1. Disentangling Gender and Relative Age Effects in Women’s and Girls’ Rugby Union

Abstract

Relative age effects (RAEs) within sports refer to the overrepresentation of athletes born earlier in the selection year and the underrepresentation of those born later in the selection year. Research examining RAEs in women’s and girls’ rugby union remains limited in comparison to the male literature, whilst the impacts of RAEs on the youth–senior transition are yet to be explored in a female sport context. As such, the purpose of this study was to examine RAEs during entry into the women’s and girls’ premiership and international rugby union pathways in England, as well as to compare them to their respective senior cohort (n = 1367): (a) U18 England Rugby Centre of Excellence Player (n = 325) vs. Senior Premiership Player (n = 868), and (b) U18 England Player (n = 49) vs. Senior England Player (n = 125). Chi-square (χ2) analyses compared birth quarter (BQ) distributions against expected distributions. The findings revealed no significant difference in BQ distributions at either youth or senior levels, as well as no significant differences in the BQ distributions of those who were likely to transition from youth to senior levels (all p > 0.05). Importantly, though, descriptive statistics showed a skewed birthdate distribution in both U18 England Rugby Centre of Excellence Player (BQ1 = 30% vs. BQ4 = 20%) and U18 England Player cohorts (BQ1 = 33% vs. BQ4 = 18%). We highlight the gender-specific mechanisms that potentially explain the variations between male and female RAEs in rugby union, including developmental differences, sport popularity, and sociocultural norms. We also warn against a ‘copy and paste’ template from the male provision to ensure the recent growth of female rugby union does not fall victim to the same RAEs in the future.

Abstract

The Biomass Proxy is a new cloud-free vegetation monitoring product that offers timely and analysis-ready data indicative of above-ground crop biomass dynamics at 10m spatial resolution. The Biomass Proxy links the consistent and continuous temporal signal of the Sentinel-1 Cross Ratio (CR), a vegetation index derived from Synthetic Aperture Radar backscatter, with the spatial information of the Sentinel-2 Normalized Difference Vegetation Index (NDVI), a vegetation index derived from optical observations. A global scaling relationship between CR and NDVI forms the basis of a novel fusion methodology based on static and dynamic combinations of temporal and spatial responses of CR and NDVI at field level. The fusion process is used to mitigate the impact on product quality of low satellite revisit periods due to acquisition design or persistent cloud coverage, and to respond to rapid changes in a timely manner to detect environmental and management events. The resulting Biomass Proxy provides time series that are continuous, unhindered by clouds, and produced uniformly across all geographical regions and crops. The Biomass Proxy offers opportunities including improved crop growth monitoring, event detection, and phenology stage detection.

Abstract

It is an old concern in public and academic debates that people are not interested in European-level issues, and even European Parliamentary election campaigns, which are the main democratic tools of the European Union (EU) to involve ordinary people into political decision-making, are mostly about national-level political topics. Moreover, even when European issues emerge, the context of its discussion is often harmful to European integration and strengthens the perceived importance of domestic politics. In the age of social media, however, users’ content preferences may significantly affect the presence of different political levels in political campaigns, but these preferences are still largely uncovered in academic literature. To fill this gap, we investigate the direct and moderated effects of European-focused Facebook posts on user engagement drawing upon a content analysis dataset including 9,688 posts of 68 parties from 12 EU countries. In line with the well-known second-order election hypothesis (Reif & Schmitt, 1980) we hypothesize a negative direct main effect. However, we also assume that this effect is moderated by several content-, and party-level factors, and when people engage with European-level contents they do it with those ones that are posted by populist parties, focused on a few divisive hot topics, and are framed with a negative tone. Moreover, we expect cross-country differences. We find that on the whole, user engagement with national-level political content prevails over the European-level, but in some countries there are no remarkable differences in user engagement patterns of the two levels. While our findings mostly confirm the second-order election hypothesis, they also demonstrate that European politics can spread within social media platforms in a less divisive and negative way than we expected. European-focused posts do not perform better when they are posted by populist parties, focused selectively on the salient issues of immigration or environment, or framed in a negative way.

Abstract

BackgroundCystic echinococcosis (CE) is a zoonotic neglected tropical disease (zNTD) which imposes considerable financial burden to endemic countries. The 2021-2030 World Health Organization's roadmap on NTDs has proposed that intensified control be achieved in hyperendemic areas of 17 countries by 2030. Successful interventions for disease control, and the scale-up of programmes applying such interventions, rely on understanding the associated costs and relative return for investment. We conducted a scoping review of existing peer-reviewed literature on economic evaluations of CE control strategies focused on Echinococcus granulosus zoonotic hosts.Methodology/principal findingsDatabase searches of Scopus, PubMed, Web of Science, CABI Direct and JSTOR were conducted and comprehensively reviewed in March 2022, using predefined search criteria with no date, field or language restrictions. A total of 100 papers were initially identified and assessed for eligibility against strict inclusion and exclusion criteria, following the Preferred Reporting Items for Systematic reviews and Meta-Analyses extension for Scoping Reviews (PRISMA-ScR) guidelines. Bibliography review of included manuscripts was used to identify additional literature. Full review of the final manuscript selection (n = 9) was performed and cost data for control interventions were extracted.Conclusions/significanceThere are very little published data pertaining to the cost and cost effectiveness of CE control interventions targeting its zoonotic hosts. Data given for costs are often incomplete, thus we were unable to perform an economic analysis and cost effectiveness study, highlighting a pressing need for this information. There is much scope for future work in this area. More detailed information and disaggregated costings need to be collected and made available. This would increase the accuracy of any cost-effective analyses to be performed and allow for a greater understanding of the opportunity cost of healthcare decisions and resource allocation by stakeholders and policy makers for effective and cost-effective CE control.

Abstract

Objectives: While opioids temporarily alleviate pain, the overshoot of balancing pain drivers may increase pain, leading to opioid induced hyperalgesia (OIH). Our goal was to find out what chronic opioid treatment does to pain tolerance as measured by the cold pressor test (CPT), an objective measure of pain tolerance, and to find an alternative effective treatment for chronic pain and FM.Materials and Methods: The setting was an academic addiction medicine service that has an embedded pain service. Patients had routine clinical care starting with an evaluation that included assessment of medical and psychiatric conditions. Participants were 55 patients with OIH and 21 patients with fibromyalgia; all had at least two CPTs. Treatment included a single dose of buprenorphine for detoxification. In this open-label case series, patients were treated with low dose naltrexone (LDN), a pure opioid receptor antagonist that, we hypothesize, treats OIH and FM by restoring endogenous opioid tone.Results: Comparing initial and last CPT times, those with OIH more than quadrupled their pain tolerance, and those with FM doubled theirs. This improved pain tolerance for OIH and FM was statistically significant (p < 0.0001 and p = 0.003, respectively) and had a large effect size (r = 0.82 and r = 0.63, respectively).Discussion: Results suggest that patients on chronic opioid therapy should have pain tolerance measured by CPT with detoxification and LDN provided to correct opioid induced hyperalgesia if found. FM may also be treated with LDN. The main limitation of the findings was lack of a randomized control group treated with placebo.

A common practice in youth rugby union is to group players based on (bi)annual age with fixed cut-off dates. The overrepresentation of players born at the start of the cut-off date and the underrepresentation of players born toward the end of the cut-off date are termed relative age effects (RAEs). The aim of this study was to examine RAEs during entry into professional and international rugby union pathways in England, as well as comparing them to their respective senior cohort: U15 Regional Academy Player (n = 1,114) vs. Senior Professional Player (n = 281) and U16–23 England Academy Player (n = 849) vs. Senior International Player (n = 48). Chi-square (χ2) analysis compared birth quarter (BQ) distributions against expected distributions. Odds ratios and 95% confidence intervals compared the likelihood of a BQ being selected. Findings revealed a significant overrepresentation of relatively older players compared with their relatively younger peers within both youth cohorts (P < 0.001; BQ1 = 42.5% vs. BQ4 = 9.6%; BQ1 = 36.5% vs. BQ4 = 15.2%). In comparison, there was no significant difference in the BQ distributions within both senior cohorts. Further, BQ4s were 3.86 and 3.9 times more likely to achieve senior professional and international levels than BQ1s and BQ2s, respectively. It is suggested that relatively younger players may have a greater likelihood of achieving expertise following entry into a rugby union talent pathway due to benefitting from more competitive play against relatively older counterparts during their development (e.g., reversal effects; the underdog hypothesis). Moreover, possible solutions (e.g., age and anthropometric banding; playing-up and playing-down) are discussed to encourage practitioners and policy makers to create the most appropriate learning environment for every player.

Abstract

Resíduos sólidos são todos os restos sólidos ou semi-sólidos das atividades humanas ou não-humanas, que embora possam não apresentar utilidade para a atividade fim de onde foram gerados, podem virar insumos para outras utilizações. Objetivou-se avaliar o comportamento da população à respeito das atitudes e percepção com relação aos resíduos sólidos na cidade de Coremas, Paraíba. Os dados referentes a atitudes e percepção dos atores sociais foram coletados na cidade de Coremas por meio de questionários, previamente estruturados com perguntas de múltipla escolha a fim de conhecer o perfil dos entrevistados através das seguintes variáveis: sexo, faixa etária, escolaridade, estado civil e renda. Foi realizada uma análise descritiva para verificar a percepção dos residentes no município segundo as variáveis relacionadas aos resíduos sólidos. A relação entre variáveis socioeconômicas e o comportamento ambiental referente aos resíduos sólidos dos moradores da cidade de Coremas foi verificada por meio do teste qui-quadrado de independência, mostrando que houve forte dependência entre essas variáveis, sugerindo que, a escolaridade, a renda e a localização dos residentes influenciaram em suas atitudes e percepções ambientais naquele município. Environmental Awareness and perception about solid waste by the residents of the City of Coremas, Paraíba Abstract: Solid wastes are all the solid or semi-solid residues of human or non-human activities that, although do not show utility for the order in which they were generated, can become to inputs for other activities. The study’s aim was to evaluate the population’s behavior about the attitudes and the perception connected to the solid wastes in the city of Coremas, Paraíba state. The data on attitudes and perceptions of the social actors were collected in the city of Coremas through questionnaires, previously structured with multiple choice questions in order to know the profile of the interviewed using the following variables: gender, age, education, marital status and income. The Descriptive analysis were performed to verify the perception of residents in the city according to the related variables to solid wastes. The relationship between socioeconomics variables and environmental performance related to solids wastes from residents of Coremas were verified by the chi-square test of independence, showing that there was a strong dependency between these variables, suggesting that the educational level, the income and the location of residents influenced in their attitudes and environmental perceptions in that city.

Abstract

Acinar cells represent the primary target in necroinflammatory diseases of the pancreas, including pancreatitis. The signaling pathways guiding acinar cell repair and regeneration following injury remain poorly understood. The purpose of this study was to determine the importance of Hepatocyte Growth Factor Receptor/MET signaling as an intrinsic repair mechanism for acinar cells following acute damage and chronic alcohol-associated injury. Here, we generated mice with targeted deletion of MET in adult acinar cells (MET-/-). Acute and repetitive pancreatic injury was induced in MET-/- and control mice with cerulein, and chronic injury by feeding mice Lieber-DeCarli diets containing alcohol with or without enhancement of repetitive pancreatic injury. We examined the exocrine pancreas of these mice histologically for acinar death, edema, inflammation and collagen deposition and changes in the transcriptional program. We show that MET expression is relatively low in normal adult pancreas. However, MET levels were elevated in ductal and acinar cells in human pancreatitis specimens, consistent with a role for MET in an adaptive repair mechanism. We report that genetic deletion of MET in adult murine acinar cells was linked to increased acinar cell death, chronic inflammation and delayed recovery (regeneration) of pancreatic exocrine tissue. Notably, increased pancreatic collagen deposition was detected in MET knockout mice following repetitive injury as well alcohol-associated injury. Finally, we identified specific alterations of the pancreatic transcriptome associated with MET signaling during injury, involved in tissue repair, inflammation and endoplasmic reticulum stress. Together, these data demonstrate the importance of MET signaling for acinar repair and regeneration, a novel finding that could attenuate the symptomology of pancreatic injury.

Abstract

The accessory beta subunit (Ca(v)β) of calcium channels first appear in the same genome as Ca(v)1 L-type calcium channels in single-celled coanoflagellates. The complexity of this relationship expanded in vertebrates to include four different possible Ca(v)β subunits (β1, β2, β3, β4) which associate with four Ca(v)1 channel isoforms (Ca(v)1.1 to Ca(v)1.4) and three Ca(v)2 channel isoforms (Ca(v)2.1 to Ca(v)2.3). Here we assess the fundamentally-shared features of the Ca(v)β subunit in an invertebrate model (pond snail Lymnaea stagnalis) that bears only three homologous genes: (LCa(v)1, LCa(v)2, and LCa(v)β). Invertebrate Ca(v)β subunits (in flatworms, snails, squid and honeybees) slow the inactivation kinetics of Ca(v)2 channels, and they do so with variable N-termini and lacking the canonical palmitoylation residues of the vertebrate β2a subunit. Alternative splicing of exon 7 of the HOOK domain is a primary determinant of a slow inactivation kinetics imparted by the invertebrate LCa(v)β subunit. LCa(v)β will also slow the inactivation kinetics of LCa(v)3 T-type channels, but this is likely not physiologically relevant in vivo. Variable N-termini have little influence on the voltage-dependent inactivation kinetics of differing invertebrate Ca(v)β subunits, but the expression pattern of N-terminal splice isoforms appears to be highly tissue specific. Molluscan LCa(v)β subunits have an N-terminal "A" isoform (coded by exons: 1a and 1b) that structurally resembles the muscle specific variant of vertebrate β1a subunit, and has a broad mRNA expression profile in brain, heart, muscle and glands. A more variable "B" N-terminus (exon 2) in the exon position of mammalian β3 and has a more brain-centric mRNA expression pattern. Lastly, we suggest that the facilitation of closed-state inactivation (e.g. observed in Ca(v)2.2 and Ca(v)β3 subunit combinations) is a specialization in vertebrates, because neither snail subunit (LCa(v)2 nor LCa(v)β) appears to be compatible with this observed property.

Abstract

Background. Although the burden of stroke in terms of mortality and disability has been well documented in previous years, data after 2000 are limited. Therefore, the aim of this paper was to identify the epidemiology of stroke in the US and EU5 nations from data published in 2000 and later. Methods. Data from literature databases and online sources were collated to identify information relating to the incidence, prevalence, and mortality of stroke from the year 2000 onwards. Results and Conclusions. Twenty-three data sources were identified. The incidence of and mortality due to stroke both increase with age and are greater in males compared to females. Stroke is a common problem and likely to worsen in the US and EU5 as their populations age. However, pre-2000 trends of decreasing stroke mortality over time have continued after 2000, reflecting a consistent improvement in the treatment and care of patients with stroke.

BackgroundWe conducted a systematic review of incidence rates in England over a sixty-year period to determine the extent to which rates varied along accepted (age, sex) and less-accepted epidemiological gradients (ethnicity, migration and place of birth and upbringing, time).ObjectivesTo determine variation in incidence of several psychotic disorders as above.Data sourcesPublished and grey literature searches (MEDLINE, PSycINFO, EMBASE, CINAHL, ASSIA, HMIC), and identification of unpublished data through bibliographic searches and author communication.Study eligibility criteriaPublished 1950-2009; conducted wholly or partially in England; original data on incidence of non-organic adult-onset psychosis or one or more factor(s) pertaining to incidence.ParticipantsPeople, 16-64 years, with first -onset psychosis, including non-affective psychoses, schizophrenia, bipolar disorder, psychotic depression and substance-induced psychosis.Study appraisal and synthesis methodsTitle, abstract and full-text review by two independent raters to identify suitable citations. Data were extracted to a standardized extraction form. Descriptive appraisals of variation in rates, including tables and forest plots, and where suitable, random-effects meta-analyses and meta-regressions to test specific hypotheses; rate heterogeneity was assessed by the I²-statistic.Results83 citations met inclusion. Pooled incidence of all psychoses (N = 9) was 31.7 per 100,000 person-years (95%CI: 24.6-40.9), 23.2 (95%CI: 18.3-29.5) for non-affective psychoses (N = 8), 15.2 (95%CI: 11.9-19.5) for schizophrenia (N = 15) and 12.4 (95%CI: 9.0-17.1) for affective psychoses (N = 7). This masked rate heterogeneity (I²: 0.54-0.97), possibly explained by socio-environmental factors; our review confirmed (via meta-regression) the typical age-sex interaction in psychosis risk, including secondary peak onset in women after 45 years. Rates of most disorders were elevated in several ethnic minority groups compared with the white (British) population. For example, for schizophrenia: black Caribbean (pooled RR: 5.6; 95%CI: 3.4-9.2; N = 5), black African (pooled RR: 4.7; 95%CI: 3.3-6.8; N = 5) and South Asian groups in England (pooled RR: 2.4; 95%CI: 1.3-4.5; N = 3). We found no evidence to support an overall change in the incidence of psychotic disorder over time, though diagnostic shifts (away from schizophrenia) were reported.LimitationsIncidence studies were predominantly cross-sectional, limiting causal inference. Heterogeneity, while evidencing important variation, suggested pooled estimates require interpretation alongside our descriptive systematic results.Conclusions and implications of key findingsIncidence of psychotic disorders varied markedly by age, sex, place and migration status/ethnicity. Stable incidence over time, together with a robust socio-environmental epidemiology, provides a platform for developing prediction models for health service planning.

45. Development of a modular microfluidic platform for customisable in vitro neuroscience models

Abstract

Central nervous system (CNS) disorders are the major public health problem in the developed world, leading to a huge societal and economic burden. These disorders are only set to increase given their long-term impact and the growing and ageing population. An increased understanding of the CNS under physiological and pathophysiological states is therefore essential to tackle the problem. However, many animal models have limited translational value, whilst traditional in vitro techniques fail to replicate the complex in vivo microenvironment. Conversely, microfluidic and organ-on-chip technologies can offer an opportunity to develop more physiologically relevant in vitro models of the CNS. These enable more precise cellular patterning and more complex network formation, ultimately providing enhanced experimental capabilities. Whilst microfluidic technology has been well established within neuroscience over the past two decades, there remains limited uptake outside labs with specialist microfabrication facilities required. Further, whilst there are some commercially available devices, there is limited customisable and reconfigurable options, restraining their potential. To increase the uptake and improve ease-of-use of the microfluidic technology, it must be readily accessible, without the need for additional complex equipment, and it must be user-friendly and reliable. A modular platform is therefore proposed, enabling user-defined device production with more flexibility to meet the needs of the individual. In this thesis a modular platform was developed consisting of multiple, individual microfluidic units that combine via a protrusion-intrusion interface on a pressure sensitive adhesive film. This allows components to be assembled in a user defined manner, enabling the creation of bespoke culture environments, making the technology customisable and appealing to novice users. Proof-of-concept experiments demonstrated functional connectivity between environmentally isolated hippocampal cultures across modules, alongside some example applications. This platform presents an opportunity to increase experimental capabilities in neuroscience, enabling the creation of bespoke circuits, and ultimately aid the development of novel therapies.
