5. Spray‐dried immobilized lipase from Staphylococcus aureusHA25 for application in detergent industry.

This study aims to produce an active lipase detergent additive dry powder using spray drying. Staphylococcus aureus HA25, growing at a pH range of 5.0–8.5, was isolated from Erzurum gogermis cheese and purified using a three‐phase partitioning technique. Optimal immobilization processing conditions were determined for 0.1% wt/wt chitosan, alginate, and chitosan/alginate concentrations of pure lipase enzyme. Morphological features of the immobilized enzyme structure were determined using scanning electron microscopy (SEM) analysis, and structural characterizations were determined using x‐ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, and thermogravimetric (TG) analysis. The results showed that the natural structure of the lipase was largely restored upon reconstitution of the spray‐dried immobilized lipase structures in water. While the free enzyme removed 52.6% of the oil added to the cotton fabric, the immobilized lipase@alginate enzyme removed ~98% of the oil added to the cotton fabric at the highest rate when used as a detergent additive. It was found that the reusability activity of chitosan@lipase, alginate@lipase, and chitosan/alginate@lipase enzymes remained at 86.4%, 92.8%, and 88.6% of their original activity, respectively. The study suggests that immobilized variations of the lipase enzyme within chitosan, alginate, and chitosan/alginate matrices may serve as a natural, secure, and efficient substitute for conventional chemical detergents, offering a non‐toxic alternative for additive materials. In Turkish cuisine, ready–to–eat vegetable salads (REVS) served with pide/lahmacun, kebab types, and tantuni from animal source in meat restaurants were evaluated since they have the potential to carry risks in terms of Public Health. The microbiological properties of REVS were investigated using agar plate method. Antimicrobial resistance of foodborne pathogens including Escherichia coli and Staphylococcus aureus was tested using Kirby–Bauer disc diffusion method. Moreover, the presence of important enteric viruses was detected by Polymerase Chain Reaction (PCR). The number of total aerobic bacteria, coliform bacteria, yeast and molds and, Staphylococcus and Micrococcus spp. ranged from less than 1 to 6.40, 1 to 6.26, less than 1–5.82 and less than 1–5.66 log10 colony forming units·grams-1 (CFU·g–1) in REVS samples, respectively. None of the REVS tested in this study contained Salmonella spp., whereas E. coli and S. aureus were isolated in 38.1% (16/42) and 2.4% (1/42), respectively. S. aureus was resistant to gentamicin, kanamycin, aztreonam, and ciprofloxacin in the disc diffusion assay, however, it was not harboring the mecA gene. E. coli strains (n=16) were resistant (100%) to aminoglycoside antibiotics and 35.7% (6/16) of the isolates were extended spectrum beta lactamase (ESBL) producing. blaTEM and blaCTXM8/25 were detected in two isolates, whereas one isolate carried blaCTXM–1 and blaTEM together by PCR. Of the REVS, two were evaluated as positive for rotavirus (4.8%), six for hepatitis A (14%), and hepatitis E virus (14%). These results indicate the high microorganism load, presence of ESBL E. coli, and viral enteric pathogens in REVS, hence it is important to perform routine hygiene practices. Ensaladas de verduras listas–para–comer (EVLC) que se sirven con pita/ lahmacun, tipos de kebab y tantuni de origen animal en los asadores de la cocina turca, ya que tienen el potencial de conllevar riesgos en términos de salud pública fueron evaluadas. Se investigaron las propiedades microbiológicas de REVS utilizando el método de placa de agar. La resistencia a los antimicrobianos de los patógenos transmitidos por los alimentos, incluidos Escherichia coli y Staphylococcus aureus, se probó mediante método de difusión por disco de Kirby–Bauer. Además, se detectó la presencia de importantes virus entéricos por Reacción en Cadena de la Polimerasa (RCP). El número de bacterias aeróbicas totales, bacterias coliformes, levaduras y mohos y Staphylococcus y Micrococcus spp. varió de menos de 1 a 6,40; 1 a 6,26; menos de 1 a 5,82 y de menos de 1 a 5,66 log10 Unidades Formadoras de Colonias·gramos-1 (UFC·g–1) en muestras EVLC, respectivamente. Ninguna muestra de EVLC analizadas en este estudio contenía Salmonella spp., mientras que E. coli y S. aureus se aislaron en el 38,1 % (16/42) y el 2,4 % (1/42), respectivamente. S. aureus fue resistente a la gentamicina, la kanamicina, el aztreonam y la ciprofloxacina en el ensayo de difusión en disco; sin embargo, no albergaba el gen mecA. Las cepas de E. coli (n=16) fueron resistentes (100 %) a los antibióticos aminoglucósidos y el 35,7 % (6/16) de los aislamientos produjeron beta lactamasa de espectro extendido (BLEE). blaTEM y blaCTXM8/25 se detectaron en dos aislados, mientras que un aislado portaba blaCTXM–1 y blaTEM juntos mediante RCP. De los EVLC, dos fueron evaluados como positivos para rotavirus (5 %), seis para hepatitis A (14 %), y virus de la hepatitis E (14 %). Today, there is a growing demand for fast, reliable and cost-effective systems for the detection, monitoring and diagnosis of food ingredients as well as contaminants in food. Quantitative/qualitative analysis of foods is carried out using traditional analytical methods such as chromatographic and spectroscopic techniques. Despite their sensitivity and accuracy, these methods are challenging due to the multi-stage and complex sample preparation procedures, requiring specialized personnel and expensive instrumentation for analysis. In addition, their labor-intensive and time-consuming nature eliminates the possibility of on-site and high-frequency monitoring of the analytes. A wide variety of nanotechnology-based new nanobiosensors are being developed in order to eliminate the difficulties posed by these techniques. In this context, in the present study, nanomaterials used in the development of nanobiosensors for use in foods, their working principles and their use in foods were examined.

The aim of this study was to investigate the effects of Cysteamine (Cys) and Fetal Calf Serum (FCS) in Synthetic Oviduct Fluid (SOF) and Charles and Rosenkrans (CR1aa) on the bovine in vitro embryo production and its ability of freeze. The oocytes were divided into two groups and allowed to mature in TCM-199, with and without cysteamine. They were divided into 4 subgroups according to whether they contain Cys and FCS in 2 different culture media (SOF and CR1aa). Accordingly, 8 groups were formed as SOF+FCS+Cys, SOF+Cys, SOF+FCS, S Dulbeco's OF, CR1aa+FCS+Cys, CR1aa+Cys, CR1aa+FCS and CR1aa. Embryos were cultured for 7-9 days and were frozen using vitrification method. Development of embryos was observed during the first 24 hr post-thaw period. The highest rate of compact morula was 20% in SOF+FCS+Cys and 19.4% in CR1aa+FCS+Cys. Toplanan oositler iki gruba ayrıldı ve sisteaminli ve sisteaminsiz TCM-199 medyumunda maturasyonuna izin verildi. 2 farklı medyumda (SOF ve CR1aa) kültüre edilmek üzere Cys ve FCS içerip içermediklerine göre 4 alt gruba ayrıldı. Buna göre 8 alt grup; SOF+FCS+Cys, SOF+Cys, SOF+FCS, SOF, CR1aa+FCS+Cys, CR1aa+Cys, CR1aa+FCS ve CR1aa olarak oluşturuldu. Ejaculates were obtained from five Saanen bucks by means of an electro-ejaculator and divided into five aliquots. Then, aliquots of ejaculates were used to create five experimental groups as follows: Group 1 (control), group 2A containing seminal plasma (SP) and bovine serum albumin (BSA; 10 mg/mL(-1)), group 2B containing BSA without SP, group 3A containing egg yolk (EY; 17.00%) and SP and finally group 3B containing EY without SP. All aliquots of semen were extended with Tris-based extender. Spermatological parameters of experimental groups were observed at 0, 6, 12 and 24 h periods during storage at 4 degrees C. Motility of semen in groups 2A and 2B was found to be best values compared to other groups. However, the motility of group 3A at 24 h was 0.00%. In conclusion, it was concluded that the addition of 10 mg/mL(-1) of BSA to Tris-based extender could be useful in the storage of Saanen goat semen regardless of SP at 4 degrees C.

Colistin is one of the most effective antibiotics against multidrug resistant Gram-negative bacteria. However, the recent emergence of plasmid-borne mobilized colistin resistance (mcr) genes is considered a serious antimicrobial resistance challenge worldwide. In this study, we report detection of anmcr-1carryingEscherichia coliisolate (named ATAVETmcr-1 Turkey) from retail raw chicken meat in Turkey. Of the 11 (from 500 total tested) phenotypically colistin-resistant isolates, 1 was shown to carry themcr-1gene by PCR. Whole-genome sequencing indicated thatmcr-1was located on a similar to 13 kb-long contig that was almost identical to the corresponding part in pZJ1635, an IncI2 plasmid encodingmcr-1in the same genetic context in anotherE. colistrain. In addition, ATAVETmcr-1 Turkey harboredbla(CTX-M-8),qnrB19,mdf(A),tet(A),sul2,aph(3 '')-Ib,aph(6)-Id, andfloRresistance genes. Phylogenetic analysis based on whole genome and multilocus sequence typing indicated that ATAVETmcr-1 Turkey was more closely related tomcr-1carryingE. coliisolates from food and human clinical samples previously reported from different parts of the world than to those from Turkey. These findings further emphasize the worldwide emergence and spread ofmcrmeditated colistin resistance in bacteria with zoonotic potential within animals and the food chain.

Sekanslama teknolojisiningelişmesiyle birlikte hücreler arası iletişimde rol oynadığı belirlenen 19-24bazlık uzunluğa sahip mikroRNA (miRNA)’lar keşfedilmiştir. Bu miRNA’larıninsanda hücrenin gelişimi ve farklılaşmasının yanı sıra metabolizmada da önemligörevleri bulunmaktadır. Mikroveziküller içerisinde taşınan miRNA’lar bu sayedeaşırı pH ve sıcaklık gibi koşullara bağlı olarak parçalanmaktan korunur. Yapılan güncel çalışmalarda miRNA ekspresyonuile gıdaların metabolizmada hücresel düzeyde faaliyetleri düzenlediğibelirlenmiştir. İlk defa diyetlerine pirinç konularak beslenen farelerdeyapılan bir çalışmayla pirinçe ait miR-168a’nın farelerin sindirim sistemitarafından absorbe edildiği belirlenmiş ve bu miRNA’nın kolesteroltransportunda rol oynayan LDLRAP1 mRNA ekspresyonunu engellediği tespitedilmiştir. miRNA’ların sindirim sistemi tarafından absorbe edildiğinin ortayakonduğu çalışmalarda, özellikle hayvansal kaynaklı miRNA’ların serum veplazmaya geçtiği ileri sürülmüştür. Sahip olduğu bu potansiyel ile miRNA’lar,diyet aracılığıyla ortaya çıkan sağlık problemlerin engellenmesinde biyolojikmarker olarak kullanılabilecektir. Gıdaların sadece içermiş oldukları yağ, şeker, protein, vitamin gibibileşenlerinin değil aynı zamanda sahip oldukları miRNA’ların metabolizmayıdoğrudan veya dolaylı yollarla etkilediği bilinmektedir. Gıda kaynaklımiRNA’ların önem kazanmasıyla birlikte güncel bir besinsel miRNA veri tabanıoluşturulmuş ve burada elma, atlantik salmon, muz, tavuk, tavuk yumurtası,mısır, sığır yağı ve sütü, anne sütü, üzüm, portakal, domuz, pirinç, domates,soya fasulyesi ve buğday gibi gıdalarla ilgili miRNA profilleri bulunmaktadır.Bu çalışmada miRNA biyosentezi, eylem mekanizması ve gıdalardaki varlığı ilemetabolizma üzerine olan etkileri tartışılmıştır., With the development ofsequencing technology, microRNAs (miRNA) have between 19-24 bases in lengthwhich play a role in cell communication was discovered. Inthe current studies, it has been determined that the foods have regulate theactivities at cellular level in metabolism by miRNA expression. ’This was firstdemonstrated by detecting rice miR-168a which absorbed by digestive system. On the other hand, thesemiRNAs were found to inhibit the mRNA expression of LDL RAP1 involved incholesterol transport. In studies where miRNAs have been absorbed by thedigestive system, it has been suggested that miRNAs, in particular animal origin,pass into serum and plasma. With the miRNAs have this potential, it can be usedas biological markers for the prevention of the health problems associated withdiet. A recent dietary miRNAdatabase was created with the increasing importance of foodborne miRNAs andthere are miRNA profiles associated with foods such as apples, Atlantic salmon,bananas, chicken meat and egg, corn, beef fat and milk, breast milk, grapes,oranges, pork, rice, tomato, soybeans and wheat in this database. In thisstudy, biosynthesis, the mechanism of action and presence in food and effectson metabolism of miRNAs were discussed.

StafilokokalEnterotoksin’lere ait bugüne kadar 24 farklı antijenik serotip tanımlanmıştırfakat stafilokokal gıda zehirlenmelerinin % 95’inden klasik enterotoksinler (A,B, C, D ve E) sorumlu tutulmaktadır. Staphlyococcusaureus (S. aureus), gıdadabulunan toksinlerinin alınımını takiben 2-4 saat sonra abdominal kramplar,bulantı, kusma ve diyare gibi zehirlenme semptomlarına neden olan ubiquiterözelliğe sahip bir mikroorganizmadır. Peynir, yüksek seviyedeki besinseliçeriğinden ötürü S. aureus’ungelişimi için uygun bir substrattır. S.aureus’un peynirde ortaya çıkışının sebebi olarak yüksek miktarda S. aureus içeren mastitisli sütlerinpastörize edilmeden peynire işlenmesi, kullanılan starter kültür aktivitesininyetersiz olması, sütün pastörizasyon sonrası kontaminasyonu ile ürününişlenmesi ve depolanması sırasındaki uygun olmayan şartlar düşünülmektedir.Birçok peynir türünün üretimi ve olgunlaşması sırasında S. aureus’un gelişimi ve bazı vakalarda enterotoksin üretimi,sanitasyon öneminden dolayı bazı araştırmacılar tarafından incelenmiştir. Buçalışmada S. aureus’un genelkarakteristikleri, peynirdeki varlığı, epidemiyolojisi ve koruma-mücadele yöntemleriderlenmiştir., Up to now, 24different antigenic serotypes of Staphylococcal enterotoxins have beendescribed, but classical enterotoxins (A, B, C, D and E) are responsible for95% of staphylococcal food poisonings.Staphlyococcus aureus (S. aureus)is an ubiquitous microorganism that causesymptoms of poisoning such as abdominal cramps, nausea, vomiting and diarrheaafter 2-4 hours following ingestion of toxins present in food. Cheese is asuitable substrate for the development of S.aureus due to its high nutritional content. The reason for the emergence ofS. aureus in cheese are the inabilityto pasteurize the milk of animals exposed to mastitis infection with highamounts of S. aureus, the inadequatestarter culture activity, the post-pasteurisation contamination and theunfavorable conditions for product processing and storage. During theproduction and maturation of many cheese species, the development of S. aureus and the production ofenterotoxin in some cases have been investigated by some researchers for itsimportance of sanitation. In this study, the general characteristics of S.aureus, the presence in cheese, epidemiology and protection-fighting methodshave been compiled.

In this research, it was aimed to determine the microbiological and chemical properties of raw cow milk in smallholder dairy farms. 49 milk samples were used in the study. Chemical properties of samples analyzed was determined to be good and these properties are met the requirements in Turkish Food Codex. However, Microbiological quality of raw milk samples collected from bulk tank milk was not adequate and significant part is not met the requirements in Turkish Food Codex., Bu çalışmada, küçük ölçekli aile tipi işletmelerde üretilen çiğ inek sütlerinde kimyasal ve mikrobiyolojik özelliklerinin tespiti yapılarak mevcut durumun ortaya konulması amaçlanmıştır. İncelenen 49 süt örneğinde yapılan mikrobiyolojik ve kimyasal analizler sonucunda, incelenen örneklerin kimyasal özelliklerin Türk Gıda Kodeksi Çiğ ve Isıl İşlem Görmüş İçme Sütleri Tebliği’ne uyduğunu, ancak çiğ süt örneklerin mikrobiyolojik kalitesinin yeterli düzeyde olmadığı, önemli bir kısmının da Türk Gıda Kodeksi kriterlerine uymadığını göstermektedir.

27. The Effects of Gonadotrophins on In Vitro Maturation of Cat Oocytes

The aim of this study was to determine the effects of FSH and LH hormones on in vitro development of cat oocytes. The ovaries from spayed cats were brought to the laboratory in +4 °C PBS solution two hours after being collected. The oocytes were recovered by slicing of ovaries and were separated into two groups: Group 1 (Control); TCM 199 + 5% FCS + 2.2 g/l NaHCO3 + 0.23 mM Na Pyruvate; and Group 2 (Treatment); TCM 199 + 5% FCS + 2.2 g/l NaHCO3 + 0.23mM Na Pyruvate + FSH (Sigma F-2293) (1 µg/ml) + LH (Sigma L-5269) (1 µg/ml). Oocytes were matured for 48 hours at 38.5 °C and 5% CO2 in a humidified atmosphere. At the end of this period oocytes were fixed, stained and evaluated for maturation criteria by phase-contrast microscope at x400 magnification. Data were analyzed statistically by Student's t - test. A total of 428 oocytes (Control: 203; Treatment: 225) were used. Eleven of the control group oocytes were at M I (5.4%), 2 (0.98%) were at anaphase I and 2 (0.98%) were at M II stage. These values for the treatment group were 25 (11.11%), 0 (0.00%) and 6 (2.66%), respectively. There was no statistical difference between the control and hormone added groups (p>0.05). 28. Freezing of Cat Semen in Straws with Different Glycerol Levels Containing Tris Extender

Cat semen was extended with 3% and 4% glycerol containing Tris extender and frozen in (0.25 ml) straws. Spermatological characteristics were examined in post-thaw semen samples. Five tom cats aged 2-3 were used. Semen was collected by electro-ejaculator under general anesthesia (xylazine-ketamine HCl combination). On a weekly basis a total of 8 ejaculates were collected from each tom cat. Spermatozoon concentration (x106/ml) was determined by the hemocytometric method. Morphologic examination was done by light microscope with Spermac® stain (x1000 magnification, immersion objective) and progressive motility was examined by hot-plate phase-contrast microscope (x200 magnification). Semen samples chosen as suitable for being frozen were extended 1:1 (semen/extender) by Tris extender containing 20% egg yolk without glycerol [Tris (hydroxymethyl-aminomethane) (2.40 g) - fructose (1.00 g) - citric acid (1.30 g) osmolarity: 285 mOsmol/kg] at +26 ºC. Extended semen was cooled to +5 ºC in 1 h and divided into 2 parts. Each volume was re-extended with glycerol containing Tris extender to obtain 3% and 4% final glycerol levels. The semen samples were left under equilibration for 30 min after glycerolization. Semen was filled into 0.25 ml straws and frozen in liquid nitrogen vapor. Frozen semen samples were stored in liquid nitrogen at -196 ºC. The application was repeated 8 times for the tom cats and 40 straws were frozen for each group (n=40). The frozen straws were thawed for 30 s at 37 ºC. During the study, examinations of motility (%) and morphology (%) (acrosome, other and total) were done at collection, dilution at +26 ºC, chilling at +5 ºC, equilibration and post-thaw points. The mean post-thaw motility, acrosome, other and total morphologic defect rates with 3% glycerol containing Tris extender were 53.00 ± 10.85%, 26.35 ± 8.55%, 14.43 ± 3.99% and 40.53 ± 11.00%, respectively. These values were 50.50 ± 13.90%, 25.45 ± 7.08%, 15.20 ± 4.36% and 40.40 ± 10.22%, respectively, with 4% glycerol containing Tris extender. The differences between the values of the 2 groups were not statistically significant (P > 0.05). At the end of the study, it was observed that 3% and 4% glycerol containing Tris extender could be used to preserve the post-thaw motility and morphology of cat semen frozen in straws.

29. The Effects of BSA and FCS on In Vitro Maturation of Bitch Oocytes

The effects of protein sources added to the medium for maturation of bitch oocytes were investigated. Ovaries from spayed bitches were transported to the laboratory in PBS at 38 ºC within an hour. Recovered oocytes were divided into three groups and left for 72 h maturation at 38.5 ºC under an atmosphere of 5% CO2 and high humidity. Group 1 (Control): TCM 199 medium, Group 2 (BSA): TCM 199 medium supplemented with 0.3% BSA, Group 3 (FCS): TCM 199 medium supplemented with 5% FCS. At the end of the maturation period, all oocytes were denuded from cumulus cells by pipetting and fixed in 1:3 acetic acid: ethanol solution for 48 h. The oocytes were then stained with 2% aceto-orcein and examined under a phased-contrast microscope at x400 magnification for maturation criteria. It was observed that 16.58% (34/205), 37.08% (79/213) and 22.81% (47/206) of the oocytes resumed meiosis in the control, BSA and FCS groups, respectively; a total number of 624 oocytes were used. In the BSA group, the rate of oocytes that resumed meiosis was significant (p < 0.05) compared to the FCS and control groups. This rate was also significant (p < 0.05) in the FCS group compared to the control group. According to these results, it can be suggested that addition of protein sources to the medium for in vitro maturation of bitch oocytes is necessarily important, and BSA gives better results than FCS does.

Peynirin olgunlaşmasında proteoliz ve lipoliz gibi çeşitli biyokimyasal değişiklikler meydana gelir. Lipoliz, özellikle Mavi ve Sert İtalyan peyniri gibi yoğun lipolizin gerçekleştiği peynir türlerinde olgunlaşma süresince gözlemlenen önemli bir biyokimyasal olaydır. Triaçilgliserolun yağ asiti ve gliserol arasındaki bağı parçalayan hidrolaz (lipaz ve esteraz) olarak ta bilinen lipolitik enzimlerin varlığıyla bu peynirlerde lipoliz gerçekleşir. Bu lipolitik enzimler başlıca küflerden ve diğer sekonder kültürlerden köken almaktadır. Peynirin olgunlaşması esnasında süt yağının triaçilgliserol yapısının lipolitik parçalanması, daha sonraki aşamalarda metil keton, tiyoester ve lakton gibi yüksek aromatik bileşiklere parçalanan serbest yağ asitlerinin (SYA) oluşumuyla sonuçlanır. Olgunlaşma esnasında salınan temel lezzet bileşenleri peynirin lezzetini doğrudan etkileyen serbest yağ asitleridir ve serbest yağ asitlerinin parçalanma ürünleri de farklı peynir türlerinde lezzeti doğrudan etkiler. Bu derlemede peynirde bulunan özellikle mikrobiyel kökenli lipoliz etkenleri; lipolize bağlı olarak oluşan serbest yağ asitleri, metilketon, ester, sekonder alkol gibi serbest yağ asitlerinin katabolizma ürünleri ve bu bileşenlerin lezzet üzerine etkisi irdelenmiştir.

Bu çalışma Doberman ve Labrador Retriever ırkı köpeklerin başlıca döl verimi özellikleri, yavrularda ölüm oranı ve beden ölçülerini saptamak amacıyla yapılmıştır. İlk kızgınlık gösterme yaşı ve ortalama gebelik süresi; Doberman ırkı köpekler için sırasıyla; 9.07 ay ve 63.00 gün, Labrador Retriever ırkı köpekler için sırasıyla; 8.7 ay ve 61.20 gün bulunmuştur. Doberman ırkı köpekler için bir batında doğan yavru sayısı 7.33 adet, iki kızgınlık arası geçen süre 6.00 ay, yavru ölümleri erkeklerde %57.65 ve dişilerde %51.23 saptanmıştır. Bu çalışmada kısraklarda son aşımı takiben 22. günde alınan kan örneklerinde hızlı progesteron tanı kiti kullanarak serum progesteron düzeyleri belirlendi ve erken gebelik tanısı olanakları araştırıldı. Materyal olarak, yaşları 5-15 arasında değişen 25 İngiliz kısrak kullanıldı. Kan örnekleri son aşımı takiben 22. günde toplandı. Örnekler 5000 devirde 20 dakika süreyle santrifüj edilerek serumlar elde edildi ve elde edilen serumlar iki eşit kısma ayrıldı. Birinci kısım serumlarda EIA yöntemine dayalı hızlı tanı kiti (Ovulation Test) kullanılarak serum progesteron düzeyleri saptandı. İkinci kısım serum radioimmunoassay yöntemiyle progesteron tayini yapılmak üzere -20 °C'de saklandı. Gebe kısraklar son aşımı takiben 22. günde B-mode real time ultrason kullanılarak belirlendi. Çalışmada her iki yöntemde de gebeliği belirleme oranı % 85.0 ve % 89.5, gebe olmayanları belirleme oranı ise her ikisi için % 100 olarak saptandı. Sonuç olarak, kısraklarda hızlı progesteron kiti 'Ovulation Test 'in gebelik tanısı için kullanılabileceği kanısına varıldı. The aim of this study was to investigate the pregnancy diagnosis rates by blood serum progesterone assays on day 22 after last mating in mares. Twenty-five thoroughbred mares aging 5-15 were used as materials. Blood samples were collected from mares on day 22 after last mating. Samples were centrifugated for 20 minutes at 5000 rpm and divided into two equal parts. Part one was used for determination of progesterone by enzymimmunoassay technique. The other part was kept at -20°C until the determination of progesterone by radioimmunoassay method. Pregnant mares were detected using B-mode real time ultrasound on day 22 after the last mating. The pregnancy and nonpregnancy diagnosis rates were 85.0%, 100 % by using EIA method and 89.5 %, 100 % by using RIA, for pregnant and non pregnant mares. As a conclusion, the rapid progesterone test Ovulation Test can be used to detect pregnancy in mares.

Bu çalışmada kısraklarda östrüsün; rektal palpasyon, ultrasonogra.fi ve hızlı kan-progesteron testi yardımıyla karşılaştırmalı olarak tespiti amaçlandı. Materyal olarak yaşları 5-15 arasında değişen saf ve yarım kan 20 İngiliz kısrak kullanıldı. Östrüs tespiti, deneme aygırı muayenesinden sonra rektal palpasyon ve ultrasonografi yardımıyla gerçekleştirildi. Follikül çapları östrüs boyunca rektal palpasyon ve ultrason kullanılarak östrüsün birinci gününden ovulasyon tespitine kadar gün aşırı ölçüldü. Follikül çapı ölçümlerini takiben kısraklardan gün aşırı olmak koşuluyla östrüsün birinci gününden ovulasyona kadar kan örnekleri alındı. Alınan örnekler 5000 devirde 20 dakika süreyle santrifüj edilerek serumlar elde edildi. Elde edilen serum iki eşit kısma ayrıldı. Birinci kısım serumda EIA (hızlı test), ikinci kısım serumda ise RIA yöntemine göre progesteron tayini yapıldı. Rektal palpasyon ve ııltrasonografık ölçümler arasındaki korelasyon (r0.92) önemli bulundu (p0.01). Serum progesteron düzeyleri EIA ile RIA yöntemi arasında % 85 oranında doğrulukta uyum gösterdi. Sonuç olarak, rektal palpasyon ve ultrasonografik muayeneler gibi hızlı kan progesteron testleriyle de östrüs tanısı yapılabileceği kanısına varıldı., In this study, oestrus detection was compared using a rapid progesterone test, rectal palpation and ultrasonography. Total of 20 thoroughbred mares aging between 5-15 were used as materials. The oestrus cycle of each mare was followed through teasing, palpation per rectum and transrectal ultrasonography. Follicle diameters were measured every other day by rectal palpation and ultrasonography from 1"' day of oestrus to ovulation. After rectal palpation and ultrasonography, 10 ml blood samples were collected every other day from the 1st day of oestrus to ovıdation. Bu çalışma Alman Kısa Tüylü Pointer ırkı köpeklerin başlıca döl verimi özellikleri, yavrularda ölüm oranı ve beden ölçülerini saptamak amacıyla yapıldı. Bu araştırmada ilk östrüs gösterme ve ilk çiftleştirme yaşı ortalama, sırasıyla; 10.99 ve 19.48 ay saptandı. Ortalama gebelik süresi 62.22 gün, iki östrüs arası geçen süre 7. 72 ay bulundu. Yavru ölümleri erkekler için % 49.90, dişiler için% 65.98 hesaplandı. Erkek ve dişiler için ergin çağda ortalama vücut ağırlığı sırasıyla 22.95 ve 21.57 kg bulundu. Baş uzunluğu, kulak uzunluğu, iki kulak arası mesafe, cidago yüksekliği, göğüs genişliği, göğüs çevresi, bilek çevresi, vücut uzunluğu, sağrı yüksekliği ve sağrı genişliği erkekler için sırasıyla; 23.91, 17. 00, 10.94, 53.00, 16.95, 64.52, 12.95, 54.31, 52.11 ve 15.44 cm, dişiler için sırasıyla; 21.58, 14.13, 11.09, 51.51, 16.41, 60. 75, 11.66, 52.50, 47.54 ve 15.36 cm bulundu. Cidago yüksekliği 1 vücut uzunluğu oranı ve sağrı yüksekliği 1 cidago yüksekliği oranı erkekler için sırasıyla; 9. 68:10 ve 9.84:10, dişiler için sırasıyla; 9.83:10 ve 9.26:10 hesaplandı. Baş uzunluğu, cidago yüksekliği, göğüs çevresi, bi/ek çevresi, vücut uzunluğu, sağrı yüksekliği ve sağrı yüksekliğinin cidago yüksekliğine oranı bakımından erkek ve dişiler arası farklılıklar önemli bulundu. 64 9. ÖZET: Çalışmada canlı materyal olarak, saf ırk, damızlık, 2 yaşlı 5 adet (Alman Kurt, Buldog, Chow chow, Sibirya Husky, New Foundland) erkek köpek kullanıldı. Araştırma süresince, parmak maniplasyonları tekniği uygulanarak haftada iki kez olmak üzere toplam 50 ejakülat alındı. Hacim, motilite, yoğunluk, canlı spermatozoa, pH ve morfolojik muayenelerden oluşan spermatolojik özellikler saptandı. Alınan sperma 2 eşit hacime ayrılarak %1 glikoz, %10 yumurta sarılı yağsız sütlü sulandırıcı ve %1.78 sitrik asit, %1.25 fruktoz'lu Tris (%3.2) sulandırıcısı ile oda ısısında 1:1 oranında sulandırıldı. 2 saatte 5°C'ye soğutulan süt ve Tris sulandırıcıları ile sulandırılmış spermalar yine iki eşit hacime ayrıldılar. Finalde, Tris sulandırıcısı %4 - %7 gliserollü ve sütlü sulandırıcı %4 - %7 gliserollü olacak şekilde gliserolizasyon işlemi uygulanarak 4 grup oluşturuldu. Beş farklı ırk köpeğe ait spermaların motilite ve anormal spermatozoa oranları her aşamada değerlendirildi. Sperma oda ısısında sulandırıldıktan sonra ve 5°C'ye soğutulduktan sonra yapılan incelemelerde sulandırıcılar arasında önemli bir fark saptanmadı. Gliserolizasyon sonrası en yüksek motilite %79.80±6.14 ile %4 gliserollü Tris sulandırıcısında bulunurken, en düşük motilite %59.70±9.65 ile %7 gliserollü süt sulandırıcısında elde edildi (n=50). En yüksek akrozomal ve toplam morfolojik bozukluklar, %7 gliserollü süt ve Tris sulandırıcılarında saptandı. 2 saatlik ekilibrasyon sonrasında da belirtilen spermatolojik özellikler aynı doğrultuda elde edildi. 0.2 mi pelletlerde dondurma ve eritme sonrası ise en yüksek motilite %68.36±1 1.71 ile %4 gliserollü sütlü sulandırıcıda elde edildi (p

Cat semen was extended with 3% and 4% glycerol containing Tris extender and frozen in (0.25 ml) straws. Spermatological characteristics were examined in post-thaw semen samples. Five tom cats aged 2-3 were used. Semen was collected by electro-ejaculator under general anesthesia (xylazine-ketamine HCl combination). On a weekly basis a total of 8 ejaculates were collected from each tom cat. Spermatozoon concentration (x106 /ml) was determined by the hemocytometric method. Morphologic examination was done by light microscope with Spermac® stain (x1000 magnification, immersion objective) and progressive motility was examined by hot-plate phase-contrast microscope (x200 magnification). Semen samples chosen as suitable for being frozen were extended 1:1 (semen/extender) by Tris extender containing 20% egg yolk without glycerol [Tris (hydroxymethyl-aminomethane) (2.40 g) -- fructose (1.00 g) - citric acid (1.30 g) osmolarity: 285 mOsmol/kg] at +26 °C. Extended semen was cooled to +5 °C in 1 h and divided into 2 parts. Each volume was re-extended with glycerol containing Tris extender to obtain 3% and 4% final glycerol levels. The semen samples were left under equilibration for 30 min after glycerolization. Semen was filled into 0.25 ml straws and frozen in liquid nitrogen vapor. Frozen semen samples were stored in liquid nitrogen at -196 °C. The application was repeated 8 times for the tom cats and 40 straws were frozen for each group (n=40). The frozen straws were thawed for 30 s at 37 °C. During the study, examinations of motility (%) and morphology (%) (acrosome, other and total) were done at collection, dilution at +26 °C, chilling at +5 °C, equilibration and post- thaw points. The mean post-thaw motility, acrosome, other and total morphologic defect rates with 3% glycerol containing Tris extender were 53.00 ± 10.85%, 26.35 ± 8.55%, 14.43 ± 3.99% and 40.53 ± 11.00%, respectively. ... The types and rates of acrosome and other (head, mid-piece and tail) abnormal spermatozoon types were determined by the Spermac® staining technique. Semen from 5 stray tom cats under the same management conditions was used. Semen collection was performed under general anesthesia by electro-ejaculator once a week for 5 weeks. After ejaculation the semen was diluted by 100 µl of 0.9% NaCl solution and stained with Spermac® stain for morphological evaluation. The morphological criteria were acrosome, other (head, mid-piece and tail) and total morphological defect rates, and were 9.20 ± 2.55%, 9.20 ± 3.49% and 18.40 ± 4.79%, respectively. It was concluded at the end of the study that the Spermac® staining technique could provide a detailed observation of cat spermatozoa, especially the acrosomal region, and could be employed in the determination of acrosomal defects. Background/Aim: The aim of the study was to determine the effects of gestation type single or twin and parity primiparous or pluriparous , on uterus involution period and on parameters that are related with uterus involution such as; caruncular diameter CD , uterine horn diameter UHD , uterine endometrial wall tickness UEWT ultrasonographically, and presence of vaginal discharge in Saanen does. Material and Method: Totally 20 Saanen does were used for the study. Ten of them delivered twin offsprings twin group , the other 10 does delivered single offspring single group . The does were further categorized as either primiparous n=7 or pluriparous n=13 . Ultrasonographic examinations were performed at postpartum pp 1st, 2nd, 4th, 7th, 10th, 13th, 19th, 25th, 28th days, except in three does ultrasonography was continued till 29th day of pp in which their uterine involution completed at 29th day pp. During ultrasonographic examinations CD, UHD, UEWT parameters always were measured on the thickest and same uterine horn continuously also presence of vaginal discharge were recorded in these days. Two-way ANOVA and chi-square tests were used for statistical analysis of data. Results and Discussion: Gestation type and parity had no significant effect on CD, UHD and involution period except for CD on day 13, in Saanen does. However, uterine endometrial wall thickness on days 10, 13 and 19 were higher in twin delivered does than single ones P, Özbilgi/Amaç: Bu çalışmanın amacı gebelik tipinin tekiz veya ikiz ve doğum sayısının primiparous veya pluriparous Saanen keçilerinde uterus involusyonu süresi üzerine ve uterus involusyonu ile ilişkili parametreler; karunkular çap KÇ , uterus kornu çapı UKÇ , uterus endometriyal duvar kalınlığı UEDK üzerine etkilerinin ultrasonografik olarak belirlenmesidir. Materyal ve Metot: Toplam 20 Saanen keçisi çalışmada kullanıldı. Bunlardan ikiz doğuran 10 tanesi, ikiz grubunu ve tekiz doğuran diğer 10 tanesi de tekiz grubunu oluşturdu. Keçiler daha sonradan primiparous n=7 ve pluriparous n=13 olarak da ayrıca gruplandırıldı. Üç keçi dışındaki keçilerde ultrasonografik muayeneler doğum sonrası ds 1., 2., 4., 7., 10., 13., 19., 25., 28. günlerde uygulandı. Üç keçide ise ultrasonografi uterus involusyonunun tamamlandığı 29. güne kadar devam etti. Ultrasonografik muayeneler esnasında uterus kornusunun en kalın olanı üzerinde KÇ, UKÇ, UEDK ölçümleri yapıldı ve her zaman aynı kornuda ölçümlere devam edildi. Ölçüm günlerinde ayrıca vaginal akıntıda da kayıt edildi. Araştırma verisinin istatistik analizinde iki yönlü varyans analizi ve ki-kare yöntemleri uygulandı. Gebelik tipi ve doğum sayısının KÇ, UKÇ ve involusyon süreleri üzerine 13. gün KÇ hariç Saanen keçilerinde önemli etkileri saptanmadı. Canine brusellosis, as a contagious disease, still has an important place in dog breeding due to inadequate diagnosis and treatment. The reason behind the failure of prevention of this disease is that people do not give enough attention to it is importance in dog breeding. In this review, it is discussed how veterinarians can succeed in the diagnosis and control of Brucella canis in dogs with paying attention to a few aspects, Bulaşıcı bir hastalık olan canine brucellozis, teşhis ve tedavinin yetersiz kalması sebebiyle günümüzde halen köpek yetiştiriciliğinde önemini korumaktadır. Hastalığa gereken önemin verilmemesi hastalığın önüne geçilememesinde en önemli nedendir. Bu yazıda, klinisyen veteriner hekimlerin birkaç hususa dikkat etmeleriyle köpeklerde Brucella canis etkeninin teşhis ve kontrolünde nasıl başarıya ulaşabilecekleri konusuna değinilmiştir.

Bu araştırmada, Rus Mersin balığının (Acipenser gueldenstaedtii B&R., 1833) spermatolojik özellikleri ve spermanın kriyoprezervasyonunda kullanılan farklı sulandırıcıların çözdürme sonrası spermatozoa motilitesi ve motil kalma süresine etkisi incelenmiştir. İstanbul Üniversitesi Su ürünleri Fakültesi Sapanca İçsu Ürünleri Üretimi Araştırma ve Uygulama Birimin'de bulunan ve ortalama ağırlığı 7702±2300 g, ortalama boyu 116,9±9,9 cm olan 9+ ile 10+ yaşa sahip toplam 12 adet anaç erkek balığın kullanıldığı çalışma, Haziran 2010 ve 2011 olmak üzere iki üreme döneminde gerçekleştirilmiştir.Her iki üreme döneminde balıklara LHRH hormonu uygulanmış ve spermatolojik özellikler (sperma miktarı, sperma pH'sı, seminal plazma ozmolaritesi, spermatozoa yoğunluğu, spermatozoa motilitesi, spermatozoa motil kalma süresi ) incelenmiştir. Dört farklı sulandırıcı (Sulandırıcı I, Sulandırıcı II, Sulandırıcı III, Sulandırıcı IV) kullanılarak dondurulan spermada çözdürme sonrası motilite ve motil kalma süreleri incelenmiştir.Taze spermada ortalama miktar 140±84 ml, sperma pH'sı 7,8±0,23, seminal plazma ozmolaritesi 88±44 mOsmkg-1, spermatozoa yoğunluğu 3,03±109/ml, spermatozoa motilitesi %84±6 ve spermatozoa motil kalma süresi 304±135 saniye olarak saptanmıştır.Çözdürme sonrası en yüksek spermatozoa motilitesi ortalama %30±12 olarak Sulandırıcı III, en düşük ise %11±6,2 olarak sulandırıcı IV ile sulandırılan örneklerde belirlenmiştir. Taze spermanın motilite yüzdesi ile sulandırıcı II, III ve IV için çözdürme sonrası motilite yüzdeleri ile elde edilen sonuçlar arasında istatistiksel olarak anlamlı bir fark bulunmuştur (p0,05), sulandırıcı II ve IV ve Sulandırıcı III ve IV arasında ise istatistiksel olarak anlamlı bir fark olduğu tespit edilmiştir (p0,05) In this study, spermatological parameters of Russian sturgeon fresh sperm and effects of different extenders on post-thaw spermatozoa motility and motility duration were investigated. 12 adult male with initial mean weight of 7702±2300 g, length 116,9±9,9 and age 9+ - 10+ obtained from Istanbul University, Faculty of Fisheries, Sapanca Inland Fish Water Production, Research and Applied Station. The study was conducted at two different reproductive periods (June 2010 and 2011).LHRH-a hormone was used on the fish and fresh sperm parameters such as sperm volume, pH, ozmolarity of seminal plasma, sperm concentration, sperm motility and duration of motility were determined at each reproductive period.Post-thaw motility and duration of motility were investigated after cryopreserved fresh sperm with different extenders (Extender I, Extender II, Extender III, Extender IV).The spermatological parameters of fresh sperm were identified and expressed as sperm volume 140±84 ml, pH 7,8±0,23, sperm concentration 3,03±109/ml, sperm motility %84±6 and duration of motility 304±135 s.Highest post-thaw motility was achived using extender III (%30±12) while lowest was with extender IV (%11±6,2).There was significant difference between fresh sperm motility and post-thaw motility for extenders II, III and IV (p0,05) whereas there was significant difference between extendersII and IV, extendersIII and IV (p0,05). 120
