum, respectively. The assays were tested on swab samples from confirmed pathogen free Atlantic salmon (Salmo salar L.) and on samples from farmed Atlantic salmon, where the presence and intensity of parasites had been established through autopsy. The COI assay was shown to be specific to E. crassum, while the 18S assay also amplified the closely related E. salvelini, a species infecting Arctic charr (Salvelinus alpinus L.) in freshwater. The COI assay detected E. crassum in all field samples regardless of parasite load while the 18S assay failed to detect the parasite in two samples. The results thus demonstrates that this non‐lethal approach can effectively detect E. crassum and can be a valuable tool in assessing the prevalence of infection in farmed salmon, aiding in treatment decisions and evaluating treatment effectiveness. Scavenger receptors participate in a wide range of biological functions after binding to multiple non‐self or altered self‐ligands. Among them, CD5 and CD6 are lymphocyte scavenger receptors known to interact with different microbial‐associated molecular patterns, and the administration of the recombinant soluble ectodomains of human CD5 (rshCD5) and/or CD6 (rshCD6) has shown therapeutic/prophylactic potential in experimental models of fungal, bacterial and echinococcal infections. The latter is a zoonosis caused by the larval stage of the cestode parasite Echinococcus granulosus sensu lato, which in humans can induce secondary cystic echinococcosis (CE) after the spillage of protoscoleces contained within fertile cysts, either spontaneously or during surgical removal of primary hydatid cysts. Herein, we have analysed the mechanisms behind the significant protection observed in the mouse model of secondary CE following prophylactic administration of rshCD5 or rshCD6. Our results show that both molecules exhibit intrinsic antiparasitic activities in vitro, as well as immunomodulatory functions during early secondary CE, mainly through Th1/Th17 cytokine bias and promotion of peritoneal polyreactive antibodies. These data support the relevance of the parasite components bound by rshCD5 and rshCD6, as well as the potential of their prophylactic administration as a useful strategy to reduce secondary CE in patients. Alveolar echinococcosis (AE) is a zoonotic parasitic disease, resulting from being infected with the metacestode larvae of the tapeworm Echinococcus multilocularis (E. multilocularis). Novel prophylactic and therapeutic interventions are urgently needed since the current chemotherapy displays limited efficiency in AE treatment. Bioengineered nano cellular membrane vesicles are widely used for displaying the native conformational epitope peptides because of their unique structure and biocompatibility. In this study, four T‐cells and four B‐cells dominant epitope peptides of E. multilocularis with high immunogenicity were engineered into the Vero cell surface to construct a membrane vesicle nanovaccine for the treatment of AE. The results showed that the nanovesicle vaccine can efficiently activate dendritic cells, induce specific T/B cells to form a mutually activated circuit, and inhibit E. multilocularis infection. Parasites are ubiquitous, yet their effects on hosts are difficult to quantify and generalize across ecosystems. One promising metric of parasitic impact uses the metabolic theory of ecology (MTE) to calculate energy flux, an estimate of energy lost to parasites. We investigated the feasibility of using metabolic scaling rules to compare the energetic burden of parasitism among individuals. Specifically, we found substantial sensitivity of energy flux estimates to input parameters used in the MTE equation when using available data from a model host–parasite system (Gasterosteus aculeatus and Schistocephalus solidus). Using literature values, size data from parasitized wild fish, and a respirometry experiment, we estimate that a single S. solidus tapeworm may extract up to 32% of its stickleback host's baseline metabolic energy requirement, and that parasites in multiple infections may collectively extract up to 46%. The amount of energy siphoned from stickleback to tapeworms is large but did not instigate an increase in respiration rate in the current study. This emphasizes the importance of future work focusing on how parasites influence ecosystem energetics. The approach of using the MTE to calculate energy flux provides great promise as a quantitative foundation for such estimates and provides a more concrete metric of parasite impact on hosts than parasite abundance alone. Parasites with complex life cycles are known to induce phenotypic changes in their intermediate hosts to increase transmission to the final host. The magnitude of these changes could increase with the number of parasites, which would be beneficial to co‐infecting parasites. Yet, adverse effects of high parasite load (i.e. many parasites in a single host) might stress both hosts and parasites (e.g. through an increased immune response). We investigated the consequences of parasite load on the transcriptional activity and morphology of the cestode Anomotaenia brevis and its intermediate host, the ant Temnothorax nylanderi. We demonstrated that many differentially expressed host genes shifted with parasite load, and their functions indicate a stronger immune response and fight against oxidative stress in heavily infected hosts. The expression of other host genes responded to infection in an all‐or‐nothing manner, as did the morphology of the host workers. However, the cestodes became smaller when they competed with other parasites for resources from a single host. Their expression profile further indicated shifts in host immune avoidance, starvation resistance and vesicle‐mediated transport. Host–parasite coevolution may lead to patterns of local adaptation in either the host or parasite. For parasites with complex multi‐host life cycles, this coevolution may be more challenging as they must adapt to multiple geographically varying hosts. The tapeworm Schistocephalus solidus exhibits some local adaptation to its second intermediate host, threespine stickleback, to which the parasite is strictly specialized. However, the tapeworm's adaptation to its first intermediate host (any of a number of copepod species) is not documented. We investigated if there was local adaptation and host specify in the tapeworm Schistocephalus solidus to its copepod first intermediate hosts. We exposed copepods from five lakes in Vancouver Island (BC, Canada) to local (i.e. same lake) and foreign tapeworms in a reciprocal exposure experiment. Results indicate that the tapeworm is not locally adapted to the copepods. Instead, we observed moderate‐effect host specificity, infection rates being higher in certain copepod species than in others. Infection rates also varied among cestode populations. These results show that although S. solidus infects multiple copepod genera, they are not equally competent hosts. Differences in S. solidus epidemiology among lakes is likely to be driven more by this partial specialization, than by local adaptation to first intermediate hosts. [ABSTRACT FROM AUTHOR]

Background: This study involving non-compliant, seized dogs entering the UK surveyed endoparasites detected in faecal samples. A focus was placed on taeniid infection as the detection of these tapeworms acts as a marker for failure of effective tapeworm treatment. Methods: Individual faecal samples taken from 65 dogs over a 24-month period were examined for helminth eggs, for protozoal oocysts and cysts, using a centrifugal flotation technique. Any sample presenting positive results for taeniid eggs had residual faeces examined using polymerase chain reaction to aid speciation of the tapeworm eggs. Additionally, a Baermann technique was used to assess faeces for lungworm larvae. Results: Patent endoparasite infection was detected in 27.7% of dog faecal samples. No sample was positive for lungworm larvae. Five dogs were co-infected with Isospora spp. and Toxocara canis. One dog sample was detected with taeniid eggs, identified as Taenia serialis. Conclusions: The taeniid-positive dog indicated that appropriate tapeworm treatment may not have occurred, reinforcing the risk to the UK of illegally imported dogs potentially introducing Echinococcus multilocularis infection. Cystic echinococcosis (CE) is one of the most important zoonotic diseases with a worldwide distribution. It is caused by the larval stage of the dog tapeworm "Echinococcus granulosussensu lato" and constitutes a major economic and public health problem in several countries. Protoscoleces are one component of this larval stage that can interact with both definitive and intermediate hosts. The aim of the present study was to investigate the potential role of using a radio‐attenuated form of these protoscoleces for immunoprophylaxis against experimental murine echinococcosis. However, mice were immunized twice at 15‐day intervals with gamma (γ) irradiated protoscoleces at doses of 0.4, 0.8, 1.2 and 1.4 kGy then challenged with the intact parasites. Macroscopic and histological analyses with cytokine measurements were performed in order to estimate the number and diameter of cysts, microscopic changes and cytokine profile. An improvement in protection against the challenge dose was observed with increasing dose, giving percentages of 47.7, 49, 55.23 and 70.6%, for the 0.4, 0.8, 1.2 and 1.4 kGy‐groups respectively. These data suggest that immunization with radio‐attenuated protoscoleces may induce satisfactory protective immunity by reducing successfully the formation of cysts, caused by challenge infection. Many anthelmintics target the neuromuscular system, in particular by interfering with signaling mediated by classical neurotransmitters. Although peptidergic signaling has been proposed as a novel target for anthelmintics, current knowledge of the neuropeptide complement of many helminth groups is still limited, especially for parasitic flatworms (cestodes, trematodes, and monogeneans). In this work, we have characterized the neuropeptide complement of the model cestode Hymenolepis microstoma. Peptidomic characterization of adults of H. microstoma validated many of the neuropeptide precursor (npp) genes previously predicted in silico, and identified novel neuropeptides that are conserved in parasitic flatworms. Most neuropeptides from parasitic flatworms lack significant similarity to those from other animals, confirming the uniqueness of their peptidergic signaling. Analysis of gene expression of ten npp genes by in situ hybridization confirmed that all of them are expressed in the nervous system and identified cryptic features, including the first evidence of dorsoventral asymmetry, as well as a new population of peripheral peptidergic cells that appears to be conserved in the trematode Schistosoma mansoni. Finally, we characterized in greater detail Attachin, an SIFamide homolog. Although its expression is largely restricted to the longitudinal nerve cords and cerebral commissure in H. microstoma, it shows widespread localization in the larval nervous system of Echinococcus multilocularis and Mesocestoides corti. Exogenous addition of a peptide corresponding to the highly conserved C‐terminus of Attachin stimulated motility and attachment of M. corti larvae. Altogether, this work provides a robust experimental foothold for the characterization of peptidergic signaling in parasitic flatworms. Currently, little is known about inhibitory substances enabling tapeworms to settle in fish intestines thereby avoiding proteolysis. Contrary to previous studies with certain host–parasite pairs, this research compares the inhibitory capacities in three tapeworm species of the same genus Proteocephalus from four different fishes (P. torulosus from dace and zope, P. sagittus from stone loach and P. cernuae from ruffe). The tapeworm extracts studied significantly reduced the activity of commercial trypsin (although to a lesser degree than the synthetic inhibitor of serine proteinases PMSF), displaying clear inter‐specific variation in worms' inhibitory ability. We also measured the proteolytic activity of the host intestinal mucosa exposed to tapeworm extracts which served as inhibitors. Based on per cent inhibition values, all tapeworm extracts significantly suppressed the mucosal proteolytic activity, with marked differences between certain host–parasite pairs. SDS‐PAGE electrophoresis of the incubation media and extracts detected in each tapeworm species 20–36 protein bands with apparent molecular weights from 10–12 to 312.5 kDa, mostly below 50 kDa. The incubation medium and extract of each parasite shared one to six bands ranging from 12 to 35 kDa, depending on its species, with only four bands common for two or more species. The band profiles suggest that in various Proteocephalus species inhibitory capacities against host proteinases can be ensured by different proteins. [ABSTRACT FROM AUTHOR]

Popular conceptions of the relationship between science and religion often emphasize the mutual compatibility but segregation of these two authoritative domains. However, along the uncertain border between these two spheres exist many questions that appear to influence and be influenced by both scientific and theological evidence. For biologists, the gruesome details of parasite biology and behavior illuminate a shadow in nature where science and theology must intersect—did a loving creator God design parasites? Here, I explore the writings and experiments of the early fathers of parasitology who established and matured a new scientific discipline while deeply integrating their theological beliefs with their scientific investigations. I argue that modern scientists can, and often should, follow in the footsteps of our scientific forefathers and intentionally allow our theological presuppositions to inspire scientific experiments, especially in these important boundary‐crossing subjects. In doing so, I believe we may advance scientific understanding of the way reality is by asking unexpected questions and uncover deeper truths about the character of God as revealed in his intricately complex and eternally fascinating creation. Members of the Torpedinidae (torpedoes) and Hypnidae (coffin ray) use electric organ discharges (EOD) to stun or kill their prey before consumption. We investigated whether EOD could also negatively affect the helminth larvae infecting these preys through a surrogate model: we applied electric discharges to individuals of blue whiting, Micromesistius poutassou, that harbored live larvae of Anisakis. Larval mortality throughout a 6‐h period was significantly higher in the treatment group, suggesting that EODs could significantly hamper helminth recruitment. We then tested whether torpedinids and hypnids ("strong‐EOD" families) harbored species‐poor helminth (cestode) assemblages compared with "weak‐EOD" Torpediniformes (Narcidae and Narkidae) and other Batoidea. Based on comparisons on estimated species diversity and mean species richness of tapeworms at host individual level we found that (i) Torpediniformes had the lowest tapeworm diversity of all Batoidea orders; (ii) Torpedo spp. consistently had the lowest mean cestode richness at host individual level, and this could not be related to other host factors influencing cestode diversity in chondrichthyans, that is body size, trophic level or dietary breath. However, a preliminary comparison between "strong‐EOD" and "weak‐EOD" Torpediniformes did not detect clear differences of cestode richness. The sub‐Alpine lakes of Switzerland, Italy and France have long been reported as an endemic area of diphyllobothriosis, a parasitic zoonosis caused by Dibothriocephalus latus. With this study, we explored the hypothesis for a relationship between the prevalence of D. latus in Perca fluviatilis and the Escherichia coli load in lake water. To do this, we identified eleven sampling sites in three areas (north, centre and south) of Lake Iseo (north Italy) to determine E. coli load and the prevalence of D. latus in P. fluviatilis. Prevalence and 95% confidence interval (CI) of D. latus infestation ranged from 0% (95% CI: 0.71–0.0) in Sarnico (southern area) to 20% (95% CI: 33.0–11.2) in Pisogne (northern area). There were significant differences in prevalence between the sites (χ2 = 31.12; p‐value =.0006) and in E. coli load (Kruskal–Wallis test; p‐value =.0005). There was decreasing gradient of E. coli load and prevalence of D. latus infestation from north to south. A significant positive correlation (r =.881; p‐value =.003) was found between E. coli load and prevalence of D. latus. Also, linear regression showed a significant relationship between E. coli load and prevalence of infestation (R2 =.775). Our findings offer an explanation for the link between E. coli load in water and D. latus prevalence. The potential factors in this link are the efficiency of the local wastewater treatment plant, the bathymetric profile of the lake and the life cycle of D. latus, which is mainly affected by light and water temperature. Tapeworms (Cestoda) represents a species rich (about 5000 species) group of flatworms (Neodermata) parasitizing all groups of vertebrates including humans, with about 1000 species parasitizing elasmobranchs and almost 500 occurring in teleosts as adults. They are common parasites of cultured fish, both as adults and larvae (metacestodes), but only few adult tapeworms are actually pathogenic for their fish hosts. In contrast, cestode larvae can be harmful for fish, especially plerocercoids migrating throughout their tissue and internal organs. Current knowledge of host–parasite relationships, including immune response of fish infected with tapeworms, is still insufficient to enable adequate control of cestodoses, and most data available were obtained several decades ago. Treatment of fish infected with adult tapeworms is effective, especially with praziquantel, whereas the treatment of metacestodes is problematic. The mechanisms enabling fish tapeworms to avoid proteolytic attacks by digestive enzymes of their fish host have been studied in less detail compared with mammalian cestodes. This study aimed to assess the inhibitory ability towards trypsin and chymotrypsin in Eubothrium rugosum, an intestinal parasite of burbot Lota lota, and establish its localization in the tapeworm. To this end, the worms were treated with Triton X‐100 followed by differential centrifugation to isolate the tegumental brush border membrane. The protease inhibitory abilities of the worms were mostly determined by their excretory/secretory products released into the incubation medium. These inhibitory abilities proved to be linked mainly with the brush border fractions. Notably, the per cent inhibition of both studied digestive enzymes (trypsin and chymotrypsin) hardly depended on the duration of the parasite exposure in the incubation medium, probably due to intermittent glycocalyx renewal. Improved knowledge on functions of the excretory/secretory proteins produced by fish tapeworms may contribute to a better understanding of host–parasite relations and development of new approaches to the treatment and prevention of diseases caused by pathogenic helminths. Aim: Trypanorhyncha cestodes comprise a wide range of heteroxenous parasites infecting elasmobranchs as definitive hosts. Limited data exist on the larval infection of these cestodes and the role of intermediate and paratenic hosts in the life cycle of these parasites. We investigated the factors that determine the occurrence and the level of infection of Grillotia plerocerci in the skeletal muscles of various benthonic sharks and analyzed the parasites through an integrative taxonomic approach. Location: Mediterranean Sea. Methods: Sharks obtained as bycatch of commercial trawling activities (i.e., Etmopterus spinax, Galeus melastomus, and Scyliorhinus canicula) were used in this study. Data from a limited number of Dalatias licha and Scyliorhinus stellaris were also included. Grillotia plerocerci were molecularly characterized using the partial 28S large subunit rDNA. Boosted regression trees were used to model the relationship between the abundance of infection with both morphological and physiological predictors in each host. Results: Plerocerci of Grillotia were detected in all shark species except S. stellaris. Host species significantly differed in terms of parasite abundance, with the highest and lowest prevalence and abundance of infection detected in G. melastomus and E. spinax, respectively. The relative influence of the traits involved in explaining the parasite abundance was related to the host size in G. melastomus, while both morphology‐ and physiology‐related traits explained the patterns observed in E. spinax and S. canicula. The 28S rDNA sequences shared an identity of ∼99.40% with a Grillotia species previously found in the Mediterranean Sea. At intraspecific level, two different genotypes were found. A first type was retrieved only from D. licha, whereas a second type was found in G. melastomus, E. spinax, and S. canicula. Main conclusions: Present results suggest that the two genotypes could be involved in different consumer‐resource systems and confirm most of the examined shark species as transport hosts of Grillotia species for unknown larger top predators. The distribution of parasites in host populations is integral to simulations of parasite populations.We compared three methods to measure parasite aggregation, namely the Hoover, coefficient of variation and I10 indices, using field data from a range of fish parasites. These included anisakid nematodes, trypanorhynch cestodes and polyopisthocotylean monogeneans from 10 previously published studies and one unpublished study.Positive correlations among the three methods were high. Standard errors and estimated biases obtained by bootstrap methods were smallest for the Hoover index. The structural response and plasticity of the cestode tegument in response to the influence of the host organism is not yet well understood. The main aims of our in vitro study were to analyse the ultrastructural mechanisms and kinetics of tegumental secretion in two cestode species, Dibothriocephalusdendriticus and Ligula interrupta, in response to the influence of fish host blood serum. The incubation of plerocercoids in the culture medium, which contained fish host blood serum, resulted in an increased number of secretory products on the tegumental surface. Our study is the first to experimentally demonstrate the formation of plerocercoid protective layers influenced by the host's internal environment factors. The mechanism of the generation of the protective layer included the following: the intensive formation of organelles in the tegumental cytons and their transfer to the distal cytoplasm of the tegument; increases in extracellular vesicles and vacuoles released on the tegumental surface; arrangement of secretory products and fine‐dispersed extracellular matrix in layers; and formation of the protective layer. The structural tegumental response included increases in the glycocalyx layer and structural changes. Our study revealed that the universal mechanism of protective layer formation was intrinsic to different tapeworms. We hypothesize that plerocercoids of cestodes parasitizing fish may use tegumental secretion in the formation of a protective layer and in the release of immunoregulator molecules to evade the host's immune response. Background: Coenurosis, known commonly as gid, is caused by a tapeworm Taenia multiceps. It is a disease of small ruminants globally but also occurs in large ruminants, especially in yak in the Himalaya. Gid is a pathological condition in young yaks, mostly below 3 years of age. The infected animal displays a circling movement with the head tilting towards the location of the cyst of a tapeworm on the cerebral surface of the brain. Objective: We conducted a study with the primary aim to gain an insight into yak herdsmen's practices to manage Coenurosis in the Laya administrative block of Bhutan. Methods: All seven villages of Laya were included for sampling. Seventy‐five out of 182 households owned yaks, and 54 yak‐owning households were selected randomly. The government livestock officials of nine yak‐rearing highland districts were also included in the study. A cross‐sectional study was conducted based on a questionnaire survey and focussed mainly on herdsmen's practices to manage gid. Two sets of questionnaires were used for yak herders and livestock officials. Each predesigned questionnaire was semi‐structured and consisted of both open– and closed‐ended questions. Results: The study revealed gid as a major cause of yak mortality. Gid occurred more in winter during migration and most herders lost one to three yaks annually. Herdsmen kept an average of two watchdogs and dewormed them once annually. Similarly, calves were also dewormed once annually. The carcasses of dead yaks were fed to dogs. Livestock officials were optimistic about controlling the disease in the future, despite the yak areas being difficult to access. Most herders had not attended the gid awareness programme. The animal health worker visited herds once annually. In absence of animal health workers, most herdsmen resorted to different practices to treat affected yaks –the most common practice being surgery. Gid was mentioned to harm herdsmen's economy. Conclusions: The study concluded that for effective management of gid in Laya, livestock agencies must create more awareness on gid, increase the frequency of visits by animal health workers to yak herds, and increase the frequency of deworming of watchdogs and calves. Proteocephalid tapeworms are the dominant group of endoparasites in siluriform catfishes in South America, but their interrelationships and actual diversity remain poorly understood. Molecular prospecting of proteocephalids found in pimelodid and other catfishes, especially in Pimelodus spp., along with thorough morphological evaluation, revealed high species diversity of these parasites. Using newly obtained sequences of the partial large subunit nuclear ribosomal RNA (lsrDNA) and complete mitochondrial cytochrome c oxidase subunit I (COI), phylogenetic relationships of the cestodes of Pimelodus catfishes were assessed and a new genus, Pseudoendorchis gen. n., proposed to accommodate P. auchenipteri comb. n. (type species; formerly Endorchis auchenipteri), P. cristata sp. n. from Pimelodella cristata (Heptapteridae), P. felipei sp. n. from Pimelodus fur, P. souzalimae sp. n. from Pimelodus maculatus and another three putative new species. Members of the new genus can be readily distinguished from all known Neotropical proteocephalids by having a large Mehlis' gland, representing at least 18% of proglottid width, and the vagina always anterior to the cirrus‐sac. Moreover, all phylogenetic, species discovery and validation analyses support validity of Pseudoendorchis. Combining the lsrDNA + COI sequence data appeared to provide the best resolution in phylogenetic analyses, which may be useful to reduce the uncertainty of the interrelationships among Neotropical proteocephalids. Aim: We test the predictions of the Stockholm Paradigm, a synthesis of eco‐evolutionary theory explaining the nature of faunal assembly, host range and parasite diversification. Faunal diversification and assembly, manifested in patterns of host colonization, co‐adaptation and parasite speciation, are predicted to emerge as a consequence of alternating episodes of ecological disruption and stability. Specifically, for a diverse cestode genus (Arostrilepis), we evaluate the number and direction of Pleistocene dispersal events across Beringia, the number and relative timing of host colonization events and the relationship between host and parasite biogeographic histories and associations through time. Location: Beringia and adjacent temperate to arctic biomes in North America and Eurasia. Taxon: Arostrilepis (Cyclophyllidea: Hymenolepididae) and its rodent hosts. Methods: Multi‐locus phylogenetic reconstruction and biogeographic ancestral range estimation. Results: Arostrilepis lineages crossed Beringia eastward into North America a minimum of four times and westward into Asia twice in association with temporally disjunct geographic expansions of three major tribes of cricetid rodents (Arvicolini, Myodini, Lemmini). Inferences of ancestral host associations support at least nine instances of host colonization involving shifts from one rodent tribe or family to another. Several previously unrecognized lineages of Arostrilepis are revealed. Main conclusions: Consistent with expectations of the Stockholm Paradigm, episodes of intercontinental dispersal were both frequent in the history of Arostrilepis and preceded a majority of inferred host‐colonization events. Events of historical geographic expansion created numerous opportunities for development of novel host–parasite associations through ecological fitting, as parasites tracked historically conserved resources available across diverse host taxa. Beringia played a major role in shaping rodent/parasite assemblages by mediating dispersal between the northern continents during glacial episodes of the Pleistocene, rather than by serving as a zone of refugial isolation. The Amur leopard, one of nine recently recognized subspecies of leopard, is still the most threatened by a stochastic procession of extinction. Evaluation of the potential danger to the conservation of the Amur leopard originating from disease urgently needs to be studied. Unfortunately, research on the potential risk to Amur leopards caused by disease is rare. In terms of parasitic diseases that affect this species, even basic data for parasitic fauna are absent. The aim of this study is to acquire this knowledge to improve the general understanding of Amur leopard parasites. Two migratory and endangered freshwater fish species, Salminus franciscanus and Brycon orthotaenia, are endemic to one of the most important basins for hydropower capacity in South America. These two fish are target species of conservation aquaculture programmes along the São Francisco River system.In order to implement effective strategies for the conservation of these species, microsatellite markers were developed and used to assess the genetic diversity and structure among wild groups. Fish were collected during the spawning season at four sampling sites, covering more than 1000 km.Polymorphism loci were assessed, and a total of 133 and 107 alleles were found for S. franciscanus and B. orthotaenia, respectively. Despite the critical conservation status, B. orthotaenia showed high levels of genetic diversity, comparable with those of other closely related species.Salminus franciscanus exhibited half of the genetic diversity of its sister species in terms of allelic diversity, indicating that this species will be more prone to genetic drift and inbreeding in conservation aquaculture programmes.The results of an analysis of molecular variance for microsatellite markers in these species (FST = 0.007 for S. franciscanus and FST = 0.003 for B. orthotaenia) and the estimated recent migration rates (>40% among sampling sites) support panmixia in both species, despite the heterogeneities of the riverscape.The results of this study and the microsatellites developed will play an important role in assisting agencies in making informed decisions on conservation aquaculture programmes. Background: Cysticercus tenuicollis (larvae of Taenia hydatigena) is a frequent cosmopolitan endoparasite of ruminants. The infestation by this parasite is underestimated since it is neither zoonotic nor inducing high economic losses in the sheep sector. Methods: This study aimed at estimating different parasitological indicators, Cysticercus tenuicollis infestation in small ruminants using a slaughterhouse‐based survey in Northeast Tunisia. A total number of 3,692 sheep and 78 goats were examined in the slaughterhouse of Menzel Temime. Results: The overall prevalence was estimated to be 2.8 (106/3692) and 8.9% (7/78) in sheep and goats, respectively. The abundance in these two species was 0.24 and 0.05 and the intensity 1.97 and 2.85, respectively. In goats, all the cysts were found in the mesentery, whilst, in sheep, the majority were in mesentery (96%) but also on the liver in 2% of the cases and in both organs (2%). There were only cysts with aqueous liquid with predominantly middle‐sized cysts (1 to 3 cm) corresponding to 63.15 and 70.34% in sheep and goats, respectively. Conclusion: Since, in small ruminants, the infestation by C. tenuicollis indicators are not high, low cost control measures should be implemented to eliminate this parasite in Northeast Tunisia. We report on two new lineages of the Eumida sanguinea complex from Great Britain and describe one of them as a new species using a multilocus approach, including the mitochondrial DNA COI‐5P and the nuclear markers ITS (ITS1, 5.8S rRNA and ITS2) and 28S rRNA. The molecular analysis placed Eumida mackiei sp. nov. in a monophyletic clade with 19.1% (COI), 10.1% (ITS) and 1.7% (28S) mean distance to its nearest neighbour. Molecular diagnoses were also applied to nine lineages within the E. sanguinea complex. This was complemented with morphometric data employing multivariate statistical analysis and the incorporation of statistical dissimilarities against three other described species from the complex. Eumida mackiei sp. nov. can be distinguished from E. notata and E. maia by the larger distance between the eyes and differences in morphometric proportions mainly in the dorsal and ventral cirri as well as in the prostomial appendages. E. sanguinea sensu stricto failed to produce a cluster of its own in the morphometric analysis, probably due to juvenile bias. Using the approach of sequencing the V3‐V4 region of the 16S rRNA gene, we have analysed the bacterial diversity associated with the distinct compartments of the gastrointestinal tract of perch (Perca fluviatilis) and cestodes (Proteocephalus sp.) parasitizing their digestive tract. The dominant microbiota associated with cestodes (Proteocephalus sp.) was represented by bacteria from the genera Serratia, Pseudomonas and Mycoplasma. By comparing the associated microbiota of perch and cestodes, a clear difference in bacterial composition and diversity was revealed between the community from the stomach content and other parts of the gastrointestinal tract of fish. Microbiota associated with cestodes was not significantly different in comparison with microbiota of different subcompartments of perch (mucosa and content of intestine and pyloric caeca) (ADONIS, p >.05) excluding microbiota of stomach content (ADONIS, p ≤.05). PICRUSt‐based functional assessments of the microbial communities of perch and cestodes indicated that they mainly linked in terms of metabolism and environmental information processing and could play an important role in the nutrition and health of host. Estimates of population size and trends are essential for effective conservation and management of wildlife populations. For harbour seals (Phoca vitulina), these data are required to fulfil statutory reporting obligations under national and international regulations.Aerial survey counts of harbour seals hauled out during their annual moult were used to estimate population sizes and trends at UK, regional (seal management unit, SMU) and local (Special Area of Conservation, SAC) scales.Results indicate that the current UK harbour seal population is similar to estimates from the late 1990s, but there were significant declines in some subpopulations and increases in others.Fitted trends suggest that the UK harbour seal population can be divided into three geographically coherent groups: South‐east populations (South‐East and North‐East England SMUs) have shown continuous increases punctuated by phocine distemper virus epidemics in 1988 and 2002; north‐east populations (East Scotland, Moray Firth, North Coast and Orkney, and Shetland SMUs) have declined since the late 1990s; north‐west populations (West Scotland, Western Isles, and South‐West Scotland SMUs) have remained stable or increased. Similar geographical population substructure is evident in recent population genetics results.Trends within SACs generally match SMU trends since 2002. Of the nine SACs designated for harbour seals, four declined (in East Scotland, Moray Firth, and North Coast and Orkney SMUs), four remained stable (in Shetland and West Scotland SMUs), and one increased (in South‐East England SMU).Large changes in relative abundance have resulted from differences in regional trends. Many parasites alter their host's phenotype in a manner that enhances their own fitness beyond the benefits they would gain from normal exploitation. Such host manipulation is rarely consistent with the host's best interests resulting in suboptimal and often fatal behavior from the host's perspective. In this case, hosts should evolve resistance to host manipulation. The cestode Schistocephalus solidus manipulates the behavior of its first intermediate copepod host to reduce its predation susceptibility and avoid fatal premature predation before the parasite is ready for transmission to its subsequent host. Thereafter, S. solidus increases host activity to facilitate transmission. If successful, this host manipulation is necessarily fatal for the host. I selected the copepod Macrocyclops albidus, a first intermediate host of S. solidus, for resistance or susceptibility to host manipulation to investigate their evolvability. Selection on the host indeed increased host manipulation in susceptible and reduced host manipulation in resistant selection lines. Interestingly, this seemed to be at least partly due to changes in the baseline levels of the modified trait (activity) rather than actual changes in resistance or susceptibility to host manipulation. Parasitic helminths have evolved strategies to evade their host's immune systems. Particularly, the early time of interactions between helminths and their hosts might be decisive for their infection success. We used the cestode Schistocephalus solidus, and its highly specific second intermediate host, the three‐spined stickleback (Gasterosteus aculeatus) to investigate parasite infection and host cellular immune responses starting 1 day postexposure (dpe). We recovered live parasites from stickleback body cavities already 24 hr after exposure. Infection rates increased up to 50% and did not change from 4 dpe onwards. Thus, not all parasites had reached the body cavity at the early time points and clearance of the parasite at later time points did not occur. Stickleback head kidney leucocytes (HKLs) did not show distinct signs of activation and lymphocyte proliferation, granulocyte‐to‐lymphocyte ratios and respiratory burst activity of infected sticklebacks did not deviate from controls significantly. The immune system was activated only late, as indicated by an increase in the total count of HKL relative to stickleback weight (HKL per mg fish), which was significantly elevated in infected fish 32 dpe. Objective: Preventive chemotherapy is the WHO-recommended control method for soil-transmitted helminthiases. In the Bolivian Chaco, 6-monthly single-dose mebendazole delivery to school-age children achieved a dramatic decrease in soil-transmitted helminthiases prevalence between 1987 and 2013. Consequently, in September 2016, preventive chemotherapy delivery was interrupted in nine rural communities. In compliance with WHO recommendations, we intensified surveillance to monitor soil-transmitted helminthiases prevalence and detect potential changes that would require interventions.Methods: We conducted two cross-sectional parasitology surveys 12 months apart (September 2016-2017) among school-age children living in the communities where preventive chemotherapy delivery had been halted. Study design, methods of sampling and sample analysis technique (direct microscopy, Kato-Katz technique) followed WHO recommendations, aiming to obtain data representative of the Bolivian Chaco ecological zone.Results: We collected 426 samples in 2016 and 520 in 2017. Soil-transmitted helminthiasis prevalence was unremarkable: 0.7% (95% CI 0-1.5%) in 2016 and 0.8% (0-1.5%) in 2017. Conversely, the prevalence of tapeworms (13% in 2016, 12% in 2017) and intestinal protozoan infections (81% in 2016 and 75% in 2017) continued to be high.Conclusions: Our findings support the role of preventive chemotherapy in reducing soil-transmitted helminthiases transmission, as otherwise poor hygienic and health conditions persist in the Bolivian Chaco. Abstract: Microtriches on the scolices and adjacent strobila of seven species of diphyllobothriidean cestodes (Bothridium pithonis, Cephalochlamys namaquensis, Dibothriocephalus latus, Duthiersia expansa, D. fimbriata, Ligula intestinalis, and Schistocephalus solidus) from different hosts (frogs, snakes, lizards, birds, and mammals) and biogeographic areas were examined using scanning and transmission electron microscopy. The basic structure of the tegument of the seven species studied does not differ markedly from that found in other cestodes. The main characteristic is the presence of electron‐dense bodies and vesicles in the distal cytoplasm. However, this study has shown differences in the morphology of microtriches even among species of the same family. Two different types of microtriches were found, filitriches and spinitriches, with the latter represented by two forms. Our study reveals that capilliform filitriches are most commonly found in Diphyllobothriidea. They were observed mainly on the strobila and the scolices of all but one studied species; individuals of L. intestinalis bore only coniform spinitriches on their surface. The same type of microtriches was found on the cirrus in D. latus. Gladiate spinitriches covered the scolex in both species of Duthiersia, and gladiate spinitriches interspersed with capilliform filitriches were observed on the anterior part of the strobila in D. fimbriata and the posterior part of the scolex in B. pithonis. Individuals of C. namaquensis were covered only by small acicular filitriches. Objective: The zoonotic helminth Taenia solium is endemic in Zambia, causing human (taeniasis and (neuro)cysticercosis) and pig (porcine cysticercosis) diseases with high health, social and economic burdens. We aimed to evaluate the impact of a health educational program intended to lead to powerful and cumulative improvements in knowledge, attitudes and practices that decrease parasite transmission and disease occurrence.Methods: Half-day health education workshops were conducted in three primary schools in the highly endemic Eastern Province of Zambia, using the computer-based T. solium educational program 'The Vicious Worm'. Questionnaires were administered before and after the educational component to determine the program's impact on knowledge uptake in primary school students.Results: In total, 99 students participated: 38 males and 61 females, with a median age of 14 years (range 10-18 years). Baseline general knowledge of T. solium, including awareness of the different human and pig disease states, and disease diagnosis, treatment and prevention, was quite high (average score 62%) and consistent across all three study areas. Participants' knowledge had significantly increased after the educational component, particularly regarding parasite transmission and disease prevention.Conclusion: Preliminary assessment of 'The Vicious Worm' indicates it is an effective tool for the short-term T. solium education of primary school students in Zambia. Follow-up studies are planned to assess the longer term impact of the program on knowledge uptake in the study neighbourhoods. Phylogenetic relationships, evolutionary history and systematics of tapeworms of the family Catenotaeniidae were studied using nucleotide sequences of the partial 28S nuclear rDNA (ca. 1,500 bp) and mitochondrial 12S–16S DNA (ca. 820 bp) genes. The tapeworm material consists of 29 species, including type species of the genera Catenotaenia Janicki, 1904, Catenotaenioides Haukisalmi, Hardman and Henttonen, 2010, Pseudocatenotaenia Tenora, Mas‐Coma, Murai and Feliu, 1980, Skrjabinotaenia Akhumyan, 1946, Meggittina Lynsdale, 1953, and Hemicatenotaenia Tenora, 1977. The basal phylogenetic structure of the Catenotaeniidae remains unresolved, but it is shown that most of the catenotaeniids in Eurasia and Africa comprise a large clade represented by species of Catenotaenia, Catenotaenioides, Skrjabinotaenia and Meggittina, parasitizing murid, cricetid, nesomyid and sciurid rodents. The results suggest that the divergence and early radiation of this clade have occurred in murid rodents (represented by Apodemus spp. and Mus musculus in the present material) in western Eurasia, followed by colonization of Africa, most likely independently of the colonization of their murid hosts between these continents. There is very little evidence of cophylogeny between hosts and parasites, suggesting that host transfers have played a major role in the divergence of catenotaeniids. In Africa, catenotaeniids have radiated in other murid and nesomyid rodents, and later colonized Madagascar and recolonized Eurasia. The results also show that the subfamily Skrjabinotaeniinae (including Skrjabinotaenia and Meggittina) is monophyletic, but the Catenotaeniinae (including Catenotaenia, Catenotaenioides, Pseudocatenotaenia and Hemicatenotaenia) is clearly non‐monophyletic. In addition, the genera Catenotaenia and Skrjabinotaenia were both found to be non‐monophyletic. Based on the phylogenetic and morphological evidence, several taxonomical changes, mainly new combinations, are proposed. Litobothrium aenigmaticum Caira, Jensen, Waeschenbach, & Littlewood 2014 is a cestode species that parasitizes pelagic thresher sharks in Taiwan and the Gulf of California. A previous study using light microscopy suggested the scolex contains four types of unusual tissues, one of which was considered to aid in adhesion of the worm to the mucosa of the spiral intestine. The function of the other tissues was unknown. Also unknown was the function of two laterally paired ducts found extending throughout the length of the worm. The goal of the present study was to use transmission electron microscopy ( TEM) to examine the internal anatomy of the scolex and cephalic peduncle of this tapeworm in more detail. Three specimens from Taiwan were examined with TEM and two with light microscopy. Transmission electron microscopy revealed that the laterally paired ducts were excretory, since they were closely associated with numerous protonephridia. The number of cell types, previously referred to as tissues, in the scolex was expanded to 11 based on a combination of features including cell size, nucleus size, ratio of the maximum diameter of the nucleus to maximum diameter of the whole cell, and types of observed organelles, as well as the presence or absence of cytoplasmic electron-dense and electron-lucent vesicles. The cytoplasm of all 11 cell types was found to include electron-dense vesicles, and all types were periodic acid-Schiff ( PAS) positive. This suggests that the electron-dense vesicles may contain glycoproteins and/or mucoproteins. As all 11 cell types occurred within the region of the scolex that is surrounded by a papilla of host mucosa at the site of tapeworm attachment, we hypothesize that one or more of these cell types may produce a substance that induces this inflammatory reaction. The specific products in the secretory vesicles and how those products are released are still unclear, as are any relationships between or among the cell types in terms of products produced. This study focuses on plerocercoids of the cestode Diphyllobothrium ditremum in brown trout Salmo trutta from the subalpine lake Øvre Heimdalsvatn in south-central Norway. Salmo trutta was the only fish species in this lake until European minnow Phoxinus phoxinus was registered in 1969. The P. phoxinus population increased substantially in the following years. In contrast with the 1969-1972 period, when plerocercoids of D. ditremum were practically absent in S. trutta, there was a high prevalence and intensity of infection in the 2013 S. trutta samples. Because the life cycle of D. ditremum involves two larval stages, in copepods and salmonids and mature worms in piscivorous birds, such as mergansers and loons, a change in feeding ecology of S. trutta or changes in population densities of copepods, fish or birds might have influenced the infection pattern. No relationships between D. ditremum infection and muscle-tissue δ15N signature or Hg concentration were found, indicating that infection is not a result of piscivory or cannibalism. Furthermore, consumption of copepods by S. trutta during summer and autumn was low. On the other hand, the number of piscivorous birds has increased, probably due to the presence of P. phoxinus as a new and numerous prey. An increased number of final D. ditremum hosts may have produced a higher output of cestode eggs, resulting in more infected copepods that in turn are consumed by S. trutta. Indirectly, P. phoxinus may therefore have caused the observed increased infection in S. trutta and thereby imposed further negative effects on S. trutta in high mountain areas. Neurocysticercosis, the central nervous system's localised form of cysticercosis, is considered to be the leading cause of epilepsy in the developing world. In Europe, the disease is mainly imported and affects both immigrants and travellers. However, autochthonous cases of cysticercosis in low-endemic countries could also originate from Taenia solium carriers (migrants or travellers) who acquired taeniasis overseas. Management of cysticercosis is a challenge for European healthcare providers as they are often hardly aware of this infection and have little familiarity in managing this disease. This study provides a summary of recommendations concerning screening, diagnosis and management of cysticercosis and T. solium taeniasis in Europe drawn up by nine experts in migrant health and imported diseases with experience in cysticercosis and T. solium taeniasis. Some fish parasites constitute severe management problems as they may cause mortality of their fish host or are important zoonoses of humans. Parasite assessments are therefore critical to keep track of infections. If conventional sampling techniques can be simplified, parasite assessments might be easier to obtain, less time-consuming and more extensive. In this study, we compare the assessed number of Diphyllobothrium spp. cysts ( CYST) with the counted number of Diphyllobothrium spp. plerocercoid larvae recovered using a conventional digestive technique ( LARV). The aim was to determine the potential of using CYST as a simplified methodology for assessing Diphyllobothrium spp. infection in salmonids. In total, 365 brown trout and 424 Arctic charr were sampled from nine lakes in subarctic Norway. Strong correlation, significant linear relationship and large amount of explained variation were found between log10 CYST and log10 LARV in both fish species. The method had a slight, but not significant tendency to work better in charr compared to trout. In addition, absolute difference between CYST and LARV increased at parasite intensities >100 indicating that the method has reduced functionality when estimating parasite intensity in heavily infected salmonid populations. However, overall, using this simplified and less time-consuming methodology, a good indication of Diphyllobothrium spp. intensity, abundance and prevalence was obtained. We suggest that this method provides a sound proxy of the Diphyllobothrium spp. burden and have the potential to be used in parasite assessment during fish monitoring and fisheries management surveys, particularly if the time and resources for detailed parasite studies are not available.
