In the context of global climate change, recent studies indicate a poleward migration trend of tropical cyclones (TCs) in the western North Pacific (WNP), while little attention has been paid to the TC cyclolysis (hereafter Lysis). With respect to two different datasets, this study identifies the poleward migration of the annual-mean latitude of TC Lysis during 1979-2018, being more significant for the intensified TCs, although this trend is suppressed by the reduction in the TC frequency over the sea. It is found that the TC migration is more like a poleward translation of the overall movement rather than the expansion of a specific phase's distance span. Subsequently, the trends of several environmental parameters related to TC development are also analyzed. The large-scale sea surface temperature warming leads to the increase of potential intensity and enhances the possibility of TC poleward migration. Through controlling the TC formation in the eastern WNP tropics, the variations of vertical wind shear and horizontal winds affect TC Lysis latitude, facilitate the TC development environment around East Asia offshore and island chain areas, and steer TCs poleward migration through the southerly wind anomalies in the area north of 308N. Regarding the cyclonic vorticity in the lower troposphere and the divergence in the upper troposphere, their influence on TC Lysis latitude is mainly by adjusting the numbers of TCs rather than directly interfering with the TC movement process. This paper studies the consensus problem of heterogeneous multi-agent systems (MASs) composed of first-order and second-order agents. To describe the communication environment realistically, the effect of the multiplicative noise on the communication network is considered. Firstly, the consensus problem is transformed into the stability problem of stochastic differential equations with multiplicative noise. Then, a stability analysis method for such stochastic differential equations is proposed by structuring the appropriate Lyapunov function. Some sufficient conditions for both mean square (m.s.) and almost sure (a.s.) consensus are obtained for leader-free/leader-following heterogeneous MASs. These sufficient conditions, expressed by simple scalar inequalities, are explicitly related to control gains, noise intensities, and network topology. We derived an analytical expression for a resonant-mode based bi-layered cantilever with distributed mass load. The behavior of mode of vibration, nodal position, frequency shift, as well as sensitivity under different mass load distributions was theoretically studied. The theoretical results suggested that asymmetric mass load distribution leads to the shift of nodes as well as the sensitive regions of a resonant-mode based cantilever. n – 1 local maximal sensitivities and n – 1 local minimal sensitivities are observed when the cantilever vibrates in the nth-order resonance. The maximal sensitivity is found at the first local maximal sensitivity and the behavior of mass load length as a function of the maximal sensitivity follows the rule of an exponent decaying function. In this paper, we presented a theoretical method for studying the behavior of magnetic field intensity and eddy current inside a magnetostriction based bilayered composite. Firstly, the mathematical model for the electromagnetic field in the composite was established. Then, the governing equation for determining the magnetic field intensity and eddy current was solved. Furthermore, the effect of the composite's conductivity on the magnetic field intensity and eddy current were discussed. Lastly, by comparing with the well known R.L. We introduce Lipschitz continuous and C1,1 geometric approximation and interpolation methods for sampled bounded uniformly continuous functions over compact sets and over complements of bounded open sets in Rn by using compensated convex transforms. Error estimates are provided for the approximations of bounded uniformly continuous functions, of Lipschitz functions, and of C1,1 functions. We also prove that our approximation methods, which are differentiation and integration free and not sensitive to sample type, are stable with respect to the Hausdorff distance between samples. In this work, we propose a novel wireless actuator which is composed of magnetostrictive material/copper bi-layer film. The actuator can be controlled to move like a snake bi-directionally along a pipe by tuning the frequency of external magnetic field near its first order resonant frequency. The governing equation for the actuator is established and the vibration mode shape function is derived. Theoretical analysis shows that motion of the actuator is achieved by asymmetric vibration mode shape, specific vibration bending deformation, and effective net total impacting force. The simulation and experimental results well confirm the theoretical analysis. This work provides contribution to the development of wireless micro robots and autonomous magnetostrictive sensors. Background: The low-phosphate-tolerant maize mutant Qi319-96 was obtained from Qi319 through cellular engineering. To elucidate the molecular mechanisms underlying the low-phosphate tolerance of this mutant, we performed comparative proteome analyses of the leaves of Qi319-96 and Qi319 under inorganic phosphate (Pi)-sufficient and Pi-deficient conditions. Results: Low-phosphorus levels limit plant growth and metabolism. Although the overall phosphorus contents of shoots were not significantly different between Qi319 and Qi319-96, the Pi level of Qi319-96 was 52.94 % higher than that of Qi319. Under low phosphorus conditions, Qi319-96 had increased chlorophyll levels and enhanced photosynthesis. The changes in starch and sucrose contents under these conditions also differed between genotypes. The proteomic changes included 29 (Pi-sufficient) and 71 (Pi-deficient) differentially expressed proteins involved in numerous metabolic processes. Proteome and physiological analyses revealed that Qi319-96 could better remodel the lipid composition of membranes and had higher V-ATPase activity levels than Qi319 under low-phosphate starvation, which enhanced the recycling of intracellular Pi, as reflected by its increased Pi levels. Chlorophyll biosynthesis was improved and the levels, and activities, of several Calvin cycle and "CO2 pump" enzymes were greater in Qi319-96 than in Qi319, which led to a higher rate of photosynthesis under low-phosphate stress in this line compared with in Qi319. Conclusions: Our results suggest that the increased tolerance of the maize mutant Qi319-96 to low-phosphate levels is owing to its ability to increase Pi availability. Resonant-mode based cantilevers are an important type of acoustic wave based masssensing devices. In this work, the governing vibration equation of a bi-layer resonantmode based cantilever attached with concentrated mass is established by using a modal analysis method. The effects of resonance modes and mass loading conditions on nodes and mass sensitivity of the cantilever were theoretically studied. The results suggested that the node did not shift when concentrated mass was loaded on a specific position. Mass sensitivity of the cantilever was linearly proportional to the square of the point displacement at the mass loading position for all the resonance modes. For the first resonance mode, when mass loading position xc satisfied 0 < xc < ~0.3l (l is the cantilever beam length and 0 represents the rigid end), mass sensitivity decreased as the mass increasing while the opposite trend was obtained when mass loading satisfied ~0.3l ≤ xc ≤ l. Mass sensitivity did not change when concentrated mass was loaded at the rigid end. This work can provide scientific guidance to optimize the mass sensitivity of a resonant-mode based cantilever. In this paper we introduce a new stable mathematical model for locating and measuring the medial axis of geometric objects, called the quadratic multiscale medial axis map of scale λ, and provide a sharp regularity result for the squared-distance function to any closed nonempty subset K of Rn. Our results exploit properties of the function Cλl (dist2(.; K)) obtained by applying the quadratic lower compensated convex transform of parameter λ [K. Zhang, Ann. Inst. H. Poincaré Anal. Non Linéaire, 25 (2008), pp. 743-771] to dist2(.; K), the Euclidean squared-distance function to K. Using a quantitative estimate for the tight approximation of dist2(.; K) by Cλl (dist2(.; K)), we prove the C1,1-regularity of dist2(.; K) outside a neighborhood of the closure of the medial axis MK of K, which can be viewed as a weak Lusin-type theorem for dist2(.; K), and give an asymptotic expansion formula for Cλl (dist2(.; K)) in terms of the scaled squared-distance transform to the set and to the convex hull of the set of points that realize the minimum distance to K. The multiscale medial axis map, denoted by Mλ(.; K), is a family of nonnegative functions, parametrized by λ > 0, whose limit as λ→ ∞exists and is called the multiscale medial axis landscape map, M∞(.; K). We show that M∞(.; K) is strictly positive on the medial axis MK and zero elsewhere. We give conditions that ensure Mλ(.; K) keeps a constant height along the parts of MK generated by two-point subsets with the value of the height dependent on the scale of the distance between the generating points, thus providing a hierarchy of heights (hence, the word "multiscale") between different parts of MK that enables subsets of MK to be selected by simple thresholding. Asymptotically, further understanding of the multiscale effect is provided by our exact representation of M∞(.; K). Moreover, given a compact subset K of Rn, while it is well known that MK is not Hausdorff stable, we prove that in contrast, Mλ(.; K) is stable under the Hausdorff distance, and deduce implications for the localization of the stable parts of MK. It is introduced that the mass sensitivity (Sm) of an acoustic wave (AW) device with a concentrated mass can be simply determined using its mode shape function: the Sm is proportional to the square of its mode shape. By using the Sm of an AW device with a uniform mass, which is known for almost all AW devices, the Sm of an AW device with a concentrated mass at different locations can be determined. Magnetoelastic sensors as an important type of acoustic wave sensors have shown great promise for a variety of applications. Mass sensitivity is a key parameter to characterize its performance. In this work, the effects of mass load distribution on the mass sensitivity of a magnetoelastic sensor under different resonance modes were theoretically investigated using the modal analysis method. The results show that the mass sensitivity and "nodal point" positions are related to the point displacement, which is determined by the motion patterns. The motion patterns are affected by resonance modes and mass load distribution. Asymmetrical mass load distribution causes the motion patterns lose symmetry and leads to the shift of "nodal point". The mass sensitivity changing with mass load distribution behaves like a sine wave with decaying amplitude and the minimum mass sensitivity appears at the first valley. This study provides certain theoretical guidance for optimizing the mass sensitivity of a magnetoelastic sensor or other acoustic wave based sensors. [ABSTRACT FROM AUTHOR]

Three different morphologies of tungsten oxides, nanoparticles, nanosheets and hierarchical microspheres, have been successfully synthesized by a facile carboxyl-directed hydrothermal process. The chelation of carboxylic groups with W(OH)6 nuclei is recognized to be the origin of the morphological change. Gas-sensing measurements reveal that the sensing performance varies with WO3 morphology, and the hierarchical WO3 not only exhibits high sensitivity and fast response but also has low operating temperature to toxic NO2. The response of hierarchical WO3 is nearly 2 times and 10 times higher than those of the nanosheets and nanoparticles, respectively. The maximum response of hierarchical WO3 reaches 319 to 10 ppm NO2 at 200 °C. A relationship between morphology and crystal defect is established based on photoluminescence analysis. It is demonstrated that the change in defect feature in crystalline WO3 is responsible for its morphology-dependent gas-sensing properties. [ABSTRACT FROM AUTHOR]

The existence of "blind point" lowers the mass sensitivity and reliability of magnetostrictive particle (MSP) based biosensors. In addition, asymmetric distribution of mass loading (e.g. bacteria) will cause the shift of "blind point" and change of mass sensitivity of an MSP based biosensor. In this work, a modal analysis method was introduced and conducted to derive the governing vibration equation for an MSP biosensor with asymmetric mass loading. The effects of asymmetric mass loading on the "blind point" shift and mass sensitivity were unveiled by solving the governing vibration equation. It was found that the plot of "blind point" shift showed an "M" shape with the valley located at a/l ~ 0.5 and two peaks located at a/l ~ 0.25 and a/l ~ 0.75. In addition, the theoretical results showed that the resonance frequency shift (Δf) depends on the value of a/l (i.e. both the attached mass and mass distribution) and their relationship is nonlinear. Based on the definition of mass sensitivity (Sm), the values of Sm were derived for the selected values of a/l. The results showed that Sm decreased initially with increasing a/l, reaching the minimum, and then increased until a/l = 1. That is, the maximum mass sensitivity is obtained when a/l equals to a critical value Xc (0 < Xc < l). Based on the result, an optimal mass loading distribution design criterion for developing a more sensitive MSP based biosensor was proposed. [ABSTRACT FROM AUTHOR]

Zero-dimensional Fe2O3 nanoparticles were successfully decorated on three-dimensional WO3 architectures to constitute a photocatalyst of Fe2O3@WO3 heterojunction. The obtained samples were characterized in detail by X-ray diffraction, scanning electron microscopy, elemental mapping, X-ray photoelectron spectroscopy and UV-Vis absorption spectra. The results indicate that rhombohedral α-Fe2O3 nanoparticles are homogeneously decorated on the surface of monoclinic WO3 architectures, and the constituted n+-n heterojunction results in "redshift" of the optical absorption. The photocatalyst of 1%Fe2O3@WO3 annealed at 400 °C exhibits the highest photocatalytic activity for degradation of Rhodamine B under visible light irradiation. The degradation obeys first-order reaction kinetics with an apparent rate constant of 0.057 min-1. It is suggested that the potential-energy difference between Fe2O3 and WO3 accelerates the separation of photogenerated electron-hole pairs, dominating the enhanced photocatalytic activity. The plant hormone salicylic acid (SA) plays critical roles in plant defense, stress responses, and senescence. Although SA biosynthesis is well understood, the pathways by which SA is catabolized remain elusive. Here we report the identification and characterization of an SA 3-hydroxylase (S3H) involved in SA catabolism during leaf senescence. S3H is associated with senescence and is inducible by SA and is thus a key part of a negative feedback regulation system of SA levels during senescence. The enzyme converts SA (with a Km of 58.29 µM) to both 2,3-dihydroxybenzoic acid (2,3-DHBA) and 2,5-DHBA in vitro but only 2,3-DHBA in vivo. The s3h knockout mutants fail to produce 2,3-DHBA sugar conjugates, accumulate very high levels of SA and its sugar conjugates, and exhibit a precocious senescence phenotype. Conversely, the gain-of-function lines contain high levels of 2,3-DHBA sugar conjugates and extremely low levels of SA and its sugar conjugates and display a significantly extended leaf longevity. This research reveals an elegant SA catabolic mechanism by which plants regulate SA levels by converting it to 2,3-DHBA to prevent SA overaccumulation. AtNAP is a NAC family transcription factor gene that plays a key role in leaf senescence but its underlying mechanisms are not known. SENESCENCE-ASSOCIATED GENE113 (SAG113), a gene encoding a Golgi-localized protein phosphatase 2C family protein phosphatase, mediates abscisic acid (ABA)-regulated stomatal movement and water loss specifically during leaf senescence. Here we report that SAG113 is a direct target gene of the AtNAP transcription factor. We found that both AtNAP and SAG113 were induced by leaf senescence and ABA. When AtNAP was chemically induced, SAG113 was also induced whereas when AtNAP was knocked out, the ABA- and senescence-induced expression of SAG113 was reduced. These data suggest that the expression of SAG113 is predominantly dependent on AtNAP. Functionally, overexpression of SAG113 restored the markedly delayed leaf senescence phenotype in atnap knockouts to wild type. Yeast (Saccharomyces cerevisiae) one-hybrid experiments and electrophoresis mobility shift assays showed that AtNAP could physically bind to the SAG113 promoter in vivo and in vitro, respectively. Site-directed mutagenesis revealed that AtNAP binds to a 9-bp core sequence of the SAG113 promoter, 5'CACGTAAGT3'. These results indicate that there is a unique regulatory chain, ABA-AtNAP-SAG113 protein phosphastase 2C, which controls stomatal movement and water loss during leaf senescence. Homozygous transgenic cotton ( Gossypium hirsutum L.) plants that accumulated glycinebetaine (GB) in larger quantities were more tolerant to salt than wild-type (WT) plants. Four transgenic lines, namely 1, 3, 4, and 5, accumulated significantly higher levels of GB than WT plants did both before and after salt stress. At 175 and 275 mM NaCl, seeds of all the transgenic lines germinated earlier and recorded a higher final germination percentage, and the seedlings grew better, than those of the WT. Under salt stress, all the lines showed some characteristic features of salt tolerance, such as higher leaf relative water content (RWC), higher photosynthesis, better osmotic adjustment (OA), lower percentage of ion leakage, and lower peroxidation of the lipid membrane. Levels of endogenous GB in the transgenic plants were positively correlated with RWC and OA. The results indicate that GB in transgenic cotton plants not only maintains the integrity of cell membranes but also alleviates osmotic stress caused by high salinity. Lastly, the seed cotton yield of transgenic lines 4 and 5 was significantly higher than that of WT plants in saline soil. Lignin content and composition are two important agronomic traits for the utilization of agricultural residues. Rice (Oryza sativa) gold hull and internode phenotype is a classical morphological marker trait that has long been applied to breeding and genetics study. In this study, we have cloned the GOLD HULL AND INTERNODE2 (GH2) gene in rice using a map-based cloning approach. The result shows that the gh2 mutant is a lignin-deficient mutant, and GH2 encodes a cinnamyl-alcohol dehydrogenase (CAD). Consistent with this finding, extracts from roots, internodes, hulls, and panicles of the gh2 plants exhibited drastically reduced CAD activity and undetectable sinapyl alcohol dehydrogenase activity. When expressed in Escherichia coli, purified recombinant GH2 was found to exhibit strong catalytic ability toward coniferaldehyde and sinapaldehyde, while the mutant protein gh2 completely lost the corresponding CAD and sinapyl alcohol dehydrogenase activities. Further phenotypic analysis of the gh2 mutant plants revealed that the p-hydroxyphenyl, guaiacyl, and sinapyl monomers were reduced in almost the same ratio compared to the wild type. Endoplasmic reticulum (ER)-associated degradation (ERAD) plays key roles in controlling protein levels and quality in eukaryotes. The Ring Finger Protein 185 (RNF185)/membralin ubiquitin ligase complex was recently identified as a branch in mammals and is essential for neuronal function, but its function in plant development is unknown. Here, we report the map-based cloning and characterization of Narrow Leaf and Dwarfism 1 (NLD1), which encodes the ER membrane-localized protein membralin and specifically interacts with maize homologs of RNF185 and related components. The nld1 mutant shows defective leaf and root development due to reduced cell number. The defects of nld1 were largely restored by expressing membralin genes from Arabidopsis thaliana and mice, highlighting the conserved roles of membralin proteins in animals and plants. The excessive accumulation of ß-hydroxy ß-methylglutaryl-CoA reductase in nld1 indicates that the enzyme is a membralin-mediated ERAD target. The activation of bZIP60 mRNA splicing-related unfolded protein response signaling and marker gene expression in nld1, as well as DNA fragment and cell viability assays, indicate that membralin deficiency induces ER stress and cell death in maize, thereby affecting organogenesis. Our findings uncover the conserved, indispensable role of the membralin-mediated branch of the ERAD pathway in plants. In addition, ZmNLD1 contributes to plant architecture in a dose-dependent manner, which can serve as a potential target for genetic engineering to shape ideal plant architecture, thereby enhancing high-density maize yields. Objective: To determine the correlation between urinary and serum placental growth factor (PlGF) and investigate the predictive value as pregnancy progresses of urinary PlGF compared with serum PlGF, soluble fms-like tyrosine kinase 1 (sFLT-1), and the sFLT-1-to-PlGF ratio for the outcome of preeclampsia in women with preexisting diabetes.Research Design and Methods: A multicenter prospective cohort study was conducted of 158 women with preexisting insulin-requiring diabetes (41 with type 1 and 117 with type 2). Urinary PlGF and serum PlGF, sFLT-1, and the sFLT-1-to-PlGF ratio were assessed four times (14, 24, 30, and 36 weeks' gestation), and the association with the outcome of preeclampsia was investigated.Results: A correlation between urinary and serum PlGF was demonstrated from 24 weeks' gestation onward (P < 0.001). At all time points, those who developed preeclampsia had lower serum PlGF levels (P < 0.05), and receiver operating characteristic curves demonstrated that serum PlGF in this cohort performed better than the serum sFLT-1-to-PlGF ratio as a predictive test for preeclampsia. Preconception HbA1c ≥6.5% (48 mmol/mol) was an important discriminative predictor for preeclampsia (P = 0.01).Conclusions: This study prospectively describes the longitudinal changes in urinary PlGF alongside serum angiogenic markers throughout pregnancy in women with preexisting diabetes. We demonstrate correlation between urinary and serum PlGF and that in women with preexisting diabetes in pregnancy, serum PlGF is a better predictor of preeclampsia than the sFLT-1-to-PlGF ratio. The newly developed planar differential mobility analyzer (DMA) serving as particle sizer can achieve higher transmission and selection precision at ambient pressure compared with conventional cylindrical DMA, and show potentials on coupling with atmospheric pressure interface mass spectrometer (API-MS) for cluster detection with an additional ion mobility dimension. In this study, we assessed the performance of a commercial planar DMA (DMA P5) integrated with the home-build recirculation system. The sizing range of the system in this work is 0-3.9 nm, although larger sizes can be measured with a sheath gas flow restrictor. The resolving power under different recirculation setups (suction mode and counter flow mode) and different sheath flow rates was evaluated using electrosprayed tetra-alkyl ammonium salts (TMAI, TBAI, THAB and TDAB). The maximum resolving power of THA+ under suction and counterflow mode are 61.6 and 84.6, respectively. The sizing resolution of DMA P5 is 7-16 times higher than conventional cylindrical DMAs. The resolving power showed approximately linear correlation with v under counterflow mode, while the resolving power of THA+ under suction mode stopped linearly increase with v when the VDMA was above 3554.3V and enter a plateau due to the interference of sample flow on the laminarity of sheath flow. The transmission efficiency of DMA P5 can reach 54.3%, about one factor of magnitude higher than the commercial DMAs. The mobility spectrum of different electrosprayed tetra-alkyl ammonium salts and the mass to charge ratio-mobility 2D spectrum of sulfuric acid clusters was also characterized with the DMA P5 (-MS) system. We investigated the expression levels and clinical significance of S100β protein in patients with vascular dementia (VD) after basal ganglia hemorrhage. From June 2014 to December 2015, in 138 patients with basal ganglia hemorrhage, we carried out the Mini-Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA) in patients on the day before the operation, the day after the operation and 9 days after the operation. Thirty-two patients (blood vessel group) had cognitive dysfunction and 106 patients (control group) had VD. One hundred thirty-eight cases of healthy adult volunteers were treated in Henan Provincial People's Hospital and were selected in the same period as healthy controls. The expression levels of serum S100β in the three groups were tested through the ELISA method and the statistical analysis was carried out. In VD patients, the serum S100β levels of patients were significantly higher than VD and healthy control groups; differences were statistically significant (P<0.05). However, there was no significant difference between those without VD after operation and the healthy control group (P>0.05). The correlation analysis was carried out with serum S100β as an independent variable and mean arterial pressure, BMI, MMSE and MoCA scores as dependent variables. Our results suggest that S100β expression levels were negatively correlated to the MMSE score (rs=-4.19) and the difference was statistically significant (P<0.05). The length of hospital stay of patients with basal ganglia hemorrhage associated with VD was significantly extended, with a mean of 23.4±2.8 days. The expression levels of S100β protein in the serum of patients with VD after basal ganglia hemorrhage was significantly increased and negatively correlated to the cognitive function of patients. Background: Glioblastoma multiforme (GBM) is the most common malignant and lethal type of primary central nervous system tumor in humans. In spite of its high lethality, a small percentage of patients have a relatively good prognosis, with median survival times of 36 months or longer. The identification of clinical subsets of GBM associated with distinct molecular genetic profiles has made it possible to design therapies tailored to treat individual patients. Methods: We compared microarray data sets from long-term survivors (LTSs) and short-term survivors (STSs) to screen for prognostic biomarkers in GBM patients using the WebArrayDB platform. We focused on FBLN4, IGFBP-2, and CHI3L1, all members of a group of 10 of the most promising, differentially regulated gene candidates. Using formalin-fixed paraffinembedded GBM samples, we corroborated the relationship between these genes and patient outcomes using methylation-specific polymerase chain reaction (PCR) for MGMT methylation status and quantitative reverse transcription PCR for expression of these genes. Results: Expression levels of the mRNAs of these 3 genes were higher in the GBM samples than in normal brain samples and these 3 genes were significantly upregulated in STSs compared to the levels in LTS samples (P,0.01). Furthermore, Kaplan-Meier analysis showed that the expression patterns of FBLN4 and IGFBP-2 serve as independent prognostic indicators for overall survival (P,0.01 and P,0.05, respectively). Conclusion: To our knowledge, this is the first report describing FBLN4 as a prognostic factor for GBM patient survival, demonstrating that increased GBM survival time correlates with decreased FBLN4 expression. Leaf angle is determined by lamina joint inclination and is an important agronomic trait that determines plant architecture, photosynthetic efficiency, and crop yield. Cytokinins (CKs) are phytohormones involved in shaping rice (Oryza sativa L.) architecture, but their role in leaf angle remains unknown. Here, we report that CK accumulation mediated by rice CK OXIDASE/DEHYDROGENASE3 (OsCKX3) controls lamina joint development and negatively regulates leaf angle. Phenotypic analysis showed that rice osckx3 mutants had smaller leaf angles, while the overexpression lines (OsCKX3-OE) had larger leaf angles. Histological sections indicated that the leaf inclination changes in the osckx3 and OsCKX3-OE lines resulted from asymmetric proliferation of the cells and vascular bundles in the lamina joint. Reverse transcription quantitative PCR, promoter-fused β-glucuronidase expression, and subcellular localization assays indicated that OsCKX3 was highly expressed in the lamina joint, and OsCKX3-GFP fusion protein localized to the endoplasmic reticulum. The enzyme assays using recombinant protein OsCKX3 revealed that OsCKX3 prefers trans-zeatin (tZ) and isopentenyladenine (iP). Consistently, tZ and iP levels increased in the osckx3 mutants but decreased in the OsCKX3 overexpression lines. Interestingly, agronomic trait analysis of the rice grown in the paddy field indicated that osckx3 displayed a smaller leaf angle and enhanced primary branch number, grain size, 1,000-grain weight, and flag leaf size. Collectively, our results revealed that enhancing CK levels in the lamina joint by disrupting OsCKX3 negatively regulates leaf angle, highlighting that the CK pathway can be engineered to reduce leaf angle in rice and possibly in other cereals. Salinity is one of the major environmental factors limiting plant growth and productivity. An H+-PPase gene, TsVP from Thellungiella halophila, was transferred into cotton (Gossypium hirsutum) in sense and antisense orientations under control of the cauliflower mosaic virus (CaMV) 35S promoter. Southern and Northern blotting analysis showed that the sense or antisense TsVP were integrated into the cotton genome and expressed. Transgenic plants overexpressing the vacuolar H+-PPase were much more resistant to 150 and 250 mM NaCl than the isogenic wild-type plants. In contrast, the plants from the antisense line (L–2), with lower H+-PPase activity, were more sensitive to salinity than the wild-type plants. Overexpressing TsVP in cotton improved shoot and root growth and photosynthetic performance. These transgenic plants accumulated more Na+, K+, Ca2+, Cl− and soluble sugars in their root and leaf tissues under salinity conditions compared with the wild-type plants. The lower membrane ion leakage and malondialdehyde (MDA) level in these transgenic plants suggest that overexpression of H+-PPase causes the accumulation of Na+ and Cl− in vacuoles instead of in the cytoplasm, thus reducing their toxic effects. On the other hand, the increased accumulation of ions and sugars decreases the solute potential in cells, and facilitates water uptake under salinity, which is an important mechanism for the increased salt tolerance in TsVP-overexpressing cotton. RNA polymerase III (pol III) products play fundamental roles in a variety of cellular processes, including protein synthesis and cancer cell proliferation. In addition, dysregulation of pol III-directed transcription closely correlates with tumorigenesis. It is therefore of interest to identify novel pathways or factors governing pol III-directed transcription. Here, we show that transcription factor (TF) GATA binding protein 4 (GATA4) expression in SaOS2 cells was stimulated by the silencing of filamin A (FLNA), a repressor of pol IIIdirected transcription, suggesting that GATA4 is potentially associated with the regulation of pol III-directed transcription. Indeed, we show that GATA4 expression positively correlates with pol III-mediated transcription and tumor cell proliferation. Mechanistically, we found that GATA4 depletion inhibits the occupancies of the pol III transcription machinery factors at the loci of pol III target genes by reducing expression of both TFIIIB subunit TFIIB-related factor 1 and TFIIIC subunit general transcription factor 3C subunit 2 (GTF3C2). GATA4 has been shown to activate specificity factor 1 (Sp1) gene transcription by binding to the Sp1 gene promoter, and Sp1 has been confirmed to activate pol III gene transcription by directly binding to both Brf1 and Gtf3c2 gene promoters. Thus, the findings from this study suggest that GATA4 links FLNA and Sp1 signaling to form an FLNA/GATA4/Sp1 axis to modulate pol III-directed transcription and transformed cell proliferation. The tolerance to drought stress of the homozygous transgenic cotton ( Gossypium hirsutum L.) plants with enhanced glycinebetaine (GB) accumulation was investigated at three development stages. Among the five transgenic lines investigated, lines 1, 3, 4, and 5 accumulated significantly higher levels of GB than the wild-type (WT) plants either before or after drought stress, and the transgenic plants were more tolerant to drought stress than the wild-type counterparts from young seedlings to flowering plants. Under drought stress conditions, transgenic lines 1, 3, 4, and 5 had higher relative water content, increased photosynthesis, better osmotic adjustment (OA), a lower percentage of ion leakage, and less lipid membrane peroxidation than WT plants. The GB levels in transgenic plants were positively correlated with drought tolerance under water stress. The results suggested that GB may not only protect the integrity of the cell membrane from drought stress damage, but also be involved in OA in transgenic cotton plants. Most importantly, the seedcotton yield of transgenic line 4 was significantly greater than that of WT plants after drought stress, which is of great value in cotton production. Root-synthesized cytokinins are transported to the shoot and regulate the growth, development, and stress responses of aerial tissues. Previous studies have demonstrated that Arabidopsis (Arabidopsis thaliana) ATP binding cassette (ABC) transporter G family member 14 (AtABCG14) participates in xylem loading of root-synthesized cytokinins. However, the mechanism by which these root-derived cytokinins are distributed in the shoot remains unclear. Here, we revealed that AtABCG14-mediated phloem unloading through the apoplastic pathway is required for the appropriate shoot distribution of root-synthesized cytokinins in Arabidopsis. Wild-type rootstocks grafted to atabcg14 scions successfully restored trans-zeatin xylem loading. However, only low levels of root-synthesized cytokinins and induced shoot signaling were rescued. Reciprocal grafting and tissue-specific genetic complementation demonstrated that AtABCG14 disruption in the shoot considerably increased the retention of root-synthesized cytokinins in the phloem and substantially impaired their distribution in the leaf apoplast. The translocation of root-synthesized cytokinins from the xylem to the phloem and the subsequent unloading from the phloem is required for the shoot distribution and long-distance shootward transport of root-synthesized cytokinins. The plant defense hormone salicylic acid (SA) is perceived by two classes of receptors, NPR1 and NPR3/NPR4. They function in two parallel pathways to regulate SA-induced defense gene expression. To better understand the roles of the SA receptors in plant defense, we systematically analyzed their contributions to different aspects of Arabidopsis (Arabidopsis thaliana) plant immunity using the SA-insensitive npr1-1 npr4-4D double mutant. We found that perception of SA by NPR1 and NPR4 is required for activation of N-hydroxypipecolic acid biosynthesis, which is essential for inducing systemic acquired resistance. In addition, both pattern-triggered immunity (PTI) and effector-triggered immunity (ETI) are severely compromised in the npr1-1 npr4-4D double mutant. Interestingly, the PTI and ETI attenuation in npr1-1 npr4-4D is more dramatic compared with the SA-induction deficient2-1 (sid2-1) mutant, suggesting that the perception of residual levels of SA in sid2-1 also contributes to immunity. Furthermore, NPR1 and NPR4 are involved in positive feedback amplification of SA biosynthesis and regulation of SA homeostasis through modifications including 5-hydroxylation and glycosylation. The aim of the present study was to identify abdominal aortic aneurysms (AAA)-associated miR-155 contributing to AAA pathology by regulating macrophage-mediated inflammation. Angiotensin II (AngII)-infused apolipoprotein E-deficient (ApoE-/-) mice and THP-1 cells model of miR-155 overexpression and deficiency were used in the experiments. The expression of miR-155 was detected by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Cytokines were evaluated using enzyme-linked immunoabsorbent assay (ELISA).Western blotting was used to measure the levels of MMP-2, MMP-9, iNOS, and monocyte chemoattractant protein (MCP)-1 proteins. Immunostaining and transwell were used to determine CD68, elastic collagen, proliferation, and migration of vascular smooth muscle cells (VSMCs). The results showed that miR-155 and cytokines were up-regulated in AAA patients or ApoE-/- mice. Overexpression of miR-155 enhanced MMP-2, MMP-9, iNOS, and MCP-1 levels, and stimulated the proliferation and migration of VSMCs. Meanwhile, inhibition of miR-155 had the opposite effect. In addition, histology demonstrated accumulation of CD68 and elastic collagen-positive areas significantly decreased in miR-155 antagomir injection group. Background: Nonspecific phospholipase C (NPC), which belongs to a phospholipase C subtype, is a class of phospholipases that hydrolyzes the primary membrane phospholipids, such as phosphatidylcholine, to yield sn-1, 2- diacylglycerol and a phosphorylated head-group. NPC plays multiple physiological roles in lipid metabolism and signaling in plants. To fully understand the putative roles of NPC genes in upland cotton, we cloned NPC genes from Gossypium hirsutum and carried out structural, expression and evolutionary analysis. Results: Eleven NPC genes were cloned from G. hirsutum, which were found on chromosomes scaffold269.1, D03, A07, D07, A08, D11, and scaffold3511_A13. All GhNPCs had typical phosphoesterase domains and have hydrolase activity that acts on ester bonds. GhNPCs were annotated as phospholipase C, which was involved in glycerophospholipid metabolism, ether lipid metabolism, and biosynthesis of secondary metabolites. These GhNPCs showed differential expression patterns in distinct plant tissues and in response to various types of stress (low-phosphate, salt, drought, and abscisic acid). They also had different types and numbers of cis-element. GhNPCs could be classified into four subfamilies. Four pairs of GhNPCs were generated by whole-genome duplication and they underwent purifying selection. Conclusions: Our results suggested that GhNPCs are involved in regulating key abiotic stress responses and ABA signaling transduction, and they may have various functional roles for different members under complex abiotic stress conditions. Functional divergence may be the evolutionary driving force for the retention of four pairs of duplicate NPCs. Our analysis provides a solid foundation for the further functional characterization of the GhNPC gene family, and leads to potential applications in the genetic improvement of cotton cultivars. The phytohormone salicylic acid (SA) plays essential roles in biotic and abiotic responses, plant development, and leaf senescence. 2,5-Dihydroxybenzoic acid (2,5-DHBA or gentisic acid) is one of the most commonly occurring aromatic acids in green plants and is assumed to be generated from SA, but the enzymes involved in its production remain obscure. DMR6 (Downy Mildew Resistant6; At5g24530) has been proven essential in plant immunity of Arabidopsis (Arabidopsis thaliana), but its biochemical properties are not well understood. Here, we report the discovery and functional characterization of DMR6 as a salicylic acid 5-hydroxylase (S5H) that catalyzes the formation of 2,5-DHBA by hydroxylating SA at the C5 position of its phenyl ring in Arabidopsis. S5H/DMR6 specifically converts SA to 2,5-DHBA in vitro and displays higher catalytic efficiency (Kcat/Km = 4.96 × 104 m-1 s-1) than the previously reported S3H (Kcat/Km = 6.09 × 10³ m-1 s-1) for SA. Interestingly, S5H/DMR6 displays a substrate inhibition property that may enable automatic control of its enzyme activities. The s5h mutant and s5hs3h double mutant overaccumulate SA and display phenotypes such as a smaller growth size, early senescence, and a loss of susceptibility to Pseudomonas syringae pv tomato DC3000. S5H/DMR6 is sensitively induced by SA/pathogen treatment and is expressed widely from young seedlings to senescing plants, whereas S3H is more specifically expressed at the mature and senescing stages. Collectively, our results disclose the identity of the enzyme required for 2,5-DHBA formation and reveal a mechanism by which plants fine-tune SA homeostasis by mediating SA 5-hydroxylation. The present study investigated the incidence, causes, treatment and prevention of limb graft occlusion following endovascular aortic repair (EVAR). A total of 66 cases of abdominal aortic aneurysm receiving EVAR at our department from January 2005 to December 2013 were enrolled. After EVAR, patients received routine antiplatelet therapy of 75 mg PLAVIX for 6 months and then 100 mg Aspirin for another 6 months by oral administration. According to previous clinical experiences, antiplatelet therapy is able to effectively reduce the incidence of iliac occlusion after EVAR. A total of 61 bifurcated grafts and 5 aortauniilac grafts (127 limbs in total) were used. Physical examination, ankle-brachial-index and computer tomographic angiography were performed at 10 days, at 3, 6 and 12 months and annually thereafter. It was found that 7 limbs in 7 patients (10.6% of patients, 5.5% of limbs) were occluded between 20 days and 12 months (average, 7.8±5.3 months) after EVAR. Acute and severe ischemia was found in 2 cases, claudication was in found 3 cases, asthenia in both legs was found in 1 case and 1 case was asymptomatic. Femoral-femoral bypass, femoral-femoral bypass and stenting, aorto-iliac/femoral bypass, thrombectomy and conservative treatment were performed in 1 patient each and thrombectomy together with stenting was performed in 2 cases. Limb graft occlusion was not rare after EVAR. Treatment of this complication included surgery and endovascular therapy such as bypass, thrombectomy and thrombolysis. Background: Fe80B20 amorphous alloys exhibit excellent soft magnetic properties, high abrasive resistance and outstanding corrosion resistance. In this work, Fe80B20 amorphous micro-fibers with HC of 3.33 Oe were firstly fabricated and the effects of annealing temperature on the structure and magnetic properties of the fibers were investigated. Methods: In this study, Fe80B20 amorphous fibers were prepared by the single roller melt-spinning method. The structures of as-spun and annealed fibers were investigated by X-ray diffractometer (XRD) (PANalytical X,Pert Power) using Cu Kα radiation. The morphology of the fibers was observed by scanning electron microscopy (SEM) (HITACHI-S4800). Differential scanning calorimetry (DSC) measurements of the fibers were performed on Mettler Toledo TGA/DSC1 device under N2 protection. Vibrating sample magnetometer (VSM, Versalab) was used to examine the magnetic properties of the fibers. The resonance behavior of the fibers was characterized by an impedance analyzer (Agilent 4294A) with a home-made copper coil. Results: The X-ray diffusion (XRD) patterns show that the fibers remain amorphous structure until the annealing temperature reaches 500°C. The differential scanning calorimetry (DSC) results show that the crystallization temperature of the fibers is 449°C. The crystallization activation energy is calculated to be 221 kJ/mol using Kissinger formula. The scanning electron microscopy (SEM) images show that a few dendrites appear at the fiber surface after annealing. The result indicates that the coercivity HC (//) and HC (⊥) slightly increases with increasing annealing temperature until 400°C, and then dramatically increases with further increasing annealing temperature which is due to significant increase in magneto-crystalline anisotropy and magneto-elastic anisotropy. The Q value firstly increases slightly when the annealing temperature rises from room temperature (RT) to 300°C, then decreases until 400°C. Eventually, the value of Q increases to ~2000 at annealing temperature of 500°C. Conclusions: In this study, Fe80B20 amorphous fibers with the diameter of 60 μm were prepared by the single roller melt-spinning method and annealed at 200°C, 300°C, 400°C, and 500°C, respectively. XRD results indicate that the fiber structure remains amorphous when the annealing temperature is below 400°C. α-Fe phase and Fe3B phase appear when the annealing temperature rises to 500°C, which is above the crystallization temperature of 449°C. The recrystallization activation energy is calculated to be 221 kJ/mol. The coercivity increases with increasing annealing temperature, which attributes to the increase of total anisotropy. Lambert-Eaton myasthenic syndrome (LEMS) is typically characterized as a paraneoplastic syndrome associated with small cell lung carcinoma. The presence of LEMS in association with other neuroendocrine lung tumors, including carcinoids or large cell lung carcinoma, is highly unusual. The present case study reports the very rare case of a patient with mediastinal small cell cancer combined with LEMS. A 53-year-old man was admitted to the Bethune First Hospital with a 3-month history of weakness in the lower extremities that had become aggravated during the previous 20 days. A positron emission tomography-computed tomography (CT) examination demonstrated multiple enlarged, hypermetabolic lymph nodes between the vena cava and trachea, indicating the presence of inflammation. No other abnormalities were identified. A mediastinal lymph node biopsy revealed a high prevalence of small cell neuroendocrine carcinoma. The immunohistochemistry results were as follows: Ki-67 (+50, meaning that 50% of the cells exhibited Ki-67 expression), cytokeratin (CK; +), cluster of differentiation (CD)56 (+), synaptophysin (+), thyroid transcriptional factor-1 (+), epithelial membrane antigen (+), neuron-specific enolase (+), P63 (weakly +), leukocyte common antigen (-), CD99 (-), vimentin (-) and CK5/6 (-). Following a diagnosis of LEMS, the patient underwent one course of cisplatin + etoposide chemotherapy, which was ineffective. Subsequently, the patient underwent a different chemotherapy regimen (cisplatin + etoposide + ifosfamide), which significantly improved the symptoms of myasthenia. Following completion of this treatment regimen, the primary lesion was not observed on a lung CT scan, and the mediastinal lymph nodes had clearly diminished. Background: Food frequency questionnaire (FFQ) is a reliable tool to estimate dietary intake in large nutritional epidemiological studies, but there is lack of a current and validated FFQ for use in urban Chinese pregnant women. This study aimed to evaluate the reproducibility and validity of a semi-quantitative FFQ designed to estimate dietary intake among urban pregnant women in a cohort study conducted in central China. Methods: In the reproducibility study, a sample of 123 healthy pregnant women completed the first FFQ at 12-13 weeks gestation and the second FFQ 3-4 weeks later. To validate the FFQ, the pregnant women completed three 24-h recalls (24HRs) between the intervals of two FFQs. Results: The intraclass correlation coefficients of two administrations of FFQ for foods ranged from 0.23 (nuts) to 0.49 (fruits) and for nutrients from 0.24 (iodine) to 0.58 (selenium) and coefficients were all statistically significant. The unadjusted Pearson correlation coefficients between two methods ranged from 0.28 (beans) to 0.53 (fruits) for foods and from 0.15 (iodine) to 0.59 (protein) for nutrients. Energy-adjusted and de-attenuated correlation coefficients for foods ranged from 0.35 (beans) to 0.56 (fruits) and for nutrients from 0.11 (iodine) to 0.63 (protein), and all correlations being statistically significant except for iodine, sodium and riboflavin. On average, 67.0% (51.2%-80.5%) of women were classified by both methods into the same or adjacent quintiles based on their food intakes, while 68.5% (56.1%-77.2%) of women were classified as such based on nutrient intakes. Extreme misclassifications were very low for both foods (average of 2.0%) and nutrients (average of 2.2%). Bland-Altman Plots also showed reasonably acceptable agreement between two methods. Extreme misclassifications were very low for both foods (average of 2.0 %) and nutrients (average of 2.2 %). Bland-Altman Plots also showed reasonably acceptable agreement between two methods. Survivin is a promising anticancer therapeutic target due to its important role in the inhibition of apoptosis of tumor cells. However, little is currently known about its role in non small cell lung cancer (NSCLC). The present study evaluated whether the downregulation of survivin expression would affect cell proliferation, cell cycle distribution, apoptosis and colony formation of NSCLC. A recombinant lentiviral small hairpin RNA (shRNA) expression vector, which specifically targeted survivin, was constructed and transfected into the A549 human NSCLC cell line. Quantitative polymerase chain reaction and western blotting were used to determine the mRNA and protein expression levels of survivin, 48 h following the knockdown of survivin expression. Cell proliferation, apoptosis, cell cycle distribution and colony formation were determined following the downregulation of survivin by shRNA. In addition, A549 cells were injected into nude mice, and the effects of shRNA targeting the survivin gene on tumor growth were assessed. Downregulation of survivin expression, using the RNA silencing approach in A549 tumor cells, significantly suppressed the proliferation and colony formation ability of the cells, and induced tumor apoptosis in vitro. The nude mice inoculated with A549 cells developed cancer, and treatment with shRNA targeting survivin markedly inhibited the growth of these cancers, with no obvious side effects. The results of the present study suggest that suppression of survivin expression by RNA interference may induce NSCLC apoptosis, and provide a novel approach for anticancer gene therapy. Abstract: One-dimensional (1D) needle-like ZnO nanorods with aspect ratio about 50 were successfully synthesized by a sodium dodecylsulphate (SDS)-assisted hydrothermal process at lower temperature (85°C). The morphology and structure of crystals were characterized by field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM) and X-ray diffraction (XRD) analysis. Experimental results show that SDS, ratio of OH−/Zn2+ and hydrothermal reaction time play important roles in the formation and oriented growth of ZnO nanorods. Gas sensors have been fabricated using 1D ZnO powders to examine the response towards 40ppm CO over a range of operating temperature from 150°C to 450°C. The highest response is 49.58 at the operating temperature of 400°C. The response was greatly enhanced by doping Nd element as surface modifier or ZrO2 as secondary oxide. The effect of doping on the ZnO structure and gas sensing has been discussed by X-ray photoelectron spectroscopy (XPS) analysis. [Copyright &y& Elsevier]

Low phosphorus (P) availability is a major limitation for plant growth. To better understand the molecular mechanism of P efficiency in maize, comparative proteome analyses were performed on the roots of the low-P-tolerant mutant 99038 and wild-type Qi-319 grown under P-sufficient (+P) or P-deficient (−P) conditions. Over 10% of proteins detected on two-dimensional electrophoresis (2-DE) gels showed expression that was altered twofold or more between the genotypes under +P or −P conditions. We identified 73 (+P) and 95 (−P) differentially expressed proteins in response to phosphate (Pi) starvation. These proteins were involved in a large number of cellular and metabolic processes, with an obvious functional skew toward carbon metabolism and regulation of cell proliferation. Further analysis of proteome data, physiological measurements and cell morphological observations showed that, compared to the wild-type, the low-P-tolerant mutant could accumulate and secrete more citrate under Pi starvation, which facilitates solubilization of soil Pi and enhances Pi absorption. The proportion of sucrose in the total soluble sugars of the low-P-tolerant mutant was significantly higher, and cell proliferation in root meristem was accelerated. This resulted in better developed roots and more advantageous root morphology for Pi uptake. These results indicate that differences in citrate secretion, sugar metabolism and root-cell proliferation are the main reasons for higher tolerance to low-P conditions in the mutant compared to the wild-type. Thus, the mutant displayed specialized P-efficient root systems with a higher capacity for mobilization of external Pi and increased cell division in the root meristem under Pi starvation. The relationship between internal phosphorus (P) concentration [P] and kinetics of phosphate (Pi) uptake was investigated in maize seedlings grown hydroponically at different Pi concentrations (0.1-1,000 µM) and in the phase of Pi deprivation (0-10 d). The results indicated when the internal [P] was higher than 85 µmol g-1 dw, apparent Km, Cmin, and Vmax were significantly (P<0.01) related to [P]s in shoots and roots; when the internal [P] was lower than 85 µmol g-1 dw, Km and Cmin were small and only Vmax was significantly related (P<0.01) to internal [P]s. Three equations were deduced from the linear regressions of the kinetic parameters and [P]s in shoots. Using these equations, the values of apparent Km, Cmin, and Vmax of Pi uptake of seedlings grown in different circumstances were calculated according to [P]s in shoots. In all the circumstances involved, for Km and Cmin, there was a parallel relationship between the values estimated by [P]s in shoots and by the Pi-depletion technique; for Vmax, the values estimated by [P]s in shoots were consistent with those obtained from Pi-depletion experiments except the period of supplying Pi to the Pi-starved seedlings over several days. These results indicated it is possible to estimate the Pi-uptake status according to shoot P concentrations in maize plants under experimental conditions, which might be helpful to estimate in-season status of Pi uptake of maize plants in the field.
