Dentin hypersensitivity is an oral health concern affecting a large percentage of the world's adult population. Occlusion of the exposed dentinal tubules is among the treatment options available, and silver diammine fluoride (SDF) is an occluding agent used for interrupting or dampening the stimulus of the dental pulp nerves that produce pain. In addition to dentin permeability testing, the evaluation of desensitizing agents occluding dentinal tubules strongly relies on microscopic techniques, such as scanning electron microscopy (SEM). Limitations of SEM are that it provides only surface images that lack detailed information on the depth of penetration and amount of material present within the treated specimen, and it is prone to sample preparation artifacts. Here, we present high-resolution X-ray computed tomography (nano-CT) as a potential method for investigating dentin specimens with occluded tubules. We studied human dentin treated with SDF as an exemplary dentinal occlusion treatment option. We evaluated the silver deposits formed on the dentin surface region near the dentinal tubules and in the tubular regions using cross-section SEM, Energy Dispersive X-ray (EDX) analysis, and nano-CT. The resulting images obtained by SEM and nano-CT had comparable resolutions, and both techniques produced images of the tubules' occlusion. Nano-CT provided three-dimensional images adequate to quantitate tubule size and orientation in space. Moreover, it enabled clear visualization of dentinal tubules in any virtual plane and estimation of the amount and depth of occluding material. The stability of nonpatterned and nanopatterned strong polyelectrolyte brushes (PEBs) is studied as a function of both brush character and the properties of a contacting liquid. High‐molecular‐weight PEBs of poly(4‐methyl vinylpyridinium iodide) (PMeVP) are synthesized using surface‐initiated radical‐chain polymerization. Nanopatterned brushes (NPBs) line with pattern sizes ranging from 50 to 200 nm are generated by patterning the initiator layer using deep‐ultraviolet photolithography followed by brush growth initiated from the patterned layer. Homogeneous PEBs with different degrees of charging and grafting densities are exposed to water and salt solutions with different temperatures for different periods. The degradation is monitored through dry‐state ellipsometry and atomic force microscopy measurements. Enhanced degrafting for more strongly swollen polymer brushes can be observed in agreement with an "entropic spring" model. Based on the results of the nonpatterned brushes, the NPBs are exposed to water at different temperatures and external salt content for varying periods of time. Counterintuitively, the NPBs show increased degrafting for smaller patterns, which is attributed to different polymer chain dynamics for nanobrushes and microbrushes. The morphology of top-down and bottom-up nanostructured neutral brushes is studied as a function of the production process and the quality of solvent to which the brushes have been exposed. Neutral brushes of poly(tert-butyl methacrylate) (PtBuMA) were grafted from the substrate by surface-initiated radical-chain polymerization (SI-RCP). Brush patterning was conducted using deep ultraviolet (DUV) photolithography following either top-down (TD) or bottom-up (BU) approaches with patterned lines as small as 200 nm. The resulting brushes were immersed in toluene or methanol, and the change of morphology of the dried brushes was monitored using tapping mode atomic force microscopy (AFM). TD nanopatterned brushes (NPBs) exhibit a box-like and BU generated brushes a lens-like shape directly after the production process. We show that the pattern width for the BU brushed depends not only on the wetting properties of the areas adjacent to the pattern but also on the process history to which the sample has been exposed, especially the solvent quality. Furthermore, we show that TD NPBs are cross-linked on the edges of the pattern because of the high-energy process conditions during brush patterning and study the behavior of the thus-obtained structures. The stability of strong polyelectrolyte brushes (PEBs) was studied in bulk and in patterned structures. Thick PEBs of poly([(2-methacryloyloxy)ethyl]trimethylammonium chloride) with thicknesses >100 nm were synthesized using single electron transfer living radical polymerization. Brush patterning was identified using deep-ultraviolet photolithography by means of either a top-down (TD) or bottom-up (BU) method, with features as small as 200 nm. The brushes were soaked in water under a range of pH or temperature conditions, and the hydrolysis was monitored through dry-state ellipsometry and atomic force microscopy measurements. BU patterns showed reduced degrafting for smaller patterns, which was attributed to increased stress relaxation at such dimensions. In contrast to the already relaxed BU-patterned brush, a TD-patterned brush possesses perpendicular structures that result from the use of orthogonal lithography. It was found that the TD process induces cross-linking on the sidewall, which subsequently fortifies the sidewall materials. This modification of the polymer brushes hindered the stress relaxation of the patterns, and the degrafting trends became irrelevant to the pattern sizes. Background: Spider silk is a tear-resistant and elastic biopolymer that has outstanding mechanical properties. Additionally, exiguous immunogenicity is anticipated for spider silks. Therefore, spider silk represents a potential ideal biomaterial for medical applications. All known spider silk proteins, so-called spidroins, reveal a composite nature of silk-specific units, allowing the recombinant production of individual and combined segments. Results: In this report, a miniaturized spidroin gene, named VSO1 that contains repetitive motifs of MaSp1 has been synthesized and combined to form multimers of distinct lengths, which were heterologously expressed as elastin-like peptide (ELP) fusion proteins in tobacco. The elastic penetration moduli of layered proteins were analyzed for different spidroin-based biopolymers. Moreover, we present the first immunological analysis of synthetic spidroin-based biopolymers. Characterization of the binding behavior of the sera after immunization by competitive ELISA suggested that the humoral immune response is mainly directed against the fusion partner ELP. In addition, cytocompatibility studies with murine embryonic fibroblasts indicated that recombinant spidroin-based biopolymers, in solution or as coated proteins, are well tolerated. Conclusion: The results show that spidroin-based biopolymers can induce humoral immune responses that are dependent on the fusion partner and the overall protein structure. Furthermore, cytocompatibility assays gave no indication of spidroin-derived cytotoxicity, suggesting that recombinant produced biopolymers composed of spider silk-like repetitive elements are suitable for biomedical applications. Phytophenols are considered to have beneficial effects towards human physiology. They are food microcomponents with potent chemopreventive properties towards the most three frequent contemporary human diseases, e.g., cardiovascular alterations, cancer and neurodegenerative pathologies. Related to this, the plasmatic form and plasmatic level of plant polyphenols in the body circulation are crucial for their efficiency. Thus, determinations of the binding process of resveratrol and of common flavonoids produced by major edible plants, berries and fruits to plasma proteins are essential. The interactions between resveratrol and albumin, a major plasma protein, were compared with those already published, involving curcumin, genistein, quercetin and other well-known food-containing polyphenols. The approaches used are usually intrinsic fluorescence intensity changes, quenching of protein intrinsic fluorescence and infrared spectroscopy. It appears that: (1) all of the studied polyphenols interact with albumin; (2) while most of the studied polyphenols interact at one albumin binding site, there are two different types of resveratrol binding sites for bovine serum albumin, one with the highest affinity (apparent KD of 4 µM) with a stoichiometry of one per monomer and a second with a lower affinity (apparent KD of 20 µM) with also a stoichiometry of one per monomer; (3) at least one binding site is in the vicinity of one tryptophanyl residue of bovine serum albumin; and (4) resveratrol binding to bovine serum albumin produces a very small structural conformation change of the polypeptide chain. The magnetic ground state of biatomic Fe chains on the reconstructed (5×1)-Ir(001) surface is a cycloidal 120° spin spiral. Spin-resolved scanning tunneling microscopy reveals a striking variation of magnetic field dependences among the chains, which we attribute to parity effects resulting from finite lengths. Numerical simulations show that the chains are divided in three symmetry classes with the exact number of atoms in the chain determining the size and direction of their net magnetic moment. In contrast to antiferromagnetic systems, the three-atom periodicity causes the effective anisotropy to alternate between out of plane, in plane, and quenched. [ABSTRACT FROM AUTHOR]

Blends of two elastomeric ethylene–octene copolymers with similar octene contents having a random (ORC) and a blocky architecture (OBC) are prepared by melt mixing. The thermal and mechanical properties of ORC, OBC and their blends are investigated by DSC, dynamic mechanical analysis and tensile tests. The morphology of the semi-crystalline samples is studied by AFM and WAXS. Two types of crystals have been observed: (i) Orthorhombic crystals forming lamellae with an estimated thickness of about 13 nm composed mainly of long polyethylene-like sequences of OBC that melt a temperature of about 120 °C and (ii) fringed micellar crystals with a thickness of 2–4 nm formed basically by short polyethylene-like sequences of ORC that have melting temperatures between 30 and 80 °C. The amorphous phase contains a relatively homogeneous mixture of segments of both components indicated by the relatively uniform shape of the loss modulus peaks from dymamic-mechanical measurements for all investigated copolymers and blends. ORC crystallization is hindered in blends as indicated by lower melting enthalpies. This might be related to the high octene content of the amorphous phase at the relevant crystallization temperature as well as geometrical constraints since ORC crystallization occurs in an already semi-crystalline polymer. The results of tensile tests show that the mechanical behavior can be tailored via blend composition and morphology of the semi-crystalline material. Vector spin chirality is one of the fundamental characteristics of complex magnets. For a one-dimensional spin-spiral state it can be interpreted as the handedness, or rotational sense of the spiral. Here, using spin-polarized scanning tunneling microscopy, we demonstrate the occurrence of an atomic-scale spin spiral in finite individual bi-atomic Fe chains on the (5 X 1) -- Ir(001) surface. We show that the broken inversion symmetry at the surface promotes one direction of the vector spin chirality, leading to a unique rotational sense of the spiral in all chains. Correspondingly, changes in the spin direction of one chain end can be probed tens of nanometers away, suggesting a new way of transmitting information about the state of magnetic objects on the nanoscale. Resveratrol produced by several plants, berries and fruits, including grapes, is one of the best known natural food microcomponents with potent chemopreventive properties towards the most severe contemporary human diseases: cardiovascular sickness, cancer and neurodegenerative pathologies. Demonstration of its mechanism of action also implies the elucidation of the steps of bioavailability and bioabsorption in cells and tissues. In order to estimate the relationships between the amounts of resveratrol taken up by food or drink intake, and the several possible benefits illustrated from in vitro/in vivo experiments and from epidemiological studies, it is essential to demonstrate step by step the route of resveratrol from plasma to the cell active site. In plasma, resveratrol was shown to interact with lipoproteins. This commentary also contains previously unpublished results about interactions between resveratrol and albumin and the enhancement of this binding in presence of fatty acids. We have previously described that resveratrol uptake by hepatic cells involves two processes – a passive one and a carrier-mediated one. Thanks to this last process, resveratrol, while tightly bound to blood proteins, could be largely delivered to body tissues. The intracellular proteic targets of resveratrol remain to be identified. Objective: The utility of continuous intra-arterial blood gas analysis (CBGA) with combined electrochemical and optode sensors has been demonstrated. More recently, a pure optode sensor with a changed sensing element architecture has become available. The aim was to determine the measurement accuracy and long-term stability of the new sensor.Design: A prospective explorative study was performed. Simultaneous measurements of intermittent blood gas analyses (IBGA) (ABL 610, Radiometer, Copenhagen) and CBGA (Diametrics Medical, High Wycombe, Bucks., UK) were compared using Bland-Altman analysis.Patients: Twenty-five patients admitted to the ICU and requiring mechanical ventilation for an expected minimum of about 96 h were included.Results: Mean monitoring time was 106.1 (range 15-231) hours. Bias and precision for PO(2 )were -0.2 kPa (1%)+/-1.8 kPa (9.5%); PCO(2): 0.03 kPa (0.6%)+/-0.44 kPa (9.3%); pH: -0.001 (0.01%)+/-0.04 (0.45%). The sensor showed no change of measurement characteristics during 4 days of measurement. However, in 69 cases continuous monitoring was interrupted (reversible sudden drops of PO(2) measurement) possibly caused by thrombotic deposition and/or sensor bending and accidental sensor retraction.Conclusions: The precision and bias of the PCO(2)- and pH-sensing elements were in line with the findings of the older sensor technology. The possibility that the PO(2) optode could offer greater accuracy than the older technology is suggested by comparisons with results reported in previous studies. In this work, an easy to up-scale method combining atmospheric plasma treatment and wet chemical coating with zwitterionic poly(sulfobetaine methacrylate) (pSBMA) was employed to modify the feed spacer surface aiming at reducing its susceptibility to biofouling. The goals of the study were: 1) to verify whether the proposed modification method is suitable for preparation of homogenous feed spacer coatings and 2) to compare the biofouling propensity of pSBMA modified and unmodified spacers. The effect of the plasma intensity and monomer concentration on the feed spacer surface composition, morphology and roughness were studied. The most favorable modification was compared with the reference material in a batch experiment and a continuously operated fouling simulator. The zwitterionic coating showed reduced adhesion of Pseudomonas fluorescens by 70% and postponed the EPS production in the short-term batch testing. However, the long-term studies performed under continuous flow conditions with nutrient dosage did not demonstrate any significant biofouling mitigation of the surface-conditioned spacers. • Plasma treated spacers were coated with zwitterionic poly(sulfobetaine methacrylate). • Feed spacers modified with poly(sulfobetaine methacrylate) showed anti-adhesion properties • Zwitterionic coating postponed EPS production by adhered bacteria. • Spacer coating did not prevent biofouling in continuous experiments. • Poly(sulfobetaine methacrylate) coating attracted calcium and magnesium ions. Direct compounding injection molding of ternary blends of polylactide (PLA), polybutylene succinate (PBS) and a hydrogenated styrene farnesene block copolymer (HSFC) was investigated for different blend compositions. Morphological analysis by atomic force microscopy and scanning electron microscopy showed a heterogeneous morphology of the ternary blends, with coexisting droplets of HSFC and PLA respectively PBS. Particle sizes analysis of the minor phases in the ternary blends showed mean values in the range of ∼1 μm, which is discussed in correlation to the rheological properties of the blend components as obtained by high pressure capillary rheometry. In result, with increasing HSFC content, the ternary blends showed a significant increase in notched Charpy impact strength (acN). In concrete, for the ternary blends with 15 wt% of HSFC a maximum acN of 15.8 kJ/m2 was found. At the same time, incorporation of the HSFC resulted only in a moderate reduction in elastic modulus and tensile strength. [ABSTRACT FROM AUTHOR]

Endothelin‐1 (ET‐1) is a member of the endothelin family of peptide hormones first discovered as endothelium‐derived mediators regulating vascular tone. ET‐1 also regulates the proliferation and differentiation of bone cells that synthesize fibroblast growth factor 23 (FGF23). FGF23 is a hormone controlling renal phosphate and vitamin D metabolism. Here, we studied the role of ET‐1 and endothelin receptor B (ETB) for FGF23 production. Fgf23 gene expression was studied in IDG‐SW3 bone cells by quantitative RT‐PCR. ETB‐expressing (etb+/+) and rescued ETB‐deficient mice (etb−/−) were studied in metabolic cages. Their serum FGF23, PTH, and 1,25(OH)2D3 concentrations were determined by ELISA, serum and urinary phosphate and Ca2+ by photometric methods. ET‐1 and ETB agonist sarafotoxin 6c suppressed Fgf23 mRNA in IDG‐SW3 cells. Serum C‐terminal and intact FGF23 as well as bone Fgf23 mRNA levels were significantly higher in etb−/− mice than in etb+/+ mice. Renal phosphate excretion was significantly higher in etb−/− mice despite lower phosphate levels. In addition, etb−/− animals exhibited calciuria and a significantly higher serum 1,25(OH)2D3 concentration compared to etb+/+ mice. In conclusion, ETB‐dependent ET‐1 signaling is a potent suppressor of FGF23 formation. The degradation of 405-nm fiber-coupled diode laser systems with more than 50 mW power was investigated in detail with focus on the effects occurring at the input end. The coupling and transmission loss of the laser light were associated with the growth of a projection and a periodic structure on the input surface. To avoid this degradation, a short launch fiber with a good surface quality was used at the input end. In this way, the power transmission was stabilized for at least one month. However, structural degradation was noticed on the output surface of the single-mode fiber. To investigate this effect, the damaged samples weremeasured after different periods of time and examined with a scanning electron microscope and with an atomic force microscope. Reproducible spherical projections with a submicron periodic structure were found in the core region. Additionally, the spectral loss of the fiber was measured, showing the formation of color centers in the deep ultraviolet along the length of the fiber. These investigations were accompanied by simulations of the growth of the structure on the output surface. Background and Aims Forisomes are specialized structural phloem proteins that mediate sieve element occlusion after wounding exclusively in papilionoid legumes, but most studies of forisome structure and function have focused on the Old World clade rather than the early lineages. A comprehensive phylogenetic, molecular, structural and functional analysis of forisomes from species covering a broad spectrum of the papilionoid legumes was therefore carried out, including the first analysis of Dipteryx panamensis forisomes, representing the earliest branch of the Papilionoideae lineage. The aim was to study the molecular, structural and functional conservation among forisomes from different tribes and to establish the roles of individual forisome subunits. Methods Sequence analysis and bioinformatics were combined with structural and functional analysis of native forisomes and artificial forisome-like protein bodies, the latter produced by expressing forisome genes from different legumes in a heterologous background. The structure of these bodies was analysed using a combination of confocal laser scanning microscopy (CLSM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM), and the function of individual subunits was examined by combinatorial expression, micromanipulation and light microscopy. Key Results Dipteryx panamensis native forisomes and homomeric protein bodies assembled from the single sieve element occlusion by forisome (SEO-F) subunit identified in this species were structurally and functionally similar to forisomes from the Old World clade. In contrast, homomeric protein bodies assembled from individual SEO-F subunits from Old World species yielded artificial forisomes differing in proportion to their native counterparts, suggesting that multiple SEO-F proteins are required for forisome assembly in these plants. Structural differences between Medicago truncatula native forisomes, homomeric protein bodies and heteromeric bodies containing all possible subunit combinations suggested that combinations of SEO-F proteins may fine-tune the geometric proportions and reactivity of forisomes. Conclusions It is concluded that forisome structure and function have been strongly conserved during evolution and that species-dependent subsets of SEO-F proteins may have evolved to fine-tune the structure of native forisomes. In the last two decades it was shown that plants have a great potential for production of specific heterologous proteins. But high cost and inefficient downstream processing are a main technical bottleneck for the broader use of plant-based production technology especially for protein-based products, for technical use as fibres or biodegradable plastics and also for medical applications. High-performance fibres from recombinant spider silks are, therefore, a prominent example. Spiders developed rather different silk materials that are based on proteins. These spider silks show excellent properties in terms of elasticity and toughness. Natural spider silk proteins have a very high molecular weight, and it is precisely this property which is thought to give them their strength. Transgenic plants were generated to produce ELPylated recombinant spider silk derivatives. These fusion proteins were purified by Inverse Transition Cycling ( ITC) and enzymatically multimerized with transglutaminase in vitro. Layers produced by casting monomers and multimers were characterized using atomic force microscopy ( AFM) and AFM-based nanoindentation. Topologically nontrivialspin textures have recently been investigated for spintronic applications. Here, we report on an ultrathin magnetic film in which individual skyrmions can be written and deleted in a controlled fashion with local spin-polarized currents from a scanning tunneling microscope. An external magnetic field is used to tune the energy landscape, and the temperature is adjusted to prevent thermally activated switching between topologically distinct states. Switching rate and direction can then be controlled by the parameters used for current injection. • XCT and 3D-image analysis of glass sphere compacts during sintering. • Coarseness C is used to parameterise local relative density distributions. • Decrease in C between initial and final sintering stages. • Intermediate stage with highest C and bimodal distribution of closed and interconnected pores. • Spatial heterogeneity of local particle/pore arrangements is biased by substrate effects. Using high-resolution X-ray computed tomography and 3D image analysis, the global relative density was determined and fluctuations in relative density were localized and quantified in a series of sintered glass bodies made of 80 μm glass spheres over the entire sintering process. A representative volume element (RVE) in the sintered body was divided into cubic volumes of 500, 250 and 125 µm edge length by an automatic segmentation routine. It was found that the expected decrease in coarseness C from the initial to the final sintering stage is interrupted by an intermediate stage with highest C and a characteristic bimodal density distribution. The bimodality of the intermediate stage is due to spatially separated denser arrangements within the RVE with closed pores and less dense arrangements with a network of interconnected or open pores. Skyrmions are topologically protected field configurations with particle-like properties that play an important role in various fields of science. Recently, skyrmions have been observed to be stabilized by an external magnetic field in bulk magnets. Here, we describe a two-dimensional square lattice of skyrmions on the atomic length scale as the magnetic ground state of a hexagonal Fe film of one-atomic-layer thickness on the Ir(111) surface. Using spin-polarized scanning tunnelling microscopy we can directly image this non-collinear spin texture in real space on the atomic scale and demonstrate that it is incommensurate to the underlying atomic lattice. With the aid of first-principles calculations, we develop a spin model on a discrete lattice that identifies the interplay of Heisenberg exchange, the four-spin and the Dzyaloshinskii-Moriya interaction as the microscopic origin of this magnetic state. Objective: The aim of the present study was to examine the impact of moderate and profound hyperventilation on regional cerebral blood flow (rCBF), oxygenation and metabolism. Materials and methods: Twelve anesthetized pigs were subjected to moderate (mHV) and profound (pHV) hyperventilation (target arterial pO2: 30 and 20 mmHg, respectively) for 30 min each, after baseline normoventilation (BL) for 1 h. Local cerebral extracellular fluid (ECF) concentrations of glucose, lactate, pyruvate and glutamate as well as brain tissue oxygenation (ptiO2) were monitored using microdialysis and a Licox oxygen sensor, respectively. In nine pigs, regional cerebral blood flow (rCBF) was also continuously measured via a thermal diffusion system. Results: Both moderate and profound hyperventilation resulted in a significant decrease in rCBF (BL: 37.9±4.3 ml/100 g/min; mHV: 29.4±3.6 ml/100 g/min; pHV: 23.6±4.7 ml/100 g/min; p<0.05) and ptiO2 (BL: 22.7±4.1 mmHg; mHV: 18.9±4.9 mmHg; pHV: 13.0±2.2 mmHg; p<0.05). A ptiO2 decrease below the critical threshold of 10 mmHg was induced in three animals by moderate hyperventilation and in five animals by profound hyperventilation. Furthermore, significant increases in lactate (BL: 1.06±0.18 mmol/l; mHV: 1.36±0.20 mmol/l; pHV: 1.67±0.17 mmol/l; p<0.005), pyruvate (BL: 46.4±7.8 μmol/l; mHV: 58.0±10.3 μmol/l; pHV: 66.1±12.7 μmol/l; p<0.05), and lactate/glucose ratio were observed during hyperventilation. (Data are presented as mean±S.E.M.) Conclusions: Both moderate and profound hyperventilation may result in insufficient regional oxygen supply and anaerobic metabolism, even in the uninjured brain. Neuropathic pain is a major problem in the treatment of cancer pain. We performed a retrospective analysis of 213 cancer patients with neuropathic pain treated by a pain service following the World Health Organization guidelines for relief of cancer pain. Of these, 79% presented with nerve compression pain, 16% with nerve injury pain, and 5% with sympathetically-maintained pain. Whereas nerve compression and nerve injury pain were caused most frequently by cancer growth, sympathetically-maintained pain was caused most frequently by cancer treatment. There were no significant differences in the use of analgesics, the mean pain intensity, or the efficacy of analgesic treatment among the three groups. Nerve injury pain and sympathetically-maintained pain were treated more frequently with adjuvant analgesics, especially antidepressants and anticonvulsants. The variety of different neuropathic pain syndromes should be separated in future studies of the efficacy of different treatment approaches. [Copyright &y& Elsevier]

Background: Prevention of inadvertent hypothermia is recommended for procedures >30 minutes because hypothermia increases the risk of myocardial ischemia, intraoperative blood loss, transfusion and wound complications. Therefore, short warming interruptions between pre-warming and intraoperative warming might result in lower hypothermia rates. The aim of this retrospective investigation was to determine whether the incidence of inadvertent intraoperative hypothermia was affected by the warming interruption.Methods: The lowest intraoperative body core temperature value and the warming interruption time were taken from anaesthesia records. Body core temperature was recorded continuously, and a patient was classified to be hypothermic if the lowest recorded temperature value was <36°C. Hypothermia rates and the correlation between warming interruption times and intraoperative hypothermia rates were calculated.Results: Five thousand eighty-four patients were analysed. The intraoperative hypothermia rate was 15.3%. Nineteen patients (0.4%) had a recorded temperature of <35.0°C. An increase in forced-air warming interruption time was significantly associated with an increase in intraoperative hypothermia rates (P < .0001). Patients with interruptions in forced-air warming >20 minutes showed significantly higher hypothermia rates than those with interruptions of ≤20 minutes (P < .0001).Conclusion: Intraoperative hypothermia rates increased significantly with longer forced-air warming interruptions between pre-warming and intraoperative warming. Summary: To avoid pathogen‐associated molecular pattern recognition, the hemibiotrophic maize pathogen Colletotrichum graminicola secretes proteins mediating the establishment of biotrophy. Targeted deletion of 26 individual candidate genes and seven gene clusters comprising 32 genes of C. graminicola identified a pathogenicity cluster (CLU5) of five co‐linear genes, all of which, with the exception of CLU5b, encode secreted proteins. Targeted deletion of all genes of CLU5 revealed that CLU5a and CLU5d are required for full appressorial penetration competence, with virulence deficiencies independent of the host genotype and organ inoculated. Cytorrhysis experiments and microscopy showed that Δclu5a mutants form pressurized appressoria, but they are hampered in forming penetration pores and fail to differentiate a penetration peg. Whereas Δclu5d mutants elicited WT‐like papillae, albeit at increased frequencies, papillae induced by Δclu5a mutants were much smaller than those elicited by the WT. Synteny of CLU5 is not only conserved in Colletotrichum spp. but also in additional species of Sordariomycetes including insect pathogens and saprophytes suggesting importance of CLU5 for fungal biology. Since CLU5a and CLU5d also occur in non‐pathogenic fungi and since they are expressed prior to plant invasion and even in vegetative hyphae, the encoded proteins probably do not act primarily as effectors. Abstract: Objectives: Collagen membranes are not limited to be occlusive barriers as they actively support bone regeneration. However, the impact of bone‐derived growth factors on their osteoconductive competence has not been examined. Methods: Twenty adult Sprague Dawley rats were included in the study. Calvaria defects with a diameter of five millimeter were created. The defect was covered with one layer of a collagen membrane previously soaked in conditioned medium of porcine bone chips or in culture medium alone. After 4 weeks, microcomputed tomography was performed. Undecalcified thin‐ground sections were subjected to light and scanning electron microscopy. Primary outcome parameter was the bone volume in the defect. Unit of analysis was the bone‐conditioned medium (BCM). Results: In the central defect area of the control and the BCM group, median new bone connected to the host bone was 0.54 and 0.32 mm³, respectively (p = .10). In the ectocranial defect area, the control group showed significantly more bone than the BCM group (0.90 and 0.26 mm³; p = .02). Based on an exploratory interpretation, the control group had smaller bony islands than the BCM group. Scanning electron microscopy and histology indicate the formation of bone but also the collagen membrane to be mineralized in the defect site. Conclusions: These results demonstrate that the commercial collagen membrane holds an osteoconductive competence in a rat calvaria defect model. Soaking collagen membranes with BCM shifts bone formation toward the formation of bony islands rather than new bone connected to the host bone. [ABSTRACT FROM AUTHOR]

High-voltage and high-current SiC bipolar diode modules are fabricated and characterized under static and dynamic conditions. The modules are built using 6 \times 6\ \mm^2 SiC chips that are fabricated on 3-in SiC substrates. Individual chips were also packaged in an ISOPLUS™ package and used to perform switching tests on the diodes. The modules have been fully characterized under static and dynamic conditions. These modules are targeted for high-voltage high-frequency applications as well as antiparallel diodes for 6.5-kV insulated gate bipolar transistors, insulated gate commutated thyristors, and injection enhanced insulated gate bipolar transistors. Single magnetic atoms on surfaces are the smallest conceivable units for two-dimensional magnetic data storage. Previous experiments on such systems have investigated magnetization curves, the many-body Kondo effect and magnetic excitations in quantum spin systems, but a stable magnetization has not yet been detected for an atom on a non-magnetic surface in the absence of a magnetic field. The spin direction of a single magnetic atom can be fixed by coupling it to an underlying magnetic substrate via the exchange interaction, but it is then difficult to differentiate between the magnetism of the atom and the surface. Here, we take advantage of the orbital symmetry of the spin-polarized density of states of single cobalt atoms to unambiguously determine their spin direction in real space using a combination of spin-resolved scanning tunnelling microscopy experiments and ab initio calculations. By laterally moving atoms on our non-collinear magnetic template, the spin direction can also be controlled while maintaining magnetic sensitivity, thereby providing an approach for constructing and characterizing artificial atomic-scale magnetic structures. The relative density of a sintered glass-powder compact varies at the local level due to its heterogeneous nature, which makes its characterization essential. In this work, high-resolution X-ray computed tomography (XCT) was used to quantitatively characterize the local relative density fluctuation of a sintered glass compact in its initial stage. The degree of heterogeneity of the microstructure was calculated taking into account the coarseness parameter. With a basis of 23,500 particles scanned in the representative volume element (RVE), the local environment was subdivided into three different groups of particles, represented by a cubic cell with a volume equivalent to that of approximately 600, 80 and 10 spherical particles with a diameter of 80 μm. Independent of the particle group analysed, we found that the maximum of the local relative density distributions, which could be described by a normal distribution function, always occurs around the relative density of the RVE. However, the fluctuation of the data showed a considerable change regarding the width, which increases as the volume of the analysed group of particles decreases, suggesting that particle packing is more heterogeneous on a small scale. In particular, we observed that the heterogeneity of the microstructure for the RVE under consideration becomes pronounced for groups of approximately 10 to 80 particles. • The limitations and preparation efforts of different microanalytical methods are discussed. • SEM on wet-etched blend film is sufficient to evaluate the bulk morphology. • ToF-SIMS imaging is a valuable method for the surface analysis of the polymer blend. • PP and HDPE distributions vary between the surface and bulk of HDPE/PP blend film. In this study, the morphology of an immiscible polymer blend system at various regions of interests was analyzed using different microanalytical methods with varying surface sensitivities. As a model immiscible polymer blend, a HDPE/PP (80/20 wt%) polymer film was used. The blend film was subjected to polarized light microscopy (PLM), scanning electron microscopy (SEM), atomic force microscopy (AFM), transmission electron microscopy (TEM) and time of flight secondary ion mass spectrometry (ToF-SIMS). The obtained results were compared regarding the sensitivities, informational values and overall applicability of the analytical methods. It was evaluated which methods can be applied for a fast analysis of the morphology (surface and bulk) of the immiscible polymer blend with low preparation efforts, which is especially important for the analysis of new materials, for example materials manufactured via recycling. It was demonstrated that PLM, as well as SEM on wet-etched material, provide sufficient information to evaluate the bulk morphology. Additionally, the presented study shows the advantage of applying ToF-SIMS imaging for the characterization of the surface of immiscible polymer blend. As expected, the domain distribution of HDPE and PP varied between the bulk and the surface of the films.
