The aim of this study was to evaluate etazolate against depression-like behavior and, learning and memory impairment induced by 6- hydroxydopamine (6-OHDA) rat model of Parkinson’s disease (PD). This aim was achieved through comparing 6-OHDA lesioned rats in the presence and absence of etazolate. The 6-OHDA was used to induce lesion as a model of PD. Etazolate was administered at a dose of 1 mg/kg/day for 14 days, starting 7 days after lesion induction. Apomorphine-induced rotation test was used to evaluate 6-OHDA-induced motor deficits, tail suspension test was used to assess depression-like symptoms, and the radial arms water maze (RAWM) was used to evaluate special learning and memory functions. Antioxidant biomarkers and BDNF protein levels were assessed in the hippocampus. Results revealed that etazolate administration significantly improved 6-OHDA-induced PD related symptoms including motor deficits, depression-like behavior and impairment of both short- and long- term memory. Moreover, etazolate significantly prevented 6-OHDA-induced reduction in oxidative stress biomarkers (GSH/GSSG ratio, GPx) and BDNF levels. In conclusion, motor dysfunction, depressive- like behavior, and learning and memory deficits in the 6-OHDA rat model of PD can be significantly prevented by etazolate. Anxiety disorders are associated with abnormalities in fear-learning and bias to threat; early life experiences are influential to the development of an anxiety-like phenotype in adulthood. We recently reported that adult mice ( Prkar1a+/-) with haploinsufficiency for the main regulatory subunit of the protein kinase A (PKA) exhibit an anxiety-like phenotype associated with increased PKA activity in the amygdala. PKA is the main effector of cyclic adenosine mono-phosphate signaling, a key pathway involved in the regulation of fear learning. Since anxiety has developmental and genetic components, we sought to examine the interaction of a genetic defect associated with anxiety phenotype and early life experiences. We investigated the effects of neonatal maternal separation or tactile stimulation on measures of behavior typical to adolescence as well as developmental changes in the behavioral phenotype between adolescent and adult wild-type (WT) and Prkar1a+/- mice. Our results showed developmental differences in assays of anxiety and novelty behavior for both genotypes. Adolescent mice showed increased exploratory and novelty seeking behavior compared to adult counterparts. However, early life experiences modulated behavior in adolescent WT differently than in adolescent Prkar1a+/- mice. Adolescent WT mice exposed to early life tactile stimulation showed attenuation of anxiety-like behavior, whereas an increase in exploratory behavior was found in Prkar1a+/- adolescent mice. The finding of behavioral differences that are apparent during adolescence in Prkar1a +/− mice suggests that long-term exposure of the brain to increased PKA activity during critical developmental periods contributes to the anxiety-like phenotype noted in the adult animals with increased PKA activity. We tested whether (1) the capacity of a reward-associated conditioned stimulus (CS) to cause conditioned activation of ventral tegmental area (VTA) dopamine (DA) neurons is associated with its capacity to elicit conditioned approach responses and (2) whether the acquisition of these capacities by a CS requires N -methyl-D-aspartate (NMDA) and muscarinic acetylcholine (mACh) receptor stimulation. Rats were trained to emit a conditioned approach response to a light CS that was previously paired with food and were treated systemically with scopolamine (a mACh receptor antagonist) or MK-801 (an NMDA receptor antagonist) either prior to each conditioning session (during which animals experienced paired CS and food presentations) or prior to the conditioned approach (CS-only) test. Brains were harvested after the CS-only test and processed for tyrosine hydroxylase (TH) (DA cells) and c-fos in the VTA. When animals received scopolamine or MK-801 treatment prior to conditioning sessions we observed significantly fewer TH-labeled (i.e., DA) cells in the VTA that expressed c-fos and significantly less conditioned approach responding during the CS-only test. Further analysis showed a correlation between the number of VTA DA cells activated and the number of conditioned approach responses. Treatments made prior to the CS-only test did not affect responding. Altogether these results suggest that the degree to which a CS elicits conditioned approach depends partially on the degree to which the CS activates VTA DA cells and that the acquisition of both of these capacities by a CS requires mACh and NMDA receptor stimulation. Acute stress influences learning and memory in humans and rodents, enhancing performance in some tasks while impairing it in others. Typically, subjects preferentially employ striatal-mediated stimulus-response strategies in spatial memory tasks following stress, making use of fewer hippocampal-based strategies which may be more cognitively demanding. Previous research demonstrated that the acquisition of rodent paired associates learning (PAL) relies primarily on the striatum, while task performance after extensive training is impaired by hippocampal disruption. Therefore, we sought to explore whether the acquisition of PAL, an operant conditioning task involving spatial stimuli, could be enhanced by acute stress. Male Long-Evans rats were trained to a predefined criterion in PAL and then subjected to either a single session of restraint stress (30 min) or injection of corticosterone (CORT; 3 mg/kg). Subsequent task performance was monitored for one week. We found that rats subjected to restraint stress, but not those rats injected with CORT, performed with higher accuracy and efficiency, when compared to untreated controls. These results suggest that while acute stress enhances the acquisition of PAL, CORT alone does not. This dissociation may be due to differences between these treatments and their ability to produce sufficient catecholamine release in the amygdala, a requirement for stress effects on memory. Recent research suggests that video game playing is associated with many cognitive benefits. However, little is known about the neural mechanisms mediating such effects, especially with regard to probabilistic categorization learning, which is a widely unexplored area in gaming research. Therefore, the present study aimed to investigate the neural correlates of probabilistic classification learning in video gamers in comparison to non-gamers. Subjects were scanned in a 3 T magnetic resonance imaging (MRI) scanner while performing a modified version of the weather prediction task. Behavioral data yielded evidence for better categorization performance of video gamers, particularly under conditions characterized by stronger uncertainty. Furthermore, a post-experimental questionnaire showed that video gamers had acquired higher declarative knowledge about the card combinations and the related weather outcomes. Functional imaging data revealed for video gamers stronger activation clusters in the hippocampus, the precuneus, the cingulate gyrus and the middle temporal gyrus as well as in occipital visual areas and in areas related to attentional processes. All these areas are connected with each other and represent critical nodes for semantic memory, visual imagery and cognitive control. Apart from this, and in line with previous studies, both groups showed activation in brain areas that are related to attention and executive functions as well as in the basal ganglia and in memory-associated regions of the medial temporal lobe. These results suggest that playing video games might enhance the usage of declarative knowledge as well as hippocampal involvement and enhances overall learning performance during probabilistic learning. In an effort to advance the understanding of brain function and organisation accompanying second language learning, we investigate the neural substrates of novel grammar learning in a group of healthy adults, consisting of participants with high and average language analytical abilities (LAA). By means of an Independent Components Analysis, a data-driven approach to functional connectivity of the brain, the fMRI data collected during a grammar-learning task were decomposed into maps representing separate cognitive processes. These included the default mode , task-positive , working memory , visual , cerebellar and emotional networks. We further tested for differences within the components, representing individual differences between the High and Average LAA learners. We found high analytical abilities to be coupled with stronger contributions to the task-positive network from areas adjacent to bilateral Broca’s region, stronger connectivity within the working memory network and within the emotional network. Average LAA participants displayed stronger engagement within the task-positive network from areas adjacent to the right-hemisphere homologue of Broca's region and typical to lower level processing (visual word recognition), and increased connectivity within the default mode network. The significance of each of the identified networks for the grammar learning process is presented next to a discussion on the established markers of inter-individual learners’ differences. We conclude that in terms of functional connectivity, the engagement of brain’s networks during grammar acquisition is coupled with one’s language learning abilities. [ABSTRACT FROM AUTHOR]

Chemotherapeutic agents are widely used to treat patients with systemic cancer. The efficacy of these therapies is undermined by their adverse side-effect profiles such as cognitive deficits that have a negative impact on the quality of life of cancer survivors. Cognitive side effects occur across a variety of domains, including memory, executive function, and processing speed. Such impairments are exacerbated under cognitive challenges and a subgroup of patients experience long-term impairments. Episodic memory in rats can be examined using a source memory task. In the current study, rats received paclitaxel, a taxane-derived chemotherapeutic agent, and learning and memory functioning was examined using the source memory task. Treatment with paclitaxel did not impair spatial and episodic memory, and paclitaxel treated rats were not more susceptible to cognitive challenges. Under conditions in which memory was not impaired, paclitaxel treatment impaired learning of new rules, documenting a decreased sensitivity to changes in experimental contingencies. These findings provide new information on the nature of cancer chemotherapy-induced cognitive impairments, particularly regarding the incongruent vulnerability of episodic memory and new learning following treatment with paclitaxel. Extinction of fear to a Pavlovian conditioned stimulus (CS) is known to be context-specific. When the CS is tested outside the context of extinction, fear returns, or renews. Several studies have demonstrated that renewal depends upon the hippocampus, although there are also studies where renewal was not impacted by hippocampal damage, suggesting that under some conditions context encoding and/or retrieval of extinction depends upon other regions. One candidate region is the retrosplenial cortex (RSC), which is known to contribute to contextual and spatial learning and memory. Using a conditioned-suppression paradigm, Experiment 1 tested the impact of pre-training RSC lesions on renewal of extinguished fear. Consistent with previous studies, lesions of the RSC did not impact acquisition or extinction of conditioned fear to the CS. Further, there was no evidence that RSC lesions impaired renewal, indicating that contextual encoding and/or retrieval of extinction does not depend upon the RSC. In Experiment 2, post-extinction lesions of either the RSC or dorsal hippocampus (DH) also had no impact on renewal. However, in Experiment 3, both RSC and DH lesions did impair performance in an object-in-place procedure, an index of place memory. RSC and DH contributions to extinction and renewal are discussed. Individuals with autism spectrum disorder (ASD) exhibit social-communication deficits along with restricted interests and repetitive behaviors (RRBs). To date, there is a lack of effective treatments to alleviate RRBs. A recent study found that treatment with the 5HT 2A receptor antagonist M100907 attenuates a reversal learning deficit in the BTBR mouse model of autism. The BTBR mouse also exhibits elevated grooming behavior which may model stereotyped motor behaviors also observed in ASD. The present study examined whether 5HT 2A receptor blockade with M100907 at either 0.01 or 0.1 mg/kg can reduce repetitive grooming in BTBR mice compared to that of vehicle-treated BTBR and C57BL6/J (B6) mice. M100907 at 0.1 mg/kg, but not 0.01 mg/kg, significantly attenuated repetitive grooming in BTBR mice compared to that of vehicle-treated BTBR mice. M100907 at either dose did not affect grooming behavior in B6 mice. To determine whether 0.1 mg/kg M100907 had a more general effect on activity in BTBR mice, a second experiment determined whether M100907 at 0.1 mg/kg affected locomotor activity in BTBR mice. M100907 treatment in BTBR and B6 mice did not alter locomotor activity compared to that of vehicle-treated BTBR and B6 mice. The present findings taken together with past results suggest that treatment with a 5HT 2A receptor antagonist may be effective in ameliorating RRBs in ASD. [ABSTRACT FROM AUTHOR]

Alterations in behaviour can arise through a number of factors, including strain and sex. Here, we explored strain and sex differences between Long–Evans (LER) and Wistar (WR) male and female rats that had been trained in a myriad of behavioural tasks. Tests included those assessing motor learning (skilled reaching task), spatial learning and memory (Morris water task), contextual learning (discriminative fear-conditioning to context) and anxiety behaviour (elevated plus maze). Following behavioural assessment, associated brain areas were examined for volumetric differences, including the hippocampus and its subregions, prefrontal cortex areas and the amygdala. LER and WR differed in their rates of performance in the skilled reaching task throughout the training period. Overall, LER outperformed WR in tasks related to contextual and spatial learning, although this was not accompanied by larger volumes of associated brain areas. Males outperformed females in spatial learning, and females outperformed males in the contextual fear-conditioning task and had an associated larger amygdalar volume, although these sexual dimorphisms were only observed within the LER strain. Overall, this study highlights differences between these two rat strains as well as highlights that larger volumetric estimates of brain areas do not always confer improved function of associated behaviours. Marijuana is one of the most highly used psychoactive substances in the world, and its use typically begins during adolescence, a period of substantial brain development. Females across species appear to be more susceptible to the long-term consequences of marijuana use. Despite the identification of inherent differences between rat strains including measures of anatomy, genetics and behaviour, no studies to our knowledge have examined the long-term consequences of adolescent exposure to marijuana or its main psychoactive component, Δ 9 -tetrahydrocannabinol (THC), in males and females of two widely used rat strains: Long–Evans hooded (LER) and Wistar (WR) rats. THC was administered for 14 consecutive days following puberty onset, and once they reached adulthood, changes in behaviour and in the volume of associated brain areas were quantified. Rats were assessed in behavioural tests of motor, spatial and contextual learning, and anxiety. Some tasks showed effects of injection, since handled and vehicle groups were included as controls. Performance on all tasks, except motor learning, and the volume of associated brain areas were altered with injection or THC administration, although these effects varied by strain and sex group. Finally, analysis revealed treatment-specific correlations between performance and brain volumes. This study is the first of its kind to directly compare males and females of two rat strains for the long-term consequences of adolescent THC exposure. Variability in fear conditionability is common, and clarity regarding the neural regions responsible for individual differences in fear conditionability could uncover brain-based biomarkers of resilience or vulnerability to trauma-based psychopathologies (e.g., post-traumatic stress disorder). In recent years, neuroimaging work has yielded a detailed understanding of the neural mechanisms underlying fear conditioning common across participants, however only a minority of studies have investigated the brain basis of inter-individual variation in fear learning. Moreover, the majority of these studies have employed small sample sizes (mean n = 17; range n = 5–27) and all have failed to meet the minimum recommended sample size for functional magnetic resonance imaging (fMRI) studies of individual differences. Here, using fMRI, we analyzed blood-oxygenation level dependent (BOLD) response recorded simultaneously with skin conductance response (SCR) and ratings of unconditioned stimulus (US) expectancy in 49 participants undergoing Pavlovian fear conditioning. On average, participants became conditioned to the conditioned stimulus (CS+; higher US expectancy ratings and SCR for the CS+ compared to the unpaired conditioned stimulus, CS−); the CS+ also robustly increased activation in the bilateral insula. Amygdala activation was revealed from a regression analysis that incorporated individual differences in fear conditionability (i.e., a between-subjects regressor of mean CS+ > CS− SCR). By replicating results observed using much smaller sample sizes, the results confirm that variation in amygdala reactivity covaries with individual differences in fear conditionability. We have recently shown that the M1 muscarinic receptor positive allosteric modulator, PQCA, improves cognitive performance in rodents and non-human primates administered the muscarinic receptor antagonist scopolamine. The purpose of the present experiments was to characterize the effects of PQCA in a model more relevant to the disease pathology of Alzheimer's disease. Tg2576 transgenic mice that have elevated Aβ were tested in the novel object recognition task to characterize recognition memory as a function of age and treatment with the PQCA. The effects of PQCA were compared to the acetylcholinesterase inhibitor donepezil, the standard of care for Alzheimer's disease. In addition, the effect of co-administering PQCA and donepezil was evaluated. Aged Tg2576 mice demonstrated a deficit in recognition memory that was significantly attenuated by PQCA. The positive control donepezil also reversed the deficit. Furthermore, doses of PQCA and donepezil that were inactive on their own were found to improve recognition memory when given together. These studies suggest that M1 muscarinic receptor positive allosteric modulation can ameliorate memory deficits in disease relevant models of Alzheimer's disease. These data, combined with our previous findings demonstrating PQCA improves scopolamine-induced cognitive deficits in both rodents and non-human primates, suggest that M1 positive allosteric modulators have therapeutic potential for the treatment of Alzheimer's disease. We tested whether learning the mapping between objects and their locations is better when actively moving the hand to these locations, to reveal the object, compared to when the hand is passively moved by a robotic manipulandum. Recall of object locations was more accurate in the active compared to passive condition. We also found that recall was less accurate when participant made active movements that were not directed to the object locations. These results indicate that the well-established active exploration advantage for spatial memory extends to location memory for objects within reach. Abstract: The hippocampal formation is essential for the processing of episodic memories for autobiographical events that happen in unique spatiotemporal contexts. Interestingly, before 2 years of age, children are unable to form or store episodic memories for recall later in life, a phenomenon known as infantile amnesia. From 2 to 7 years of age, there are fewer memories than predicted based on a forgetting function alone, a phenomenon known as childhood amnesia. Here, we discuss the postnatal maturation of the primate hippocampal formation with the goal of characterizing the development of the neurobiological substrates thought to subserve the emergence of episodic memory. Distinct regions, layers and cells of the hippocampal formation exhibit different profiles of structural and molecular development during early postnatal life. The protracted period of neuronal addition and maturation in the dentate gyrus is accompanied by the late maturation of specific layers in different hippocampal regions that are located downstream from the dentate gyrus, particularly CA3. In contrast, distinct layers in several hippocampal regions, particularly CA1, which receive direct projections from the entorhinal cortex, exhibit an early maturation. In addition, hippocampal regions that are more highly interconnected with subcortical structures, including the subiculum, presubiculum, parasubiculum and CA2, mature even earlier. These findings, together with our studies of the development of human spatial memory, support the hypothesis that the differential maturation of distinct hippocampal circuits might underlie the differential emergence of specific “hippocampus-dependent” memory processes, culminating in the emergence of episodic memory concomitant with the maturation of all hippocampal circuits. Abstract: Neurohormones such as testosterone (TE) are important in modulation of learning and memory. In the present study, we investigated the interactive effects of pre-training bilateral intra-hippocampal infusions of testosterone and H-89, a selective PKAII inhibitor, on spatial acquisition in the Morris water maze (MWM). Different doses of TE (20, 40 and 80μg/side) and H-89 (5 and 10μM/side) were administered 30min before start of the training each day. Control animals received bilateral intra-hippocampal infusions of DMSO as vehicle for TE and H-89. Animals were trained for 4 days and each day included one block of four trials. The results of this study showed that bilateral infusion of TE (40 and 80μg/side) or H-89 (10μM/side) impaired spatial learning as indicated by significant increases in escape latency and traveled distance compared to the control group. Although pre-training bilateral infusions of a low concentration of either TE (20μg/side) or H-89 (5μM/side) into the CA1 region of the hippocampus did not affect learning capabilities, but the combination of the low doses of the drugs led to significant deficits in spatial acquisition. Overall, our data suggest that spatial acquisition was affected by PKAII inhibition or TE administration. Moreover, when co-administered, these drugs had a negative synergistic impact on acquisition. Abstract: Post-operative cognitive dysfunction (POCD) is a clinical phenomenon that has drawn significant attention from the public and scientific community. Age is a risk factor for POCD. However, the contribution of general anesthesia/anesthetics to POCD and the underlying neuropathology are not clear. Here, we showed that 18-month-old male Fisher 344 rats exposed to 1.2% isoflurane, a general anesthetic, for 2h had significant learning and memory impairments assessed at 2–4 weeks after isoflurane exposure. These isoflurane effects were attenuated by intravenous lidocaine (1.5mg/kg as a bolus and then 2mg/kg/h during isoflurane exposure), a local anesthetic that has neuroprotective effect. Exposure to isoflurane or isoflurane plus lidocaine did not change the neuronal and synaptic density as well as the expression of NeuN (a neuronal protein), drebrin (a dendritic spine protein), synaptophysin (a synaptic protein), activated caspase 3 and caspase-activated DNase in the hippocampus at 29 days after isoflurane exposure when cognitive impairment was present. Isoflurane and lidocaine did not affect the amount of β-amyloid peptide, total tau and phospho-tau in the cerebral cortex as well as interleukin-1β and tumor necrosis factor-α in the hippocampus at 29 days after isoflurane exposure. Thus, isoflurane induces learning and memory impairment in old rats. Lidocaine attenuates these isoflurane effects. Isoflurane may not cause long-lasting neuropathological changes. Abstract: Paradoxical sleep deprivation (PSD) may alter subsequent learning and memory capacity. There are differences in both the intensity and direction of responses of the male and female species to the same environmental stimuli and experimental conditions. In the present study, we examined the extent of the effects of PSD for 72h on spatial learning and memory, anxiety-like behavior, corticosterone levels, and the body weight in male as well as in intact and ovariectomized (OVX) female Wistar rats. Multiple platform method was used for PSD induction. Spatial learning and memory and anxiety-like behavior were determined using Morris water maze (MWM) task and open field test, respectively. The data showed that PSD could not significantly affect subsequent spatial learning and short-term memory in male rats, while it significantly impaired the performance of the intact and OVX female rats. The PSD-intact and -OVX female rats showed more memory impairment than the PSD-male animals. Those impairments do not appear to be due to elevated stress level, since the plasma corticosterone did not significantly change following PSD induction. The open field data showed that PSD significantly reduced anxiety-like behavior in all experimental groups. In addition, PSD had a reducing effect on the mean body weight of female groups. Such results suggest that the female rats are more vulnerable to the deleterious effects of sleep loss on cognitive performance. [Copyright &y& Elsevier]

Abstract: Accumulating evidence has implicated neuropeptides in modulating recognition, learning and memory. However, to date, no study has investigated the effects of neuropeptide Trefoil factor 3 (TFF3) on the process of learning and memory. In the present study, we evaluated the acute effects of TFF3 administration (0.1 and 0.5mg/kg, i.p.) on the acquisition and retention of object recognition memory in mice. We found that TFF3 administration significantly enhanced both short-term and long-term memory during the retention test, conducted 90min and 24h after training respectively. Remarkably, acute TFF3 administration transformed a learning event that would not normally result in long-term memory into an event retained for a long-term period and produced no effect on locomotor activity in mice. In conclusion, the present results provide an important role of TFF3 in improving object recognition memory and reserving it for a longer time, which suggests a potential therapeutic application for diseases with recognition and memory impairment. Abstract: Visuo-spatial learning and memory were assessed in male and female mice of 13 inbred strains on a small diameter mouse version of the Barnes maze surrounded by a wall and intra-maze visual cues. Mice completed acquisition and reversal training to assess learning, followed by a probe test to assess memory for the spatial location of the escape hole. The C57BL/6J and CAST/EiJ strains showed better learning performance than the other strains. A/J and 129/SvImJ strains showed poor learning performance, which may be due to their low rates of exploration. No differences in memory were found between strains in the probe test. Males showed better learning performance than females in the DBA/2J and C3H/HeJ strains, but there were no sex differences in the other strains. However, mice may not have used visuo-spatial cues to locate the escape hole in this maze, as (1) all strains primarily used the non-spatial serial/thigmotaxic search strategy, (2) no strains showed a reversal effect when the escape hole location was moved, and (3) learning and memory performance were not correlated with measures of visual ability. Multivariate and univariate analyses of variance indicated that mice with good visual ability performed better than mice with poor visual ability, but the effect sizes were small. The small diameter of the maze and the presence of a wall around the edge of the maze may promote thigmotaxis in mice, increasing the use of a non-visual search strategy, thereby reducing the influence of vision on performance and decreasing the sensitivity of this maze design to detect strain differences in visuo-spatial learning and memory. Abstract: It has long been known that emotions can modulate learning and memory processes in humans and non-human mammals. Here we will review evidence from clinical, neuropsychological, neuroimaging and animal research suggesting an important role of emotions for the establishment of long-term episodic memories in the mammalian brain. In the first part of the review the neuroanatomical and neurochemical foundations of the interaction between brain areas generating emotions, such as the amygdala, and those allowing the association of multi-dimensional stimuli into an episodic memory, such as the hippocampus, are delineated. Patients with emotional and affective disorders show changes in memory performance in dependence of the positive or negative valence of the stimulus material. Furthermore, these patients often exhibit a reduced ability to access specific memories of life events with a striking lack of specific detail. Therefore, in the second part the clinical literature on memory impairments observed in patients with emotional and affective disorders, including post-traumatic stress disorder, schizophrenia and major depression, with a special emphasis on episodic memory function, is discussed. Finally, the relationship between memory deficits in Alzheimer''s disease and neurodegeneration in brain systems mediating emotions is reviewed. [Copyright &y& Elsevier]

Abstract: Background: Asenapine is a new pharmacological agent for the acute treatment of schizophrenia and bipolar disorder. It has relatively higher affinity for serotonergic and α2-adrenergic than dopaminergic D2 receptors. We evaluated the effects of asenapine, risperidone, and olanzapine on acute and subchronic psychotomimetic-induced disruption of cued reversal learning in rats. Methods: After operant training, rats were treated acutely with d-amphetamine (0.75mg/kg intraperitoneally [i.p.]) or phencyclidine (PCP; 1.5mg/kg i.p.) or subchronically with PCP (2mg/kg i.p. for 7 days). We assessed the effects of acute coadministration of asenapine, risperidone, or olanzapine on acute d-amphetamine- and PCP-induced deficits and the effects of long-term coadministration of these agents (for 28 additional days) on the deficits induced by subchronic PCP. Results: Deficits in reversal learning induced by acute d-amphetamine were attenuated by risperidone (0.2mg/kg i.p.). Acute PCP-induced impairment of reversal learning was attenuated by acute asenapine (0.025mg/kg subcutaneously [s.c.]), risperidone (0.2mg/kg i.p.), and olanzapine (1.0mg/kg i.p.). Subchronic PCP administration induced an enduring deficit that was attenuated by acute asenapine (0.075mg/kg s.c.) and by olanzapine (1.5mg/kg i.p.). Asenapine (0.075mg/kg s.c.), risperidone (0.2mg/kg i.p.), and olanzapine (1.0mg/kg i.p.) all showed sustained efficacy with chronic (29 days) treatment to improve subchronic PCP-induced impairments. Conclusion: These data suggest that asenapine may have beneficial effects in the treatment of cognitive symptoms in schizophrenia. Abstract: The influences of complex housing, T-maze, and Grice box training on dendritic morphology of the prefrontal cortex (PFC) and primary somatosensory (Par 1) were investigated in the rat. Golgi–Cox analyses demonstrated that all learning paradigms produced alterations in PFC connectivity, albeit differently. Furthermore, the effects of experience on dendritic morphology varied by region, hemisphere, and lamina and often resulted in opposing changes within each. For example, complex housing produced a time-dependent decrease in the dendritic fields of layer (L) V neurons in region 3 of the cingulate cortex (Cg3) in the medial PFC (mPFC) neurons and increases in LIII of the dorsal agranular insular (AID) region of the orbitofrontal cortex. In contrast, T-maze training produced increases in Cg3 LV and decreases in AID LIII spine density. Of interest, the influence of experience was not reflected equally between hemispheres. For example, T-maze training produced an increase in Cg3 LV and AID LIII branch order and length in the right hemisphere, and an increase in Cg3 LIII branch order in the left hemisphere. Additionally, correlation analysis of task performance and dendritic morphology indicated an opposing influence of experience within the different laminae of the Cg3. For example, whereas, performance was negatively correlated with Cg3 LV dendritic branch order/length, there was a positive correlation with Cg3 LIII dendrites. Complex housing induced changes in the Par 1were also time -dependent but were only apparent after a prolonged period of exposure. In addition to demonstrating learning-specific modifications in dendritic connectivity within the PFC, the results illustrate varying patterns of change that likely reflect task-dependent requirements on the PFC. Abstract: Recent evidence indicates that stress modulates multiple memory systems, favoring caudate nucleus-based stimulus–response learning at the expense of hippocampus-based spatial learning. Whether this is due to a facilitating effect of stress on stimulus–response learning, an impairing effect on spatial learning, or both, is not known. To answer this question, mice were either subjected to restraint stress, injected with vehicle or corticosterone or left untreated before training in two circular hole board tasks that could discriminate spatial and stimulus–response strategies. Stress, vehicle and corticosterone injection all impaired learning performance in the spatial task. Conversely, performance in the stimulus–response task was not affected by stress or corticosterone injection, although performance was generally lower than in the spatial task. Irrespective of the treatment, mice had to overcome the preference to use their spatial memory system to achieve the stimulus–response task. These findings suggest that (i) the caudate nucleus-based memory system is less stress sensitive than the hippocampus-based system and may thus dominate behavior in situations of stress and (ii) that multiple memory systems may compete for control of behavior even in tasks that can solely be solved by one system. Abstract: Our study focuses on the physiological effects of repetition on learning and working memory using an adaptation of Luria''s Memory Word-Task (LMWT). We assess the hemodynamic response in dorsolateral prefrontal cortex (DLPFC) of 13 healthy subjects while completing LMWT. Free word recalls were acquired at the beginning, middle and end of the task. Behavioral results showed that all subjects could recall the complete word list by the 10th trial, which was considered as successful task accomplishment. We observed an attenuation of stimulus-evoked neural activity in prefrontal neurons. Our findings show that the temporal integration of efficient verbal learning is mediated by a mechanism known as neural repetition suppression (NRS). This mechanism facilitates cortical deactivation in DLPFC once learning is successfully completed. This cortical reorganization is interpreted as a progressive optimization of neural responses to produce a more efficient use of neural circuits. Abstract: In this study, we aimed at comparing the effect of the social versus the physical enrichment of the environment on inflammatory pain. Hence, a rat model of carrageenan-induced knee inflammation was used. Four housing conditions were investigated: a physically enriched environment (PE), a socially enriched environment (SE), an enriched environment (EE) (i.e. physically and socially enriched) and a restricted environment (RE) (i.e. non-physically or socially enriched housing). Mechanical allodynia was assessed using the von Frey test preoperatively and at day post-operative (DPO) 1, 3, 7, 10, 14, 17, 21, 24 and 28. Besides, anxiety was evaluated at DPO29, using the Elevated Plus-Maze test. Results show that RE housing resulted in a duration of mechanical allodynia of 4 weeks and of only 3 weeks in EE housing. Housing in a physically enriched environment also resulted in a reduction of the duration of mechanical allodynia of 1 week. Finally, if housed in a SE, the mechanical allodynia lasted for 3 weeks and an half. From these data, we conclude that both physical and social aspects of the environment are involved in the reduction of inflammatory pain duration, although the PE has a larger effect than the SE in this experimental setting. Interestingly, an inter-dependent relationship was noted between the PE and SE. Moreover, no significant difference in the rat anxiety was measured between groups, suggesting that the pain outcomes are likely not biased by the mean of potential housing condition-induced anxiety. Abstract: Nitric oxide (NO) is sought to be an intracellular messenger in the central nervous system and its implication in learning and memory is well documented. Compounds that inhibit nitric oxide synthase (NOS), the key synthesizing enzyme, block cognition, though discrepant findings, in this context, have also been reported. The present study was designed to investigate in the rat: (a) the effects on recognition memory exerted by low doses of the NOS inhibitor L-NAME and (b) the ability of this compound in modulating different mnemonic processes (acquisition, storage and retrieval). For this aim, the object recognition task was selected. In a first study, pre- or post-training systemic administration of L-NAME (1, 3 and 10mg/kg, i.p.) did not disrupt animals’ performance in this recognition memory paradigm. Subsequently, L-NAME (1 and 3, but not 10mg/kg, i.p.) antagonized delay-dependent deficits in the object recognition task suggesting that L-NAME affected acquisition, storage and retrieval of information. Abstract: In this report, we demonstrate that chronic prenatal exposure to a moderate dose of caffeine disrupts novel object recognition and radial arm maze behaviors in adult male and female rats. Pregnant dams were administered either tap water or 75mg/L caffeinated tap water throughout gestation. Oral self-administration in the drinking water led to an approximate maternal intake of 10mg/kg/day, equivalent to 2–3 cups of coffee/day in humans based on a metabolic body weight conversion. In adulthood, the offspring underwent testing on novel object recognition, radial arm maze, and Morris water maze tasks. Prenatal caffeine exposure was found to impair 24-h memory retention in the novel object recognition task and impair both working and reference memory in the radial arm maze. However, prenatal caffeine exposure did not alter Morris water maze performance in either a simple water maze procedure or in an advanced water maze procedure that included reversal and working memory paradigms. These findings demonstrate that chronic oral intake of caffeine throughout gestation can alter adult cognitive behaviors in rats. [Copyright &y& Elsevier]

Abstract: Several executive functions rely on the medial prefrontal cortex (mPFC) in the rat. Aspiration and neurotoxic lesions of the mPFC impair reversal learning in adult rats. Systemic administration of MK-801, an NMDA-receptor antagonist, impairs T-maze reversal learning in weanling rats but the role of mPFC NMDA-receptor antagonism in this effect is not known in either adult or young animals. This set of studies showed that mPFC NMDA receptors are specifically involved in T-maze discrimination reversal in weanling rats. In Experiment 1, 26-day-old rats (P26) demonstrated a dose-dependent impairment following bilateral mPFC administration of either 2.5 or 5.0μg MK-801 or saline (vehicle) during the reversal training phase only. In Experiment 2, P26 rats were trained on the same task, but four groups of rats received bilateral mPFC infusions during acquisition only (MK–SAL), reversal only (SAL–MK), both phases (MK–MK), or neither phase (SAL–SAL). MK-801 impaired performance only when infused during reversal. This suggests that NMDA-receptor antagonism in the mPFC is selectively involved in reversal learning during development and this may account for the previously reported effects of systemic MK-801 on T-maze discrimination reversal in weanling rats. Abstract: The combined effects of high fat diet (HFD) and chronic stress on the hippocampus-dependent spatial learning and memory were studied in rats using the radial arm water maze (RAWM). Chronic psychosocial stress and/or HFD were simultaneously administered for 3 months to young adult male Wister rats. In the RAWM, rats were subjected to 12 learning trials as well as short-term and long-term memory tests. This procedure was applied on a daily basis until the animal reaches days to criterion (DTC) in the 12th learning trial and in memory tests. DTC is the number of days that the animal takes to make zero error in two consecutive days. Groups were compared based on the number of errors per trial or test as well as on the DTC. Chronic stress, HFD and chronic stress/HFD animal groups showed impaired learning as indicated by committing significantly (P <0.05) more errors than untreated control group in trials 6 through 9 of day 4. In memory tests, chronic stress, HFD and chronic stress/HFD groups showed significantly impaired performance compared to control group. Additionally, the stress/HFD was the only group that showed significantly impaired performance in memory tests on the 5th training day, suggesting more severe memory impairment in that group. Furthermore, DTC value for above groups indicated that chronic stress or HFD, alone, resulted in a mild impairment of spatial memory, but the combination of chronic stress and HFD resulted in a more severe and long-lasting memory impairment. The data indicated that the combination of stress and HFD produced more deleterious effects on hippocampal cognitive function than either chronic stress or HFD alone. [Copyright &y& Elsevier]

Abstract: There is a wealth of information indicating that the hippocampal formation is important for learning and memory consolidation. The hippocampus is very sensitive to ageing and developmentally stressful factors such as prenatal malnutrition, which produces anatomical alterations of hippocampal pyramidal cells as well as impaired spatial learning. On the other hand, there are no reports about differential effects of postnatal malnutrition, installed at birth and maintained all through life in young and aged rats, on learning and memory of active avoidance, a task with an important procedural component. We now report that learning and long-term retention of this task were impaired in young malnourished animals, but not in young control, senile control, and senile malnourished Sprague–Dawley rats; young and senile rats were 90 and 660 days of age, respectively. Extinction tests showed, however, that long-term memory of the malnourished groups and senile control animals is impaired as compared with the young control animals. These data strongly suggest that the learning and long-term retention impairments seen in the young animals were due to postnatal malnutrition; in the senile groups, this cognitive alteration did not occur, probably because ageing itself is an important factor that enables the brain to engage in compensatory mechanisms that reduce the effects of malnutrition. Nonetheless, ageing and malnutrition, conditions known to produce anatomic and functional hippocampal alterations, impede the maintenance of long-term memory, as seen during the extinction test. Abstract: The striatal dopamine signal has multiple facets; tonic level, phasic rise and fall, and variation of the phasic rise/fall depending on the expectation of reward/punishment. We have developed a network model of the striatal direct pathway using an ionic current level model of the medium spiny neuron that incorporates currents sensitive to changes in the tonic level of dopamine. The model neurons in the network learn action selection based on a novel set of mathematical rules that incorporate the phasic change in the dopamine signal. This network model is capable of learning to perform a sequence learning task that in humans is thought to be dependent on the basal ganglia. When both tonic and phasic levels of dopamine are decreased, as would be expected in unmedicated Parkinson''s disease (PD), the model reproduces the deficits seen in a human PD group off medication. When the tonic level is increased to normal, but with reduced phasic increases and decreases in response to reward and punishment, respectively, as would be expected in PD medicated with L-Dopa, the model again reproduces the human data. These findings support the view that the cognitive dysfunctions seen in Parkinson''s disease are not solely either due to the decreased tonic level of dopamine or to the decreased responsiveness of the phasic dopamine signal to reward and punishment, but to a combination of the two factors that varies dependent on disease stage and medication status. Abstract: Objective: In addition to memory impairment, a tendency to get lost is among the initial symptoms in patients with Alzheimer disease (AD). At least two kinds of wayfinding strategies, egocentric and allocentric, have been proposed. It is believed that people may form a cognitive map after repeated movement in a specific environment, and are able to use it as an aid to navigation. In the present study, we investigated the cognitive maps in early AD patients and their application in a computer-generated arena (CGA). Methods: We invited very mild AD (CDR 0.5) patients and normal controls (NCs) to participate in the current study. Hand-drawing tests were used to assess their supposedly previously formed cognitive maps of familiar environments, and CGA was used to measure their new environment learning as well as the application of the old map. Results: Nineteen patients (8 females, mean age 67.6 years old, education 9.7 years, and MMSE 24) and 18 NCs (10 females, mean age 66.4 years old, education 8.8 years, and MMSE 27) completed the study. In the hand-drawing map part, both groups did equally well. In the new environment learning, NCs did better than the AD group on the third of six trials. As for the old environment navigation experiment, the AD group spent more time than the NCs in finding the target, but showed no difference to NC regarding the path traveled in the target quadrant. Conclusion: Early AD patients maintain their ability to use a cognitive map and keep pretty good allocentric representation of their familiar environments as well as NC do, but probably both groups do not routinely use their cognitive map to navigate in everyday life properly. Abstract: The nucleus accumbens (NAc) is important for the ability of motivationally significant stimuli to guide behavior. To further delineate its role in appetitive Pavlovian conditioning, we tested the hypothesis that the NAc contributes to memory consolidation and expression for a goal-tracking version of Pavlovian conditioned approach (PCA) in rats. We found that neither post-training reversible inactivation with the GABA receptor agonists muscimol and baclofen nor inhibition of protein synthesis with anisomycin (ANI) in either the core or shell regions of the NAc had an effect on approach to a reward port in response to a reward-predictive cue (conditioned stimulus, CS+). In contrast, pre-test reversible inactivation of both the core and shell decreased conditioned responding during the CS+. Unlike inactivation of the core, however, reversible inactivation of the shell also produced an increase in responding during the CS− and the inter-trial interval. This suggests that the NAc is not involved in the consolidation of goal-tracking PCA, but that once the memory is formed, the core is required for expression of the CS–unconditioned stimulus (US) association and the shell is required to inhibit conditioned approach behavior at times when the CS+ is not presented. Abstract: In this review we discuss the evidence that drug addiction, defined as a maladaptive compulsive habit, results from the progressive subversion by addictive drugs of striatum-dependent operant and Pavlovian learning mechanisms that are usually involved in the control over behaviour by stimuli associated with natural reinforcement. Although mainly organized through segregated parallel cortico-striato-pallido-thalamo-cortical loops involved in motor or emotional functions, the basal ganglia, and especially the striatum, are key mediators of the modulation of behavioural responses, under the control of both action-outcome and stimulus-response mechanisms, by incentive motivational processes and Pavlovian associations. Here we suggest that protracted exposure to addictive drugs recruits serial and dopamine-dependent, striato-nigro-striatal ascending spirals from the nucleus accumbens to more dorsal regions of the striatum that underlie a shift from action-outcome to stimulus-response mechanisms in the control over drug seeking. When this progressive ventral to dorsal striatum shift is combined with drug-associated Pavlovian influences from limbic structures such as the amygdala and the orbitofrontal cortex, drug seeking behaviour becomes established as an incentive habit. This instantiation of implicit sub-cortical processing of drug-associated stimuli and instrumental responding might be a key mechanism underlying the development of compulsive drug seeking and the high vulnerability to relapse which are hallmarks of drug addiction. Abstract: The plus-maze apparatus figured prominently in the historical debate between cognitive and stimulus–response habit learning theorists concerned with the fundamental question of “what” animals learn. An important feature of this task is that variants of the training procedure can be arranged to allow for an assessment of the relative use of cognitive/place or habit/response learning mechanisms. This brief review describes findings from several neurobiological studies published primarily over the past decade that have re-introduced the plus-maze to investigate the role of the dorsal striatum in learning and memory. Converging evidence from research using brain lesion, pharmacological, and molecular/genetic approaches is described supporting the hypothesis that the dorsolateral striatum plays a selective role in response learning in the plus-maze. Within a multiple systems framework of memory organization, factors that can influence the relative use of place and response learning in the plus-maze are also considered, including the nature of the visual environment, reinforcement/training parameters, and emotional state of the organism. Response learning in the plus-maze may be considered an exemplar task useful for investigating the neurobiological bases of dorsal striatal involvement in habit learning and memory. Abstract: Traumatic brain injury (TBI) is a devastating disease that commonly causes persistent mental disturbances and cognitive deficits. Although studies indicate that oxidative stress and functional deficits occurring after TBI are interrelated events, the knowledge of the mechanisms underlying the development of such cognitive deficits has been limited. Thus, in the present study, we investigated the effect of fluid percussion brain injury (FPI) on a spatial learning task and levels of oxidative stress markers, namely, protein carbonylation and thiobarbituric acid-reactive substances (TBARS) and Na+,K+-ATPase activity 1 or 3 months after FPI in rats. Statistical analysis revealed that FPI increased the scape latency and mean number of error in Barnes maze test 1 and 3 months after FPI. We also found that protein carbonylation and TBARS content increased in the parietal cortex 1 and 3 months after FPI. In addition, 3 months after FPI, protein carbonylation levels increased both in ipsilateral and contralateral cortices of FPI animals. Indeed, statistical analysis revealed a decrease in Na+,K+-ATPase activity in the cerebral cortex of 1 month FPI animals. Furthermore, the decrease in enzyme activity found 3 months was larger, when compared with 1 month after FPI. These results suggest that cognitive impairment following TBI may result, at least in part, from increase of two oxidative stress markers, protein carbonylation and TBARS that occurs concomitantly to a decrease in Na+,K+-ATPase activity. Abstract: We recently found that the interfering peptide Tat-3L4F is able not only to disrupt the protein–protein interaction of PTEN (phosphatase and tensin homologue deleted on chromosome 10) with the serotonin 5-HT2C receptor in the rat ventral tegmental area (VTA) but also to suppress the conditioned place preference induced by cannabinoid and nicotine without significant effects on anxiety, feeding behavior and motor activity. It is unknown, however, whether Tat-3L4F affects learning and memory. Using a Morris water maze test, we show here that while the cannabinoid HU210 significantly inhibited the performance of spatial learning without significant effects on long-term memory, Tat-3L4F did not induce significant effects on the acquisition and retrieval of spatial memory. These results indicate that Tat-3L4F can suppress the rewarding effects of abused drugs without significant effects on learning and memory. [Copyright &y& Elsevier]

Abstract: We tested whether N-methyl-d-aspartate (NMDA) receptors in the dorsal hippocampus (DH) are critical for the acquisition of trace fear conditioning using conditioned hypoalgesia (CHA), decrease in pain reactivity, as the conditioned response (CR) instead of commonly used freezing. Infusions of the NMDA receptor antagonist, dl-2-amino-5-phosphonovaleric acid (APV) into DH prior to conditioning resulted in impaired CHA, measured with the radiant heat tail flick test, only in the trace-conditioning group when they were tested during the trace interval. The same infusion had no effect on CHA in the delay-conditioned animals. The results support that NMDA receptors in DH are critically involved in associating the CS with the US across a temporal gap. In addition, temporal specificity of the CR was revealed as CHA was induced only in the temporal vicinity of the US used for the training. [Copyright &y& Elsevier]

Abstract: Animals can use both allocentric and egocentric strategies to learn a spatial task. Our results suggest that allocentric cues are more dominant than idiothetic cues in guiding navigation. Animals do not necessarily learn an egocentric strategy automatically, instead they probably hold just one solution to any particular task at a time until forced to learn an alternative strategy. Further, with overtraining animals do not always switch from allocentric to an egocentric learning strategy perhaps challenging suggestions of a stored hierarchy of strategies. Abstract: The purpose of the experiments was to examine the memory-related effects of nicotine using the mouse elevated plus maze. It has been shown that the acute doses of nicotine (0.1 and 0.5mg/kg) significantly decreased the time of transfer latency (TL2) on the retention trial, indicating that nicotine improved memory processes. Similarly, acute doses of the CB1 cannabinoid receptor antagonist AM 251 (0.5, 1, 1.5 and 3mg/kg) significantly decreased TL2 values. WIN55,212-2, a non-selective CB cannabinoid receptor agonist, at any dose tested (0.25, 0.5 and 1mg/kg), did not provoke any effect in this model. Moreover, the acute injection of both WIN55,212-2 (0.25 and 0.5mg/kg) and AM 251 (0.25mg/kg), prior to injections of nicotine (0.1 and 0.5mg/kg), significantly prevented nicotine-induced memory improvement. The results of this study provide clear evidence that the endogenous cannabinoid system participates in the responses induced by nicotine on memory-related behaviour in mice. Abstract: The effect of two chronic motor training paradigms on the ability of the lumbar spinal cord to perform an acute instrumental learning task was examined in neonatally (postnatal day 5; P5) spinal cord transected (i.e., spinal) rats. At ∼P30, rats began either unipedal hindlimb stand training (Stand-Tr; 20–25min/day, 5days/week), or bipedal hindlimb step training (Step-Tr; 20min/day; 5days/week) for 7 weeks. Non-trained spinal rats (Non-Tr) served as controls. After 7 weeks all groups were tested on the flexor-biased instrumental learning paradigm. We hypothesized that (1) Step-Tr rats would exhibit an increased capacity to learn the flexor-biased task relative to Non-Tr subjects, as locomotion involves repetitive training of the tibialis anterior (TA), the ankle flexor whose activation is important for successful instrumental learning, and (2) Stand-Tr rats would exhibit a deficit in acute motor learning, as unipedal training activates the ipsilateral ankle extensors, but not flexors. Results showed no differences in acute learning potential between Non-Tr and Step-Tr rats, while the Stand-Tr group showed a reduced capacity to learn the acute task. Further investigation of the Stand-Tr group showed that, while both the ipsilateral and contralateral hindlimbs were significantly impaired in their acute learning potential, the contralateral, untrained hindlimbs exhibited significantly greater learning deficits. These results suggest that different types of chronic peripheral input may have a significant impact on the ability to learn a novel motor task, and demonstrate the potential for experience-dependent plasticity in the spinal cord in the absence of supraspinal connectivity. Abstract: Forelimb/hand motor cortex injury in rodents and primates causes impairments in skilled paw/hand movements that includes a period of movement absence followed by functional recovery/compensation. Although the postsurgical period of movement absence has been attributed to “shock” or “diaschisis”, the behavior of animals during this period has not been fully described. Here, rats were trained to reach for single food pellets from a shelf and then the vasculature of the forelimb region of the sensorimotor cortex contralateral to the reaching limb was removed. A control group received a posterior parietal cortex devasularization. Frame-by-frame video analysis of reaching behavior showed that the stages of the acquisition of skilled reaching and the stages of recovery after motor cortex stroke were similar. The animals sequentially learn three relationships or “oppositions” between a body part and the food target. The oppositions are invariant relationships but each can be achieved with movements that can vary from reach to reach and between rats. A snout-pellet opposition organizes the movements of orienting, a paw-pellet opposition organizes limb transport and grasping the pellet in the digits, and a mouth-pellet opposition organizes limb withdrawal and the release of the food into the mouth. The three oppositions and the movements that they recruit were disrupted after motor cortex damage, but not parietal cortex damage. The oppositions were reestablished after stroke in the order in which they were acquired prior to stroke. Enduring impairments were more noticeable in transport and withdrawal oppositions. That the stages of recovery from motor cortex stroke parallel those of initial acquisition are discussed in relation to contemporary explanations of diaschisis and the contribution of motor cortex to motor learning. Abstract: Methyl-β-carboline-3-carboxylate (or β-CCM) is a benzodiazepine receptor ligand with inverse agonist properties. Two strains of mice were selected, one for sensitivity (BS) and one for resistance (BR) to a convulsive dose of β-CCM. These two strains were then shown to differ in several biochemical, pharmacological and behavioral characteristics; specifically BS mice were less anxious than BR mice. The present work provides evidence of differences in the learning abilities of the two strains. Three different learning tasks were used: spatial delayed discrimination on the 4-hole board, a learned choice between a lit and a dark compartment in a T-maze, and place-learning in an 8-arm radial maze. Abstract: It is now well accepted that besides its roles in motor control, the cerebellum is involved in non-motor functions with emotional aspects. Consistent with this view, several studies highlighted that the cerebellar Lurcher mutant mice (+/Lc), with motor impairments, also exhibited altered emotional reactivity, previously interpreted in term of behavioural disinhibition. In this study, we investigated the effects of a classical anxiolytic on such disinhibition. For that, behaviours of +/Lc and control (+/+) mice injected with NaCl or chlordiazepoxide (CDP 5 and 7.5mg/kg) were evaluated in the elevated plus-maze test. The motor impact of the drug (7.5mg/kg only) was also evaluated in the hole-board and unstable platform tests. Our results showed that, compared to the +/+ mice, CDP injection greatly influenced the anxious-related behaviours in the +/Lc mice by reducing their preference to the open areas in the elevated plus-maze test. Furthermore, we found that injection of CDP at the dose of 7.5mg/kg aggravated motor coordination deficit, altered motor learning capabilities in the mutants and provoked equilibrium disturbances in the non-mutant mice in the unstable platform test. These results indicated that CDP was able to reduce behavioural disinhibition in the cerebellar +/Lc mice and were discussed in term of implication of the cerebellar connections into CDP-sensitive neural circuitries involved in both emotional and motor processes. Abstract: Some rats of the WAG/Rij (Wistar Albino Glaxo from Rijswijk) and Wistar strain are susceptible for audiogenic (convulsive) seizures. In the present study, behavior of susceptible and non-susceptible rats from the WAG/Rij strain, genetically predisposed to absence epilepsy, and outbred Wistar strain, genetically not predisposed to absence epilepsy, was compared to assess the level of anxiety (in the open field, light–dark choice and elevated plus-maze tests) and the level of depression (in the sucrose consumption and forced swimming tests). Increased level of anxiety was found only in audiogenic susceptible rats both from WAG/Rij and Wistar strain, but increased level of depression was found only in WAG/Rij rats independently of their susceptibility to audiogenic seizures. The results suggest that enhanced level of depression in WAG/Rij strain rats is associated with absence epilepsy but enhanced level of anxiety with susceptibility to audiogenic seizures. Abstract: Weaning is one of the most important events in the early stage of life, and recently we have found that precocious weaning augments anxiety and aggressiveness in mice. Maternal behavior has been reported to be transmitted from one generation to the next; that is, female pups that received intensive maternal care showed higher maternal behavior in their adulthood. In the present study, the following three experiments were conducted to understand maternal behavior transmission in early-weaned mice that were separated from the dam on postnatal day 14. First, the maternal behavior observed from the postpartum day 15 to 21, which was deprived in the early-weaned mice, were analyzed. Mothers spent 3% of their time on licking/grooming and arched-back nursing of their pups on postpartum day 15, and the time spent on these behaviors was gradually decreased until postpartum day 21; however, they spent 50% of their time attending to their pups through postpartum days 15–21. Simultaneously, the behavior of the pups was monitored, and it was found that the early-weaned mice had higher activity and lower resting behavior over the period from postnatal day 15 to 21. Secondly, the early- and normally weaned female mice were subjected to an elevated plus maze test at the age of 8 weeks to assess their anxiety level. The early-weaned mice showed a lower frequency of entering the open arms, and a shorter duration of time spent within them, as compared to the normally weaned mice, suggesting that early-weaned females had a higher anxiety level. In the third experiment, the two groups of female mice were paired with adult male mice for 2 weeks, and the mother''s maternal behavior was analyzed. The early-weaned female mice showed lower frequency of licking/grooming and arched-back nursing of their pups as compared to the normally weaned mice, whereas the time of mother-off pups and attending to pups were not different between groups. These results suggest that early-weaning manipulation deprives offspring of a certain level of maternal care, and as a consequence, the offspring show higher anxiety levels and lower maternal behavior in their own adulthood. Abstract: Wild-type and mutant mice lacking expression of α5, α7, β2, β3, or β4 neuronal nicotinic cholinergic receptors (nAChRs) were compared on a signaled nose poke task, a multi-phased task used to measure appetitive learning and impulsivity. In the early phases of training, mutants of all nicotinic lines did not differ compared to wild types in the days to reach criterion when mice were required to nose poke for a sucrose reward on FR1 or FR3 schedules, or in their ability to respond to an auditory clicker to receive a sucrose reward. However, mutants lacking α7 nAChRs, but not lines lacking other nAChRs, showed impairments when task difficulty was increased such that an auditory stimulus was presented on a variable schedule and mice were required to withhold their responses until the presentation of the auditory cue to obtain a reward. α7 mutants were impaired compared to wild types in appetitive learning as measured by the percentage of conditioned responses but overcame their deficits with extensive training for 10 days. However, when efficiency ratios were used to measure impulsivity, α7 mutants exhibited lower efficiency ratios even after 10 days of training. These results support a role of the α7 nicotinic receptor in mediating appetitive learning and suggest a potential role for the α7 nAChRs in the regulation of behavioral disinhibition. Abstract: Habituation (a form of non-associative learning) was measured by assessing locomotion in novel activity monitors in CB1 receptor knockout mice and juxtaposed to habituation measured in muscarinic M2, M4, and double M2/M4 receptor knockout mice. M2 and M2/M4, but not M4, receptor knockout mice appeared to have an impaired ability to habituate, whereas CB1 receptor knockout mice showed enhanced habituation compared to wild-type animals. We conclude that CB1 receptor gene invalidation improves habituation tentatively through an increase in cholinergic neurotransmission. [Copyright &y& Elsevier]
