Analytics services provided by marketplace platforms have become increasingly important for sellers seeking market insights. In this paper, we examine a scenario in which an analytics service plays a vital role in enhancing sellers' understanding of market size and improving their decision-making. Using a game-theoretic model, we analyze the pricing strategies of the platform and the adoption strategies of sellers for the analytics service. Our study identifies two distinct effects of analytics services: the competition effect and the accuracy effect. Specifically, the competition effect manifests in opposing ways across different market scenarios, with a competition-intensifying effect in lowdemand markets and a competition-weakening effect in high-demand markets. Consequently, sellers using an analytics service command lower prices in low-demand markets and higher prices in highdemand markets. More interestingly, our results reveal that offering an analytics service could potentially hurt the total market demand, subsequently impacting the platform's revenue from the marketplace service and potentially leaving the platform worse off. Additionally, driven by both the accuracy and competition effects, adopting an analytics service may adversely affect seller profitability and consumer surplus without necessarily improving overall welfare. Moreover, the transaction fee for the marketplace service plays a crucial role in the interplay between the analytics and marketplace services. Specifically, in low-demand (high-demand) markets, as the transaction fee increases, platforms should consider reducing (increasing) the subscription fee to encourage more (fewer) sellers to adopt the analytics service, thereby enhancing overall market demand and increasing revenue from the marketplace service. Our findings also suggest that platforms should refrain from offering analytics services in high-demand markets when the transaction fee is relatively high. Furthermore, policymakers (sellers) should be mindful of the potential negative consequences associated with the adoption of analytics services in high-demand (low-demand) markets. Electrochemical ammonia synthesis (EAS) presents an attractive alternative to the Haber–Bosch process due to the benefits of energy saving, low carbon emission, environmental friendliness, and so on. However, the competing hydrogen evolution reaction (HER) severely limits the yield, selectivity, and current efficiency of NH3. Although the accumulation and self‐aggregation of active hydrogen (H*) are the primary causes of the competing HER, it also serves as the critical active species and intermediate for the multistep hydrogenation and deoxygenation processes. Therefore, the sensible regulation of the H* generation and consumption are essential for enhancing EAS performance. And it is significant to thoroughly review the current strategies for H* control. Herein, a comprehensive introduction of H* to provide a fundamental understanding of its role in electrochemical reactions, including generation, conversion, identification, and quantification protocols is first proposed. In addition, the role and control strategies of H* in EAS are carefully summarized with a particular focus on regulating H* generation and consumption to enhance the activity, selectivity, and Faradaic efficiency. Finally, the remaining challenges and perspectives are discussed. In the past, diatoms were considered the primary food source for copepods in both wild and aquaculture settings. However, recent studies have found that diatoms have defense mechanisms against predators, making them unsuitable as copepod feed. This study assessed the impact of addition of 100 μg L−1 of silicate to an inorganic nutrition formula containing 700 μg L−1 nitrogen, 100 μg L−1 phosphorus, and 100 μg L−1 iron. Our objective was to assess the impact of increased diatom abundance on copepod production. The experiment was carried out in 1000 L outdoor tanks over a period of 20 days, with adult Pseudodiaptomus annandalei copepods, introduced into each tank on the second day at an initial density of 10 ind. L−1. The results indicated that adding silicate reduced the prevalence of Chlorophyta, replaced by a higher proportion of Dinophyta, and eventually dominated by diatoms. While silicate had a positive effect on diatom culture, it had a negative effect on copepod production. Copepods are proven nutritious food sources for the mariculture/larviculture industry, however, unreliable methods for mass production of copepods are a major bottleneck. In this study, we modified a previously reported inorganic fertilization method (N: 700 μg L−1 and P: 100 μg L−1) by the addition of iron (Fe: 10 μg L−1, using FeSO4·7H2O) (+Fe treatment) and compared its suitability for copepod culture (Pseudodiaptomus annandalei) to the original method (control). The experiment was conducted outdoors in 1000 L tanks for 15 days. The addition of iron prolonged the growth phase of the phytoplankton and resulted in the production of significantly more small phytoplankton (0.45–20 μm, average 2.01 ± 0.52 vs. 9.03 ± 4.17 µg L−1 in control and +Fe, respectively) and adult copepods (control: 195 ± 35, +Fe: 431 ± 109 ind L−1), whereas copepodid-stage was similar between treatments (control: 511 ± 107 vs. +Fe: 502 ± 68 ind L−1). Although adding iron increased the cost of production by 23% compared to the control, the estimated net profit was 97% greater. We concluded that inorganic fertilization, with the addition of iron (Fe: 10 μg L−1), could be an effective method for the mass production of copepods for larviculture. [ABSTRACT FROM AUTHOR]

As a frequent complication of inflammatory bowel disease (IBD), Clostridium difficile infection (CDI) was confirmed to not only aggravate the symptoms of IBD but also result in unexpected outcomes, including death. With the increasing prevalence rate of IBD and the updating of CDI diagnosis, the incidence of CDI in IBD patients is also seen rising. Although a detection method consisting of glutamate dehydrogenase immunoassay or nucleic acid amplification test and then toxin A/B enzyme immunoassay was recommended and widely adopted, the diagnosis of CDI in IBD is still a challenge because of the overlap between the symptoms of CDI in IBD and CDI itself. Vancomycin and fidaxomicin are the first-line therapy for CDI in IBD; however, the treatment has different effects due to the complexity of IBD patients’ conditions and the choice of different treatment schemes. Although the use of fecal microbial transplantation is now in the ascendant for IBD management, the prospects are still uncertain and the prevention and treatment of the recurrence of CDI in IBD remain a clinical challenge. Breast cancer is the most common form of cancer in women in need for better treatment. Sophoridine, a plant-derived quinoxidine alkaloid, has been shown to exhibit pharmacological effects on several diseases including lung and gastric cancers. To further extend these beneficial effects we have explored the role of sophoridine in breast cancer progression. Our results show that sophoridine suppressed the proliferation of breast cancer cells and inhibited the migration as well as invasion. In addition, sophoridine inhibited the angiogenesis of breast cancer cells. Mechanically, our data confirmed that sophoridine suppressed the vascular endothelial growth factor receptor 2/extracellular signal-regulated kinase 2 pathway, and therefore suppressed breast cancer progression. Copepods are commonly used as live feed for cultured fish larvae, but the current mass production method using organic fertilizers cannot meet the market demand for copepods. We evaluated the feasibility of applying an inorganic fertilization method, which is currently in use in freshwater and marine larviculture, to the mass production of copepods. For 30 days, and with five replicates of each treatment, we made comparative daily measurements of various parameters of (1) copepod cultures fertilized with commercially available condensed fish solubles (organic fertilization) and (2) other cultures in which the concentration of inorganic phosphorus was maintained at 100 μg P L−1 and that of inorganic nitrogen at 700 μg N L−1 (inorganic fertilization). With inorganic fertilization, pH fluctuated over a smaller range and much less filamentous algae grew in the tanks. The mean production of copepod nauplii over the course of the study was similar between the two treatments, but the combined density of copepodites and adult copepods was significantly higher with inorganic fertilization. Compared to commercial zooplankton products, copepods cultured with inorganic fertilization were smaller, were mixed with fewer (almost none) non-copepod contaminants, were also pathogen-free, and could be produced at the cheapest cost per unit output. Based on these results, we conclude that the inorganic fertilization method can profitably be adopted by commercial copepod producers to meet the demand from fish farmers, especially for small-sized copepods. Background: Neurodegenerative diseases are a group of unexplained disorders of the central nervous system, and studies have shown that a large number of genetic and environmental factors are associated with these diseases. Since these diseases show significant gender differences in epidemiology, sex hormones are thought to be strongly associated with these diseases. In this study, we used Mendelian randomization to explore the causal relationship between sex hormones and the risk of developing neurodegenerative diseases. Methods: We obtained genetic instrumental variables for sex hormones (sex hormone-binding globulin [SHBG], estradiol levels [EL], and bioavailable testosterone [BT]) separately through the Integrative Epidemiology Unit (IEU) database (https://gwas.mrcieu.ac.uk/). We analyzed the causal relationship of each with the risk of developing neurodegenerative diseases (Amyotrophic Lateral Sclerosis [ALS], Parkinson's disease [PD], and Alzheimer's disease [AD]) using inverse variance weighted (IVW) in Mendelian randomization. Data were then analyzed for sensitivity. Results: BT was negatively associated with the risk of developing ALS (odds ratio [OR] = 0.794; 95% confidence interval [95% CI] = 0.672-0.938; p = 0.006). EL and SHBG were not associated with a risk for developing neurodegenerative diseases (ALS, PD, AD). Conclusions: Elevated BT is associated with a reduced risk of developing ALS. Further research is needed to investigate the underlying mechanisms of action for this correlation and how it can be used as a potential target of action to reduce the risk of developing ALS. Pentraxin 3 (PTX3) is a soluble pattern recognition receptor playing an important role in immune response and inflammation. Lipopolysaccharide (LPS) stimulation can significantly induce PTX3 expression and secretion in adipocytes. Appropriate regulation of PTX3 secretion is critical for inflammatory homeostasis. Using chemical inhibitors of conventional and unconventional protein secretion, we explored the mechanisms that control LPS-stimulated PTX3 secretion in 3T3-L1 adipocytes. Inhibiting the conventional protein secretion blocked LPS-stimulated PTX3 secretion, resulting in cellular PTX3 accumulation in adipocytes. We also detected PTX3 in exosomes from LPS-treated adipocytes; inhibiting exosome trafficking attenuated PTX3 secretion. However, only 4.3% of secreted PTX3 was detected in exosomes compared to 95.7% in the non-exosomal fractions. The fractionation of isolated exosomes by the iodixanol density gradient centrifugation confirmed that a small portion of secreted PTX3 overlapped with exosomal markers in small extracellular-vesicle fractions. We conclude that PTX3 is secreted mainly through conventional protein secretion, and a small percentage of PTX3 is released in exosomes from LPS-stimulated adipocytes. [ABSTRACT FROM AUTHOR]

Gas insulated metal enclosed switchgear (GIS) has been widely used in the world because of its compact structure and adaption to the environment. However, due to frequent operation and other reasons, GIS disconnector is prone to mechanical failure with poor contact, leading to abnormal vibration signals. In this paper, a GIS disconnector contact fault simulation platform is built to collect vibration signals in three states: normal contact, moderate poor contact and severe poor contact of disconnector contact. The Ensemble Empirical Mode Decomposition (EEMD) method is used to decompose the vibration signals, and the IMF decomposition signals are screened by Kurtosis criterion (a numerical statistic method). Finally, the vibration signal features of GIS under different conditions are obtained by reconstructing the screening signals. The results indicate that, under the normal contact, the frequency of GIS vibration signal mainly presents the distribution of 100 Hz and its integer multiple frequency component, when there is a contact defect, the high frequency component in the vibration signal spectrum increases significantly, and the tripling frequency component is present, and the amplitude of component will increase with the severity of the fault. The analysis of vehicle speeds at the moment of traffic accidents is becoming increasingly important. To solve difficult calculations arising from video analysis when the shooting direction of the camera is the same as the driving direction of the vehicles, an improved plane homography method is proposed. This method is based on detecting the frame rate of and extracting all of the key image frames from a surveillance video. Next, a normalizing approach for the control points is proposed to reduce the ill-conditions of matrix calculations when the perspective image is rectified to an orthographic image. The information in the rectified image is then measured and recorded by extracting the mark point information. Finally, the corresponding curve of time and speed for the examined vehicle is created, which can be used to analyze the average braking deceleration of the accident-causing vehicle. This method can directly determine the speed of a vehicle using videos of a traffic accident without any reference to the dimensions of the vehicle itself. Considering an actual traffic accident as an example, the driving trajectories and velocity curves for two vehicles are obtained using this method. The core of the tumor microenvironment in the hematological system is formed by bone marrow stromal cells (BMSCs). In the present study, we explored the interaction between the urokinase plasminogen activator (uPA) system and the leukemia bone marrow microenvironment (BMM). We established BMSCs-HL60 and HS-5-K562 co-culture models in direct contact mode to simulate the BMM in leukemia. In BMSCs-HL60 co-culture model, the expression levels of uPA, uPA receptor (uPAR), plasminogen activator inhibitor 1 (PAI-1) and vascular endothelial growth factor (VEGF) in BMSCs were higher than those in mono-cultured BMSCs. Matrix metalloproteinase (MMP)-9 (MMP-9) was up-regulated in co-cultured HL60 cells. In HS-5-K562 co-culture model, only uPA, PAI-1, and VEGF-A were up-regulated in HS-5 cells. The levels of the uPA protein in the co-culture supernatant were significantly higher than that of mono-cultured BMSCs or HS-5 cells. Our findings demonstrate that the co-culture stimulates the production of uPA, uPAR, PAI-1, MMP-9, and VEGF-A by BMSCs. A switchable access to ethenylphosphorylamides and chloroethenylphosphorylamides from phosphorylamides and alkenes has been disclosed herein. Multiple myeloma (MM) accounts for over twenty percent of hematological cancer-related death worldwide. Long non-coding RNA (lncRNA) H19 is associated with multiple tumorigenesis and is increased in MM, but the underlying mechanism of H19 in MM is unclear. In this study, the expression of H19, miR-152-3p and BRD4 in MM patients was evaluated by qRT-PCR and Western blotting. Colony formation and flow cytometry analysis were used to determine the effects of H19 and miR-152-3p on MM cell proliferation, apoptosis and cell cycle. Luciferase reporter assay was conducted to confirm the interaction among H19, miR-152-3p and BRD4. Nude mice xenograft model was established, and the cell proliferation and apoptosis were evaluated by immunohistochemistry (IHC) staining and TUNEL assay. We found that levels of H19 and BRD4 were up-regulated, and the expression of miR-152-3p was down-regulated in MM patients. Dual luciferase reporter assay showed H19 targeted miR-152-3p to promote BRD4 expression. Knockdown of H19 repressed proliferation, and enhanced apoptosis and cell cycle G1 arrest by up-regulating miR-152-3p in MM cells. Furthermore, H19 knockdown suppressed the growth of xenograft tumor, and reduced Ki-67 and BRD4 level, as well as increased cell apoptosis in xenograft tumor tissues. Taken together, H19 knockdown suppresses MM tumorigenesis via inhibiting BRD4-mediated cell proliferation through targeting miR-152-3p, implying that H19 may be a promising biomarker and drug target for MM. The nucleus is enclosed by a double-membrane structure, the nuclear envelope, which separates the nucleoplasm from the cytoplasm. The outer nuclear membrane is continuous with the endoplasmic reticulum (ER), whereas the inner nuclear membrane (INM) is a specialized compartment with a unique proteome. In order to ensure compartmental homeostasis, INM-associated degradation (INMAD) is required for both protein quality control and regulated proteolysis of INM proteins. INMAD shares similarities with ER-associated degradation (ERAD). The mechanism of ERAD is well characterized, whereas the INMAD pathway requires further definition. Here we review the three different branches of INMAD, mediated by their respective E3 ubiquitin ligases: Doa10, Asi1-3, and APC/C. We clarify the distinction between ERAD and INMAD, their substrate recognition signals, and the subsequent processing by their respective degradation machineries. The removal of C2H2 and C2H6 from C2H4 streams is of great significance for feedstock purification to produce polyethylene and other commodity chemicals but the simultaneous adsorption of C2H6 and C2H2 over C2H4 from a ternary mixture has never been realized. Herein, a robust metal–organic framework, TJT‐100, was designed and synthesized, which demonstrates remarkably selective adsorption of C2H2 and C2H6 over C2H4. Breakthrough experiments show that TJT‐100 can be used as an adsorbent for high‐performance purification of C2H4 from a ternary mixture of C2H2/C2H4/C2H6 (0.5:99:0.5) to afford a C2H4 purity greater than 99.997 %, beyond that required for ethylene polymerization. Computational studies reveal that the uncoordinated carboxylate oxygen atoms and coordinated water molecules pointing towards the pore can trap C2H2 and C2H6 through the formation of multiple C−H⋅⋅⋅O electrostatic interactions, while the corresponding C2H4–framework interaction is unfavorable. A robust porous metal–organic framework was synthesized and utilized for the highly selective separation of C2H4 from a ternary mixture of C2 hydrocarbons. Abstract Two new luminescent Zn(II) metal–organic frameworks of [Zn(H 2 BPTC)](H 2 O) 2 (1) and [Zn 4 (H 2 BPTC) 2 (SO 4) 2 ][(CH 3) 2 NH 2 ] 4 (H 2 O) 4 (2) were constructed using zinc salts and 3,3′,5,5′‑biphenyltetracarboxylic acid (H 4 BPTC) as building blocks. The resulted MOFs were characterized by IR spectroscopy, thermogravimetric analysis and single crystal X-ray crystallography, and they exhibit different structural topologies: 1 presents a three dimensional (4,4)-connected network with {42.84} topology, while 2 can be described as a (4,6)-connected network with (32.62.72)(34.42.64.75) topology. These differences show that counter-anions play an important role in modulating structural and luminescent properties. Both 1 and 2 possess strong luminescence in suspensions, which can be selectively quenched by some specific nitroaromatics, Fe3+ ion and acetone. Graphical abstract Two luminescent Zn(II) MOFs have been synthesized through the strategy of anion-induced self-assembly of MOFs. Those two MOFs show different sensitivity and selectivity responses for NACs, Fe(III) ion, and toxic solvents. Lipocalin-2 (LCN2) has been previously characterized as an adipokine regulating thermogenic activation of brown adipose tissue and retinoic acid (RA)-induced thermogenesis in mice. The objective of this study was to explore the role and mechanism for LCN2 in the recruitment and retinoic acid-induced activation of brown-like or 'beige' adipocytes. We found LCN2 deficiency reduces key markers of thermogenesis including uncoupling protein-1 (UCP1) and peroxisome proliferator-activated receptor gamma coactivator 1α (PGC-1α) in inguinal white adipose tissue (iWAT) and inguinal adipocytes derived from Lcn2-/- mice. Lcn2-/- inguinal adipocytes have attenuated insulin-induced upregulation of thermogenic gene expression and p38 mitogen-activated protein kinase (p38MAPK) signaling pathway activation. This is accompanied by a lower basal and maximal oxidative capacity in Lcn2-/- inguinal adipocytes, indicating mitochondrial dysfunction. Recombinant LCN2 was able to restore insulin-induced p38MAPK phosphorylation in both WT and Lcn2-/- inguinal adipocytes. Rosiglitazone treatment during differentiation of Lcn2-/- adipocytes is able to recruit beige adipocytes at a normal level, however, further activation of beige adipocytes by insulin and RA is impaired in the absence of LCN2. Further, the synergistic effect of insulin and RA on UCP1 and PGC-1α expression is markedly reduced in Lcn2-/- inguinal adipocytes. Most intriguingly, LCN2 and the retinoic acid receptor-alpha (RAR-α) are concurrently translocated to the plasma membrane of adipocytes in response to insulin, and this insulin-induced RAR-α translocation is absent in adipocytes deficient in LCN2. The C/EBPα transcription factor is required for myelopoiesis, with prior observations suggesting additional contributions to B lymphopoiesis. Cebpa expression is evident in common lymphoid progenitor (CLP) and preproB cells but is absent in proB and preB cells. We previously observed that marrow lacking the Cebpa +37 kb enhancer is impaired in producing B cells upon competitive transplantation. Additionally, a Cebpa enhancer/promoter-hCD4 transgene is expressed in B/myeloid CFU. Extending these findings, pan-hematopoietic murine Cebpa enhancer deletion using Mx1-Cre leads to expanded CLP, fewer preproB cells, markedly reduced proB and preB cells, and reduced mature B cells, without affecting T cell numbers. In contrast, enhancer deletion at the proB stage using Mb1-Cre does not impair B cell maturation. Further evaluation of CLP reveals that the Cebpa transgene is expressed almost exclusively in Flt3+ multipotent CLP versus B cell-restricted Flt3- CLP. In vitro, hCD4+ preproB cells produce both B and myeloid cells, whereas hCD4- preproB cells only produce B cells. Additionally, a subset of hCD4- preproB cells express high levels of RAG1-GFP, as seen also in proB cells. Global gene expression analysis indicates that hCD4+ preproB cells express proliferative pathways, whereas B cell development and signal transduction pathways predominate in hCD4- preproB cells. This study investigated the capability of near-infrared spectroscopy (NIRS) to predict the concentration of Zygosaccharomyces rouxii in apple and kiwi fruit juices. The yeast was inoculated in fresh kiwi fruit juice (n = 68), reconstituted kiwi juice (n = 85), and reconstituted apple juice (n = 64), followed by NIR spectra collection and plate counting. A principal component analysis indicated direct orthogonal signal correction preprocessing was suitable to separate spectral samples. Parameter optimization algorithms increased the performance of support vector machine regression models developed in a single variety juice system and a multiple variety juice system. Single variety juice models achieved accurate prediction of Z. rouxii concentrations, with the limit of quantification at 3 to 15 CFU/mL (R2 = 0.997 to 0.999), and the method was also feasible for Hanseniaspora uvarum and Candida tropicalis. The best multiple variety juice model obtained had a limit of quantification of 237 CFU/mL (R2 = 0.961) for Z. rouxii. A Bland-Altman analysis indicated good agreement between the support vector machine regression model and the plate counting method. Cement stabilization is a widely applied technique in improving the workability and geotechnical properties of soft clay. This paper investigated the influence of cement stabilization on the soil–water characteristic curve (SWCC) and the deformation behavior of unsaturated cement–stabilized soft clay. The combined effect of cementation and partial saturation on the hydromechanical behavior of cement-stabilized soils was discussed. The SWCC of stabilized soil was measured in the full suction range under drying and wetting paths using the pressure plate method, the filter paper method, and the vapor equilibrium technique. The deformation behaviors of cement-stabilized soft soil were investigated by conducting suction-controlled oedometer tests. Furthermore, scanning electron microscope micrographs and mercury intrusion porosimetry tests were performed to investigate the effects of cement content on the evolution of soil pore structures. The test results showed that the water retention capacity of the stabilized soft clay increased with increasing cement content. The SWCC showed a significant hysteretic behavior in the boundary effect zone and transition zone, whereas the hysteretic effect was unapparent in the residual zone. The increased cement content had a more significant hysteretic effect on the SWCC. The deformation resistance capacity increased as the cement content and matric suction increased. However, the effect of matric suction on deformation resistance capacity decreased as the cement content increased. With the increase in cement content, the macropores were converted to small interaggregate pores, the pore spaces were distributed more uniformly, and the particle surface became rougher, resulting in higher water retention capacity and significant hysteresis of the SWCC. We theoretically study the influence of degree of polarization (DOP) of thermal light on the contrast-to-noise ratio (CNR) of the reflective ghost imaging (RGI), which is a novel and indirect imaging modality. An expression for the CNR of RGI with partially polarized thermal light is carefully derived, which suggests a weak dependence of CNR on the DOP, especially when the ratio of the object size to the speckle size of thermal light has a large value. Different from conventional imaging approaches, our work reveals that RGI is much more robust against the DOP of the light source, which thereby has advantages in practical applications, such as remote sensing. Background: Balanced translocations may cause the loss of genetic material at the breakpoints and may result in failure of spermatogenesis. However, carriers of reciprocal translocation may naturally conceive. Genetic counseling of male carriers of translocations remains challenging. This study explores the clinical features of carriers of chromosome 5 translocations, enabling informed genetic counseling of these patients. Results: Of 82 translocation carriers, 9 (11%) were carriers of a chromosome 5 translocation. One case had azoospermia, while three cases had experienced recurrent spontaneous abortions, two cases had each experienced stillbirth, and three cases produced a phenotypically normal child confirmed by amniocentesis. A literature review identified 106 patients who carried chromosome 5 translocations. The most common chromosome 5 translocation was t(4,5), observed in 13 patients. Breakpoint at 5p15 was observed in 11 patients. All breakpoints at chromosome 5 were associated with gestational infertility. Conclusion: In genetic counseling, physicians should consider chromosome 5 and its breakpoints. Taxol is currently a valuable anticancer drug; however, the accumulated mixture of taxoids can vary greatly among Taxus species. So far, there is very little genomic information for the genus Taxus, except for Taxus baccata. Transcriptome analysis is a powerful approach to explore the different regulatory mechanisms underlying the taxoid biosynthesis pathway in Taxus species. First, we quantified the variation in the taxoid contents between Taxus media and Taxus mairei. The contents of paclitaxel and 10- deacetylpaclitaxel in T. media are higher than that in T. mairei. Then, the transcriptome profiles of T. media and T. mairei were analyzed to investigate the altered expressions. A total of 20,704 significantly differentially expressed genes (DEGs), including 9865 unigenes predominantly expressed in T. media and 10,839 unigenes predominantly expressed in T. mairei, were identified. In total, 120 jasmonic acid-related DEGs were analyzed, suggesting variations in 'response to JA stimulus' and 'JA biosynthetic process' pathways between T. media and T. mairei. Furthermore, a number of genes related to the precursor supply, taxane skeleton formation and hydroxylation, and C13-side chain assembly were also identified. Grass carp (Ctenopharyngodon idellus) is an important worldwide commercial freshwater culture species. However, grass carp reovirus (GCRV) causes serious hemorrhagic disease in fingerlings and yearlings of fishes. To understand the molecular pathogenesis of host cells during GCRV infection, intensive proteomic quantification analysis of lysine acetylation in Ctenopharyngodon idella kidney (CIK) cells was performed. Using dimethylation labeling-based quantitative proteomics, 832 acetylated proteins with 1391 lysine acetylation sites were identified in response to GCRV infection, among which 792 proteins with 1323 sites were quantifiable. Bioinformatics analysis showed that differentially expressed lysine acetylated proteins are involved in diverse cellular processes and associated with multifarious functions, suggesting that extensive intracellular activities were changed upon viral infection. In addition, extensive alterations on host-protein interactions at the lysine acetylation level were also detected. Further biological experiments showed that the histone deacetylases (HDAC) inhibitor suberoylanilide hydroxamic acid (SAHA) could significantly suppress the GCRV replication. To our knowledge, this is the first to reveal the proteome-wide changes in host cell acetylome with aquatic virus infection. The results provided in this study laid a basis for further understanding the host response to aquareovirus infection in the post-translational modification aspect by regulating cell lysine acetylation conducive to viral replication. Taxus species are well-known for paclitaxel, which exhibits antitumor activities and is used for treating various cancers. Although most Taxus species are widespread in many areas, few studies have characterized the variation in metabolism among different Taxus species. Using an integrated approach involving 'tandem mass tag' labeling and liquid chromatography-tandem mass spectrometry (HPLC-MS), proteomes of T. media and T. mairei were investigated and 4078 proteins were quantified. Screening and classification of differentially expressed proteins revealed many metabolism-associated proteins. In detail, four enzymes involved in the flavonoid biosynthesis pathway were predominantly expressed in T. mairei. Four enzymes associated with supplying precursors for paclitaxel biosynthesis and three cytochrome P450 taxoid oxygenases were preferentially expressed in T. media compared with T. mairei. Furthermore, variations in taxoid contents between T. media and T. mairei were determined using HPLC-MS analysis. Variations in flavonoid contents between T. media and T. mairei were determined by HPLC analysis. The present study proposed a modified decision-making trial and evaluation laboratory (DEMATEL) method. This innovation method involves collecting the repeated or identically defined technical keywords of patent techniques related to light-emitting diode (LED) bicycle light to determine the ratios of the normalized numerical values of these technical keywords by using one technical domain as the primary domain and another as a variable. The values obtained are then converted to mutual influence levels on a scale of 0 to 4, replacing the conventional expert questionnaire. In this study, in accordance with the operational steps of the decision-making trial and evaluation laboratory method, a general relational influence matrix, direct and indirect relationships diagram, and values of centrality (D + R) and causality (D − R) were obtained. A causal diagram was therefore created. The causal diagram was drawn using values of (D + R) and (D − R) as the two axes and facilitated determining the levels of mutual influence between technical domains. In accordance with the proposed modified decision-making trial and evaluation laboratory method, this study collected patents related to LED bicycle light; moreover, the normalized numerical values of key technical, part/component, and function words that appeared in these patents were calculated. Furthermore, clusters of technical and part or component words were defined in accordance with the first-layer technical category. The second-layer technical categories and functional categories were subsequently defined under the first-layer technical categories to establish the technique–function matrix, thereby dividing the techniques related to LED bicycle light into seven main technical domains. This study then analyzed patent life span. Patent life span was calculated using the announcement date of related patents. Finally, this study investigated the development potential of each technical domain of LED bicycle light by conducting a combined analysis of causal diagram obtained by modified DEMATEL method, activity trend chart of techniques, and patent life spans. The soluble urokinase‑type plasminogen activator receptor (suPAR) and the urokinase‑type plasminogen activator receptor (uPAR) have been proposed as useful biomarkers of tumor progression. Recently, suPAR was associated with chemoresistance in lung cancer. However, its clinical significance in leukemia has not previously been investigated. The present study examined the plasma levels of suPAR and the expression of the uPAR on bone marrow (BM) cells in 86 patients with leukemia at diagnosis prior to chemotherapy and 26 normal subjects (control group). The plasma suPAR levels were measured using ELISA, whilst uPAR expression was assayed by flow cytometry analysis. In addition, cell surface uPAR expression on K562 and multidrug‑resistant K562/ADM cell lines was studied by western blotting. On admission and follow-up, the levels of suPAR in patients with leukemia were significantly increased compared with controls. Systemic levels of suPAR were strongly associated with the numbers of white blood cells. A case was defined as uPAR‑positive (uPAR+) if >20% of the gated cells expressed uPAR. In comparison with 26 healthy BM samples that were negative for uPAR expression, 48 (55.8%) of the 86 leukemia patients were uPAR+. uPAR expression on the cell surface of multidrug‑resistant K562/ADM cells was increased compared with that on K562 cells. In conclusion, plasma suPAR expression may be a useful marker for subtype classification of patients with leukemia and cell surface uPAR may be associated with resistance to chemotherapy or disease progression. Objective: Secreted frizzled-related proteins (SFRPs) as Wnt signaling antagonists have been found to be dysregulated by promoter hypermethylation in several cancers including acute myeloid leukemia (AML). This study aimed to investigate the methylated status of SFRPs promoter region and its clinical relevance in Chinese non-M3 AML patients. Methods: SFRPs methylation in 139 primary non-M3 AML patients was determined using methylation-specific real-time quantitative polymerase chain reaction. Results: The frequency of aberrant methylation was as follows: 30.2% for SFRP1, 27.3% for SFRP2, 5.0% for SFRP4, and 1.4% for SFRP5. Hypermethylation of at least one SFRP gene occurred in 51.8% (72/139) of non-M3 AML patient samples, which was significantly higher compared to normal control (0/21) (P<0.001). Hypermethylation of SFRP1 was potentially associated with N/K-RAS mutations (P=0.043), and the frequency of SFRPs methylation was higher in patients ≥50 years compared to those <50 years, especially for SFRP2 (P<0.05). Furthermore, both whole cohort and cytogenetically normal (CN) patients with high SFRPsmethylated group showed a shorter overall survival (OS) compared to those with low group (P=0.036 and P=0.035, respectively). Moreover, Cox regression multivariate analysis revealed that SFRPs hypermethylation acts as an independent prognostic biomarker among both whole cohort (hazard ratio =1.804, P=0.026) and CN (hazard ratio =2.477, P=0.023) patients. In leukemic cell line HL60 treated with 5-aza-2'-deoxycytidine, the alteration of SFRP1/2 expression inversely correlated with change in SFRP1/2 methylation (r=-0.975, P=0.005 and r=-0.975, P=0.005, respectively). A tendency of negative correlation was observed between SFRP1 expression and its promoter methylation in AML patients (r=-0.334, P=0.038). Conclusion: These findings suggested that hypermethylation of SFRP1/2 was a frequent event and silenced SFRP1/2 expression in AML. Supporters of net neutrality have often argued that more competition among Internet service providers (ISPs) is beneficial for an open Internet and that the market power of the ISPs lies at the heart of the net neutrality debate. However, the joint effects of the competition among ISPs and among content providers (CPs) have yet to be examined. We study the critical linkage between ISP competition and CP competition, as well as its policy implications. We find that even under competitive pressure from a rival ISP, an ISP always has the incentive and the ability to enforce charging CPs for priority delivery of content. Upending the commonly held belief that when facing direct competition CPs will always support the preservation of net neutrality, we find that, under certain conditions, it is economically beneficial for the dominant CP to reverse its stance on net neutrality. Bogie is the most important component in a running gear of a high-speed train. Advantages and disadvantages of its mechanical performance can directly influence safety and comfort of train running. Failure of key components on the bogie always leads to serious vibration and performance decrease of each train component, and can even cause serious accidents such as de-railing and overturning. Therefore, it is very necessary to adopt an efficient and accurate intelligent diagnosis method. Based on the previous researches, the paper proposed a deep neural network method to make systematic and complete diagnosis of bogie faults. Firstly, a deep neural network was used to diagnose standard bearing system faults, and the results were compared with those obtained by traditional neural networks such as FANN (Firefly Artificial Neural Network), PSONN (Particle Swarm Optimization Neural Network) and GANN (Genetic Artificial Neural Network) to highlight advantages of the deep neural network. Then, based on vibration data extracted by a multi-body dynamic model of the high-speed train, the deep neural network was used to diagnose 8 conditions of the bogie systematically. The fault diagnosis was repeated for 10 times. Results obtained for the 10 times present obvious advantages of the deep neural network which could obtain the average diagnosis rate of 98.3 % and had the diagnosis standard deviation of 0.71. Finally, training process of fault diagnosis was extracted. Results show that the deep neural network could converge to a critical value at the quickest speed. The present study investigated the anti-aging effects of melatonin on the myocardial mitochondria of D-galactose-aged rats and associated mechanisms. A total of 30 male Sprague-Dawley (SD) rats were randomly divided into three equal groups: An accelerated aging group that received 125 mg/kg/day D-galactose; a melatonin-treated group of D-galactose-aged rats that received 10 mg/kg/day melatonin; and a control group receiving normal saline. ATP, ADP and AMP levels in the left ventricular myocardium of rats were determined by high performance liquid chromatography and the total adenylic acid number (TAN) was subsequently calculated. Bax, Bcl-2, and cytochrome c (cyt-c) protein expression levels in myocardial mitochondria and cytoplasm were quantified by western blot analysis. In the melatonin-treated group, ATP levels were significantly higher when compared with the untreated control group and the accelerated-ageing group (0.068 vs. 0.052 and 0.058; P=0.002 and P=0.045, respectively), and TAN was significantly increased in the melatonin-treated group when compared with controls (P=0.011). In addition, cyt-c levels in the cytoplasm, but not in the mitochondria, were significantly higher in the accelerated-aging group compared with the control and melatonin-treated groups (P=0.001 and P=0.002, respectively). Bcl-2 and Bax ratios were significantly higher in the control and melatonin-treated groups when compared with the accelerated-aging group (P=0.004 and P=0.032, respectively). The role of long non-coding RNAs (lncRNAs) in atrial fibrillation remains to be fully elucidated. The current study performed microarray analysis to investigate differential lncRNA expression profiling in atrial samples from the pulmonary vein and the surrounding left atrial area (LA-PV) and from the left atrial appendage (LAA) in 16 patients with atrial fibrillation (AF). Microarray analysis identified 94 lncRNAs that were differentially expressed between the LA-PV and LAA in patients with AF. AK055347 was one of lncRNAs with the most significant alterations observed. Knockdown of AK055347 inhibited cell viability of H9C2 cardiomyocytes, accompanied by downregulation of Cyp450 and ATP synthases. In addition, microarray analysis identified that MSS51 was a target of AK055347. Knockdown of AK055347 inhibited the expression of MSS51 in H9C2 cells. The observations of the current study suggest that lncRNAs are differentially expressed in the LA-PV and LAA in patients with AF. Vertical jet grouted columns by drilling holes through the embankment of busy highways are applied to mitigate the problem of excessive postconstruction settlement on the east coast of China. Short-term ground heave due to vertical jet grouted columns is focused, investigated, and analyzed via field monitoring of a construction project in Lianyungang, China. Field results show that heaves in ground surfaces were observed to be 219 mm at most. Theoretical analysis by cavity expansion was performed to investigate the soil displacement due to vertical jet grouted columns. It was found that ground surface heave decreased with smaller grout pressure, larger geostatic pressure, and a larger distance from the column center. Intrauterine adhesion (IUA), also known as Asherman's syndrome, is a common disease for among women. The extent of adhesion and pre-surgery hormone therapy greatly affects the function of uterine cavity. This current study investigates the association of different doses of estrogen before transcervical resection of adhesions (TCRA) surgery and clinical outcome in serious IUA. About 120 newly diagnostic serious IUA patients who underwent TCRA were randomly divided into three study groups: Estradiol valerate (progynova) 3 or 9 mg per diet before surgery and the control group. Follow-up hysteroscopy checkups were taken in 1- and 3-month post-operation. The effective power of 9 mg group was significantly higher than other groups. The 9 mg group achieved the best menstrual recovery rate in all study groups compared with the other two groups in 6 months post-operatively (p < 0.05). Introduction: The treatment of subclinical hypothyroidism in elderly persons is controversial. Previous studies have shown that there are interactions between kidney and thyroid function, but data regarding interventions that target thyroid function in elderly patients are scarce. We aimed to investigate the impact of thyroid hormone therapy on the estimated glomerular filtration rate (eGFR) in elderly patients.Material and Methods: Ninety elderly patients aged ≥ 65 years with subclinical hypothyroidism were followed for 36 months in our case-control study. The changes in the eGFR in patients with and without thyroid hormone replacement therapy were compared. The adverse effects during the treatment period were noted.Results: The eGFR of both groups was similar at the beginning of the study (43.4 ±6.1 vs. 42.8 ±5.9 ml/min/1.73 m(2); p = 0.62). With the decline in thyroid stimulating hormone levels after treatment, the eGFR of the treatment group significantly improved compared with the control group (45.8 ±4.8 vs. 35.8 ±5.3 ml/min/1.73 m(2); p < 0.001); the eGFR increased rapidly over the first 6 months and then plateaued. No patients withdrew from the study, but the therapeutic dose was decreased in two patients due to angina pectoris.Conclusions: Thyroid hormone therapy preserved renal function in elderly patients. We model the luminosity-dependent projected and redshift-space two-point correlation functions (2PCFs) of the Sloan Digital Sky Survey (SDSS) Data Release 7 Main galaxy sample, using the halo occupation distribution (HOD) model and the subhalo abundance matching (SHAM) model and its extension. All the models are built on the same high-resolution Nbody simulations. We find that the HOD model generally provides the best performance in reproducing the clustering measurements in both projected and redshift spaces. The SHAM model with the same halo-galaxy relation for central and satellite galaxies (or distinct haloes and subhaloes), when including scatters, has a best-fitting X2 around 2-3. We therefore extend the SHAM model to the subhalo clustering and abundance matching (SCAM) by allowing the central and satellite galaxies to have different galaxy-halo relations. We infer the corresponding halo/subhalo parameters by jointly fitting the galaxy 2PCFs and abundances and consider subhaloes selected based on three properties, the mass Macc at the time of accretion, the maximum circular velocity Vacc at the time of accretion, and the peak maximum circular velocity Vpeak over the history of the subhaloes. The three subhalo models work well for luminous galaxy samples (with luminosity above L*). For low-luminosity samples, the Vacc model stands out in reproducing the data, with the Vpeak model slightly worse, while the Macc model fails to fit the data. Small- and intermediate-scale galaxy clustering can be used to establish the galaxy-halo connection to study galaxy formation and evolution and to tighten constraints on cosmological parameters. With the increasing precision of galaxy clustering measurements from ongoing and forthcoming large galaxy surveys, accurate models are required to interpret the data and extract relevant information. We introduce a method based on high-resolution N-body simulations to accurately and efficiently model the galaxy two-point correlation functions (2PCFs) in projected and redshift spaces. The basic idea is to tabulate all information of haloes in the simulations necessary for computing the galaxy 2PCFs within the framework of halo occupation distribution or conditional luminosity function. It is equivalent to populating galaxies to dark matter haloes and using the mock 2PCF measurements as the model predictions. Besides the accurate 2PCF calculations, the method is also fast and therefore enables an efficient exploration of the parameter space. As an example of the method, we decompose the redshiftspace galaxy 2PCF into different components based on the type of galaxy pairs and show the redshift-space distortion effect in each component. We have recently characterized the role of lipocalin 2 (Lcn2) as a new adipose-derived cytokine in the regulation of adaptive thermogenesis via a non-adrenergic pathway. Herein, we explored a potential non-adrenergic mechanism by which Lcn2 regulates thermogenesis and lipid metabolism. We found that Lcn2 is a retinoic acid target gene, and retinoic acid concurrently stimulated UCP1 and Lcn2 expression in adipocytes. Lcn2 KO mice exhibited a blunted effect of all-trans-retinoic acid (ATRA) on body weight and fat mass, lipid metabolism, and retinoic acid signaling pathway activation in adipose tissue under the high fat diet-induced obese condition. We further demonstrated that Lcn2 is required for the full action of ATRA on the induction of UCP1 and PGC-1α, expression in brown adipocytes and the restoration of cold intolerance in Lcn2 KO mice. Interestingly, we discovered that Lcn2 KO mice have decreased levels of retinoic acid and retinol in adipose tissue. The protein levels of STRA6 responsible for retinol uptake were significantly decreased in adipose tissue. The retinol transporter RBP4 was increased in adipose tissue but decreased in the circulation, suggesting the impairment of RBP4 secretion in Lcn2 KO adipose tissue. Moreover, Lcn2 deficiency abolished the ATRA effect on RBP4 expression in adipocytes. All the data suggest that the decreased retinoid level and action are associated with impaired retinol transport and storage in adipose tissue in Lcn2KOmice. Herein, we reported for the first time that gold nanoparticles-catalyzed 3,3′,5,5′-tetramethylbenzidine-H 2 O 2 system can serve as an ultrasensitive colorimetric pH indicator. Gold nanoparticles acted as a catalyst and imitated the function of horseradish peroxidase. The absorbance at 450 nm of the yellow-color product in the catalytic reaction exhibited a linear fashion over the pH range of 6.40–6.60. On the basis of this property, we constructed a novel sensing platform for the determination of urea, urease, and urease inhibitor. The limit of detection for urea and urease was 5 μM and 1.8 U/L, respectively. The half-maximal inhibition value IC 50 of acetohydroxamic acid was found to be 0.05 mM. Two new flavonoids, 1 and 2, together with two known flavonoids, tephrosin ( 3) and 12a-hydroxy- α-toxicarol ( 4), were isolated from the whole herb of Derris eriocarpa How. The structures and absolute configurations of the new compounds were elucidated on the basis of their MS, NMR, and ECD data. The structures of the known compounds were established by extensive spectroscopic ( MS, 1D- and 2D- NMR) analyses and comparison with the literature data. All compounds were isolated from D. eriocarpa for the first time. Compound 3 showed modest inhibitory activities against the growth of human cancer cells HEL and A549 with the IC50 values of 15.03 ± 0.62 and 13.27 ± 0.39 μ m, respectively. [ABSTRACT FROM AUTHOR]

We describe here a fluorometric assay for sensitive detection of oligonucleotides, based on a target recycling amplification strategy driven by toehold-mediated strand displacement reaction and on exonuclease III (Exo Ш)-assisted fluorescence background suppression strategy. The network consists of a pair of partially complementary DNA hairpins (HP 1 and HP 2 ) with 3′ overhang ends, between which the spontaneous hybridization is kinetically hindered by the stems. The target DNA is repeatedly used to trigger a recycling progress between the hairpins, generating numerous HP 1 –HP 2 duplex complexes. Exo III was then employed to digest the double strand parts of the residual hairpins and the intermediate products. The fluorescent dye, SYBR Green I, binds to the double-strand DNA products and emits strong fluorescence to achieve sensitive detection of the target DNA with the detection limit of 5.34 pM. Moreover, this proposed strategy showed high discrimination efficiency towards target DNA against mismatched DNA and was successfully applied in the analysis of human serum sample. Inspired by dual-signaling ratiometric mechanism which could reduce the influence of the environmental change, a novel, convenient, and reliable method for the detection of mercury ions (Hg 2+ ) based on Y-shaped DNA (Y-DNA) was developed. Firstly, the Y-DNA was formed via the simple annealing way of using two different redox probes simultaneously, omitting the multiple operation steps on the electrode. The Y-DNA was immobilized on the gold electrode surface and then an obvious ferrocene (Fc) signal and a weak methylene blue (MB) signal were observed. Upon addition of Hg 2+ , the Y-DNA structure was transformed to hairpin structure based on the formation of T-Hg 2+ -T complex. During the transformation, the redox MB gets close to and the redox Fc gets far away from the electrode surface, respectively. This special design allows a reliable Hg 2+ detection with a detection range from 1 nM to 5 μM and a low detection limit down to 0.094 nM. Furthermore, this biosensor exhibits good selectivity and repeatability, and can be easily regenerated by using l -cysteine. The influence of detector response speed on the contrast-to-noise ratio (CNR) of reflective ghost imaging (RGI) is studied. To mimic the situation of a slow response detector, the illuminating speckle patterns are replaced with the sum of uncorrelated speckle patterns for each measurement. An expression for the CNR of RGI with added speckle patterns is derived. By employing a light projector to provide spatially incoherent structured illumination in a computational ghost imaging system, we perform computational RGI based on added speckle patterns. The experimental results show that when up to 40 uncorrelated speckle patterns are added together in each measurement, the CNR of computational RGI obtained from 5000 effective measurements remains almost the same as the conventional computational ghost imaging. The reason for this is that the image quality of RGI depends on the kurtosis of intensity fluctuation of speckle field instead of the contrast ratio of speckle pattern. We present the measurements and modelling of the small-to-intermediate scale (~0.1-25 h-1 Mpc) projected and three-dimensional redshift-space two-point correlation functions (2PCFs) of local galaxies in the Sloan Digital Sky Survey Data Release 7. We find a clear dependence of galaxy clustering on luminosity in both projected and redshift spaces, generally being stronger for more luminous samples. The measurements are successfully interpreted within the halo occupation distribution (HOD) framework with central and satellite velocity bias parameters to describe galaxy kinematics inside haloes and to model redshift-space distortion effects. In agreement with previous studies, we find that more luminous galaxies reside in more massive haloes. Including the redshift-space 2PCFs helps tighten the HOD constraints. Moreover, we find that luminous central galaxies are not at rest at the halo centres, with the velocity dispersion about 30 per cent that of the dark matter. Such a relative motion may reflect the consequence of galaxy and halo mergers, and we find that central galaxies in lower mass haloes tend to be more relaxed with respect to their host haloes. The motion of satellite galaxies in luminous samples is consistent with their following that of the dark matter. For faint samples, satellites tends to have slower motion, with velocity dispersion inside haloes about 85 per cent that of the dark matter. We discuss possible applications of the velocity bias constraints on studying galaxy evolution and cosmology. A complete set of diabolo optical antenna arrays with different waist widths and periods was fabricated on a sapphire substrate by using a standard e-beam lithography and lift-off process. Fabricated diabolo optical antenna arrays were characterized by measuring the transmittance and reflectance with a microscope-coupled FTIR spectrometer. It was found experimentally that reducing the waist width significantly shifts the resonance to longer wavelength and narrowing the waist of the antennas is more effective than increasing the period of the array for tuning the resonance wavelength. Also it is found that the magnetic field enhancement near the antenna waist is correlated to the shift of the resonance wavelength. [ABSTRACT FROM AUTHOR]
