Microcystins (MCs) are hepatotoxic heptapeptides identified in cyanobacterial bloom-impacted waters and soils. However, their environmental fate in soils is poorly understood, preventing reliable site assessment. This study aims to clarify the variant-specific adsorption, desorption, and dissipation of MC-LR and MC-RR in agricultural soils. Results revealed that their adsorption isotherms followed the Freundlich model (R2 ≥ 0.96), exhibiting a higher nonlinear trend and lower adsorption capacity for MC-LR than for MC-RR. The soils had low desorption rates of 8.14-21.06% and 3.06-34.04%, respectively, following a 24 h desorption cycle. Pairwise comparison indicated that soil pH and clay played key roles in MC-LR adsorption and desorption, while organic matter and cation exchange capacity played key roles in those of MC-RR. MC-LR dissipation half-lives in soils were 27.18-42.52 days, compared with 35.19-43.87 days for MC-RR. Specifically, an appreciable decrease in MC concentration in sterile soils suggested the significant role of abiotic degradation. This study demonstrates that the minor structural changes in MCs might have major effects on their environmental fates in agricultural soil and indicates that the toxic effects of MCs should be of high concern due to high adsorption, low desorption, and slow dissipation.

Perfluorooctanesulfonate (PFOS) as an accumulative emerging persistent organic pollutant in crops poses severe threats to human health. Lettuce varieties that accumulate a lower amount of PFOS (low-accumulating crop variety, LACV) have been identified, but the regarding mechanisms remain unsolved. Here, rhizospheric activation, uptake, translocation, and compartmentalization of PFOS in LACV were investigated in comparison with those of high-accumulating crop variety (HACV) in terms of rhizospheric forms, transporters, and subcellular distributions of PFOS. The enhanced PFOS desorption from the rhizosphere soils by dissolved organic matter from root exudates was observed with weaker effect in LACV than in HACV. PFOS root uptake was controlled by a transporter-mediated passive process in which low activities of aquaporins and rapid-type anion channels were corrected with low expression levels of PIPs (PIP1-1 and PIP2-2) and ALMTs (ALMT10 and ALMT13) genes in LACV roots. Higher PFOS proportions in root cell walls and trophoplasts caused lower root-to-shoot transport in LACV. The ability to cope with PFOS toxicity to shoot cells was poorer in LACV relative to HACV since PFOS proportions were higher in chloroplasts but lower in vacuoles. Our findings provide novel insights into PFOS accumulation in lettuce and further understanding of multiprocess mechanisms of LACV.

An asymmetric [3 + 2] cycloaddition of vinyl ethylenecarbonates (VECs) and (E)-3-arylvinyl substituted benzo[d] isothiazole 1,1-dioxides has been developed using the Pd complex of a bidentate phosphoramidite (Me-BIPAM) as the catalyst, providing a wide variety of chiral multistereogenic vinyltetrahydrofurans in good yields with excellent diastereo- and enantioselectivities (up to >20:1 dr, 99% ee).

Enantioselective copper-catalyzed cascade inter- and intramolecular amidation was achieved between ethynyl benzoxazinanones and α-halohydroxamates in the presence of an indapybox ligand. The one-pot cascade transformation was triggered by the attack of hydroxamates to dipolar copper–allenylidene intermediates, followed by a nucleophilic annulation reaction. Thus, a series of exo-methylene 3-aminoindoline derivatives were obtained in good yields with high enantioselectivities under mild reaction conditions.

Two new compounds (1 and 2), belonging to C-20 oxygenated ent-kauranes-type diterpenoids, were identified from the aerial parts of Isodon serra. Their structures were elucidated by extensive analysis of HRESI-MS and NMR spectroscopic data. Both these two compounds possess a common 7,20-epoxy-ent-kauranes skeleton with a hydroxyl group rarely occurring at C-13. Compounds 1 and 2 were evaluated for their cytotoxic activity against Hela-60 and HepG2 as well as the antibacterial activity against Staphylococcus aureus, Bacillus cereus and Escherichia coli.

One new 3,24-dinor-2,4-seco-ursane triterpene, rosanortriterpene C (1), together with four known compounds including 24-norursane-type nortriterpenes (2-3), 24-noroleanane-type nortriterpene (4), ursane-type triterpene (5), was isolated from the fruits of Rosa laevigata var. leiocapus. The new structure was elucidated through comprehensive spectroscopic analysis, including one dimensional (1D) and 2D NMR data, as well as electrospray ionization high resolution (HR-ESI) MS and IR spectrometry. Compounds 1-5 showed moderate anti-inflammatory activities against the production of nitric oxide (NO) in RAW264.7 cells stimulated by lipopolysaccharide (LPS) with IC50 values of 10.35 ± 0.92, 14.28 ± 1.20, 5.04 ± 1.43, 29.29 ± 3.64, and 14.37 ± 0.59 µM, respectively.

Echinacea purpurea polysaccharide (EPP) was obtained by modern separation technology and sulfated EPP (sEPP) was prepared by sulfation modification. The immunological effects of EPP and sEPP were compared on chicken bone marrow-derived dendritic cells (chBM-DCs). The results showed that the surface marker expression of CD11c and CD80 was increased after chBM-DCs were cultured with three dosage of sEPP, especially in sEPPM group. Three dosage of sEPP, EPPL and LPS could significantly enhance the effects of chBM-DCs on the proliferation of allogenic mixed lymphocytes. Life-threatening electrolyte imbalance is not uncommon in preemies. Differential diagnosis is important for immediate treatment. The syndrome of pseudohypoaldosteronism (PHA) is characterized by increased aldosterone secretion associated with clinical signs of hypoaldosteronism reflecting mineralocorticoid resistance. There are type I, type II, and secondary type of PHA. Most secondary PHA reported in the pediatric population result from urinary infection and obstructive uropathy and extremely rarely from gastrointestinal fluid loss. Seven preemies accepted jejunostomy or ileostomy, and they suffered from high output stoma. Electrolyte imbalance with bodyweight loss or cardiac event was noted. We found a high level of aldosterone and renin and diagnosed them with secondary PHA due to excessive gastrointestinal losses. After stomal reversal, aldosterone and renin level became normalized, and electrolyte was corrected. This study reports the finding of secondary pseudohyperaldosteronism (hyponatremia, hyperkalemia, and metabolic acidosis) in a series of cases with intestinal resection and ostomy of different causes. Early stomal reversal was recommended.

Three new pterocarpans, named abrusprecatins A-C (1–3), along with three known ones, namely medicarpin (4), maackiain (5), and 4-hydroxy-3-methoxy-8,9-methylenedioxypterocarpan (6) were isolated from the aerial parts of Abrus precatorius. The structures of these compounds were established by extensive analysis of mass spectrometric data, 1 D and 2 D NMR spectroscopic data. In addition, the absolute configurations were determined by a combination of single crystal X-ray diffraction analysis and circular dichroism spectroscopy.

A phytochemical investigation on the leaves of Callistemon viminalis resulted in the isolation of two unusual compounds, callistemonols A (1) and B (2). Callistemonol A (1) possesses a novel skeleton of a furan ring fusing both an α,β-triketone and a phloroglucinol unit, while callistemonol B (2) is an acylphloroglucinol derivative featuring two methyl substituents on a five-membered ring and an isovaleryl side chain. Their structures were fully characterized on the basis of extensive spectroscopic analysis, including 1D and 2D NMR parameters, as well as the IR and HRESIMS data. Callistemonol A (1) represents an example of a natural dibenzofuran with two phenyl moieties, and a plausible biogenetic pathway to generate this novel dibenzofuran through a C-C bond-forming radical SAM enzyme is proposed. Moreover, antimicrobial assays, in conjunction with time-killing and biophysical studies, revealed that 1 and 2 exert potent bactericidal activities against a panel of methicillin-resistant pathogenic microbes.

Single extraction procedures (SEPs) have been extensively conducted to determine Cd bioavailability (Cd-Bav) in soils. However, whether SEPs can simultaneously predict Cd accumulation in crop grains and bioaccessibility (Cd-Bac) in soils remains unclear. To assess their suitability, the Cd-Bav in 20 contaminated soils (containing 0.27–56.59 mg/kg Cd) determined by four SEPs (including DTPA, EDTA, HOAc and HCl) was compared with Cd concentrations in crop grains (wheat and rice) and Cd-Bac in soils (based on SBET and PBET assays). The results indicated that both Cd-Bav (0–103.2%) and Cd-Bac (0–110.4%) in soils varied greatly with the methods used. The Cd-Bav obtained from chelators (DTPA and EDTA) was generally greater in low-Cd soils but lower in high-Cd soils as compared to those obtained from acid solutions (HOAc and HCl). Regression analysis revealed that bioavailable Cd concentrations in soils were linearly correlated with Cd concentrations in wheat grains (R2 = 0.88–0.91); however, no significant correlation was found for rice grains. The Cd-Bac in soils was significantly correlated with Cd-Bav obtained from HOAc (R2 = 0.55–0.59) or HCl (R2 = 0.60–0.68), but not with those obtained from chelators (DTPA and EDTA). Our data suggest that SEPs, particularly the HCl method, have great potential to simultaneously predict Cd accumulation in wheat grains and Cd-Bac in contaminated soils.

Negative 99mTc-pertechnetate uptake of the thyroid bed indicates the absence or a small volume of remnant thyroid tissue (RTT) after total thyroidectomy (TT). The aim of this study is to evaluate the predictive value of negative 99mTc-pertechnetate scintigraphy for excellent response (ER) to radioactive iodine therapy (RIT) in low- to intermediate-risk differentiated thyroid cancer (DTC) patients. One-hundred and eighty-nine low- to intermediate-risk DTC patients who underwent TT, RIT with a single dose of 30 mCi and suppressive therapy with thyroid-stimulating hormone (TSH) from July 2015 to February 2016 in our hospital were retrospectively evaluated. 99mTc-pertechnetate thyroid scintigraphy was performed just before RIT and images were reported dichotomously as negative or positive. The response of patients was assessed for 23.2 ± 3.8 months after RIT and dichotomized as excellent response (ER) or non-excellent response (NER). 99mTc-pertechnetate uptake, age at diagnosis, gender, multifocality, T stage, N stage, preablative stimulated thyroglobulin (ps-Tg), and TSH were explored as potential predictors for ER. 80.68% (71/88) of patients with negative 99mTc-pertechnetate uptake achieved ER. When patients were evaluated according to different ps-Tg levels, we found that 94.83% (55/58) of patients with ps-Tg < 1 ng/ml and negative 99mTc-pertechnetate uptake achieved ER. Multivariate Cox regression analysis revealed that ps-Tg (P = 0.0001) and 99mTc-pertechnetate uptake (P = 0.0473) were independent predictors for ER. In addition to ps-Tg, negative 99mTc-pertechnetate uptake is also a significant independent predictor for an excellent response in low- to intermediate-risk patients. It may be possible to omit RIT in patients with ps-Tg < 1 ng/ml and concurrent negative 99mTc-pertechnetate uptake.

High speed development of the mining industry has lead to serious heavy metal pollution problems around mining areas in China, such as contaminating soil, and threatening the health of ecosystems and local residents. Plants long-term grown in mining areas largely show high adaptive capacity to the environment and facilitate the reclamation of degraded lands. In this study, we tested whether and how nitrogen form affect the adaptive capacity to soil contamination between legume (Robinia pseudoacacia) and nonlegume (Rhus chinensis) plants, typical phytoremediation species for soil environments polluted by heavy metals in China. All growth parameters and leaf catalase (CAT) and peroxidase (POD) of plant species were significantly greater in control than in contamination soil, in legume than in nonlegume, and in 50% NO3− (nitrate)+ 50% NH4+ (ammonium) and 100% NH4+ than in without and 100% NO3− treatments. The negative effect of soil contamination on all growth parameters was significantly stronger in nonlegume than in legume plant. Legume plant produced greater total mass, root mass and leaf mass in 50% NO3− + 50% NH4+ and 100% NH4+. Furthermore, the positive effect was not influenced by soil contamination. The greater physiological response to soil contamination in nonlegume plant and mixed addition of NO3− and NH4+ greatly relieved heavy metal stress on plants, especially in nonlegume plant. Mixed addition of NO3− and NH4+ greatly increased transfer factor for Pb and Zn and the positive effect significantly greater in nonlegume than in legume plant. Mixed addition of NO3− and NH4+ greatly increased the performance and adaptation of legume in contamination environment. To some extent, multiple N form addition in soil contamination environment may facilitate high plant adaption in phytoremediation, and using legume plants with mixed addition of NO3− and NH4+ is largely an important restoration mode for degraded mining areas.

Sixteen known lignans were isolated from the 95% alcohol extract of the whole plant of Serissa japonica (Thunb.) Thunb., including nine furofurans (1–9), three tetrahydrofurans (10–12) and four arylnaphthalenes (13–16). In the present report, compounds (+)-epipinoresinol (1), (+)-1-hydroxy-6-epipinoresinol 4,4″-di-O-methyl ether (3), (−)-pinoresinol (4), (+)-8-hydroxypinoresinol (6), pseuderesinol (7), (+)-1-hydroxysyringaresinol (8), (−)-(7′S,8S,8′R)-4,4′-dihydroxy-3,3′,5,5′-tetramethoxy-7′,9-epoxylignan-9′-ol-7-one (10), wikstrone (11), 7'-(+)-oxomatairesinol (12), (+)-cycloolivil (13), (+)-isolariciresinol (14), 5-methoxy-(+)-isolariciresinol (15) and cyclolignans (16) were reported from the Serissa genus for the first time, and compounds (+)-lirioresinol A (2) and (−)-lirioresinol B (5) were firstly isolated from the plant. Their structures were elucidated on the basis of extensive spectroscopic and chemical analyses. Moreover, the chemotaxonomic significance of the isolated compounds is discussed.

The growing incidence of microcystins (MCs) in the environment has become an issue of global concern for the high ecological and human health risks. Herein, a comparative adsorption of three MCs (MC-LR, MC-YR and MC-RR) by spent mushroom substrate (SMS)-derived biochars from contrasting pyrolytic conditions (temperature: 600/300 °C; and gas steam: CO2/N2) was surveyed to better understand the mechanisms and factors affecting the adsorption performance. For biochar preparation, 600 °C and CO2 led to greater levels of aromaticity, ash, SBET, and porosity, while 300 °C and N2 created more surface functional groups. The adsorption of MCs by biochars was a pH-dependent and endothermic physisorption process, following the pseudo-second-order kinetics (R2 = 0.99) and linear isotherm model (R2 > 0.88). The distribution coefficients Kd (0.98–19.2 L/kg) varied greatly among MCs (MC-YR > MC-RR > MC-LR) and biochars (BC600 > BN600 > BC300 > BN300), which depends on the combined effects of hydrophobicity, electrostatic attraction, H-bonding, cation bridging, and the amounts of adsorption sites on biochars. Higher ash, SBET, and total pore volume of BC600 facilitated the adsorption capacity for MCs relative to other biochars. Furthermore, the co-adsorption efficacy for MCs (Kd = 1.09–8.86 L/kg) was far below those for the single adsorption, indicating strong conflicts among competing MCs. This study sheds light on the roles of pyrolytic temperature and gas steam in biochar properties, and elucidates the mechanisms and factors affecting the adsorption performance of different MCs, which lays a foundation for MCs removal from water.

A novel PAE-hydrolyzing esterase (named Hyd) gene was screened from the genomic library of Rhodococcus sp. 2G and was successfully expressed in heterologous E. coli, which was defined as a new family of esterolytic enzymes. The purified Hyd could efficiently degrade various PAEs, displaying high activity and stability with a broad range of pH (4–10) and temperature (20–60 °C). Interaction mechanism of Hyd with dibutyl phthalate (DBP) was investigated by integrated multi-spectroscopic and docking simulation methods. Fluorescence and UV–vis spectra revealed that DBP could quench the fluorescence of Hyd through a static quenching mechanism. The results from synchronous fluorescence and CD spectra confirmed that the DBP binding to Hyd triggered conformational and micro-environmental changes of Hyd, which were characterized by increased stretching extent and random coil, and decreased α-helix and β-sheet. Molecular docking study showed that DBP could be bound to the cavity of Hyd with hydrogen bonding and hydrophobic interaction. A novel and distinctive catalytic mechanism was proposed: two key residues Thr190 and Ser191 might catalyze the hydrolysis of DBP, instead of the conserved catalytic triad (Ser-His-Asp) reported elsewhere, which was confirmed by site-directed mutagenesis.

Microbial fuel cell (MFC), a promising bio-electrochemical reactor could decompose organics in wastewater by redox processes of electro-active microorganism in anode and produce bio-energy, and the total MFC performance could mainly rely on electrochemical performance anode. Here, biomass carbon derived from municipal sludge was employed as low-cost and high-performance bio-anode for enhancing bioelectricity generation and wastewater treatment in MFC simultaneously. The electrochemical tests demonstrated that the large electrochemical active surface area, strong conductivity, and good biocompatibility in sludge carbon (SC) electrode resulted in higher power density (615.2 mW m−2) and lower power loss (5.4%) than those of none carbon (NC) electrode in long term operation. After 30-cycle of continuous running, the low loss of chemical oxygen demand (COD) removal was achieved up to 5.2%, which was smaller than that of NC electrode (14.1%), indicating that the MFC with SC anode could effectively treat wastewater and keep stable redox processes in anode electrode. After the formation of biofilm, the charge transfer resistance of SC electrode (16.38 Ω) was 72.4% lower than that of NC electrode (59.35 Ω). High-throughput analysis of biofilm exhibit Proteobacteria was the dominant electro-active bacteria, and the modification of SC could slightly change the bacterial community. Therefore, resource utilization of natural wastes provided the novel concept of anode catalyst fabrication for MFC in enhancing electron transfer, power output and wastewater decomposition.

Rosanortriterpenes A–B (RTA and RTB), two nortriterpenoids, are characteristic constituents in the fruits of Rosa laevigata var. leiocapus. However, pharmacological studies on these compounds are still scarce. In the present study, we aim to investigate the anti-inflammatory mechanisms associated with the effects of RTA–B in RAW264.7 macrophages and LO2 cells by detecting cell viabilities, nitric oxide (NO) production, tumour necrosis factor-alpha (TNF-α), and interleukin-6 (IL-6) production. Simultaneously, the anti-inflammatory action mechanisms of these two compounds were illustrated through western blot assay. Besides, the antihepatic fibrosis activities of these compounds have also been explored. The results demonstrated that RTA and RTB inhibited the production of NO, TNF-α, and IL-6 and suppressed liver fibrosis. RTA and RTB treatment also greatly inhibited the activation of NF-kappaB (NF-κB) pathway. Our study confirmed the promising anti-inflammatory and anti-liver fibrosis actions of RTA–B, suggesting that they might be developed as alternative and promising drugs for the treatment of hepatic inflammatory and fibrotic diseases.

Frequent cyanobacterial blooms in the eutrophic waters produce a variety of toxins such as the monocyclic heptapeptide microcystins, greatly harming aquatic ecosystems and human health. However, little information of microcystins in agricultural fields is known. This field study of three common microcystin variants (MC-LR, MC-RR, and MC-YR) in vegetables (n = 161), soils (n = 161) and irrigation water samples (n = 23) collected from southern China regions affected by cyanobacteria blooms, shows their prevalence with total concentrations up to 514 μg/L water, 187 μg/kg soil (dry weight) and 382 μg/kg vegetable (fresh weight). Myrotheium verrucaria strain DJTU-sh7 had the highest laccase activity of 216.2 U/ml, which was increased to above 300 U/ml after optimization. DJTU-sh7 had the best decolorizing effect for three classes of reactive dyes. The DJTU-sh7-containing fungal consortium displayed the robust decolorizing ability. Both color removal efficiency and chemical oxygen demand were increased in the consortium mediated biotransformation. Transcriptome changes of M. verrucaria elicited by azo dye and phenolic were quantified by the high throughput transcriptome sequencing, and the activities of the selected oxidases and reductases were determined. The possible involvement of oxidases and reductases, especially laccase, aryl alcohol oxidase, and ferric reductase in the biotransformation of dye and phenolic compounds was revealed at both transcriptomic and phenotypic levels. Revealing the transcriptomic mechanisms of fungi in dealing with organic pollutants facilitates the fine-tuned manipulation of strains in developing novel bioremediation and biodegradation strategies. Over the years, plant-derived substances have been developed as effective substitutes for synthetic insecticides because of their safety and environmental friendliness. In this study, we investigated the insecticidal properties of four polyacetylenes isolated from Bidens pilosa, a traditional edible herb, against Plutella xylostella (L.), Spodoptera litura, Mythimna separata (Walker), Spodoptera exigua (Hubner), Helicoverpa armigera and Ostrinia furnacalis (Guene’e). The polyacetylenes 1-4 showed potent insecticidal activities with LD50 values ranging from 0.133 to 4.084 μg/larva in the contact toxicity assay and with LC50 values ranging from 0.070 to 2.561 mg/mL in the stomach toxicity assay. Among these four compounds, compound 1 exhibited the highest insecticidal activity, comparable with the activity of commercial pesticide pyrethrins. The results of in vitro glutathione S-transferases (GSTs) and acetylcholinesterase (AChE) assays indicated these polyacetylenes exerted inhibitory effects on GSTs or (and) AChE in P. xylostella. Meanwhile, their anti-inflammatory properties were investigated using the q-PCR assay. These compounds proved to be non-cytotoxic against human keratinocytes (HaCaT) in MTT assay. These findings suggested that compounds 1-4 have the potential to be developed as skin-safe insecticidal agents.

The 7918 Fe deposit in the North Altun orogenic belt, NW China, is composed of strata-bound orebodies between Late Cambrian basalts and marble, associated with intensive skarn-type alteration. Basalts close to the orebodies are altered, with gradational contacts to skarns. Variations of major and trace elements and δ57Fe values of low-Ti and high-Ti basalts reflect the role of both magmatic evolution and hydrothermal alteration. Negative correlations between δ57Fe and TFeO/TiO2 and between Na2O and TFeO/TiO2 of high-Ti basalts are explained by lighter Fe isotopes leached from basalts during water–rock reaction. Hence, we considered basalts as the main source of Fe. Magnetite precipitation was related to a retrograde skarn stage. Early-precipitated magnetite has lower incompatible trace elements (Lu, Zr, Hf, Nb, Ta, and Yb), higher compatible trace elements (Co and Ni), and lower δ57Fe values than late-precipitated magnetite. Hydrothermal fluids equilibrated with early-precipitated magnetite (−0.11‰ to 0.16‰) and late-precipitated magnetite (0.22‰ to 0.27‰) have δ57Fe values of −0.38‰ to −0.11‰ and −0.05‰ to 0.00‰, respectively. The isotopically heavier late-phase fluids indicate an increase of oxygen fugacity, in which Fe2+ was mostly oxidized to Fe3+. This is also consistent with reduced V concentrations from early-precipitated to late-precipitated magnetite. Our new data and previous geochronological results suggested that the 7918 Fe deposit records a continuous process of volcanic eruption, hydrothermal alteration, and Fe mineralization, which was related to a deep “magma chamber” formed at a subduction-related setting.

A better understanding of microcystin elimination in soil will boost the development of effective strategies to mitigate associated contamination. This study addressed the role and mechanism by which earthworm Eisenia fetida take part in the elimination of microcystin-LR (MC-LR) in soil. The degradation kinetics of MC-LR exhibited good fits with the availability-adjusted first-order kinetic model (R2 > 0.97), where >71% of the toxin was rapidly eliminated within the initial eight days (t1/2 = 2.34–3.07 days). E. fetida significantly accelerated MC-LR elimination, with a less t1/2 of 1.09–1.12 days; however, E. fetida did not further improve the final elimination efficiencies on Day 29 (82.59–93.57% with E. fetida vs. 82.96–92.34% without E. fetida). In soil, E. fetida accelerated MC-LR elimination mainly via biodegradation arising from elevated total organic carbon, enhanced enzyme activities (urease and sucrase) and ameliorations of bacterial bioactivity, diversity and abundance. Indigenous bacteria of Methylophilus and Methylovorus, as well as earthworm-derived Flavobacterium, were considered as potential bacterial degraders for MC-LR. These findings elaborate on the variations in soil properties and soil bacterial community structure and diversity that occurs during MC-LR elimination and lay a foundation for controlling MC-LR contamination in the soil environment.

Phthalate acid esters (PAEs) are environmentally ubiquitous and have aroused a worldwide concern due to their threats to environment and human health. Di-n-butyl phthalate (DBP) is one of the most frequently observed PAEs in the environment. In this study, a novel bacterium identified as Pseudomonas sp. YJB6 that isolated from PAEs-contaminated soil was determined to have strong DBP-degrading activity. A complete degradation of DBP in 200 mg/L was achieved within 3 days when YJB6 was cultivated at 31.4 °C with an initial inoculation size of 0.6 (OD600) in basic mineral salts liquid medium (MSM), pH 7.6. The degradation curves of DBP (50–2000 mg/L) fitted well the first-order kinetics model, with a half-life (t1/2) ranging from 0.86 to 1.88 d. The main degradation intermediates were identified as butyl-ethyl phthalate (BEP), mono-butyl phthalate (MBP), phthalic acid (PA) and benzoic acid (BA), indicating a new complex and complete biodegradation pathway presented by YJB6. DBP might be metabolized through de-esterification, β-oxidation, and hydrolysis, followed by entering the Krebs cycle of YJB6 as a final step. Strain YJB6 was successfully immobilized with sodium alginate (SA), polyvinyl alcohol (PVA), and SA-PVA. The immobilization significantly improved the stability and adaptability of the cells thus resulting in high volumetric DBP-degrading rates compared to that of the freely suspended cells. In addition, these immobilized cells can be reused for many cycles with well conserved in DBP-degrading activity. The ideal DBP degrading ability of the free and immobilized YJB6 cells suggests that strain YJB6, especially the SA-PVA+ YJB6 promises great potential to remove hazardous PAEs.

One new ent-kaurane diterpene derivative (1), along with four known diterpenes, was isolated from the stems of Eurya chinensis R.Br. The structure of the new compound was established by extensive analysis of mass spectrometric and 1D and 2D NMR spectroscopic data. Compound 1 showed moderate anti-inflammatory activities with IC50 value of 8.12 μM. This is the first example of diterpenoids with 4-hydroxy-4-(2-hydroxyethyl)-1-hydroxyl-cyclohexanoyl substituent.

Methicillin-resistant Staphylococcus aureus (MRSA) poses a serious threat to global public health, because it exhibits resistance to existing antibiotics and therefore high rates of morbidity and mortality. In this study, twenty-one natural product-based acylphloroglucinol congeners were synthesized, which possessed different side chains. Antibacterial screening against MRSA strains revealed that acyl moiety tailoring is a prerequisite for the antibacterial activity. Moreover, the lipophilicity, rather than the magnitude of the hydrophobic acyl tail dominates variability in activity potency. Compound 11j was identified as a promising lead for the generation of new anti-MRSA drug development. It was discovered by optimization of the side chain length in light of the potency, the breadth of the antibacterial spectrum, the rate of bactericidal action, as well as the membrane selectivity. Compound 11j exerted profound in vitro antibacterial activity against the MRSA strain (JCSC 2172), and its MIC was 3-4 orders of magnitude lower than that of vancomycin. A preliminary mode of action study of compound 11j at the biophysical and morphology levels disclosed that the mechanism underlying its anti-MRSA activity included membrane depolarization and, to a lesser extent, membrane disruption and cell lysis.

Respiration chambers share one analyzer working in parallel, and methane (CH4) concentrations have to be measured at certain intervals. The maximum and minimum values in the kinetics of CH4 emissions can be missed during the interval between measurements, which may influence the quantification of CH4 emissions. Chambers must be opened for morning feeding and cleaning, which causes a loss of CH4 data. Calculation methods are needed to estimate the lost CH4 emission data, which may influence the estimated amount of daily CH4 emissions. In this study, we measured the CH4 emissions of 10 growing Chinese Holstein dairy heifers in respiration chambers. Methane concentrations were measured every 0.5 min to obtain the 23-h kinetics of CH4 emissions, which were further selected at different intervals between measurements (i.e., 5, 30, 60, 120, 180, and 240 min) to evaluate the effects of interval on quantification of CH4 emissions. The missing 1-h kinetics of CH4 emissions before feeding were not measured, and 2 calculation methods were used to estimate the missing 1-h kinetics of CH4 emissions: mean value of measuring period (the mean method) and the nearest value of measurement just before chamber opening (the nearest method). The results showed that the rates of CH4 emission from 10 heifers varied from 4.56 to 11.42 g/h. The increment of intervals decreased maximum rate of CH4 emission and increased minimum rate of CH4 emission. Interval caused less than 5% of the difference in measuring CH4 emissions. Although the mean method had greater estimated daily CH4 emission than the nearest method, the difference was within 3%. The interval between measurements (≤3 h) and calculation method had little influence on enteric CH4 emission measurements.

The metal-organic framework MOF-5 was synthesized by self-assembling of Zn(NO3)2·7H2O and H2BDC using DMF as solvent by the direct precipitation method and loaded with Fe2+ by the equivalent loading method at room temperature to prepare Fe(II)/MOF-5 catalyst and the microstructure, phases, and pore size of which was characterized by IR, XRD, SEM, TEM, and BET. It was found that Fe(II)/MOF-5 had high specific surface and porosity like MOF-5 and uniform pore distribution, and the pore size is 1.2 nm. In order to study the catalytic activity and reaction conditions of Fe(II)/MOF-5, it was used to catalyze the hydroxylation reaction of phenol with hydrogen peroxide. The results showed that the dihydroxybenzene yield of 53.2% and the catechol selectivity of 98.6% were obtained at the Fe2+ content of 3 wt.%, the mass ratio of Fe(II)/MOF-5 to phenol of 0.053, the reaction temperature of 80°C, and the reaction time of 2 h.

A self-designed oxygen-kerosene ablation system was employed to research the ablation properties of silicone rubber-based insulation material under different oxygen-rich conditions, that is, 0%, 5.00%, 7.71%, 17.01%, and 18.50%. The morphology of posttest specimens was analyzed via a scanning electron microscope (SEM), and the ablation rates were calculated. Experimental results showed that when the particle concentration was a fixed value, the mass and the linear ablation rates increased first and then decreased with the rise of oxygen content; the maximum values were 0.572 g/s and 0.933 mm/s, respectively. Under high oxygen-rich conditions, the formation of silicone rubber restricted the further increase of the ablation rates, filling more gaps of the reaction layer with liquid silicone rubber. Meanwhile, the thickness of the liquid glass layer attached to the surface was increased, which acted as a buffer against the impacts and erosion of particles and retarded the consumption and recession of materials. Excess oxygen spread to the edge of the ablation pit and reacted. Finally, these led to the increase in diameter of the ablation pit and the decrease of two types of ablation rates.

Longistylin A (LLA) is an abundant stilbene isolated from the leaves of Cajanus cajan (L.) Millsp. However, the antibacterial effect of LLA is not yet understood. Therefore, in this study, a detailed investigation of the antibacterial effect of LLA, particularly against methicillin-resistant Staphylococcus aureus (MRSA), was conducted. In vitro, LLA exhibited strong antibacterial activity against MRSA with a minimum inhibitory concentration (MIC) of 1.56 µg/mL and displayed much more rapid bactericidal activity (3-log decrease in MRSA survival within 8 h) than vancomycin. A membrane-targeting experiment suggested that the antibacterial activity of LLA is associated with perturbation of the bacterial membrane potential and increased membrane permeability. Notably, LLA had relatively weak cytotoxicity to murine macrophages [50% cytotoxic concentration (CC50) = 8.61 ± 0.57 µg/mL]. In vivo, topical treatment of a skin injury with LLA improved wound healing and closure in an MRSA-infected wound healing mouse model. After 3 days treatment, LLA decreased MRSA bacterial counts in the wounded region, reduced the accumulation of immune cells at the injury site, and alleviated induction of the inflammatory cytokines tumour necrosis factor-alpha (180.74 ± 10.78 pg/mL vs. 606.57 ± 68.99 pg/mL) and interleukin-6 (87.25 ± 10.19 pg/mL vs. 280.58 ± 42.27 pg/mL) in serum.

Two new ent-kaurane diterpenes (1–2), together with five known analogs, were isolated from the stems of Eurya chinensis. Compound 3 exhibited noticeable anti-inflammatory activity as denoted by inhibiting LPS-induced nitric oxide (NO) production in RAW264.7 cells with an IC50 value of 7.82 μM. Compound 4 showed potent cytotoxic activity against human cancer cell lines NCI-H46, HepG2 and SW480 with IC50 values ranging from 7.45 to 8.54 μM.

Three new dimeric kavalactones, designated as diyangonins A–C (1–3), along with two known analogs were isolated from the roots of Piper methysticum. Their structures were elucidated by means of extensive analysis of their 1D, 2D NMR, and mass spectroscopic data. All these dimers possess a skeleton featuring a cyclobutane ring connecting two kavalactone units in head-to-tail or head-to-head mode. Compounds 1–5 were evaluated for their cytotoxic activities against human tumor cell lines.

A novel kind of AgNPs catalyst was synthesized by in-situ reduction method using poly(2-acrylamido-2-methyl propyl sulfonic acid-co-2-hydroxyethyl methclate) [short as P(AMPS-co-HEMA)] hydrogel as matrices and AgNO3as a metal precursor. The structure of the composite hydrogel was characterized by Scan Electrical Microscope (SEM) and Fourier Transform Infrared Spectroscopy (FTIR). The results showed that AgNPs were binded with end group and most of the particles were isolated and uniformly distributed on the P(AMPS-co-HEMA). The catalytic properties in the reduction reaction of 4-nitrophenol(shorted as 4-NP) to 4-aminophenol(shorted as 4-AP) were studied in detail, and the result showed that conversion rate and conversion efficiency attained 97.56% and 0.9671 mmol/g·min when the amount of AgNPs was 9mg, [NaBH4] was 2.0×10−1mol/L, [4-NP] was 2.0×10−2mol/L and the total volume of solution was 50 mL, respectively.

Analysis method using orthogonal experimental design factor of four levels of three experiments, electroforming solution components were verified. On the current efficiency of the sample was calculated, its micro-hardness, tensile strength and stress were measured; and its XRD tests, analysis of the micro-strain, grain size and position electrical castings dislocation density. Four factors shows that the optimum level of nickel sulfate 350g / L, nickel chloride, 15g / L, the current density 3A/dm2, THF was 0.5g / L, saccharin was 0.4g / L.

The compound 29-(4-methylpiperazine)-luepol (M22), a novel derivative of lupeol has shown anti-proliferative effects against the human non-small cell lung cancer A549 cell line. M22 showed significant anti-proliferative activity at 6.80 μM and increased accumulation of G1 cells and effectively suppressed expression of the G1 arrest-related genes cyclins D1 and E1, CDK2 and CDC25A. This was further confirmed by Western blotting demonstrating decreased cyclin D1 and CDC25A protein levels. Furthermore, M22 caused induction of apoptosis that downregulated the anti-apoptotic BCL-2 gene and increased expression of BAX, CASP3 and CASP9 as well as the APAF1 gene. The effect of caspase-induced apoptosis was confirmed by an increase in reactive oxygen species (ROS), loss of mitochondrial membrane potential (MMP). Taken together, our findings indicated that M22 possessed potent anti-proliferative and apoptotic activities.

Recently, the position of shale gas is more and more important in the world energy structure. The lithology of Xiamaling Formation in Beijing area mainly include siltstone, silty shale and shale. The organic carbon content is relatively high, averaging 2.28%. The kerogen type of organic matter is dominated by II1 and II2 types, and thermal evolution level is relatively high, averaging 2.05%. Intergranular micropore is well developed, and most of the micropore diameter is 1-7µm, so the reservoir condition is good. The content of the brittle mineral is relatively high, so it can be easily fractured in production. The content of the clay mineral is relatively high, so it has a good adsorption capacity. Through wild prospecting and geochemical analysis for samples, this paper systematically assesses the Xiamaling Formation shale in Beijing area, which can provide basic stata for further shale gas study in Beijing area.

The kinetic resolution of 2-substituted-2,3-dihydro-4-pyridones was realized via a Pd-catalyzed allylic substitution reaction using a commercially available (S)-P-Phos as a ligand, affording optically active dihydropyridones and C-allylated dihydropyridones in high yields and good enantioselectivities with the S-factor up to 43. With this protocol, a catalytic asymmetric total synthesis of indolizidine (-)-209I was realized for the first time.

Δ8-sphingolipid desaturase and Δ6-fatty acid desaturase share high protein sequence identity. Thus, it has been hypothesized that Δ6-fatty acid desaturase is derived from Δ8-sphingolipid desaturase; however, there is no direct proof. The substrate recognition regions of Δ6-fatty acid desaturase and Δ8-sphingolipid desaturase, which aid in understanding the evolution of these two enzymes, have not been reported. A blackcurrant Δ6-fatty acid desaturase and a Δ8-sphingolipid desaturase gene, RnD6C and RnD8A, respectively, share more than 80 % identity in their coding protein sequences. In this study, a set of fusion genes of RnD6C and RnD8A were constructed and expressed in yeast. The Δ6- and Δ8-desaturase activities of the fusion proteins were characterized. Our results indicated that (1) the exchange of the C-terminal 172 amino acid residues can lead to a significant decrease in both desaturase activities; (2) amino acid residues 114–174, 206–257, and 258–276 played important roles in Δ6-substrate recognition, and the last two regions were crucial for Δ8-substrate recognition; and (3) amino acid residues 114–276 of Δ6-fatty acid desaturase contained the substrate recognition site(s) responsible for discrimination between ceramide (a substrate of Δ8-sphingolipid desaturase) and acyl-PC (a substrate of Δ6-fatty acid desaturase). Substituting the amino acid residues 114-276 of RnD8A with those of RnD6C resulted in a gain of Δ6-desaturase activity in the fusion protein but a loss in Δ8-sphingolipid desaturase activity. In conclusion, several regions important for the substrate recognition of Δ8-sphingolipid desaturase and Δ6-fatty acid desaturase were identified, which provide clues in understanding the relationship between the structure and function in desaturases.

A semi-interpenetrating network hydrogel with pore structure (P-S-IPN) was prepared by solution polymerization with 4-acetyl acryloyl ethyl acetate (AAEA) and crylic acid (AA) as monomers, polyvinylpyrrolidone (PVP) as the second network and polyving akohol 2000(PEG2000) as a pore-foaming agent. Gold nanoparticle composite hydrogel (GNPs gel) was obtained through in-situ reduction reaction by using P-S-IPN as a template. P-S-IPN exhibits excellent mechanical strength, the maximum compressive strength is 14.0 Mpa. The influence of the dosage of PEG2000 on the swelling/deswelling properties of P-S-IPN and GNPs gel was investigated. The pore structures could shorten the time that P-S-IPN takes to reach swelling equilibrium from 170min to 75min, improve the ability of P-S-IPN to absorb AuCl4-. The swelling/deswelling properties of GNPs gel under an applied voltage showed when the concentration of NaCl solutions exceed 0.025mol/L, the hydrogel swells, when the concentration of NaCl solutions is below 0.025mol/L, it deswells.

In order to extract tea polyphenols (TPs) by water under room temperature, the mechanochemical methodology was used for leaching process, and then TPs was purified from leaching solution by ethanol and ammonium sulfate aqueous two-phase system. The optimum parameters of mechanochemical methodology leaching TPs were determined by response surface methodology (RSM), which optimum parameters were: Na2CO3content 25~27wt%, solid material particle size D9540~45μm, liquid/solid mass ratio55~60, leaching time 16~18min. The TPs leaching rate was 16.46%. When ammonium sulfate mass composition0.2, ethanol mass composition0.35 as the composite of the extraction system, TPs maximum extraction rate was 93.4%. The maximum yield of TPs was 15.25% on the average, with average purity was 96.54% ultimately. Under such technology, water is only solvent used in TPs leaching at room temperature, and aqueous two-phase system extraction system under mild conditions, avoid the use of toxic solvent, is a green technology for extracting TPs.

The purpose of this work was to elucidate the characteristics and the expression profiles of porcine Opn4 during developmental stages and different times of the day. The coding sequence of Opn4 gene was cloned. The mRNA expression levels of Opn4 gene in the retina of the swines during developmental stages and different times of the day were examined. The results showed that the swine ORF of Opn4 gene was 1 437 bp in length which encodes 478 amino acids residues with the molecular formula of C2398H3705N623O651S23. The expression level of Opn4 in the day was significantly higher than that in the night (P 0.05). In total, results showed that the Opn4 gene can regulate the biological rhythm of the swines. Furthermore, it play an important role in mediate the responsibility to the light in the postnatal swines.

Development of precious metal-free catalysts for oxygen reduction reaction (ORR) is of great significance for the practical application of fuel cells. In this article, we report the finding that the C–N composites containing Fe and/or Co (C–PANI, Fe/C–PANI, Co/C–PANI, and FeCo/C–PANI), produced by directly pyrolyzing the Fe- and/or Co-doped polyaniline (PANI) precursors, show efficient electroactivity for ORR both in acidic and in alkaline media. Among the prepared catalysts, Fe/C–PANI and Co/C–PANI present the most positive ORR onset potential that is 0.61 V (vs. SCE) in 0.5 mol L−1 H2SO4 solution or −0.1 V (vs. SCE) in 1 mol L−1 NaOH solution. In addition, the Fe/C–PANI catalyst shows the largest limiting-diffusion current density for ORR both in acidic and in alkaline media. A four-electron reaction of ORR on the Fe/C–PANI and Co/C–PANI catalysts is more dominant than a two-electron reaction. Fe/C–PANI catalyst also presents good electrocatalytic activity stability for ORR both in acidic and in alkaline media.

The three kinds of WC-12Co coatings were sprayed by High Velocity Oxygen/Air Fuel Spray (HVO/AF) in three different spray conditions, HVOF、HVO-AF and HVAF. The high velocity flame flow was produced by HVO/AF with the burning of kerosene and propellant which was mixed by oxygen and air at different ratio, it can be controlled from 1400°C to 2800°C.The microstructure was investigated by SEM, the phase composition was tested by XRD, and the microhardness and elastic modulus of coatings were investigated. The results indicate that the coatings with better adhesive strength and little porosity than conventional ones are dense and have a large value of elasticity and a high microhardness which is 0.4 times harder than the conventional coatings. The coatings are mainly composed of WC and Co, and there are also traces of W2C and Co6W6C. As temperature decreased, the decomposition of WC is being reduced.

The optical properties of the structures with silver nanoparticles embedded in the silica layer atop the silicon substrate are simulated by the finite-difference time-domain method. The effects of nanoparticle size, period, silica layer thickness, and the angle of incidence of the illuminated light on optical transmissions are studied. It is found that there is the red-shift for the maximum of the total light transmitting into the silicon substrate as the silica layer thickness increases. The electric field intensity distributions and the average power densities for the structure with largest optical transmission is studied, and the strong electric field intensities are found in the silica regions surrounding to the silver nanoparticles, which can help the light energy going into the silicon substrate. By controlling the structure parameters, the optical transmissions of the structures with the silica layer can have higher optical transmissions than the cases without the silica layer. The silica layer plays the role as the graded refractive index layer between the air and the silicon substrate, and the light power from the incident wave can transmit into the silicon substrate with less optical reflections for choosing a suitable silica layer thickness. A guideline to design the structures with high optical transmissions for the solar spectra is given. This study cannot only be useful for the solar cells applications, but also other antireflection applications.

Δ6-fatty acid desaturase is an important enzyme in the catalytic synthesis of polyunsaturated fatty acids. Using domain swapping and a site-directed mutagenesis strategy, we found that the region of the C-terminal 67 amino acid residues of Δ6-fatty acid desaturase RnD6C from blackcurrant was essential for its catalytic activity and that seven different residues between RnD6C and RnD8A in that region were involved in the desaturase activity. Compared with RnD6C, the activity of the following mutations, V394A, K395I, F411L, S436P, VK3945AI and IS4356VP, was significantly decreased, whereas the activity of I417T was significantly increased. The amino acids N, T and Y in the last four residues also play a certain role in the desaturase activity.
