The Raton Basin of southern Colorado and northern New Mexico is an actively produced hydrocarbon basin that has experienced increased seismicity since 2001, including the August 2011 Mw5.3 Trinidad normal faulting event. Following the 2011 earthquake, regional seismic observations were used to relocate 21 events, including the 2011 main shock, two foreshocks, and 13 aftershocks. Additionally, interferometric synthetic aperture radar (InSAR) observations of both the 2011 event and preevent basin deformation place constraint on the spatial kinematics of the 2011 event and localized basin subsidence due to ground water or gas withdrawal. We find that the 2011 earthquake ruptured an 8–10 km long segment of a normal fault at depths of 1.5–6.0 km within the crystalline Precambrian basement underlying the Raton Basin sedimentary rocks. The earthquake also nucleated within the crystalline basement in the vicinity of an active wastewater disposal site. The ensuing aftershock sequence demonstrated statistical properties expected for intraplate earthquakes, though the length of the 2011 earthquake is unexpectedly long for an Mw5.3 event, suggesting that wastewater disposal may have triggered a low stress drop, otherwise natural earthquake. Additionally, preevent and postevent seismicity in the Raton Basin spatially correlates to regions of subsidence observed in InSAR time series analysis. While these observations cannot discern a causal link between hydrocarbon production and seismicity, they constrain spatial relationships between active basin deformation and geological and anthropogenic features. Furthermore, the InSAR observations highlight the utility of space‐based geodetic observations for monitoring and assessing anthropogenically induced and triggered deformation. Project ABSOLUTE (advanced battery solid oxide fuel cell linked unit to maximize efficiency), aims to combine a sodium-nickel chloride battery and an intermediate temperature solid oxide fuel cell (IT-SOFC) to form an all-electric hybrid package that surpasses the efficiency and performance of a purely fuel cell driven vehicle, as well as extending the range of a purely battery driven electric vehicle. This paper discusses the project background, the ABSOLUTE hybrid concept, the methodology adopted, the vehicle types and drive cycles that best suit the hybrid and system control considerations. Results from a battery and IT-SOFC system model are presented.

ABSTRACTTritrichomonas foetusis a serious veterinary pathogen, causing bovine trichomoniasis, a sexually transmitted disease leading to infertility and abortion. T. foetusinfects the mucosal surfaces of the reproductive tract. Infection with T. foetusleads to apoptotic cell death of bovine vaginal epithelial cells (BVECs) in culture. An affinity-purified cysteine protease (CP) fraction yielding on sodium dodecyl sulfate-polyacrylamide gel electrophoresis a single band with an apparent molecular mass of 30 kDa (CP30) also induces BVEC apoptosis. Treatment of CP30 with the protease inhibitors TLCK (Nα-p-tosyl-l-lysine chloromethyl ketone) and E-64 [l-trans-epoxysuccinyl-leucylamide-(4-guanido)-butane] greatly reduces induction of BVEC apoptosis. Matrix-assisted laser desorption ionization-time-of-flight MALDI-TOF mass spectrometry analysis of CP30 reveals a single peak with a molecular mass of 23.7 kDa. Mass spectral peptide sequence analysis of proteolytically digested CP30 reveals homologies to a previously reported cDNA clone, CP8 (D. J. Mallinson, J. Livingstone, K. M. Appleton, S. J. Lees, G. H. Coombs, and M. J. North, Microbiology 141:3077-3085, 1995). Induction of apoptosis is highly species specific, since the related human parasite Trichomonas vaginalisand associated purified CPs did not induce BVEC death. Fluorescence microscopy along with the Cell Death Detection ELISAPLUSassay and flow cytometry analyses were used to detect apoptotic nuclear condensation, DNA fragmentation, and changes in plasma membrane asymmetry in host cells undergoing apoptosis in response to T. foetusinfection or incubation with CP30. Additionally, the activation of caspase-3 and inhibition of cell death by caspase inhibitors indicates that caspases are involved in BVEC apoptosis. These results imply that apoptosis is involved in the pathogenesis of T. foetusinfection in vivo, which may have important implications for therapeutic interference with host cell death that could alter the course of the pathology in vivo.

The clinical, criminological and demographic characteristics of a Special Hospital population of male patients convicted of homicide are described. The results confirm much previous work on mentally abnormal homicide. Typically the patients come from a disadvantaged social background with poor employment records, a prior history of aggressive behaviour and a diagnosis of schizophrenia. Victims were usually known to the patient and often came from within his circle of family and friends. Only 25% of the patients were receiving treatment at the time of the offence. These findings are discussed in the light of the current literature.

In this paper we report the complete bovine β-lactoglobulin gene sequence, consisting of seven exons within a total of 4724 base pairs. The inferred amino acid sequence agrees completely with that determined directly for the ? variant. The bovine β-lactoglobulin gene sequence shows 89% homology to the corresponding ovine sequence, with complete conservation of gene organization and splice sites, though a significant proportion of CG dinucleotides have been lost since the species diverged. Comparison with the previously reported cDNA sequence reveals a number of nucleotide substitutions in addition to those which distinguish the A and ? variants. Three of these are silent mutations, however it appears that the occurrence in the cDNA sequence of a Val residue at position 105 may represent a new variant provisionally designated β-lactoglobulin K, derived from β-lactoglobulin A.

We present a comparative study of time-integrated four-wave-mixing and femtosecond emission under resonant excitation on excitons in weakly disordered GaAs quantum wells. At highest exciton densities when dephasing dominates the spectral width (homogeneous broadening), we find that the rise time of the incoherent luminescence signal is given by TIii 2/2. At lowest densities, optical coherence times approach the exciton radiative lifetime (15 to 20 ps). This confirms our previous result that coherent resonant Rayleigh scattering is responsible for the short rise time of the excitonic emission. We also show clear evidence for dephasing due to exciton–phonon interaction, as the rise time of the emission decreases dramatically when the sample temperature is increased.

Significant disproportionation between the mono- and 2,5-bis-metalaled intermediates resulting from metalation of p-DMB has been observed. The best procedure for the generation of each of these intermediates involves use of an “incremental” amount of TMEDA.1

Time-resolved photoluminescence experiments are performed to study carrier relaxation in strain-induced (GaIn)As quantum dot structures. We observe a sub-picosecond onset of the photoluminescence from the lowest quantum-dot electron–hole transition after optical excitation of higher-energetic barrier states. This shows that carrier capture into the quantum-dot ground state can be extremely fast at high electron–hole densities. The recombination lifetime of the lowest quantum-dot transition and the interlevel relaxation time are extracted from the decay of the photoluminescence transients.

Regulation of dolichyl phosphate-linked oligosaccharide assembly has been studied during the course of diethylstilbestrol-induced chick oviduct differentiation. Oviduct membranes from treated chicks form 4.6 times as much GlcNAc-P-P-Dol and GlcNAc2-P-P-Dol upon incubation with UDP-[14C]GlcNAc and MgCl2 than do membranes from untreated chicks. Assembly of oligosaccharide-lipid was studied by incubation of membranes with purified exogenous [14C]GlcNAc2-P-P-Dol and GDP-Man. Man transfer required a divalent cation (10 mM Mg2+) and detergent (0.5% Nonidet P-40 is optimal) and occurs in the presence of amphomycin (500 micrograms/ml). The apparent Km for GDP-Man is 1 microM and for [14C]GlcNAc2-P-P-Dol is 0.45 microM. The products are a series of sequentially formed dolichyl pyrophosphate-linked saccharides up to Man5GlcNAc2, the first of which is Man beta 1,4GlcNAc2. The same products are formed either in the presence or absence of amphomycin. Conversion of GlcNAc2-P-P-Dol to higher oligosaccharides is stimulated 3-fold by estrogen treatment of chicks. Similarly, the conversion of partially purified exogenously added Man beta-[14C]GlcNAc2-P-P-Dol is 4.6-fold higher after diethylstilbestrol treatment.

We have purified a human non-pancreatic phospholipase A2that is present in platelets and is enriched in rheumatoid synovial fluid. The enzyme is calcium-dependent, has a pH optimum of 8–10, and shows a striking preference for substrate presented in the form of Escherichia colimembranes. In the E. coliphospholipase A2assay the phospholipase exhibits an apparent specific activity of 300 µmol/mg/min. Using oligonucleotide probes based on amino-terminal sequence data, we cloned the corresponding human gene from a genomic DNA library and expressed the gene in animal cells. The protein was secreted from the cells in an active form. The deduced amino acid sequence of the human protein consists of 124 amino acids, contains structural features common to all known phospholipase A2s, and has a half-cystine pattern that is characteristic of the snake venom group II enzymes.

The possibility that the insulin receptor of adipocytes undergoes cycling was examined by a method involving pronase digestion at 12 degrees C, followed by insulin binding studies to determine receptor location and quantity. In the absence of insulin treatment, the amount of internal receptors (i.e. protected from pronase) was 10% of total receptor content. Following a 30-min insulin treatment (0.1 microM) at 37 degrees C, the internal receptor content increased 2-fold (206 +/- 12% of control, 100%). This effect was rapid, and maximum internalization was approached by 5 min of insulin treatment. Warming pronase-digested cells to 37 degrees C allowed the internal receptors to move to the cell surface. This movement was rapid also, and expansion of the internal pool by insulin pretreatment provided a 2.4-fold increase in the reinsertion of cell-surface receptors (238 +/- 28% of nontreated cells, 100%). Insulin-pretreated and nontreated cells had approximately 13 and 6%, respectively, of their original cell-surface receptor content, i.e. their content before pronase digestion. These receptors appeared intact after the cycling process, as judged by affinity labeling and sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the receptor and its binding subunit. The ability of the recycled receptor to respond to insulin was examined by studies of glucose incorporation into lipids and the inhibition of isoproterenol-stimulated lipolysis. Cells pretreated with insulin and allowed to recycle (e.g. 13% of normal receptor content) were 2-3-fold more responsive and 7-fold more sensitive to subsequent insulin stimulation than nontreated cells (e.g. 6% of normal receptor content), indicating that the recycled receptors are biologically active and coupled to cellular effector systems.

Phosphate (Pi) reabsorption in renal proximal tubules involves Na+/Pi cotransport across the brush border membrane; its transport rate is influenced by the Na(+)-coupled transport of other solutes as well as by pH. In the present study, we have expressed a cloned rat renal brush border membrane Na+/Pi cotransporter (NaPi-2) in Xenopus laevis oocytes and have analyzed its electrophysiologic properties in voltage- and current-clamp studies. Addition of Pi to Na(+)-containing superfusates resulted in a depolarization of the membrane potential and, in voltage-clamped oocytes, in an inward current (IP). An analysis of the Na+ and/or Pi concentration dependence of IP suggested a Na+/Pi stoichiometry of 3:1. IP was increased by increasing the pH of the superfusate; this phenomenon seems to be mainly related to a lowering of the affinity for Na+ interaction by increasing H+ concentration. The present data suggest that known properties of Pi handling at the tubular/membrane level are "directly" related to specific characteristics of the transport molecule (NaPi-2) involved.

The oxidant H2O2 has many insulin-like effects in rat adipocytes. To determine whether these effects could be mediated by the tyrosine kinase activity of the insulin receptor, the ability of H2O2 to stimulate receptor phosphorylation in intact adipocytes and partially purified insulin receptors has been examined. Phosphorylation of the beta subunit of the insulin receptor was increased approximately 2-fold by treatment of intact cells with 3 mM H2O2, a concentration that maximally stimulates 2-deoxyglucose uptake. Stimulation of receptor phosphorylation was rapid, reaching maximal levels within 5 min, and preceded activation of glucose transport. Phosphoamino acid analysis of insulin receptors from H2O2-treated adipocytes showed that 32P incorporation into phosphotyrosine and phosphoserine residues of the beta subunit was enhanced. Furthermore, partially purified receptors from H2O2-treated cells exhibit increased tyrosine kinase activity, as measured by phosphorylation of the peptide Glu80Tyr20. In contrast, the direct addition of H2O2 to partially purified insulin receptors did not stimulate tyrosine kinase activity or insulin receptor autophosphorylation. This was not due to breakdown of H2O2 or oxidation of ATP or the required divalent cations. To define the factors involved in H2O2's effect, we have examined receptor phosphorylation in fat cell homogenates and purified plasma membranes. Although insulin stimulated receptor phosphorylation in both of these systems, H2O2 was only effective in the cell homogenates. These data demonstrate that, under certain conditions, H2O2 stimulates insulin receptor phosphorylation and tyrosine kinase activity, suggesting that the insulin-like effects of H2O2 may be mediated by stimulation of insulin receptor phosphorylation. This does not appear to be a direct effect of H2O2 on the insulin receptor and requires nonplasma membrane cellular constituents.

A novel sustained release tablet formulation of tiaprofenic acid, that has been designed to disintegrate into discrete pellets in the gastrointestinal tract, has been evaluated using a combination of gamma scintigraphy and pharmacokinetics. The performance of the product in vivo was very satisfactory, in that the disintegration of the tableted pellet formulation was rapid. The gastrointestinal transit behaviour of pellets was affected by food, but food did not affect the subsequent absorption of the released drug.
