Grape anthracnose caused by Elsinoë ampelina (Shear) is one of the most serious fungal diseases that lead to the quality reduction and yield losses of grape (Vitis vinifera ‘Red Globe’) berries. In the present study, metabolome and transcriptome analyses were conducted using grape berries in the field after infection with E. ampelina at 7, 10, and 13 days to identify the metabolic properties of berries. In total, 132 metabolites with significant differences and 6,877 differentially expressed genes were detected and shared by three comparisons. The analyses demonstrated that phenylpropanoid, flavonoid, stilbenoid, and nucleotide metabolisms were enriched in E. ampelina-infected grape berries but not amino acid metabolism. Phenolamide, terpene, and polyphenole contents also accumulated during E. ampelina infection. The results provided evidence of the enhancement of secondary metabolites such as resveratrol, α-viniferin, ε-viniferin, and lignins involved in plant defense. The results showed the plant defense-associated metabolic reprogramming caused by E. ampelina infection in grape berry and provided a global metabolic mechanism under E. ampelina stimulation. Vitis vinifera 87-1 plants infected by grapevine fleck virus (GFkV) and grapevine rupestris stem pitting-associated virus (GRSPaV) were used as the plant materials for virus elimination treatment. This study evaluated the effects of ribavirin at different concentrations (15 and 25 µg mL−1; R15 and R25, respectively), thermotherapy (37°C; T), and the combination of ribavirin and thermotherapy (R15+T and R25+T) on eliminating viruses from grapevine plants in vitro. Both R15 and R25 had phytotoxic effects and weakened plant growth. Thermotherapy positively affected the growth of grapevine plants. Plant height was significantly greater in T, R15+T, and R25+T than in CK, R15 and R25. The proportion of dead plants after T, R15+T, and R25+T was 51.4, 11.4, and 8.6%, respectively. The survival rates of regenerated plants after all treatments were >68.0%. Ribavirin concentration and treatment time were related to the regeneration of shoot tips and elimination efficiencies of the two viruses. The survival rates of plants after R15+T for 30, 40, and 50 days were 97.3, 90.7, and 74.4%, respectively. The elimination rates of GRSPaV from plants in the three time quantum were 55.6, 84.6, and 93.8%, respectively. The elimination rate of GFkV was 23.9% higher in R25 (35/44) than in R15 (25/45), and that of GRSPaV was 7.0% higher in R25 than in R15. The combination of thermotherapy and chemotherapy was found to have a positive effect on the eradication of GFkV and GRSPaV, and R25+T for 50 days was able to completely eliminate the two viruses from in vitro grapevines.

BACKGROUND: Powdery mildew affects grapevine growth and development and reduces grapevine fruit yield and quality. Plant U-box (PUB) E3 ubiquitin ligases play important roles in ubiquitin/proteasome-mediated protein degradation during plant development and in the plant defense response. OBJECTIVE: We cloned the VvPUB17 gene from Vitis vinifera and analyzed that VvPUB17 enhanced the resistance of grapevine to powdery mildew through the SA signal pathway. METHODS: Pathogen inoculation of Arabidopsis thaliana and grapevine plants was carried out by the tableting method. Gene expression was analyzed by quantitative real-time PCR. Sequence analysis and in vitro ubiquitination experiments show the structure and characteristics of VvPUB17. Exogenous salicylic acid, methyl jasmonate, ethylene and powdery mildew induced the expression of VvPUB17 in Arabidopsis leaves to verify the resistance of VvPUB17 to powdery mildew. RESULTS: Sequence analysis and in vitro ubiquitination experiments show that VvPUB17 contains U-box and Armadillo repeats (ARM repeat) and has E3 ubiquitin ligase activity dependent on the conserved U-box motif. Transgenic plants showed elevated levels of key genes related to the SA defense response pathway and high concentrations of salicylic acid. CONCLUSIONS: VvPUB17 functions as an E3 ubiquitin ligase that enhances the resistance of grapes to powdery mildew through the SA signal pathway.

Background The European grapevine moth, Lobesia botrana (Denis and Schiffermüller) (Lepidoptera: Tortricidae) is currently the most damaging pest in many viticultural regions across the Mediterranean basin and elsewhere. Its feeding activity also enhances the development of secondary infections by Botrytis cinerea - and other fungi - to wine grapes. The gram-positive bacterium Bacillus thuringiensis (Bt) has been reported to partially control larval populations of L. botrana, while it requires to be digested by the insect to cause infection. Entomopathogenic fungi (EPF) are possibly capable of acting synergistically with Bt to increase its efficacy against insect pests. Results The hypothesis of synergy or antagonism between Bt and EPF for the control of L. botrana was tested in two bioassays: A) Insects fed on Bt diet and subsequently some groups were sprayed by conidia of Beauveria bassiana or Paecilomyces fumosoroseus, and B) Grapes were sprayed by Bt, or B. bassiana, or combination of the two, and then untreated insects were placed to feed on the grapes. In both bioassays, combination treatments performed better than single treatments, indicating additive action or synergy. The Bt and B. bassiana combination treatment (Bt diet for 30 h and then sprayed with conidia of B. bassiana) resulted in 91% larval mortality while the single Bt and B. bassiana treatments caused 28% and 34% mortality respectively. Such results indicated synergism. Combination treatment on grapes also caused significantly higher mortality on L. botrana larvae, compared to single treatments. The median lethal time (LT50) was estimated as 8.43 days for the single Bt treatment, 7.87 days for the single B. bassiana treatment and 6.3 days for the combination Bt + B. bassiana treatment. Conclusions Absence of antagonism as well as additive action or synergy were indicated by the results. Larval populations of the pest can be effectively controlled by using microbial biocontrol agents. Further research is needed to investigate the biotic and abiotic factors that affect interactions between insect hosts and entomopathogenic organisms. However, the entomopathogens used in the present study showed remarkable action and may be included parallelly in control strategies against vineyard pests.

Early leaf removal around the cluster zone is a common technique applied in cool climate viticulture, to regulate yield components and improve fruit quality. Despite the increasing amount of information on early leaf removal and its impact on total soluble solids, anthocyanins, and polyphenols, less is known regarding aroma compounds. In order to verify the hypothesis that defoliation, applied before or after flowering, could impact the biosynthesis of thiol precursors, we performed a two year (2013 and 2014) experiment on Sauvignon blanc. We provided evidence that differential accumulation of thiol precursors in berries is affected by the timing of defoliation, and this impact was related to modifications in the biosynthetic pathway. Furthermore, the possible interaction between leaf removal treatment and seasonal weather conditions, and its effect on the biosynthesis of volatile precursors are discussed. Our results suggested that in Sauvignon blanc the relative proportion of 4-S-glutathionyl-4-methylpentan-2-one (G-4MSP) and 3-S-glutathionylhexan-1-ol (G-3SH) precursors can be affected by defoliation, and this could be related to the induction of two specific genes encoding glutathione-S-transferases (VvGST3 and VvGST5), while no significant effects on basic fruit chemical parameters, polyphenols, and methoxypyrazines were ascertained under our experimental conditions.

Red grape (Vitis vinifera L.) is easy to get perishable and wrinkles/wilt, brown in color, and soft fruit flesh. one of the benefits of edible coating is that it can protect products from microbiological damage. The ability of aloe vera as an antimicrobial can be a potential ingredient for making edible coatings. Besides, the use of aloe vera as an edible coating is also very easy and affordable (cheap). The purposes of this study are to determine the effect of glycerol addition and the best treatment of aloe vera edible coating on the quality of the red grape. The experiment used a completely randomized design (CRD) as an experimental design with 4 treatment factors, namely the concentration of glycerol (P1=0%, P2=3%, P3=5%, P4=7%). Each concentration was repeated three times. The parameters observed were physicochemical characteristics (weight reduction, pH, total titrated acid), hedonic quality test (color, aroma, texture). The results showed that the treatment of 3% glycerol was the best treatment since it could maintain the condition of the fruit well during storage, especially weight reduction, aromas, and textures. Parameters that highly affect the quality of red grapes are texture and weight loss. Keywords: Aloe vera, Edible coating, Glycerol, Red grape.

The metabolic impacts of endophytes on grape cells have been addressed in many studies. However, to date, no reports have investigated these phenomena in vine leaves, which also permit examination under well-controlled whereas more represent the situation of endophytes within grapevines. In the current work, we first established a method to test the symbiotic efficiency of fungal endophytes in vitro using aseptic grapevine (Vitis vinifera L.) leaves. Next, we compared the symbioses of varied fungal strains as well as physio-chemical changes triggered by these symbioses, in grapevine leaves from two different cultivars ‘Rose honey’ (RH) and ‘Cabernet sauvignon’ (CS). Our results demonstrated that all the tested fungal strains were able to achieve successful infection and symbiosis in leaves from both cultivars. The symbiotic efficiencies of the tested fungi in grapevine leaves varied considerably from one fungal strain to another, perhaps due to host-selectivity. Concerning the cultivars, most of the fungal strains that were tested appeared similarly detrimental and similar symbiotic rates between leaves of the two cultivars. Meanwhile, the symbioses of most fungal strains clearly promoted the detected physio-chemical traits compared to the control. Most of these fungal strains, such as RH7, RH12, RH32, and others, conferred greater physio-chemical effects in RH leaves than in CS leaves. Several fungal strains (RH34, RH49 and MDR36) exhibiting high symbiotic efficiency and lower host damage, while initiating greater physio-chemical responses in grape leaves of both cultivars, may potentially be developed for further applications. The results of this study demonstrate that, in addition to grape cells, grape leaves are also able to respond metabolically to endophytes, and also provide a more efficient approach to select candidate endophytes for grape quality management in viticulture.

The aim of this study was to characterize the changes in berry anatomy during the development of grape (Vitis vinifera L., and Vitis vinifera × Vitis labrusca) with different shapes. Paraffin sectioning was used to examine the structural parameters of the cells. The results revealed that, with the development of berries, the transverse and longitudinal diameters of the flesh cells gradually increased, revealing certain regularity. However, the transverse and longitudinal diameters of the epidermal and sub-epidermal cells were different between varieties, reflecting the specificity of the varieties. The growth of the transverse and longitudinal diameters of the berries was found to be completed in the early stage of development. A combination of correlation analysis and size analysis for each cell layer revealed that, due to the small cross-sectional area of the epidermal and sub-epidermal cells, the influence of these cells on the transverse and the longitudinal diameters of the berries would be small. In conclusion, the longitudinal and transverse diameters of the grape berries were mainly determined by the longitudinal and transverse diameters of the flesh cells. The different shapes of the grape berries could mainly be attributed to the different growth rates of the flesh cells in the longitudinal and transverse directions. These different rates of growth led to different lengths and widths of the berry.

BACKGROUND: HD-Zip genes encode several large and highly conserved protein families of transcription factors that play important roles in plant development and responses to environmental stress. To date, information about the involvement of HD-Zip in grape to drought response is limited. OBJECTIVE: The production of grapes is limited by a range of biotic and abiotic stresses, which cause significant losses in yield every year as well as a reduction in fruit quality. Identification and analysis of stress related genes in grapes are very important for cultivating more robust varieties with environmental stress resistance METHODS: We isolated a homeodomain-leucine zipper gene (HD-Zip), VvHDZ4, from grape, Vitis vinifera L. “Yatomi Rose”, and characterized VvHDZ4 based on analyses of its expression patterns, subcellular localization, transcription activity, and overexpression. RESULTS: We found that VvHDZ4 was highly expressed in roots and leaves, localized to nucleus, and activated the expression of reporter genes in yeast. The overexpression (OE) of VvHDZ4 led to enhanced drought stress tolerance in Solanum lycopersicum L. “Ailsa Craig” (tomato). The drought-tolerant phenotypes of the OE lines exhibited decreases in electrolyte leakage and rate of water loss, higher photosynthetic production and net photosynthesis rate, and showed up-regulation of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX), all of which are key antioxidant enzymes known to be activated during drought stress. Moreover, we observed the up-regulation of four additional drought stress indicator genes, SIDREB2A, SIAREB, SIRD29 and SIERD10, in the OE lines. VvHDZ4 improved drought tolerance in tomato, and the modes of action possibly consist of reducing membrane damage, increasing photosynthetic productivity, and modulating the expression of stress defense genes. CONCLUSIONS: This work increases our understanding of the important roles HD-Zip transcription factors in the responses of plants to the environment, especially abiotic stress.

In 2012, dormant canes of a proprietary wine grape (Vitis vinifera L.) accession were included in the collection of the University of California–Davis Foundation Plant Services. No virus-like symptoms were elicited when bud chips from propagated own-rooted canes of the accession were graft-inoculated onto a panel of biological indicators. However, chlorotic ringspot symptoms were observed on sap-inoculated Chenopodium amaranticolor Coste & A. Rein and C. quinoa Willd. plants, indicating the presence of a mechanically transmissible virus. Transmission electron microscopy of virus preparations from symptomatic C. quinoa revealed spherical, nonenveloped virions about 27 nm in diameter. Nepovirus-like haplotypes of sequence contigs were detected in both the source grape accession and symptomatic C. quinoa plants via high-throughput sequencing. A novel bipartite nepovirus-like genome was assembled from these contigs, and the termini of each RNA segment were verified by rapid amplification of complementary DNA ends assays. The RNA1 (7,186-nt) of the virus encodes a large polyprotein 1 of 231.1 kDa, and the RNA2 (4,460-nt) encodes a large polyprotein 2 of 148.9 kDa. Each of the polyadenylated RNA segments is flanked by 5′- (RNA1 = 156-nt; RNA2 = 170-nt) and 3′- (RNA1 = 834-nt; RNA2 = 261-nt) untranslated region sequences with >90% identities. Maximum likelihood phylogenetic analyses of the conserved Pro-Pol amino acid sequences revealed the clustering of the new virus within the genus Nepovirus of the family Secoviridae. Considering its biological and molecular characteristics, and based on current taxonomic criteria, we propose that the novel virus, named grapevine nepovirus A, be assigned to the genus Nepovirus.

Grapevine (Vitis vinifera L.) is a major fruit crop with commercial importance worldwide. It is highly susceptible to a large number of pathogens that cause diseases, resulting in loss of production. The timely estimation of symptom severity can become a major milestone in disease management. Due to the advancement in the field of computer vision, automatic estimation of symptom severity is made possible. This paper proposes a fuzzy logic-based method for the estimation of symptom severity in grapevine. The symptomatic area is segmented from the leaf images using a fuzzy membership function. Additionally, consideration of the lesion spots along with the symptomatic area is proposed to evaluate the symptom severity using the fuzzy inference system. The performances of the existing techniques are studied and compared and it is shown that the proposed automatic method for estimation of symptom severity yields the best results with an accuracy of 95.33%.

Background and aims: In Vitis vinifera L., the same genotype can express different phenotypic characteristics depending on the environmental conditions, e.g. soil deepness. Wood anatomy, specifically xylem vessel traits, provide information about the plant’s eco-physiological responses to the environment. Slight changes in vessel diameter and density may impact plant hydrosystem functionality, since large vessels are more efficient in the volume of transported water compared to narrower ones, although the latter are more effective in avoiding stress-induced embolism. The aim of this study was to analyze variations in the wood hydraulic structure of three grapevine cultivars, induced by soils with strong contrast in depth, texture and rock volume, providing evidence of their adaptative capacities. Methods: Anatomical and growth traits of each annual growth ring were measured in 8-year-old plants of Bonarda, Malbec and Tempranillo cultivars growing in contrasting depths of soils. Results: Bonarda exhibited no differences in wood productivity between soils with different depths, showing the ability to modulate the earlywood vessel lumen area. Malbec and Tempranillo did show differences in wood productivity between the two types of soils, with major changes in the trade-off between vessel density and lumen area in Tempranillo, while in Malbec there were few changes in the vessel traits. Conclusions: Xylem hydraulic characteristics of the grapevine stems varied in response to soil environment and cultivar. Larvae of order Lepidoptera have biting mouth parts and almost fed on leaves. The insects of Lepidopterous were divided into three groups depended on number of the host plants, the monophagous, the oligophagous and the polyphagous. Sense organs are responsible for the number of host plants and insects select for hosts to fed on. In this study, the head capsule of larvae was cut at the end of larval stage of the mulberry silkworms and the cotton leaf worms to examine by scanning electron microscope for the number and type of sense organs on head capsule of larvae of both insects. The total number of sense organs on the head capsule of the mulberry silkworm was 82 where they were more than that of the cotton leaf worm where they were only38 sense organs. In addition, When mouth parts of the mulberry silk worm were covered with nail polish then the larvae were offered five different species of plant leaves ,Abelmoschus esculentus (okra), Vitis vinifera (grapes), Ricinus communis (castor bean plant),Cucurbita moschata (Pumpkin) and Morus alba (mulberry) to fed on. The treated larvae couldn’t recognize mulberry leaves to feed on. The larvae fed on small part of grapes and okra leaves then stopped to continue feeding. Vomiting and diarrhea were happened to larvae then died.

Grapevine is one of the most cultivated fruit plant among economically relevant species in the world. It is vegetatively propagated and can be attacked by more than 80 viruses with possible detrimental effects on crop yield and wine quality. Preventive measures relying on extensive and robust diagnosis are fundamental to guarantee the use of virus-free grapevine plants and to manage its diseases. New phenotyping techniques for non-invasive identification of biochemical changes occurring during virus infection can be used for rapid diagnostic purposes. Here, we have investigated the potential of Raman spectroscopy (RS) to identify the presence of two different viruses, grapevine fan leaf virus (GFLV) and grapevine rupestris stem pitting-associated virus (GRSPaV) in Vitis vinifera cv. Chardonnay. We showed that RS can discriminate healthy plants from those infected by each of the two viruses, even in the absence of visible symptoms, with accuracy up to 100% and 80% for GFLV and GRSPaV, respectively. Chemometric analyses of the Raman spectra followed by chemical measurements showed that RS could probe a decrease in the carotenoid content in infected leaves, more profoundly altered by GFLV infection. Transcriptional analysis of genes involved in the carotenoid pathway confirmed that this biosynthetic process is altered during infection. These results indicate that RS is a cutting-edge alternative for a real-time dynamic monitoring of pathogens in grapevine plants and can be useful for studying the metabolic changes ensuing from plant stresses.

This article is licensed under a Creative Commons Attribution 4.0 International License., Warm and semi-arid climates are characterized by rainfall scarcity, resulting in the frequent use of low-quality water for irrigation. This work was undertaken to study the effects of water stress and saline irrigation on yield and grape composition of Monastrell grapevines grafted onto 1103P rootstock. The experiment was carried out during three consecutive seasons in a commercial vineyard located in Jumilla (SE Spain) with a loamy-sandy soil. Rainfed vines were compared with five watering regimes including a Control, irrigated with standard water, and four treatments that combined two different schedules for irrigation initiation (pre- and post-veraison) with saline water obtained by adding two types of salts (sulphates and chlorides). Vines from treatments with more severe water stress (i.e., rainfed) showed lower yields and vegetative growth. Moreover, the Rainfed treatment clearly modified grape composition when compared with the Control treatment by increasing berry phenolic content. The application of saline water slightly affected vine performance and grape composition regardless of the type of salts added to the irrigation water. Indeed, the watering regime had a greater effect on yield, vegetative growth and grape composition than the use of different saline waters. Our results suggest that, in the mid-term (3 years), and with a vineyard soil with good drainage, the use of saline waters is not detrimental to vine performance, but does not improve grape composition. Further research is required to assess the long-term effects of saline water application, particularly in view of the important accumulation of chlorides and sodium in leaf tissues observed in vines watered with salty water at the last season of this experiment., Open Access funding provided thanks to the CRUE-CSIC agreement with Springer Nature. Grapes and wine are widely consumed in the world, yet their mineral content can be influenced by many factors such as the mineral composition of soils, viticulture practices and environmental conditions. In this context, considering the importance of Zn in the human physiology, the enrichment of Moscatel and Castelão grapes (white and red variety, respectively) with this nutrient prompted this study; further assessment of tissue deposition and some implications for wine production. Using two foliar fertilizers (ZnO or ZnSO4, at 150, 450 and 900 g ha−1), decreases in net photosynthesis and stomatal conductance occurred in both varieties, suggesting that the physiological threshold of Zn toxicity was reached without visible symptoms. Following foliar spraying with both fertilizers, the content of Zn in leaves of the Castelão and Moscatel varieties showed higher values in all treatments relative to the control. Moreover, in grapes this tendency occurred only in Castelão. Concerning Cu, Fe, Ca, K, S and P, some significant differences also happened in leaves and grapes among treatments. At harvest, the indexes of Zn enrichment in grapes increased between 2.14- and 8.38-fold and between 1.02- and 1.44-fold in Castelão and Moscatel varieties, respectively. Zinc in the dried skin of Castelão only increased with ZnO and ZnSO4 sprayed at 900 g ha−1 (ca. 2.71- and 1.5-fold relative to the control, respectively), but in Moscatel a clear accumulation trend could not be found. The dry weight of grapes ranged (in %) between 16 and 23 (but did not vary significantly among treatments of each variety or in each treatment between varieties), and total soluble solids (e.g., mainly soluble sugars and proteins) and color parameters showed some significant variations. Through winemaking, the contents of Zn increased in both varieties (1.34- and 3.57-fold, in Castelão and Moscatel, respectively) and in all treatments, although non-significantly in Castelão. It is concluded that, to increase the contents of Zn in grapes without reaching the threshold of toxicity, ZnO or ZnSO4 can be used for foliar spraying of Castelão and Moscatel varieties until 900 g ha−1 and that winemaking augments the level of this nutrient.

Carignan is a black grape cultivar widely planted throughout the western Mediterranean Basin. The grape faces significant viticultural hazards such as soil salinization, which affects about 6% of the world’s total land area. The search for salt tolerance genotypes to be introduced in crossbreeding programs and obtaining new cultivars is a key factor. The seed germination and salt tolerance of Carignan were studied from different coastal vineyards across the Mediterranean Basin, and as well as whether the distance from the sea affected germination and salt tolerance. Carignan seeds, independently of the temperature and distance from the sea, germinated more than 50% under 125 mM NaCl concentrations. Seed recovery was elevated, including the capacity of gemination after high salt exposure (500 mM NaCl). The results on germination behavior related to the distance from the sea showed that all tested vineyards, except for the one farthest from the sea, had similar germination responses. The optimum germination condition to select salt-tolerant accessions is alternating temperatures 25/10 °C and 125 mM NaCl. Thanks to the ability of the Carignan to germinate in a saline substrate and their capacity for recovery, it could be useful to crossbreeding programs, for integrating as rootstock selection or for the improvement of cultivars through sexual reproduction.

The present study was carried out to assess the effect of grape seed oil supplemented diet on non- specific immunity of Cyprinus carpio against the pathogenic bacteria Pseudomonas aeruginosa. Fish weights 41.69±2.25 g and total length 13.65±1.24 cm were randomly distributed into four treatments (two replications for each treatment). Fish groups were fed four dissimilar diets up to 40 days. The first group (T1) was fed with 0.2% grape seed oil, the second group (T2) was fed supplemented with 0.5% grape seed oil, the third group (T3) was supplemented with 1% grape seed oil and the forth group (T4) was served as the control group which fed basal diet without supplementing with grape seed oil. Forty-six fishes in all treatment groups were challenged intramuscularly with P. aeruginosa on day 41. The fishes in experimental groups were challenged intramuscularly with 0.2 ml P. aeruginosa at a concentration of 108 CFU/ml and after challenge (14 days), the different parameters were determined including nitroblue tetrazolium % (NBT), myeloperoxidases % (MPO), phagocytic activity (%) and serum lysozyme activity (U/min). An experiment was conducted to ascertain the effect of individual fruit wrapping on fruit quality and colour development of Flame Seedless variety of grapes during 2018–19 fruiting seasons. Uniform size clusters of Flame Seedless were bagged with white colour and red colour non-woven bags, butter paper bags and LDPE sheets at 40 per cent colour break stage during first of June. At time of attaining full colour, fruits were harvested from all the replication during morning hours and taken for analysis.

In this study, it was aimed to determine the effects of V. vinifera L seed extract against the negative effects of performance parameters, organ weights and blood parameters due to the administration of gentamycin in broilers. A total of 240 Ross 308 chickens were divided into 4 groups with 6 replications of 10 birds per cage. Treatment groups: Control group was fed a basal diet only; group V was fed a diet supplemented with grape seed extract (100 mg/kg); group G was fed a basal diet and administered gentamycin (50 mg/kg body weight intramuscularly); group V+G was fed a diet supplemented with grape seed extract, and gentamycin in levels as in group V and G. The highest weight gain and feed intake at the 3rd, 4th, 5th and 6th weeks was observed in group C, and the lowest, statistically significant, was in group G. The highest carcass weight was in group C. The highest aspartate aminotransferase and alanine aminotransferase levels were in group G. The lowest red blood cell value was in group C. Liver weight was increased significantly in group G. In this study, it was observed that the use of gentamycin harmed the health of broiler hens, but these effects could be reduced partially with the grape seed extract added to the feed. For this reason, grape seed extract may be used as a feed additive with healing effect.

Flavonoids are important polyphenols, and widely distributed in plants. Anthocyanidin reductase (ANR), a precursor enzyme in (-)- epicatechin synthesis, could play an important role in anthocyanins biosynthesis. In the present study, the ANR gene was cloned from Vitis vinifera L. cv. Cabernet Sauvignon. Compared with VvANR (GenBank Accession No. NM_001280956.1), they differed in only 4 bp and shared similarity of 99.6%. The ANR protein belongs to the cl33424 Superfamily. In addition, many light-responsive elements and abiotic stress-related elements (ABRE, MYB, WRKY, etc.) were found in the promoter region of the ANR gene. STRING analysis predicted that VvMYBPA1 and VvMYBPAR may be involved in regulating the expression of the ANR gene. Different UV-C irradiation times were used for Cabernet Sauvignon berries during grape berry development, and the comparison analysis showed that UV-C irradiation improved significantly the accumulation of catechin and epicatechin during 20–50 days after full bloom, especially, under UV-C irradiation for 5 min. Furthermore, UV-C irradiation for 5 min had a greater effect on the expression of the ANR gene, the accumulation of catechin and epicatechin, and the enzymatic activity of ANR than UV-C irradiation for 10 min. Through the virus-induced gene silencing system, the ANR-silenced grape leaves were chlorotic, in which the contents of catechin and epicatechin were both significantly lower than those in the control, showing a very significant difference, respectively. In summary, our results demonstrated that the ANR gene played a crucial role in the accumulation of epicatechin in grapes, and its expression was upregulated by UV-C irradiation.

Background: Drought stress affects grapevine growth and development and reduces berry yield and quality. Identifying genes that are involved in the plant response to drought stress will enable the development of new grape strains that are tolerant to drought. Objective: We cloned the VvibZIP14 gene from Vitis vinifera and analyzed its role in drought resistance. Methods: Gene expression was analyzed by quantitative real-time PCR. Subcellular localization was assessed with a transient expression assay. The transactivation activity of the protein was evaluated in yeast. The physiologic role of VvibZIP14 was analyzed by overexpressing VvibZIP14 in Arabidopsis following drought stress. Hydrogen peroxide accumulation in Arabidopsis was visualized by diaminobenzidine staining. Results: Drought stress caused the accumulation of VvibZIP14, which was localized in the nucleus and had transcriptional activity. Transgenic plants showed improved resistance to drought stress and reduced electrolyte leakage compared to plants overexpressing empty vector, whereas chlorophyll content, photosystem II maximal photochemical efficiency, and net photosynthetic rate were higher. Catalase, peroxidase, and superoxide dismutase activities were also increased in VvibZIP14-overexpressing plants subjected to drought stress. Conclusions: VvibZIP14 functions as a transcription factor that confers resistance to drought stress in grape by enhancing the antioxidant response.

The aim of the study was to determine the effect of a live mycorrhizal vaccine on the dynamics of grapevine’s plant nutrient supply. The experiment was performed on the Graševina variety (Vitis vinifera L.), grafted on an SO4 rootstock in the conditions of the Kutjevo Vineyards. The training system was Guyot, with a 10‐bud load. In the root system’s immediate vicinity, 20 ml of mycorrhizal suspension containing approximately 2,000 propagation units of a live mycorrhizal mycelium were inoculated. From the viticultural aspect, the research was conducted in two climatically very different years. The first research year (2014) was characterized by an abundant precipitation and less sunny days, resulting in a later technological grape maturity, amounting to 34 days when compared to the year 2015, which was extraordinarily warm and arid during the grape ripening period. In both research years, in the conditions of the soil’s low supply with the organic matter, phosphorus, and potassium, a mycorrhizal symbiosis influenced the higher content of phosphorus and potassium in the Graševina’s dry leaf matter. In a year experiencing more precipitation and less heat, a higher nitrogen content was found in the vine leaves manifesting a mycorrhizal symbiosis. In the dry and warm conditions, the calcium content was increased during the entire period of grape ripening, and a justifiably higher content of zinc, manganese, and iron was found in the period immediately preceding a technological grape ripeness. A positive influence of mycorrhiza on the yield, sugar content, and total acidity was not determined.

Kaolin particle film (KPF) is an aqueous formulation of chemically inert mineral particles that can be sprayed on the surface of crops to form a protective film, resulting in increased fruit yield and quality. In this work, the effects of kaolin-based, foliar reflectant particle film on grape composition and volatile compounds in ‘Meili’ (Vitis vinifera L.) grapes were investigated under different growth stages over two growing seasons. The 100-berry weight and titratable acid content were decreased, and the sugar and soluble solid contents were increased in grapes of plants treated with kaolin over 2 years. Compared with grapes from plants not sprayed with kaolin, the levels of total phenol, flavonoid, flavanol, tannin, and anthocyanins of grapes from plants treated with kaolin for 2 years were mostly increased. High-performance liquid chromatography (HPLC) analysis also revealed an increased content of monomeric anthocyanin and changed anthocyanin composition. However, there was little effect on the volatile compounds in the grapes. These results demonstrate that KPF can facilitate the accumulation of sugar and phenolics, thereby improving grape quality even in a humid climate.

Excessive continuous noises exposure changes the male hormone system which leads to formation of oxidative stress and results in disrupt of semen quality. This condition can be reduced by the use of antioxidants. Grape seed is one of the antioxidants that contains phenol components that have Resveratrol compounds. This research aimed to observe the effect of noise exposure on the number of spermatozoa of male mice given grape seed extract. The method of the research was a post-test only control group design with research subjects of 30 male Mus musculus L. (Swiss Webster) mice divided into 5 groups: group A (treatment control) was not given noise exposure and grape seed extract, group B (negative control) was not given noise exposure and was given grape seed extract, groups C, D, E (treatment group) were given noise exposure with sequential intensities of 65 dB, 85 dB, 105 dB. The treatment was given for 33 days. The results showed that noise exposure with different intensities of 65 dB, 85 dB, 105 dB reduced the spermatozoa count of male Swiss Webster mice even with the administration of grape seed extract. One Way Anova test was used to analyze the data with p-value of 0.001. Conclusion: There were differences in spermatozoa count between the control group and the treatment group. Provision of noise exposure with a value above the threshold limit reduces the number of male Swiss Webster mice spermatozoa given with grape seed extract. Background: Scientists have used herbs nowadays for curing many diseases because they are safer and to overcome the side effect of the chemical drugs. Grape is one of the ancient herbs that used for diseases of the heart and blood vessels, high blood pressure, high cholesterol, skin care and many other conditions. Grape seeds have significantly higher concentrations of polyphenols which has anticoagulant and antithrombotic effect. Materials and Methods: In this study 20 normal blood samples from healthy individuals with age range (19-38) years were enrolled in this study]. Prothrombin time (PT) and activated partial thromboplastin time (APTT) tests were performed before adding grape seed extraction (GSE) (as control) and after adding GSE with different concentrations (25%, 50% and 75%). Results: The results revealed that grape seed extract has an anticoagulant effect as proven by the increase of Prothrombin time and Activated partial thromboplastin time results of the blood samples in different concentrations of the extract. The GSE showed a high statistical significant (P= 0.000) in all concentrations of both PT and APTT tests. Conclusion: This study suggests that GSE has a strong anticoagulant effect; so it can potentially be used as a supplementary anticoagulant agent to prevent thrombosis and cardiovascular diseases.

Pierce's disease (PD) in grapevine (Vitis vinifera) is caused by the bacterial pathogen Xylella fastidiosa. X. fastidiosa is limited to the xylem tissue and following infection induces extensive plant‐derived xylem blockages, primarily in the form of tyloses. Tylose‐mediated vessel occlusions are a hallmark of PD, particularly in susceptible V. vinifera. We temporally monitored tylose development over the course of the disease to link symptom severity to the level of tylose occlusion and the presence/absence of the bacterial pathogen at fine‐scale resolution. The majority of vessels containing tyloses were devoid of bacterial cells, indicating that direct, localized perception of X. fastidiosa was not a primary cause of tylose formation. In addition, we used X‐ray computed microtomography and machine‐learning to determine that X. fastidiosa induces significant starch depletion in xylem ray parenchyma cells. This suggests that a signalling mechanism emanating from the vessels colonized by bacteria enables a systemic response to X. fastidiosa infection. To understand the transcriptional changes underlying these phenotypes, we integrated global transcriptomics into the phenotypes we tracked over the disease spectrum. Differential gene expression analysis revealed that considerable transcriptomic reprogramming occurred during early PD before symptom appearance. Specifically, we determined that many genes associated with tylose formation (ethylene signalling and cell wall biogenesis) and drought stress were up‐regulated during both Phase I and Phase II of PD. On the contrary, several genes related to photosynthesis and carbon fixation were down‐regulated during both phases. These responses correlate with significant starch depletion observed in ray cells and tylose synthesis in vessels., During Pierce's disease, Xylella fastidiosa triggers transcriptional changes in Vitis vinifera grapevines and induces major physiological responses, including tylose formation and starch depletion.

BACKGROUND: The pathway of anthocyanin biosynthesis, and its alterations leading to berry colour modification, are well known in grape skin. This variability could affect both quantity and quality of pigment accumulation. OBJECTIVE: The present work is focused on 15 grapevine cultivars selected to represent a high variability in the phenotypical colour traits in order to highlight new polymorphisms related to the flavonoid pathway. METHODS: Twenty-one genes involved in the biosynthetic pathway of anthocyanins were studied via targeted resequencing and were correlated with phenotypic data (anthocyanin profiles and spectroscopy indices). RESULTS: Single nucleotide polymorphism (SNP) and InDel (insertion/deletion) polymorphisms were detected. Out of 1751 polymorphic loci, 68% were SNPs and 32% were InDels (568). Cluster analysis and SPLS-DA were used to investigate the genetic relationships among the cultivars, confirming the large range of phenotypical variability. Statistically significant correlations were detected between accumulation of cyanidin-based anthocyanins and genetic polymorphisms in two structural genes and one transcription factor putatively involved in the anthocyanin biosynthetic pathway. CONCLUSIONS: The understanding of the polymorphisms related to the anthocyanin accumulation could support future selection of new pink table grape varieties with increased appeal on the consumers.

The seedlessness of grape derived from stenospermocarpy is one of the most prized traits of table or raisin grapes. It is controlled by a complex genetic system containing one major dominant gene and multiple minor recessive genes. Here, we collected dense variation data from high-depth resequencing data of seeded, seedless, and wild relative grape genomes sequenced to > 37x mean depth. Variant calls were made using a modified variant calling pipeline that was suitable for highly diverse interspecific grape accessions. The modified pipeline enabled us to call several million more variants than the commonly recommended pipeline. The quality was validated by Sanger sequencing data and subsequently supported by the genetic population structure and the phylogenetic tree constructed using the obtained variation data, results of which were generally consistent with known pedigree and taxonomic classifications. Variation data enabled us to confirm a major dominant gene and identify minor recessive loci for seedlessness. Incidentally, we found that grape cultivar Rizamat contains an ancestral chromosomal region of the major gene in Sultanina, a predominant seedlessness donor cultivar. Furthermore, we predicted new candidate causal genes including Vitvi01g00455, Vitvi08g01528, and Vitvi18g01237 associated with the minor seedless-regulating loci, which showed high homology with genes that regulate seed development in Arabidopsis. This study provides fundamental insights relevant to variant calling from genome resequencing data of diverse interspecific hybrid germplasms such as grape and will accelerate future efforts aimed at crop improvement.

Grape (Vitis vinifera) cluster compactness is an important trait due to its effect on disease susceptibility, but visual evaluation of compactness relies on human judgement and an ordinal scale that is not appropriate for all populations. We developed an image analysis pipeline and used it to quantify cluster compactness traits in a segregating hybrid wine grape (Vitis sp.) population for 2 years. Images were collected from grape clusters immediately after harvest, segmented by color, and analyzed using a custom script. Both automated and conventional phenotyping methods were used, and comparisons were made between each method. A partial least squares (PLS) model was constructed to evaluate the prediction of physical cluster compactness using image-derived measurements. Quantitative trait loci (QTL) on chromosomes 4, 9, 12, 16, and 17 were associated with both image-derived and conventionally phenotyped traits within years, which demonstrated the ability of image-derived traits to identify loci related to cluster morphology and cluster compactness. QTL for 20-berry weight were observed between years on chromosomes 11 and 17. Additionally, the automated method of cluster length measurement was highly accurate, with a deviation of less than 10 mm (r = 0.95) compared with measurements obtained with a hand caliper. A remaining challenge is the utilization of color-based image segmentation in a population that segregates for fruit color, which leads to difficulty in differentiating the stem from the fruit when the two are similarly colored in non-noir fruit. Overall, this research demonstrates the validity of image-based phenotyping for quantifying cluster compactness and for identifying QTL for the advancement of grape breeding efforts.

Purpose: The effect of abusing chemical biocides in controlling pests and diseases has drawn the attention of policymakers to the development of methods potentially available in nature for this purpose. Research method: In the present study, the inhibitory effects of four different essential oils against Botrytis cinerea were tested at various concentrations (0, 200, 400, 600, and 800 μL L-1) in in vitro and in vivo. Main findings: The in vitro results showed that the growth of B. cinerea was completely inhibited by the application of anise oil at concentrations of 800 μL L-1. The in vivo results indicated that treated fruits with marjoram oil had more total soluble solids, and anthocyanin content in comparison to anise, chamomile, and black caraway oil. Furthermore, among essential oils, treated fruits with black caraway essential oils had the lowest pH, while anise, chamomile, and marjoram oil had the highest pH. The highest anthocyanin content and pH were obtained at 200 and 400 μL L-1 concentration and lowest values were found at a control treatment (respectively). The most total soluble solids were observed at control treatment and the lowest values were recorded at 600 μL L-1. The application of each essential oil decreased the percentage loss in fresh weight significantly and increased the storage-life of the fruit. Limitations: Higher cost of application was a limitation. Originality/Value: This research confirms the antifungal effects of anise, fennel, chamomile, and marjoram essential oils both in vitro and in vivo on grape fruits postharvest. Therefore, these essential oils could be an alternative to chemicals to control postharvest phytopathogenic fungi on grape fruits.

This study was conducted to determine the chilling requirement of the Abjosh grape (local cultivar). The experiment was carried out based on randomized complete block design with three replications. The grape cuttings were treated at 5°C for (0, 100, 200, 250, 300, 350, 400, and 500 hrs) as chilling treatments at Samangan Higher Education Institute in the autumn of 2018. The traits such as the percentage of bud breaking, number of days to the first bud breakage, number of days to 50% of bud breaking, and number of days to the last bud breaking were measured. The results showed that the percentage of bud breaking increased significantly, as the chilling hrs (hours) increased. By the increasing of cold duration, the percentage of bud breaking in 500 hrs increased to 80% whereas, number of days to the first bud breaking, 50% of bud breaking and the number of days to the last bud breaking were decreased to 13, 14, and 16 days, respectively. Though, there were no significant differences between the 400 and 500 cold hrs in all indicators. The findings revealed that at least 300 hrs of cold duration is essential to reach an acceptable percentage of bud breaks, but further elaboration is needed.

Grapes are known to synthesize resveratrol, a stilbene phytoalexin, associated with cancer chemopreventive activity and cardioprotection. The effect of ultrasound (US) abiotic elicitor treatments on trans-resveratrol content in Kalecik Karasi fresh and frozen grape skin was determined. D-optimal point change design was used under RSM for the experimental design of US treatment. The optimization was solved with the help of the Pareto areas and the optimal input variable values were determined by the desirability function with fuzzy similar perceivable ratio method. The optimum conditions of US treatment for fresh grape skin were determined as follows: incubation time—24 h, US application method-(P01), US frequency—20 kHz, US treatment time—60 min and ultrasonic intensity (UI)—1.15 W cm−2. The trans-resveratrol content (0.18 ± 0.01 mg/g) in the untreated grape skin significantly increased with optimum US treatment (3.58 ± 0.08 mg/g), increasing production to 19.9 times.

BackgroundBud dormancy is a strategic mechanism plants developed as an adaptation to unfavorable environments. The grapevine (Vitis vinifera) is one of the most ancient fruit vine species and vines are planted all over the world due to their great economic benefits. To better understand the molecular mechanisms underlying bud dormancy between adjacent months, the transcriptomes of ‘Rosario Bianco’ grape buds of 6 months and three nodes were analyzed using RNA-sequencing technology and pair-wise comparison. From November to April of the following year, pairwise comparisons were conducted between adjacent months.ResultsA total of 11,647 differentially expressed genes (DEGs) were obtained from five comparisons. According to the results of cluster analysis of the DEG profiles and the climatic status of the sampling period, the 6 months were divided into three key processes (November to January, January to March, and March to April). Pair-wise comparisons of DEG profiles of adjacent months and three main dormancy processes showed that the whole grapevine bud dormancy period was mainly regulated by the antioxidant system, secondary metabolism, cell cycle and division, cell wall metabolism, and carbohydrates metabolism. Additionally, several DEGs, such asVvGA2OX6andVvSS3, showed temporally and spatially differential expression patterns, which normalized to a similar trend during or before April.ConclusionConsidering these results, the molecular mechanisms underlying bud dormancy in the grapevine can be hypothesized, which lays the foundation for further research.

Grape (Vitis vinifera L.) is globally cultivated as commercial fruit crop usually used for fruit purpose or industrial product. The objective of the current review is to review and identify the research gap on the effect of different growth media and vitrification on shooting and rooting performance of grape. Factors affecting rooting of grape cuttings can be internal or external factors. Currently, grapevines are very sensitive to disease in the conventional method of propagation. Even if tissue culture is recommended for healthy propagation of the grape varieties, still factors affecting the growth of the plant verifications were reported. Background Cactus pear (Opuntia ficus-indica (L.) Miller) is an important food source for human beings but its consumption and marketability are limited due to the presence of spines and glochids on the fruit surface. Fresh-cut, ready-to-eat cactus pears have higher preference than the whole fruits. However, fresh-cut fruits have tissue wounding and quick deterioration, which decrease the marketability of the products. Therefore, present study was aimed to improve the storage quality of ready-to-eat cactus pears with the application of environmentally friendly bio-materials. Methods Fruits of present study were hand collected by a traditional way from a farm located in Yayla village of Northern Cyprus. Fruits were then peeled to prepare fresh-cut prickly pears for further experiments. Fruit peeling was carried out in three steps: (i) slicing off and discarding the both ends of the prickly pears, (ii) making one long vertical slice down the body, and (iii) peeling back the skin by sliping our finger into the slice and preparing a peeled prickly pear without skin. The six treatments of present study are: (1) control—dipping the fruits into distilled water, (2) covering the fruits with Vitis vinifera leaves, (3) dipping the fruits into jelly, (4) dipping the fruits into Aloe vera gel, (5) dipping the fruits into Portulaca oleracea extract and (6) dipping the fruits into cactus gel. Totally 108 fruits (replications) were used for each treatment and 18 fruits from each treatment were selected for quality analysis with 3-days interval (3, 6, 9, 12, 15 and 18 days). Fruits of each treatment (6 from same treatment) were placed into an open plastic box (dimensions: 20 * 13 * 5 cm; material: PET) and was covered with a stretch film (LLDPE, 10 µm) for storage. The fruit boxes were stored at storage rooms adjusted to 5 ± 1 °C and 55 ± 5% relative humidity. Results Results showed that, all treatments have varying positive effects on the prevention of weight loss. Among them, the highest positive effect was received from A. vera gel treatment (0.98%), and is followed by cactus gel (1.15%), V. vinifera leaf (1.39%) and jelly (1.61%) at the end of the experiments (18 days). A. vera gel and cactus gel were also found to have highest influence on the prevention of off-odor and decay incidence; and to protect the sensory and visual quality of the ready-to-eat cactus pears. Conclusions Results suggested that the postharvest storage duration of ready-to-eat cactus pears might be extended from 9 to 15 days with the use of Aloe vera gel or cactus gel.

Abscisic acid (ABA) is an essential phytohormone that regulates plant growth and development, particularly in response to abiotic stress. The ABA receptor PYR/PYL/RCAR (PYL) family has been identified from some plant species. However, knowledge about the PYL family (VvPYLs) in grape (Vitis vinifera) is limited. This study aims to conduct genome-wide analyses of VvPYLs. We successfully identified eight PYL genes from the newest grape genome database. These VvPYLs could be divided into three subfamilies. Exon-intron structures were closely related to the phylogenetic relationship of the genes, and PYL genes that clustered in the same subfamily had a similar number of exons. VvPYL1, VvPYL2, VvPYL4, VvPYL7, and VvPYL8 were relatively highly expressed in roots. VvPYL1, VvPYL3, VvPYL7, and VvPYL8 were expressed in response to cold, salt, or polyethylene glycol stress. VvPYL6 was up-regulated by cold stress for 4 hours, and the expression of VvPYL2 was 1.74-fold greater than that of the control under cold stress. VvPYL8 was up-regulated 1.64-, 1.83-, and 1.90-fold compared with the control when treated with salt, PEG, or cold stress after 4 hours, respectively. Additionally, abiotic stress-inducible elements exist in VvPYL2, VvPYL3, VvPYL7, and VvPYL8, indicating that in these four genes, the response to abiotic stress may be regulated by cis-regulatory elements. The transcriptional levels of VvPYL1 and VvPYL8 significantly increased from fruit set to the ripening stage and decreased in the berry when treated by exogenous ABA. The eight VvPYL genes have diverse roles in grape stress responses, berry ripening, or development. This work provides insight into the role of VvPYL gene families in response to abiotic stress and berry ripening in grape.

Background: Oral pathogens can affect a large population and the general health of individuals. Based on the evidence, the grape seed extract (GSE) contains herbal compounds that have the ability tosuppress oral pathogens associated with caries and fungal diseases. In this regard, the current studyaimed at evaluating the effect of GSE on Streptococcus mutans and Candida albicans. Methods: The grape (Vitis vinifera L.) seed was used in this in-vitro experimental study. After thepreparation of methanolic GSE, its effect on S. mutans and C. albicans was assessed at 0.25-256 mg/mL concentrations. Then, the minimum inhibitory concentration (MIC) and minimum bactericidalconcentration (MBC) were determined by macro broth dilution methods. Finally, data were analyzedby SPSS, version 20. Results: The results showed that GSE inhibits the growth of S. mutans and C. albicans. In addition, theMIC and MBC of the extract against S. mutans and C. albicans were 2 and 8 mg/mL, as well as 32 and64 mg/mL, respectively. Conclusions: In general, GSE had a significant antimicrobial effect on S. mutans, while it only affected C. albicans at high concentrations.

Pharmaceutical, food and cosmetic industries have shown great interest in grape seed extract due to its premium antioxidant activity with protective effects on human health. However, global climate change has already negatively affected the biochemistry and functionality of such nutraceutical crops. The present study was conducted to reveal the effects of sustained deficit irrigation on certain features of seed extracts obtained from the grape cultivar ‘Prima’ grafted on six genetically different rootstocks during two consecutive years. In both years, seed weight of ‘Prima’ grapes grafted on 5 BB significantly decreased in response to water treatments, although the other rootstocks did not impair the seed weight, except for 44–53 M in 217. Significant decreases in antioxidant activity of the scion cultivar occurred in response to DI when grafted on 5 BB in both years and in 44–53 M in 2017. However, the use of 99 R, 110 R and 140 Ru did not markedly impair the seed antioxidant activity in response to DI condition in both years. In both years, seed anthocyanins of FI vines were always higher than those of DI vines. In 2017, there were significant decreases in anthocyanins due to DI in vines grafted on 5 BB, 44–53 M, 99 R and 110 R. These rootstocks also led significant decreases in anthocyanins of the second year, while the use of 140 Ru and 1613 C rootstocks did not lead significant changes in anthocyanins in both two years. In 2017, total phenolics increased in response to DI, except for the vines grafted on 44–53 M. Rootstock-dependent changes in total phenolics in response to irrigation treatments were insignificant for the vines grafted on 5 BB and 44–53 M in both two experimental years. Averaged data showed that antioxidant activity showed significantly positive correlation with anthocyanins and total phenols. The findings are anticipated to guide future projects on enhancement of the quality and quantity of precious seconder metabolites with sustainable precision viticulture techniques to match the globally increasing demands on bioactive compounds.

The use of resistant rootstocks is an inevitable trend in the development and production of grapes (Vitis sp.). The present study analyzed differences in the metabolites in grape seeds of different rootstock combinations (1103P, 5C, SO4, 3309C, 140R, and control) grafted onto ‘Cabernet Sauvignon’ (CS) wine grape (Vitis vinifera) scions (control, CS/CS, self-rooted grafting vines) using liquid chromatography–mass spectrometry (LC-MS) and nontargeted metabolomic techniques. Principal component analysis (PCA), partial least squares discriminant analysis (PLS-DA), and orthogonal-partial least squares discriminant analysis identified 30 significant metabolites and 22 metabolic pathways in the seeds of CS that differed significantly from the control seeds. This study revealed that rootstocks influence metabolite concentrations and metabolic pathways (alanine–aspartate–glutamate pathway, arginine-proline pathway, and the tricarboxylic acid cycle) in the scion onto which they are grafted. The rootstocks increased the concentration of delphinidin-3-(6-acetylglucoside), peonidin 3-(6-p-coumarylglucoside), L-threonine, and D-tartaric in CS seeds. Appropriate rootstock combinations can be used to improve the quality of grape seeds by changing the concentrations of amino acids, organic acids, polyphenols, and vitamin B. This study provides a theoretical basis for selecting grape rootstocks and provides important insights for improving the quality of commercial products derived from grape seeds.

Background and Aims Optical leaf sensing of plant nitrogen (N) status enables fertilisation based on plant demand. The aim of this study was to assess the suitability of leaf reflectance indices and of a handheld device for the estimation of N and chlorophyll (Chl) in Vitis vinifera L. cv. Riesling. Methods and Results Riesling leaves from a long‐term fertilisation trial were analysed for nitrogen and pigments and their spectral reflectance in the Vis/near‐infrared range was recorded for two consecutive seasons. In the second season the leaves were also measured with a Chl fluorescence based portable device (Dualex). Published reflectance indices and fluorescence indices were regressed against the concentration of pigment and N. Temporal changes in the Chl/N ratio complicated the indirect assessment of N via Chl. Red edge inflection points and several three‐ to four‐waveband indices allowed for a good prediction of leaf N. Fluorescence indices allowed for reliable estimation of N and Chl. Conclusions While leaf pigments can be analysed by simple two‐band indices, accurate N estimation requires more complex indices such as three‐band indices or indices calculated from derivative spectra. Significance of the Study Data from this study will assist growers to monitor non‐destructively the spatial and temporal variability of grapevine N status and thereby, improve fertilisation.

This study was conducted to determine the effects of three different bud load levels (15 buds, 25 buds and 30 buds m-2) and two different leaf removal (25% and 50% on each vine shoot) treatments on number of bursted bud, midday leaf water potential (?mid), stomatal conductance (gs), total soluble solids (TSS) and titratable acidity (TA) of own-rooted ‘Sultana Seedless’ (Vitis vinifera L.) in 2016 and 2017 vegetation season in Alaşehir, Turkey.
