SIRT6, a member of the sirtuin family of histone deacetylases, belongs to the class Ⅲ longevity proteins and exhibits NAD+-dependent deacetylase and mono-ADP-ribosyltransferase activities. SIRT6 is primarily located in the cell nucleus and plays a pivotal role in regulating genomic stability and relative gene expression, participating in the control of key processes such as energy metabolism and aging. Given its crucial role in maintaining cellular homeostasis and organismal health, SIRT6 has emerged as a potential therapeutic target, sparking significant research interest in the development of targeted modulators. Activating the longevity protein with drugs may provide therapeutic strategies for age-associated diseases, including aging, metabolic syndrome, inflammation, and reproductive health issues. The review elaborates the structural characteristics, enzymatic activities, and biological functions of SIRT6, as well as the mechanisms of action, pharmacological activities, and clinical applications of various SIRT6 activators.

The effects of three oxidation processes, ozone (O3) oxidation, O3/potassium peroxymonosulfate (PMS) oxidation, and O3/hydrogen peroxide (H2O2) oxidation, on the treatment of phosphating wastewater were compared. Additionally, the quality of effluent was analyzed through measurements of COD, UV254, and three-dimensional fluorescence spectra, highlighting the influence of various advanced oxidation processes on the wastewater effluent quality and the removal pathways. The results showed that increasing the ozone dosage and adding appropriate amounts of PMS and H2O2 could enhance the removal of organic contaminants from phosphating wastewater. With initial COD of (130±20) mg/L, reaction time of 120 minutes, and ozone dosage of 7.65 mg/min, the COD removal rate was only 37% by the ozonation process, the COD removal rate reached 45.65% by the O3/PMS oxidation process with PMS mass concentration of 2 g/L, the COD removal rate reached 58.3% by the O3/H2O2 oxidation process with 0.5 g/L dosage of H2O2 (30% concentration). Among the three methods, the O3/H2O2 process demonstrated the highest efficacy in removing COD from phosphating wastewater, as the synergistic production of more highly oxidative free radicals by O3 and H2O2 enhancd the oxidation capacity of the system. All three processes effectively degraded aromatic compounds and fluorescent substances in wastewater, while the individual O3 oxidation and the O3/PMS process mainly degraded substances resembling proteins into fulvic and humic-like materials, and the O3/H2O2 oxidation process could further degrade these fulvic and humic-like substances.

ObjectiveTo explore the causes of related medical damage risks and preventive measures by analyzing the identification results of medical damage in 20 urological death cases.MethodsA retrospective analysis was conducted on 20 death cases of medical damage identification involving urology diagnosis and treatment accepted by the Judicial Appraisal Center, School of Forensic Medicine, Guizhou Medical University, and Academy of Forensic Science from 2010 to 2023.ResultsAmong the 20 cases, the male-to-female ratio was 1.5∶1. The age of the identified persons ranged from 37 to 84 years old, with an average age of 59.2 years. There were 21 medical institutions involved, with 81.0% being tertiary medical institutions. The statistical results of the causes of death showed that 60.0% deaths were due to infectious shock or hemorrhagic shock.ConclusionThe causative potency analysis of medical damage in urological death cases is closely related to the patients’ diseases, the death consequences of medical damage, and medical errors. Analyzing characteristics of typical cases can improve the comprehensive analysis ability of forensic examiners in similar cases, help them understand the rationality and compliance of medical behaviors in different situations, in order to make more accurate and comprehensive judgments.

Achieving the best wideband stealth performance under the limited weight constraint is the goal pursued in stealth aircraft radar absorbing coating（RAC） application schemes design. To meet the need for the integrated application of various kinds of RACs on stealth aircraft， an automatic optimization method for the application programme of RAC was established. Firstly， the electromagnetic fields of the model under different frequencies， azimuths and polarizations were simulated based on the finite difference time domain method. The total electromagnetic field was obtained by normalization and weighted calculation of the electromagnetic fields.Then， the total electromagnetic field was divided according to the specific direction and quantity. Each partition was loaded with the corresponding RAC. The reflection coefficient of the RAC was calculated by the transfer matrix method to superimpose the effect of the RAC on the electromagnetic field. The near-far field transformation was performed on the total electromagnetic field after loading with RAC. In this way， fast computation of radar cross section （RCS） was implemented. Based on the improved simplex method and Karush-Kuhn-Tucker condition， an automatic optimization method is established. The objective function was reducing RCS of the target or reducing weight of the absorbing coating. Finally， taking traveling wave plate as an example， the application plan of three thicknesses of RACs was optimized. The goal of optimization was to reduce total weight of the coating. Compared with the 1.5 mm coating plan， the optimization program can reduce the weight by 50%， with the RCS of traveling wave plate increasing by on more than 1 dB. The optimization effect is remarkable.

This study explores the environmental changes in the disturbed area of coal mining and presents the driving factors of ecological self-restoration. Specifically, we measured the soil nutrient and soil enzyme indicators in the mining area and non-mining area from 2014 to 2017 and 2019, and evaluated the stoichiometric characteristics of soil carbon (C), nitrogen (N), and phosphorus (P). The experimental results show that the total phosphorus concentration gradually recovered with the increase of mining years, while organic matter, available potassium, and urease first increased and then decreased with the increase of mining years. There were significant differences in total phosphorus, C/N ratio, N/P ratio, and C/P ratio between the working face and the subsidence disturbance zone. Random forest analysis showed that soil sucrase, urease, available potassium, total nitrogen, total phosphorus, C/P ratio, C/N ratio, and water-soluble salts were the key factors influencing soil nutrient cycling in different time periods; while C/N ratio, N/P ratio, total phosphorus, alkaline phosphatase, and water-soluble salts were the key factors influencing soil nutrient cycling at different locations. Mining years of the working face has differing influence path on soil factors and soil enzymes. This study could provide theoretical proof for the artificial guided self-restoration plan in coal mining subsidence areas and for ecological restoration in western mining areas.

IntroductionPortal hypertension combined with hypersplenism (PHcH) is the main cause of hypocytosis and esophagogastric variceal hemorrhage in patients with liver cirrhosis. Activated macrophages that destroy excess blood cells are the main cause of hypersplenism, but the activating pathway is not very clear. This study aims to investigate the activation types of splenic macrophages and their activation mechanisms, to provide experimental evidence for the biological treatment of splenomegaly, and to find a strategy to improve liver fibrosis and inflammation by intervening in splenic immune cells. This study revealed the occurrence of M2-like polarization of macrophages and upregulation of c-Myc gene expression in the PH spleen.MethodsRNAseq, protein chip, western blot, and chip-seq were performed on macrophages and the in vitro MEK inhibitor rafametinib was used. Carbon tetrachloride and thioacetamide induced mouse cirrhosis models were separately constructed.Resultsc-Myc gene knockout in splenic macrophages reduced M2-like polarization and exacerbated liver fibrosis inflammation. c-Myc activated the MAPK signaling pathway and upregulated the expression of IL-4 and M2-like related genes in PH hypersplenism through the MEK-ERK-c-Myc axis. In addition, the c-Myc gene exerted anti-inflammatory effects by upregulating IL-4-mediated signal transduction to promote M2-like differentiation and anti-inflammatory cytokine secretion.ConclusionsActivation of MEK‐ERK‐c‐MYC signaling pathway promotes splenic M2-like macrophage polarization to inhibit PHcH-liver cirrhosis. Therefore, the induction of macrophage depolarization might represent a new therapeutic approach in the cure of PH hypersplenism, making c-Myc a potential candidate for macrophage polarization therapy.

Sand and dust storms are significant natural disasters which affect East Asia and China in spring, occurring from March to May. Performances of CMA-CUACE/Dust, an operational Asian sand and dust storm numerical forecasting system of CMA since 2006, are analyzed. It’s found that the model overestimates in Central Asia, underestimates in northern Mongolia, and diffuses too quickly in downwind areas far away from the source area, especially in Northern China, Korean Peninsula and Japan, resulting in low peak values or less lingering time there for very extreme sand and dust storm events. A scale-adaptive dust emission scheme is applied by resolving the mean wind speed of the model grid into sectional one which can account for values larger than the mean value by Weibull integration function. This significant aspect is crucial because the dust emission is the third power of the wind in the dust emission scheme. New wind erosion database is also adopted which consists of the updated desertification by using twenty-year surface data and new parameters deduced from site observations in the heart of Gobi Desert, determining the size distribution of the emitted dust together with the soil texture data sampling from main deserts in China.After evaluation for the strongest sand and dust episode of 13-17 in March 2021 in East Asia in the past decade, and the consistent run in the same operational environment from 1 March to 31 May in 2023, it is found that the updated CMA-CUACE/Dust effectively improves disadvantages such as overestimation in Central Asia, underestimation in northern Mongolia, and rapid dissipation in China. The predicted peak dust concentration of the extreme episode closely matches observations in China both in the source area and in Shanghai after a 4-day transportation. Threat score (TS) of three-month forecast run also indicates that the improved model shows good consistency and continuity in forecast results across various forecast lengths. TS for 1-5 days with different forecast lengths is significantly higher than that of the previous operational system and surpasses those of Korean dust model—ADAM (the Asian Dust Aerosol Model). Furthermore, the missing rate is significantly reduced, while the false alarm rate remains almost unchanged. TS for episodes beyond the level of sand and storms are all above 0.3, with some exceeding 0.5. All these findings show that the improved model performs much better than the previous one.

Cold, as a primary external factor in cold areas, necessitates the body’s autonomous adaptation to uphold optimal living conditions. The repercussions of cold on the body are both far-reaching and profound, with metabolic equilibrium adjustments playing a pivotal role. This article, rooted in the TCM principle of Yin-Yang balance, delves into the metabolic intricacies and adaptive responses to the human body in cold environments. The effects manifest in heat-producing tissues, systemic substance consumption, the blood substance concentrations, liver function, and metabolic rhythms. The article subsequently presents TCM recommendations for maintaining health in cold climates. It concludes by advocating the exploration of metabolic homeostasis changes as a key avenue for investigating the metabolic traits s of populations in cold regions. We posit that such insights will enhance comprehension of the metabolic shifts in cold region populations and advance the evolution of regional medicine.

Endocrine therapy plays a crucial role in hormone receptor-positive breast cancer and is closely related to the patient’s menopausal status. This manuscript aimed to explore two key questions in assessing menopausal status of breast cancer patients. Firstly, for premenopausal patients receiving ovarian function suppression (OFS) therapy, persistently elevated estradiol (E2) levels post-treatment may be attributed to either “true” or “pseudo” elevation, with caution needed due to potential interference from endocrine therapy drugs such as fulvestrant, abemaciclib, exemestane and tamoxifen. Secondly, when determining whether patients under OFS therapy have reached physiological menopause, factors such as the impact of chemotherapy on menopausal status and the complexity of hormone levels around menopause need to be considered. It is recommended to consider switching treatments based on the patient’s age and original endocrine therapy, and to regularly monitor levels of estradiol and follicle-stimulating hormone (FSH). In challenging cases, a comprehensive evaluation can be conducted by incorporating markers such as anti-Mullerian hormone (AMH), inhibin B (INHB) and androgen.

ObjectivesCalculate the carbon emission efficiency of listed thermal power companies, compare and analyze the reasons why the efficiency ranks at the bottom and the top driving factors, group thermal power companies according to different conditions, and explore the influencing factors of efficiency differences.MethodsBased on the improved three-stage undesirable-slack based measure (SBM)-data envelopment analysis (DEA) model to more accurately measure the total-factor carbon emission efficiency of China listed thermal power companies during 2016-2022.ResultsThe carbon emission efficiency of listed thermal power companies rises annually and lowers when the effects of the external environment are taken into account. The increasing environmental subsidies from government may lead to excessive investment of capital, personnel and coal consumption in the thermal power companies. The increasing in regional R&D investment has a beneficial impact on company assets, personnel, and coal consumption. The most effective utilization of company assets and employees management will be promoted by a better regional economic level, but wasteful consumption of coal for power generation will occur.ConclusionsThe listed thermal power companies with central control, low coal consumption rate and carbon emissions trading have higher technology gap rate and better overall carbon emission efficiency.

This paper takes the sluice of the main channel on the east side of East Airport Road, Jinwan Coastal Business District, Zhuhai City, Guangdong Province as an example. Given the wind load on the sluice structure, the paper compares the calculation methods of the design standards in China, the UK, the US, and Japan and analyzes the differences in calculation methods among the four countries. Based on the numerical simulation of computational fluid dynamics, it studies the distribution of wind load on sluice structure in different wind directions, standard wind pressure, and extreme weather such as typhoon. With the structural finite element method, the force characteristics of sluice structure are analyzed under different wind load conditions, which provide some reference for the wind resistance design of sluice structure in coastal areas. The research results show that the wind load value of the Chinese standard considering the wind vibration coefficient is basically the same as that of the UK, the US, and Japanese standards. The wind direction coefficient of the sluice is between 0.75 and 1.03, and the wind load can be multiplied by a certain wind reduction factor according to the angle between the sluice and the dominant wind direction. The shape factor of wind load on the leeward side of the sluice structure is larger than that of general buildings. It is suggested to define the sluice as wind-sensitive building for wind load calculation. Under extreme weather such as typhoons, the wind load of the sluice structure should be multiplied by a certain amplification coefficient considering the wind vibration coefficient, and the stiffness of the sluice structure should be appropriately increased to satisfy the requirements of normal use. Considering the superimposed effect of offshore tide level and storm surge, it is recommended to multiply the bearing capacity of the sluice structure foundation by a safety factor of not less than 1.15.

Oil-immersed power transformer is an important part of power grid, and its reliable operation plays a vital role in pomler system security. Aiming at the problem that the insulation state of transformer cannot be assessed quantitatively after long-term operation, the accelerated aging and damp tests of oil-paper insulation model are carried out in this paper. The influence of aging and damp of oil-paper insulation on its recovery voltage curves is explored. The particle swarm optimization (PSO) is used to improve the parameter prediction method of extreme learning machine (ELM), which realizes the quantitative assessment of aging and moisture of oil-paper insulation based on the characteristic parameters of the recovery voltage curve. By comparing the physical and chemical performance analysis of oil-paper insulation models, it is shown that the prediction accuracy of PSO-ELM method is much higher than that of traditional ELM method. The absolute error range for predicting the moisture content of oil-paper insulation the degree of polymerization (DP) of pressboard is less than ±0.4% or ±30, respectively.

Pelvic exenteration is often required for primary rectal cancer beyond total mesorectal excision (PRC-bTME) and locally recurrent rectal cancer (LRRC). Some patients with radical resection can achieve long-term survival, but they need to face risks, such as huge surgical trauma, serious perioperative complications, permanent loss of organ function and decline in quality of life. Preoperative evaluation of PRC-bTME and LRRC should emphasize multidisciplinary collaboration and develop individualized diagnosis and treatment strategies. The principles of function preservation and risk-benefit balance in surgery oncology should be followed, and R0 resection should be emphasized. Perioperative complications, surgical trauma and organ function loss should be minimized to achieve the best quality control and balance point. This consensus was formulated by the Colorectal Cancer Committee of the Chinese Medical Doctor Association and the Gastrointestinal Surgery Committee of China International Exchange and Promotive Association for Medical and Health Care. The draft was formed based on the summary of domestic and foreign research progress and expert experience. After discussion, review and modification by experts, an anonymous voting was conducted for each major opinion, and in-depth verification was carried out according to the principles of evidence-based medicine. Finally, the Chinese expert consensus on the pelvic exenteration with primary rectal cancer beyond total mesorectal excision planes and locally recurrent rectal cancer (2023 edition) was formed. This consensus mainly summarizes the indications and contraindications of pelvic exenteration (PE) for PRC-bTME and LRRC, preoperative diagnosis and evaluation, perioperative treatment, as well as the resection scope, surgical methods, reconstruction of related organs, safety and complications of PE, postoperative follow-up and other issues, in order to provide guidance for PE in patients with PRC-bTME and LRRC.

Deep brain stimulation (DBS) has been widely used for Parkinson's disease, essential tremor, dystonia, drug - resistant epilepsy and other diseases. The operation procedure of this surgical method is complicated, and it also has certain complications in clinical application, including surgical complications such as intracranial hemorrhage and infection, hardware complications such as lead fracture and displacement, and stimulation complications such as paresthesia and muscle spasm, in which lead fracture and erosion and infection of the skin behind the ear are common and troublesome complications. In clinical practice, we found that lead fracture and erosion and infection of the skin were mainly located behind the ear. Since 2012, Beijing Tiantan Hospital, Capital Medical University had applied the retroauricular bone grinding groove technology in DBS, which significantly reduced the incidence of lead fracture and erosion and infection of the skin. Therefore, the retroauricular bone grinding groove technology can effectively solve the above problems. This paper mainly introduces the retroauricular bone grinding groove technology and the clinical application attention matters, providing references for colleagues.

Pipeline transportation is an economical and effective way for transferring carbon dioxide (CO2) to the sea, serving as a key procedure for an offshore carbon capture, utilization and sequestration (CCUS) project, as well as a core technology for the large-scale construction of CCUS projects in China. This study clarifies the advantages of China in the construction of offshore CCUS projects, typical offshore carbon pipeline scenarios, and typical offshore CO2 transportation modes. It also reviews the technologies and projects in China and abroad regarding offshore CO2 transportation via pipelines. The current technologies relevant to offshore CO2 pipeline transportation are systematically reviewed. Specifically, the process technologies include CO2 fluid state analysis and flow assurance; corrosion evaluation, monitoring, and early warning; real-time monitoring of pipe leakage; and release of high-pressure CO2 and its environmental impacts. The material technologies include the fracture of pipeline materials and its mitigation, high corrosion-resistant and sealing materials, key corrosion-control techniques for the long-term operation of pipelines, and corrosion risk evaluation of CO2 injection wells. Further efforts should focus on the following aspects: material selection systems for the complex conditions during offshore CO2 pipeline transportation, full-chain intelligent management and digital twin technologies for CO2 pipelines, key technologies regarding the whole life-time operation of subsea CO2 pipelines, and evaluation and assurance techniques for the transferred transportation pipelines. Furthermore, the following suggestions are proposed to promote the high-quality development of the offshore CO2 pipeline transportation system in China: (1) promoting the planning of offshore CO2 pipeline networks, (2) expanding interdisciplinary innovations, (3) establishing standards systems that applicable to both onshore and offshore scenarios, and (4) encouraging the participation of diversified technology service enterprises.

Abstract [Objective] The study aims to explore the effects of salt stress on the growth and secondary metabolites of Periploca sepium seedlings, and preliminarily understand the salt tolerance range of P . sepium and the induction effect of salt stress on secondary metabolites of P . sepium . [Methods] This experiment was conducted with the seedlings of P . sepium cultivated for 30 days to determine the changes in biomass, total steroids components, cardiac glycosides (periplocin, periplocymarin, and periplogenin), and 4-methoxy salicylaldehyde content of P . sepium seedlings treated with different salt concentrations (0, 0.2%, 0.5%, 1.0%, 2.0%). [Results] The P . sepium seedlings grew well under low concentration of salt stress, and the dry and fresh biomass of the aboveground and underground parts were increased to varying degrees. The dry to fresh ratio was gradually increased with the increase in salt stress levels. The content of the major effective components in plants, such as total steroids and cardiac glycosides, showed a significant upward trend under moderate salt stress, while 4-methoxy salicylaldehyde was significantly increased under high salt stress. [Conclusion] P . sepium has strong salt tolerance, and salt stress within a certain range increases the content of major secondary metabolites in the roots and stems of P. sepium seedlings.

Online social networks, especially Twitter (X), have become focal points for illicit activities, providing unique criminal investigation opportunities. This paper introduces an innovative methodology that uses social media sentiment analysis to predict criminal activities. One major challenge in sentiment analysis is the uneven distribution of sentiment classes, where traditional models often fail to accurately classify instances of the minority class due to the overwhelming presence of majority class data. To tackle this issue, we propose a model that combines a reinforcement learning (RL) algorithm with a scope loss function. The RL approach uses a reward mechanism that assigns a more significant value to correctly predicting minority class instances over majority class ones. The scope loss function ensures an optimal balance between utilizing known data and exploring new data, thus maintaining a delicate equilibrium between accuracy and generalizability. Our model employs a series of convolutional neural networks (CNNs) to extract significant features from textual content, which are then utilized for sentiment classification. We also incorporate an advanced artificial bee colony (ABC) optimization technique to refine the model's hyperparameters. The effectiveness of our approach was empirically tested using two distinct datasets: one consisting of crime incident reports from the Chicago Police Department covering the period from September 2019 to July 2024 and another comprising tweets containing crime-related terms related to Chicago. The predictive capabilities of our proposed model were benchmarked against existing models, demonstrating superior performance with accuracies of 96.411% and 94.088%, respectively. This breakthrough highlights the potential of integrating sentiment analysis with reinforcement learning to significantly enhance the predictive accuracy of crime-related activities in online social networks, offering valuable insights for law enforcement and criminal investigation applications.

Chestnut rose fruit, known as "Three King fruits", has high edible value and medicinal value. Polyphenols are the second major functional component of chestnut rose fruit with pharmacological and health functions such as antioxidant, regulation of glucose and lipid metabolism, anti-inflammatory and bacteriostatic, antialcoholism and liver protection, and have great potential for food development. This review summarized the research progress of the classification, extraction, qualitative and quantitative analysis, and bioactivity of chestnut rose fruit polyphenols in recent years and prospected the future development direction.

Treatment of breast cancer with low expression of human epidermal growth factor receptor 2 (HER2; HER2-low) has drawn much attention in recent years. With the proven therapeutic effect of trastuzumab deruxtecan (T-DXd) in patients with HER2-low (immunohistochemistry [IHC] 1+, or IHC2+/in situ hybridization [ISH]-) breast cancer, HER2-low may become a new subtype of targeted therapy for breast cancer. The expert committee formulated this consensus based on the current clinical studies and clinical medication experience. The current consensus is the collaborative work of an interdisciplinary working group, including experts in the fields of pathology and oncology. The purpose of this consensus was to guide the clinical diagnosis and treatment of HER2-low breast cancer, thereby prolonging the overall survival of patients.

Long noncoding RNAs (lncRNAs) are important regulators of bone metabolism. In this study, lncRNA microarray analysis was used to identify differentially expressed lncRNAs in differentiated osteoclasts. lncRNA-Gm5532 is highly expressed during osteoclast differentiation. lncRNA-Gm5532 knockdown impairs osteoclast formation and bone resorption. Mechanistic experiments show that lncRNA-Gm5532 functions as a competing endogenous RNA (ceRNA) and acts as a sponge for miR-125a-3p, which promotes TNF receptor-associated factor 6 (TRAF6) expression. miR-125a-3p mimics suppress osteoclast differentiation and TAK1/NF-κB/MAPK signaling. In summary, our study reveals that lncRNA-Gm5532 functions as an activator in osteoclast differentiation by targeting the miR-125a-3p/TRAF6 axis, making it a novel biomarker and potential therapeutic target for osteoporosis.

Electromagnetic metamaterial is an artificial composite composed of periodic subwavelength microstructure，which has a strong conduction regulation or absorption effect on electromagnetic wave，and it has been widely studied in the field of stealth design of aviation weapons and equipment. In this paper，the concept of electromagnetic metamaterial is introduced firstly，and the latest research progresses of electromagnetic control metamaterial，electromagnetic absorbing metamaterial，active tunable metamaterial and intelligent metamaterial are reviewed. Then，the stealth mechanism and the application research status of electromagnetic deflection metamaterial，electromagnetic absorbing metamaterial and frequency selective metamaterial are introduced. It is analyzed that the rich stealth mechanism and the strong designability are the main advantages of electromagnetic metamaterials as distinct from the traditional absorbing materials. For the development of electromagnetic metamaterial，this paper puts forward three suggestions from expanding the absorbing spectrum，enhancing the absorbing performance，and smart tunable of absorbing ability，specifically including the expansion of absorbing spectrum to infrared，laser and ultraviolet band，further improving the broadband absorbing ability，and intelligent adjusting the absorbing frequency band.

Aimed at the problem that the accuracy of tool remaining life prediction was reduced due to the abnormal monitoring data in machining process, a data anomaly detection method based on self-paced learning was proposed. First, a multi-layer perceptron model was established to correlate the tool processing monitoring data with the tool remaining life. Next, in the process of updating model weight, the model weight parameters were fixed first, and the loss threshold of abnormal samples was obtained by predicting loss fitting Gaussian distribution. Then, the loss function based on the self-paced learning method was constructed to update model parameters iteratively. At the end of the model training, abnormal samples were divided according to the loss threshold. Finally, the actual processing monitoring data of turbine rotor groove were used to verify the proposed method, and compare with the local anomaly factor algorithm, the density-based clustering algorithm, the K-means algorithm, the isolated forest algorithm, and the one-class support vector machines.

At present, the performance demand of power communication service for bearer network is constantly increasing. Software Defined Optical Network (SDON) technology drives the integration of power communication and SDON technology with its advantages of openness and softwareization. The index system and evaluation method established based on traditional optical transmission network cannot reflect the characteristics of SDON architecture and its influence on bearer performance of power communication network service. In this paper, a new index system is constructed based on the performance requirements, influencing factors and the influence of different SDON architectures on the service performance of power communication business. The decision matrix was determined according to the contribution degree of the index to each attribute. The Ordered Weighted Average-Extended-Technique for Order of Preference by Similarity to Ideal Solution (OWA-E-TOPSIS) multi-attribute decision making method was proposed to realize the index screening. The weight distribution of the selected indicators is carried out by Analytic Hierarchy Process (AHP), and the examples are collected. After the pretreatment by utility function method, the index value aggregation is carried out. By comparing the evaluation results of each instance, the feasibility of the index system and evaluation method are verified, which provided a basis for the application of new technology and new business support of electric power communication network.

Intrahepatic cholangiocarcinoma (iCCA) is a highly fatal malignancy with rapidly increasing incidence and mortality worldwide. Currently, gemcitabine-based systemic chemotherapy is the main clinical therapeutic regimen; however, its efficacy is poor, and its mechanism has not been elucidated. In this study, we use a Seahorse Extracellular Flux analyser to measure glycolysis capacity (extracellular acidification rate, ECAR) and oxygen consumption rate (OCR). The glucose uptake or lactic acid content is detected, and the effects of saikosaponin D, an active compound derived from Bupleuri Radix (a traditional Chinese medicine for soothing the liver and relieving depression), on gemcitabine cytotoxicity in norepinephrine-stimulated iCCA cells are analysed. We find that adrenergic signaling plays a fundamental role in chronic stress-induced therapeutic resistance in iCCA. Norepinephrine (NE) and epinephrine (E) enhance the proliferation of iCCA cells and interfere with the response to gemcitabine through activation of the β2-adrenergic receptor (ADRB2). Furthermore, we find that NE upregulates the expressions of several drug efflux-related genes (such as ABCG2 and MDR1) and promotes glycolysis in iCCA cells. In addition, saikosaponin D reverses the poor response of iCCA cells to gemcitabine by downregulating ADRB2 level. Furthermore, saikosaponin D inhibits drug efflux and glycolysis in iCCA cells by regulating the expressions of MDR1, ABCG2, HK2, and GLUT1. Collectively, saikosaponin D enhances the antitumor effect of gemcitabine by controlling glucose metabolism and drug efflux by inhibiting the ADRB2 signaling. Therefore, the combination of saikosaponin D and gemcitabine may be a potential therapeutic strategy for the treatment of iCCA.

Our study evaluated the risk and benefits of active strategies including ultra-early surgery and hematoma evacuation through a transsylvian-transinsular approach for moderate basal ganglia hemorrhage.

Objective To explore the mechanism of Artemisia annua L. (A. annua) in treatment of Mycobacterium abscessus (M. abscessus) infection with aid of network pharmacology and molecular docking. Methods The active components of A. annua were obtained from the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform (TCMSP) system, and the targets of A. annua were predicted using PharmMapper and SwissTargetPrediction. Targets related to M. abscessus infection were obtained from GeneCards database and OMIM database. After obtaining the common targets of them, a target protein-protein interaction (PPI) network was constructed using the STRING database and Cytoscape software. Then Metascape database was employed to perform Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. The main potential active components of A. annua were screened out for molecular docking with the core targets. The extracts of A. annua and the main potential active ingredients screened out were tested in vitro to explore its minimum inhibitory concentration (MIC), its combined efficacy with common anti-M. abscessus antibiotics in vitro, and its effect on the viability of M. abscessus. Results A total of 48 targets of A. annua were found to be involved in the pathogenesis of M. abscessus infection. A total of 6 targets, MMP9, MAPK3, CASP3, IL2, CASP1, and MMP2, were predicted to be the core targets of A. annua in the treatment of this disease. KEGG enrichment analysis showed that A. annua may treat this disease through IL-17 signaling pathway, neutrophil extracellular trap formation, Toll-like receptor signaling pathway, tumor necrosis factor signaling pathway, MAPK signaling pathway and other inflammatory and immune-related signaling pathways. Molecular docking results indicated that A. annua had a high affinity with these core targets. Tamarixetin, artemetin, dihydroartemisinin and Artemisia annua L. were screened to have antibacterial activity against M. abscessus in vitro, with a MIC value of 200, 100, 100 and 100 μg/mL, respectively. After combined with the exacts of A. annua, the MIC value of clarithromycin, azithromycin, amikacin, imipenem, and cefoxitin was all decreased than that of their single use, with a declining multiple ranging from 2 to 8 times. Conclusion A. annua may act on M. abscessus infection through multiple targets and multiple pathways. It has antibacterial effect in vitro and shows a synergistic effect with some antibiotics against the bacteria.

Objective To summarize the clinical characteristics of posterior reversible encephalopathy syndrome (PRES) in children with renal disease in order to provide reference for early prevention, diagnosis and treatment and improving prognosis. Methods A retrospective analysis was conducted on the clinical characteristics, auxiliary examinations, medication use and prognosis of children with renal disease complicated with PRES confirmed in the First Affiliated Hospital of Henan University of Chinese Medicine from 2015 to 2022, and the relevant literatures were reviewed. Results Thirteen children aged 3 to 14 (8.38±3.36) years old were enrolled, including 9 males and 4 females. There were 10 cases (76.92%) of infection before the onset of the disease, 10 cases (76.92%) of lower serum albumin levels than normal, 4 cases (30.77%) accompanied by varying degrees of hypertension, 10 cases (76.92%) treated with glucocorticoids (GC) combined with immunosuppressive agents, 2 cases (15.38%) treated with GC and 1 case (7.69%) treated with immunosuppressive agents. The main clinical manifestations were seizures, disturbance of consciousness, nausea and vomiting, dizziness, headache and ocular discomfort. The lesions were mostly distributed in bilateral frontal lobe, parietal lobe, temporal lobe, occipital cortex and subcortex. After treatment, the clinical manifestations and imaging findings returned to normal in a short period of time. Conclusion The incidence of PRES in children with renal disease is related to GC and immunosuppressive therapy, and cyclic lipopeptide antibiotics, hypoproteinemia and hypertension may increase the risk of disease. PRES usually is reversible and has a good prognosis, but delayed treatment can worsen progressively and even cause irreversible craniocerebral nerve damage.

Objective·To compare the pregnancy outcomes of infertile women with bilateral severe hydrosalpinx receiving neosalpingostomy or salpingectomy.Methods·The single-center prospective cohort study from 2005 to 2012 focused on pregnancy outcomes of infertile women aged 20‒40 years, with bilateral severe hydrosalpinx, undergoing bilateral neosalpingostomy or salpingectomy in International Peace Maternal and Child Health Hospital, Shanghai Jiao Tong University School of Medicine. The choice for treatment was based on a shared decision approach, and the participants were divided into the neosalpingostomy group and salpingectomy group. After registration of baseline characteristics, including age, birth place, reproductive history, preoperative hysterosalpingography results, surgical findings, and pregnancy outcomes, women were followed up on an annual basis until July 2020 for the occurrence of live birth by outpatient follow-up or telephone questionnaire. Intention-to-treat analysis and per-protocol analysis were applied to compare the pregnancy outcomes. Kaplan-Meier analysis and COX proportional hazard model were used to analyze the reproductive outcomes. In addition, subgroup analysis was performed based on age stratification. The main outcome measures were live birth rate, cumulative live birth rate, and factors affecting live birth. Secondary outcome measures included the mode of conception, time to live birth, biochemical pregnancy rate, clinical miscarriage rate, and ectopic pregnancy rate.Results·A total of 113 women were included in the analysis, 58 women underwent bilateral neosalpingostomy, and 55 women underwent bilateral salpingectomy. The study demonstrated that in infertile women with bilateral severe hydrosalpinx, bilateral salpingectomy achieved higher cumulative live birth rate than bilateral neosalpingostomy (76.36% vs 62.07, HR=2.18,95%CI 1.37‒3.45). In the neosalpingostomy group, 34.48% (20/58) live births were obtained after in vitro fertilization treatment, and 27.59% (16/58) live births were obtained through spontaneous conception which mainly occurred within 3 years after initial neosalpingostomy, while all live births in the salpingectomy group were obtained after assisted reproductive therapy. It is of great significance to integrate traditional native music into the piano teaching system in colleges and universities. Under the guidance of the CIPP concept, the study takes piano performance teaching in a city art college as an example, constructs the evaluation index system of piano performance education and local music culture, respectively, and assigns the indexes. The development level of piano performance education and local music culture is measured using the gray GM (1,1) comprehensive evaluation method, by which the degree of coupling and coordination between the two systems is measured, and the coupling development level of piano performance education and local music culture is explored. The Gray correlation model is then used to analyze the influencing factors of the coupling development of the two. The overall coupling development of piano performance education and local music culture in the sample art colleges is at the primary level of coordination, with 26.67% of the courses in the lower intermediate level of coordinated development and 26.67% of the courses in the high level of coordinated development, and nearly 50% of the courses in the barely coordinated and dysfunctional development. Content and form innovation, curriculum innovation, music culture presentation, and creative activity atmosphere are the most important influences on the coupled development of the two systems, with correlations above 0.9. Diversified teaching contents and methods need to be actively sought to improve the quality of piano teaching and enhance the effect of local music culture integration.

Carbon emissions from the power sector account for about one-third of China’s total carbon emissions, and carbon emission reduction in the power sector is crucial to the realization of the “double carbon” goal. This paper proposes a prediction model for grid carbon emission factor based on the combination of multiple linear regression models and the GM(1,1) model. The grid carbon measurement model is built using the theory of carbon emission flow, and the grid carbon emission accounting model is built using the consumption side. The average grid carbon emission factor is calculated by dividing the grid carbon emission factor into three different dimensions. The multiple linear regression model is used to study the correlation between the independent variables and the dependent variables, and the specific values of each regression factor are predicted by combining with the GM(1,1) model, and it is judged whether the prediction model is reasonable or not. Among the contributions of the increase in carbon emissions from the power grid, the contribution of electricity consumption increased from 2010 to 2014 and reached a peak of 2.9824 million tons in 2014, and the carbon emission factor value of the power grid gradually decreased from 0.719kg CO2/(kW·h) in 2010 to 0.593kg CO2/(kW·h) in 2022. The MR-GM(1,1) model is applied to the prediction of carbon emissions from power grids, and the absolute error of the results is within 15,000 tons, and the maximum relative error is only 2.42%. The calculation and prediction of carbon emission factors of the power grid can help power grid enterprises to clarify the trend of carbon emission, which is conducive to the realization of the low-carbon goal of “carbon neutral” and “carbon peak”.

Point cloud segmentation, as a key link in 3D point cloud data processing, can realize power transmission and distribution positioning, object identification, classification, and feature extraction, which helps to refine power grid management. In this paper, firstly, dense point cloud transmission and distribution 3D digital corridor modeling is carried out. Alignment splicing and noise reduction are carried out after obtaining the original dense point cloud. Contour line extraction, geometric modeling, and texture mapping are realized after processing the data to ultimately realize the transmission and distribution of 3D digitization. Then, the conversion formula for the pixel coordinate system and world coordinate system is derived to extract features from point clouds. Finally, a distance-based feature fusion method is designed to extract spatial features from point clouds and use the joint attention layer to segment them by fusing RGB pixel information. The original dense point cloud of a transmission and distribution digital corridor is segmented using the model presented in this paper for application after testing the dataset. It is found that the under-segmentation ratio of this paper’s algorithm is 0.96%, 3.44%, and 2.87% for the three scenarios of regular, irregular, and multi-targets, respectively, which is much lower than that of RANSAC+ECS with FCM + ECS. The intersection and concatenation ratios of this paper’s algorithm for the scenarios of irregular geometry as well as multi-target objects are 91.49% and 89.56%. It is much higher than 64.31% and 72.17% for RANSAC + ECS and 76.85% and 60.91% for FCM + ECS, which illustrates that this paper’s algorithm has a significant advantage in performance. In this study, the target point cloud can be segmented with high accuracy from the dense point cloud of a 3D model of power transmission and distribution with a large amount of data, effectively avoiding the phenomenon of under-segmentation and over-segmentation and contributing to the accurate control of power grid data.

Amidst the ongoing evolution and substantial reforms within the electric power market, the development of an auxiliary service pricing model grounded in game theory emerges as crucial. This study delineates the construction of an electricity auxiliary service pricing model, utilizing a dual mechanism approach: a cooperative game-based electricity price formation mechanism and a Stackelberg game-based time-sharing pricing mechanism. Furthermore, it incorporates demand response technology to conduct a detailed analysis of optimization results and the applicability of the proposed electricity service pricing model. The pricing scheme designated as Scheme 4, derived from the proposed model, demonstrates notable superiority in terms of economic efficiency and environmental sustainability when juxtaposed with three alternative schemes. Specifically, Scheme 4 yields a net profit of $13,267.6, achieves clean energy utilization amounting to 89.67 MWh, and minimizes wind abandonment to 17.35 MWh, outperforming all other considered scenarios in these metrics. Operational analysis reveals that the model's execution time varies between 15 and 50 seconds across different sample sizes, exhibiting minimal fluctuations. Additionally, the Monte Carlo simulations consistently produce values inferior to the objective function value of the developed model, with the discrepancy narrowing from 38 to 20, indicating the model's robust adaptability.

To reduce the emission of fine particulate matter (PM) from gasoline engines, a gasoline particulate filter (GPF) filtration model that considers the fractal characteristics of both particulate matter and filters is designed and its filtration characteristics are analyzed based on several GPF filtration models. Based on the GPF filtration mechanism, a new mathematical model of fiber efficiency filtration is established after correcting the classical filtration formula based on correcting the particle size of particles and considering the relationship between the porosity of the fiber filter and the fractal dimension of the filter. Simulation of the filtration efficiency of the established mathematical filtration model is carried out to obtain visual filtration efficiency curves. The filtration efficiency and pressure drop of the model are analyzed, and the effects of parameters such as particle fractal dimension, filter fractal dimension, filter thickness, flow rate, and fiber diameter on filtration efficiency are also comprehensively analyzed. The results show that when the fractal dimension of particles is in the range of 1.8-2.8, the most penetrating particle size decreases with the increase of the fractal dimension of particles. When the particle size is small, the effect of fractal dimension on filtration efficiency is not obvious. When the particle size is larger than the most penetrating particle range, the larger the fractal dimension, the filtration efficiency increases obviously. Considering the relationship between the fractal dimension of the filter body and the porosity and the influence of each on the filtration efficiency, the optimal fractal dimension of the filter body should be between 1.82 and 1.925, and the thickness of the filter has some influence on the filtration efficiency. The slower the gas flow rate is, the better the trapping effect will be. However, considering the problem of automobile emission, a lower exhaust speed can be chosen. Increasing the fiber diameter can improve the trapping efficiency of the model, but will reduce the porosity of the trapping collective. Therefore, the fiber diameter should not be too large under the combined action.

This paper first explores the composition of visual elements in modern digital media artworks, extracts the graphical element features of visual elements by improved SIFT algorithm, and classifies and recognizes the graphical elements using by SVM algorithm. Secondly, the extracted and categorized graphical elements are represented by Laplace feature vector correlation spectra in combination with spectral graph theory to study the mutual relationships between the graphical elements. Finally, some graphic elements in modern digital media artworks are used as examples to explore the performance and interrelationship of graphic feature extraction, recognition, and classification. The results show that the vector eigenvalues of spectral graph theory are categorized into [0], (0,100], (100,200], [200, ∞), and the corresponding interrelationships are one-to-one, one-to-many, many-to-one, many-to-many, respectively.

Introduction: The hot water chemical flooding technology effectively improves the fluidity ratio and enhances the horizontal and vertical oil displacement efficiency of the reservoir through the synergistic effect of thermal energy and chemicals, which can further enhance the recovery rate on the basis of throughput thermal recovery.Methods: Based on the geological reservoir parameters of Bohai B oilfield, an oil recovery system evaluation experiment and scheme design study were carried out. Injection parameters such as hot water temperature, chemical agent concentration, and chemical agent dosage that affect the oil recovery effect of hot water chemical flooding were optimized and designed. The development characteristics and effects of two thermal recovery technologies, namely multi thermal fluid throughput and hot water chemical flooding, were compared and analyzed.Results: The results show that with the increase of temperature, the effect of chemical agents on improving the water-oil mobility ratio and reducing the viscosity of crude oil is weakened, and the thermal energy plays a leading role in enhancing oil recovery, but when the temperature is higher than 115°C, the improvement of oil recovery is slowed down, and the comprehensive economic indicators decline rapidly. As the concentration of chemical agent increases, the enhanced oil recovery increases monotonously. When the concentration is 1500 mg/L, the comprehensive economic indicators reach the peak and then decline, indicating that the efficiency of chemical agent decreases. The total amount of chemical agent shall be controlled within 0.25 ~ 0.30 PV. The case calculation shows that hot water chemical flooding can expand the effective radius from 80 m to 400 m, more effectively use the reserves between injection and production wells, increase the oil recovery by 4.8 percentage points, and increase the oil yield by 31.7 m3/t per ton of agent.Discussion: It is suggested that Bohai B Oilfield can adopt hot water chemical flooding after the multiple thermal fluid huff and puff to continuously guarantee the oilfield production.

Baicalin is a small molecule medication used to treat hepatitis. Our research group discovered that administering baicalin orally to mice following fecal microbiota transplantation from patients resistant to ICIs supported anti-PD-1 activity. However, the precise mechanisms behind this effect are presently unknown. In this present study, ATB-treated C57BL/6 J mice received FMT from patients with advanced NSCLC amenable to αPD-1. Additionally, subcutaneous LLC cells were injected into the mice. Baicalin oral gavage and αPD-1 injection were administered to the mice on days 3 and 9 after tumour inoculation. 16 S rRNA, metabolomics, and flow cytometry were utilized to clarify the mechanisms of baicalin's relief of immunosuppression. The results indicated that oral administration of baicalin enriched bacteria such as Akkermansia and Clostridia_UCG-014, resulted in an increase in SCFAs, which improved the ratio of PD-1+ (CD8+ T cell/Treg) and promoted the levels of IFN-γ+ CD8+ T cells and TNF-α+ CD8+ T cells within the tumour microenvironment. In conclusion, baicalin regulates the metabolites of the gut microbiota to improve the PD-1+ (CD8+ T cell/Treg) balance and circumvent anti-PD-1 resistance. This is achieved through the regulation of short-chain fatty acids.

Objective: The aim of this study was to investigate the endometrial proteomic profiles of patients with polycystic ovary syndrome (PCOS) with and without insulin resistance (IR). Method of Study: We collected 40 endometrial samples, including PCOS-IR (n = 21), PCOS-non-IR (n = 12), and control (n = 7). Data-independent acquisition (DIA)-based proteomics method is used to identify the expressed proteins among the three groups. The correlation between pregnancy outcomes and identified proteins was analyzed by Lasso regression. Results: A total of 5331 proteins were identified, while 275 proteins were differentially expressed in the PCOS vs. control group and 215 proteins were differentially expressed in the PCOS-IR vs. PCOS-non-IR group. Platelet degranulation, neutrophil degranulation, and very long-chain fatty acid catabolic processes have been found to play important roles in the endometrium of patients with PCOS-IR. Lasso regression analysis found that ACTR1A, TSC22D2, CKB, ABRAXAS2, and TAGLN2 were associated with miscarriage in patients with PCOS. Introduction: Through the combination of hot water and chemical agent, the low-heat synergistic chemical profile control and flooding technology can not only reduce the risks of sand production and string damage caused by high-temperature thermal recovery technology, but also further improve the recovery factor on the basis of hot water flooding.Methods: Based on laboratory testing, the synergism mechanism of thermal energy and chemical agent is studied.Results: The research results showed that in the near wellbore zone of the injection well, crude oil viscosity reduction mainly relies on thermal energy, and chemical agents assist in improving the water oil mobility ratio and reducing the risk of cross flow. After entering the deep formation, due to the decrease in temperature, the thermal energy effect is weakened, but the chemical agents still aggregate at the oil-water interface and continue to act on heavy oil to reduce viscosity. The higher the temperature, the greater the role of heat energy, and the weaker the role of chemical agents. The higher the injection concentration of chemical agent, the more obvious the leading role of chemistry. Based on the comprehensive evaluation index, the optimized temperature is 80°C and the injection concentration is 1500 mg/L. The scheme study of Bohai L Oilfield shows that compared with cold water flooding, hot water flooding can only improve oil recovery by 1.2 percentage points, and low heat collaborative chemical profile control flooding technology can further improve oil recovery by 4.7 percentage points on the basis of hot water flooding, with obvious oil increase effect.Conclusion: It is suggested that Bohai L Oilfield should apply low-heat coordinated chemical profile control and flooding technology to improve the development effect of the oilfield, and provide new technical ideas for safe and economic development of offshore heavy oil.

Intelligent backfilling mining meets the industry development trend and need of green and intelligent mining in coal mines. But there is currently a lack of systematic research and practical engineering applications on key technologies of intelligent backfilling mining. In order to solve this problem, with the core of intelligent upgrading of solid backfilling mining faces, the primary goal of intelligent control of backfilling rate, and the goal of building intelligent backfilling mines, the key technologies of intelligent backfilling mining are systematically studied. The technologies include intelligent pretreatment and delivery control technology for coal-based solid waste, the underground intelligent jigging separation technology combined with real-time monitoring and control of ore deposits, intelligent control of waste discharge speed, and mixed separation control, the multi-source coal-based solid waste collection, storage and transportation technology based on real-time monitoring and precise feedback control of multi-source coal-based solid waste, and the intelligent solid backfilling and mining technology which integrates intelligent backfilling monitoring, intelligent flow control, backfilling hydraulic support, and electrohydraulic control of the working face. Through the linkage and coordination of various key intelligent backfilling mining technologies, Xingdong Mine has formed an intelligent backfilling mine with coal-based solid waste pretreatment, underground coal gangue separation, multi-source coal-based solid waste storage and transportation, and solid backfilling mining as the core. The results of engineering practice show that the capacity of intelligent backfilling working faces has been greatly improved after the application of intelligent backfilling mining technology. The working faces' output increases from 36000 t per month to 72000 t per month. The number of personnel per shift is reduced by 8-10. The time of one cycle operation is reduced by about 2.5 h, and the efficiency increases by about 50%. The problem of coal-based solid waste discharge has been comprehensively solved.

By using the Pohozǎev manifold, we obtain the existence of high energy solutions of the aforementioned equation without the well-known Ambrosetti-Rabinowitz type condition.

In order to alleviate the problem of mine thermal damage, this paper proposes to put cold water pipe into the surrounding rock of roadway, and reduce the temperature of surrounding rock by intercepting the high temperature through the cold water pipe. According to the similarity principle, a cold water interception and cooling experimental system for high temperature surrounding rock was built. Experiments were conducted by changing cold water velocity, inlet time and surrounding rock temperature, etc. Combined with COMSOL simulation, the influence of different diameters and spacing layouts of cold water pipe on cold water interception and cooling effect of roadway surrounding rock was analyzed. The results show that when other conditions remain unchanged, the cooling range of surrounding rock increases with the increase of cold water velocity, inlet time and temperature difference between surrounding rock and cold water. When the diameter of the cold water pipe is 10 cm and the vertical distance is 2.5 m, the average temperature of the roadway wall after cooling is 23.4 ℃.

In this paper, the deep hole and shallow hole temperature measurement technology is used to measure the rock temperature in No.13 mining area of Yangchangwan Coal Mine, and the cooling circle range of 937 m elevation roadway for the return air downhill in No.13 mining area and 430 m elevation roadway for the auxiliary transportation downhill in No.13 mining area are determined. The COMSOL software is used to analyze the surrounding rock temperature distribution, and the reliability of the surface borehole temperature measurement data is verified according to the temperature measurement results. Combined with the ground borehole temperature measurement data and the deep hole and shallow hole temperature measurement results, the ground temperature gradient and the heat damage area are defined, which provides a basis for the subsequent heat damage treatment.

Objective To explore the molecular mechanism of amifostine in relieving cisplatin nephrotoxicity. Methods CCK-8 assay was used to detect the inhibitory effect of amifostine on cisplatin cytotoxicity in HK-2 and primary renal proximal tubular epithelial cells. The expression of PKM2 was analyzed by online database tabula muris. The proportion of apoptotic cells was detected by flow cytometry. The expression of PKM2 gene was detected by fluorescence quantitative PCR and Western blotting. PKM2 gene expression was knocked down by small RNA interference. Results Amifostine significantly enhanced the resistance of renal proximal tubular epithelial cells to cisplatin toxicity. In cisplatin treated cells, with the increase of the concentration of amifostine, the cell viability was increased from 0.37 to 0.45 to 0.77 (P < 0.001). In addition, with the increase of amifostine concentration, PKM2 gene expression in HK-2 cells was increased in turn. The expression of PKM2 gene was increased to 8.76 times higher at 1 μmol/L of amifostine (P < 0.001). PKM2 gene expression in HK-2 cells was increased with the increase of amifostine concentration. When PKM2 gene expression was depleted in HK-2 cells, and the protective effect of amifostine on cisplatin cytotoxicity was significantly inhibited. The proportion of apoptotic cells was reduced and the levels of apoptotic markers Cleaved-PARP and Cleaved-Caspase3 induced by cisplatin were significantly inhibited by amifostine. Single-cell data analysis indicated that the expression of PKM2 in renal proximal tubular epithelial cells was the lowest compared with other cell subsets in renal tissue. Conclusion Amifostine inhibits cell apoptosis induced by cisplatin by up-regulating PKM2 in renal proximal tubular epithelial cells, and thus alleviates the cytotoxicity of cisplatin.
