The conservation and maintenance of a monumental stone building is a complex aim where different disciplines are involved. First step is concerning the determination of the state of conservation of stone material present, than determine its modification on time, as such and after conservation treatments applied on: cleaning, protecting, strengthening. In order to fulfill such objectives suitable parameters must be selected and the most appropriate diagnostic techniques for their quantitative evaluation operated. In this context, the determination of the surface water absorption, moisture content, colour variation and mechanical properties are important parameters for the control, and the monitoring over time, of the state of conservation of the monumental stone surfaces. These parameters are strongly related not only to the stone characteristics but also to the evaluation of products’s performance: efficiency and durability. Their rate of variation, determined in monitoring campaigns, is fundamental for elaborate by properly predictive model a schedule maintenance protocol.

Abstract Background Marine sediments represent one of the planet’s largest carbon stores. Bottom trawl fisheries constitute the most widespread physical disturbance to seabed habitats, which exert a large influence over the oceanic carbon dioxide (CO2) sink. Recent research has sparked concern that seabed disturbance from trawling can therefore turn marine sediments into a large source of CO2, but the calculations involved carry a high degree of uncertainty. This is primarily due to a lack of quantitative understanding of how trawling mixes and resuspends sediments, how it alters bioturbation, bioirrigation, and oxygenation rates, and how these processes translate into carbon fluxes into or out of sediments. Methods The primary question addressed by this review protocol is: how does mobile bottom fishing affect benthic carbon processing and storage? This question will be split into the following secondary questions: what is the effect of mobile bottom fishing on: (i) the amount and type of carbon found in benthic sediments; (ii) the magnitude and direction of benthic-pelagic carbon fluxes; (iii) the biogeochemical, biological, and physical parameters that control the fate of benthic carbon; and (iv) the biogeochemical, biological, and physical parameters that control the fate of resuspended carbon. Literature searches will be conducted in Web of Science, SCOPUS, PROQUEST, and a range of grey and specialist sources. An initial scoping search in Web of Science informed the final search string, which has been formulated according to Population Intervention Comparator Outcome (PICO) principles. Eligible studies must contain data concerning a change in a population of interest caused by mobile bottom fishing. Eligible study designs are Before and After, Control and Impact, and Gradient studies. Studies included at full-text screening will be critically appraised, and study findings will be extracted.Extracted data will be stored in an Excel spreadsheet. Results will be reported in narrative and quantitative syntheses using a variety of visual tools including forest plots. Meta-analysis will be conducted where sufficient data exists.

Abstract Background Endothelial cellular senescence is emerging as a key mechanism of age-related vascular dysfunction. Disruption of the endothelium glycocalyx and shedding of the syndecan (SDC) ectodomains have been associated with several age-related diseases. Although SDC4 is highly expressed in endothelial cells, its levels and shedding in senescent endothelial cells and vascular endothelial dysfunction associated with aging are still unknown. Methods To assess whether SDC4 expression was affected by inflammatory conditions, we evaluated its levels in young human umbilical vein endothelial cells (HUVECs) treated with TNF-α at a concentration of 50 ng/mL for 24 h and in cells undergoing replicative senescence. Plasma levels of SDC4 were evaluated in two previously recruited cohorts of (i) subjects with type 2 diabetes (T2D, n = 110) followed for a median of 16.8 years and age- and gender-matched healthy subjects (n = 100), and (ii) middle-aged subjects with mild-to-moderate dyslipidemia. Binomial logistic regression was used to assess whether SDC4 levels could be prognostic for major adverse cardiovascular events (MACE). Results In the in vitro study, we showed that HUVECs, when exposed to TNF-α or undergoing replicative senescence, exhibited elevated expression levels of SDC4 and matrix metallopeptidase 9 (MMP-9), as well as increased shedding of SDC4 into the extracellular microenvironment, in comparison to actively proliferating young HUVECs. Analysis of human samples revealed that patients with T2D without complications exhibited higher SDC4 levels compared to healthy controls and those with T2D vascular complications. In particular, patients with a history of major adverse cardiovascular events (MACE) had lower SDC4 levels. The longitudinal evaluation revealed that higher SDC4 levels predict the onset of new MACE during a 16.8-year follow-up. In the second cohort, no significant association was observed between SDC4 and endothelial dysfunction, assessed by flow-mediated dilation (FMD) or nitric oxide metabolites. SDC4 levels correlated positively with C-reactive protein (CRP) in both cohorts and with PAI-1 in the cohort of patients with T2D. Conclusion Overall, we conclude that the shedding of SDC4 from endothelial cells increases under acute (TNF-α treatment) and chronic (senescence) inflammatory conditions and that increased circulating SDC4 levels are associated with systemic inflammation in pathological aging.

Marine sediments are among the largest carbon reservoirs on the planet and play a key role in the global cycling of organic matter. Bottom fisheries are the most widespread anthropogenic physical disturbance to seabed habitats, prompting NGOs and governments to act on regulating mobile bottom-contacting fishing gear. However, the scientific evidence of the effects of bottom trawling on sediment biogeochemistry is highly diverse and presents contrasting results. Here we present a global harmonized dataset of 71 independent studies that assess the effects of demersal fisheries on sedimentological (i.e. grain size, porosity) and biogeochemical (i.e. organic carbon, phytopigments, nutrient fluxes) properties: the Demersal fishery Impacts on Sedimentary Organic Matter (DISOM) database (Paradis, 2023; https://doi.org/10.3929/ethz-b-000634336). We identify considerable gaps, namely in the geographical extension of the data; coverage of environmental predictors (i.e. seasons); fishing descriptors such as the availability of true controls, quantification of fishing effort, and distribution of fishing gear types; and biogeochemical variables that study the remineralization of organic matter. Future studies should address these data gaps to enhance the comprehensiveness of the dataset. With this harmonized database, we aim to allow researchers to explore the effects of demersal fisheries in variable environmental settings to disentangle the effects of this disturbance and provide efficient management strategies.

Previous research has already examined the relationship between Future Anxiety, a construct recently introduced in Italy, and mental health in young adults, although possible mediating variables in this relationship have so far never been investigated. The present study attempts to fill this gap by exploring the incidence of Future Anxiety on psychological distress (i.e., Stress, Anxiety and Depression) in a group of 302 young Italian adults (18–30 years; M = 21.9; SD = 2.6; 49.0% males; 51.0% females), presenting and evaluating the simultaneous mediating effect of Intolerance of Uncertainty and Non-Pathological Worry. Findings highlighted how Future Anxiety had a positive and significant direct effect on Stress and Depression, but not on Anxiety. In the three serial mediation models proposed, Intolerance of Uncertainty and Non-Pathological Worry mediated the relationship between Future Anxiety and mental health outcomes. The results also confirmed the hypothesized serial mediation effect by highlighting how young adults with greater Future Anxiety experienced more Intolerance of Uncertainty, which positively affected Non-Pathological Worry levels and, in turn, exacerbated psychological distress. Finally, results indicated that female participants experienced more Stress, Anxiety, and Depression in relation to Future Anxiety compared to males. Starting from the review of main references on this subject, the results discussed provide new insights for understanding youth psychological distress. Finally, practical implications for the design of supportive interventions for this study’s target group are proposed.

Lipid overload or metabolic stress has gained popularity in research that explores pathological mechanisms that may drive enhanced oxidative myocardial damage. Here, H9c2 cardiomyoblasts were exposed to various doses of palmitic acid (0.06 to 1 mM) for either 4 or 24 h to study its potential physiological response to cardiac cells. Briefly, assays performed included metabolic activity, cholesterol content, mitochondrial respiration, and prominent markers of oxidative stress, as well as determining changes in mitochondrial potential, mitochondrial production of reactive oxygen species, and intracellular antioxidant levels like glutathione, glutathione peroxidase and superoxide dismutase. Cellular damage was probed using fluorescent stains, annexin V and propidium iodide. Our results indicated that prolonged exposure (24-hours) to palmitic acid doses ≥ 0.5 mM significantly impaired mitochondrial oxidative status, leading to enhanced mitochondrial membrane potential and increased mitochondrial ROS production. While palmitic acid dose of 1 mM appeared to induce prominent cardiomyoblasts damage, likely because of its capacity to increase cholesterol content/ lipid peroxidation and severely suppressing intracellular antioxidants. Interestingly, short-term (4-hours) exposure to palmitic acid, especially for lower doses (≤ 0.25 mM), could improve metabolic activity, mitochondrial function and protect against oxidative stress induced myocardial damage. Potentially suggesting that, depending on the dose consumed or duration of exposure, consumption of saturated fatty acids such as palmitic acid can differently affect the myocardium. However, these results are still preliminary, and in vivo research is required to understand the significance of maintaining intracellular antioxidants to protect against oxidative stress induced by lipid overload.

Development and aging significantly impact the cellular levels of Coenzyme Q (CoQ), which is associated with both pathological and physiological conditions. Aim of this study was to describe the CoQ status throughout the lifetime of Drosophila melanogaster, a well-established model in aging studies. CoQ9 and CoQ distribution was analysed across different body segments and various life stages in both male and female flies. The results indicate that CoQ9 is the predominant isoform in every phase of flies’ life cycle, with the highest concentrations observed in the thorax. We noted distinct trends in CoQ distribution during aging, which varied according to sex and body segments (head, thorax, and abdomen). Supplementation with two concentrations of CoQ9 and CoQ10 (15 μM and 75 μM) for 2 weeks induced a segment- and sex-specific CoQ uptake. Although 75 μM CoQ10 was more effective in modulating the CoQ status, lifelong treatment with this concentration did not affect the longevity of the flies.

Abstract This study explores the impact of a wind storm on sediment resuspension and marine biogeochemical dynamics. Additionally, the storm took place during an expedition researching bottom trawling, enabling the direct comparison of certain natural and fisheries-related disturbances. The storm was initiated by a decline in atmospheric pressure and a 2 h period of gale force winds, which was followed by over 40 h of elevated bottom currents. Storm induced turbidity, potentially a cumulative post-fishing impact, was remarkably higher compared to what was observed in a recent trawling event. Storm-induced mixing and movement of water masses led to decreased silicate and increased phosphate concentrations in the water column, accompanied by lower salinity and higher fluorescence. The erosion depth of the seabed averaged around 0.3 cm during the peak turbidity period. Trawl-induced erosion in the area has been measured at over twice that depth, and has been linked to intermittent reductions in near-bed oxygen levels. In contrast, storm-induced turbidity coincided with increased oxygen due to wave mixing, suggesting inherent differences in how trawling and storms can oxidize reduced substances. These findings suggest that storms have a greater regional impact, whereas the local impacts of bottom trawling on biogeochemistry can be more significant.

The Russian–Ukrainian conflict is affecting mental health even in communities that are not directly involved in the war; added to this is the escalating conflict in the Middle East and its dangerous spread, which brings the war back to the center of the contemporary social and economic horizon. The present study aims to explore the psychological impact of war in a sample of 310 Italian young adults (18–30 years; M = 22.0; SD = 2.6) while exploring the relationship between Fear of War and psychological distress and evaluating the mediating effects of Future Anxiety and Intolerance of Uncertainty in this relation. Findings highlighted how Fear of War positively and significantly affects Stress, Anxiety, and Depression, and, at the same time, how it fuels both Future Anxiety and Intolerance of Uncertainty. These constructs, in addition to positively affecting the mental health outcomes considered, mediate the relationship between Fear of War and youth psychological distress with a significant indirect effect observed in all three mediation models performed. Finally, significantly higher levels of psychological distress, Fear of War, and Future Anxiety are reported in women than in men. The findings are discussed with reference to the recent literature on the psychological impact of war and on contemporary youth psychological distress, indicating the importance of educational policies and targeted interventions aimed at supporting this target in coping with multiple contemporary collective stressors.

BackgroundHepatoblastoma (HB) is the most common pediatric hepatic malignancy. Despite the progress in HB treatment, investigating HB pathomechanisms to optimize stratification and therapies remains a focal point to improve the outcome for high-risk patients.MethodsHere, we pointed to explore the impact of these mechanisms in HB. An observational study was performed on liver samples from a cohort of 17 patients with a diagnosis of HB and two normal liver samples. The in vitro experiments were executed on the Huh6 human HB cell line treated with the FAK inhibitor TAE226.ResultsOur results highlight a significant up-regulation of mRNA and protein expression of FAK in livers from HB with respect to normal livers. The increased protein expression of total and Tyr397 phosphorylated FAK (pTyr397FAK) was significantly correlated with the expression of some epigenetic regulators of histone H3 methylation and acetylation. Of note, the expression of pTyr397FAK, N-methyltransferase enzyme (EZH2) and tri-methylation of the H3K27 residue correlated with tumor size and alpha-fetoprotein (AFP) levels. Finally, TAE226 caused a significant reduction of pTyr397FAK, epigenetic regulators, AFP, EPCAM, OCT4, and SOX2, in association with anti-proliferative and pro-apoptotic effects on HB cells.ConclusionOur results suggest a role of FAK in HB that requires further investigations mainly focused on the exploration of its effective diagnostic and therapeutic translatability.

IntroductionThe importance of the response-effect trait dichotomy in marine benthic ecology has garnered recent attention. Response traits, characterising species responses to environmental variations, have been a dominant focus in the development of ecological indicators for ecosystem health assessment. In contrast, effect traits, expressing effects of organism activities on the ecosystem, still do not benefit from an equal interest in spite of the complementary facet that they provide to complete our understanding of functional diversity and ecosystem vulnerability. In this study, we explore the consequences of disturbance by bottom trawl fisheries on benthic effect trait composition.MethodsTo this end, we used different contexts of environmental and trawling conditions from thirteen case studies in European waters and apply the same analytical procedure to derive a gradient that solely account for trawling-induced disturbance (Partial RLQ analysis).ResultsBottom trawling was found to be a selective force of benthic effect trait composition in a majority of case studies. In general, tube-dwelling species were more typical of low trawling frequencies, whereas deep burrowing species were more resistant at high trawling frequencies. Although we report significantly deleterious effects of trawling on benthic ecosystem functions, the effect trait pattern along the gradient was never related to life span, a key response trait generally assumed to express recoverability following disturbance. Furthermore, we show that trends in species multi-functionality and community functional diversity can be negative or positive along the trawling intensity gradient.DiscussionWe discuss the relevance of these results in light of recent developments in the framework of response and effect trait dichotomy, and provide guidelines of trait data analysis in the context of trawl fisheries impact on the sea floor. Our findings emphasize the importance of fundamental concepts from functional ecology in this context and represent a first step toward an assessment of trawling effect more oriented on benthos-mediated biogeochemical processes.

Em universidades multicampi, garantir a acessibilidade dos estudantes a projetos em outros campi é um desafio. Neste trabalho é relatada uma experiência interdisciplinar e multicampi em um projeto de extensão da UFVJM, durante os anos de 2021 e 2022. No total, estiveram envolvidos 45 alunos, de 3 campi e 10 cursos distintos, em atividades conduzidas de forma remota, tais como, a criação de conteúdo textual e material audiovisual, culminando também na realização de um workshop online, trabalhos de conclusão de curso e publicação de artigos científicos. As atividades remotas ampliaram a acessibilidade ao projeto e ofereceram flexibilidade de horários para professores e estudantes, além de permitir a interação entre discentes de diferentes cursos. No entanto, houveram desafios devido à comunicação ineficiente durante o processo de seleção de estudantes, a falta de acesso à internet e recursos computacionais de alta qualidade.

There is an increasing interest in the use of nutraceuticals and plant-derived bioactive compounds from foods for their potential health benefits. For example, as a major active ingredient found from cruciferous vegetables like broccoli, there has been growing interest in understanding the therapeutic effects of sulforaphane against diverse metabolic complications. The past decade has seen an extensive growth in literature reporting on the potential health benefits of sulforaphane to neutralize pathological consequences of oxidative stress and inflammation, which may be essential in protecting against diabetes-related complications. In fact, preclinical evidence summarized within this review supports an active role of sulforaphane in activating nuclear factor erythroid 2-related factor 2 or effectively modulating AMP-activated protein kinase to protect against diabetic complications, including diabetic cardiomyopathy, diabetic neuropathy, diabetic nephropathy, as well as other metabolic complications involving non-alcoholic fatty liver disease and skeletal muscle insulin resistance. With clinical evidence suggesting that foods rich in sulforaphane like broccoli can improve the metabolic status and lower cardiovascular disease risk by reducing biomarkers of oxidative stress and inflammation in patients with type 2 diabetes. This information remains essential in determining the therapeutic value of sulforaphane or its potential use as a nutraceutical to manage diabetes and its related complications. Finally, this review discusses essential information on the bioavailability profile of sulforaphane, while also covering information on the pathological consequences of oxidative stress and inflammation that drive the development and progression of diabetes.

Background: The COVID-19 pandemic as well as the social, relational, and economic transformations of the lockdown had a strong traumatic impact on the mental health of the word population, even more so on specific targets such as young adults. The spread of what was an unknown virus and the lockdown experience have raised concerns for the preservation of people’s physical and mental health and their private, relational, and work life. Objectives: The current study presents the construction, development, and validation process of the Lockdown Young Adult Concerns Scale (LYACS), an instrument assessing the level of two principal concerns during the pandemic lockdown, i.e., Loss of Life Control (CLLC) and Infection/Contagion (CIF). Methods: After a pilot study that defined the dimensions, selected the items, and carried out Exploratory Factor Analysis (EFA) on a sample of 100 subjects (M = 24; DS = 3.9), a new sample of 259 Italian young adults (ages 18-35 yrs.; M = 24; SD = 3.8) was used to assess the psychometric validity of LYACS through Confirmatory Factor Analysis (CFA), reliability, convergent and divergent validity. Further descriptive analyses of the final version of scale as well as Two-way Analyses of Variance (ANOVA) on selected sociodemographic variables were conducted. Results: The outcome of Confirmatory Factor Analysis, which resulted in high goodness of fit (χ2/df = 2.0; GFI = .98; TLI = .98; RMSEA = .06; SRMR = .04), supports two identifiable factors reflecting the theoretically-based constructs of LYACS, thus supporting the EFA results. Furthermore, internal consistency as well as convergent and divergent validity analyses allow the scale to be considered a reliable and valid instrument for the present investigation. Finally, variance analysis shows that there are significant differences among the factor levels regarding gender, occupation, perception of the home space during lockdown, and having/not having worked on Concern about Loss of Life Control (CLLC), and between gender and confinement cohabitation on Concern about Infection/Contagion (CIF). Conclusion: Overall, the reported results show good psychometric proprieties for the scale and shed new light on how Italian young adults lived the COVID-19 pandemic.

Dynamic, sandy environments are generally less vulnerable to mechanical stress compared to silty, low-energy habitats. Biogenic reef communities, however, may provide an exception to this. This study explores the physical, biological, and biogeochemical effects of bottom trawl fishing on a coastal ecosystem dominated by the tube-building polychaete Lanice conchilega. Two specific gear types, both used to exploit North Sea sole (Solea solea), were compared: electric pulse trawls and tickler chain-rigged beam trawls. We detected a ∼1 cm bathymetric deepening after trawling associated with significant losses in benthic chlorophyll a caused by both fishing gears. Tickler chain trawls significantly reduced sediment oxygen consumption (57 %), total organic matter mineralization (56 %), denitrification (61 %), nitrification (60 %), and total benthos densities (52 %), while pulse trawls had no statistically significant impact on these parameters. Before trawling, significant relationships could be found between L. conchilega and very fine sand fractions, oxygen and nitrate fluxes, macrobenthos densities, and species richness; however, the trawl disturbances from both gears disrupted these connections. Our results suggest a reduced average effect for pulse compared to tickler chain beam trawls for several ecological and biogeochemical characteristics, though their impact was still significant for L. conchilega and associated species. This study also suggests that faunal-mediated ecosystem functions in habitats dominated by L. conchilega may be sensitive to relatively shallow sediment penetration from trawl gears and should be considered when assessing habitat vulnerability.

Abstract Probiotics are live microorganisms that confer several beneficial effects to the host, including enhancement of bone mineralization. However, probiotic action on bone regeneration is not well studied and therefore we analysed various effects of probiotic treatment on the caudal fin regeneration of zebrafish. Morphological analysis revealed an increased regenerated area with shorter and thicker lepidotrichia segments after probiotic treatment. Fourier transform infrared spectroscopy imaging analysis highlighted the distribution of phosphate groups in the regenerated fins and probiotic group showed higher amounts of well-crystallized hydroxyapatite. At the midpoint (5 days post amputation) of regeneration, probiotics were able to modulate various stages of osteoblast differentiation as confirmed by the upregulation of some key marker genes such as runx2b, sp7, col10a1a, spp1 and bglap, besides suppressing osteoclast activity as evidenced from the downregulation of ctsk. Probiotics also caused an enhanced cell cycle by regulating the expression of genes involved in Retinoic acid (rarga, cyp26b1) and Wnt/β-catenin (ctnnb1, ccnd1, axin2, sost) signaling pathways, and also modulated phosphate homeostasis by increasing the entpd5a levels. These findings provide new outlooks for the use of probiotics as a prophylactic treatment in accelerating bone regeneration and improving skeletal health in both aquaculture and biomedical fields.

Oxidative stress (OS) is implicated in several chronic diseases. Extra-cellular superoxide dismutase (ec-SOD) catalyses the dismutation of superoxide anions with a protective role in endothelial cells. In chronic kidney disease (CKD), OS and thyroid dysfunction (low fT3 syndrome) are frequently present, but their relationship has not yet been investigated. This cohort study evaluated ec-SOD activity in CKD patients during haemodialysis, divided into “acute haemodialytic patients” (AH, 1–3 months of treatment) and “chronic haemodialytic patients” (CH, treated for a longer period). We also evaluated plasmatic total antioxidant capacity (TAC) and its relationships with thyroid hormones. Two basal samples (“basal 1”, obtained 3 days after the last dialysis; and “basal 2”, obtained 2 days after the last dialysis) were collected. On the same day of basal 2, a sample was collected 5 and 10 min after the standard heparin dose and at the end of the procedure. The ec-SOD values were significantly higher in CH vs. AH in all determinations. Moreover, the same patients had lower TAC values. When the CH patients were divided into two subgroups according to fT3 levels (normal or low), we found significantly lower ec-SOD values in the group with low fT3 in the basal, 5, and 10 min samples. A significant correlation was also observed between fT3 and ec-SOD in the basal 1 samples. These data, confirming OS and low fT3 syndrome in patients with CKD, suggest that low fT3 concentrations can influence ec-SOD activity and could therefore potentially contribute to endothelial oxidative damage in these patients.

The return of war to Europe with the Russo-Ukrainian conflict generated mental health effects even in countries not directly involved in the war. The present study describes the Italian adaptation and validation of the Fear of War Scale (FOWARS), i.e., a 13-item Likert scale built by a Romanian research team and exploring the fear of war. For the Italian adaptation, a sample of 150 young Italian adults (aged 18–30, M = 21.7; SD = 2.2) has been collected. Exploratory Factor Analysis conducted with PCA confirmed the bifactorial structure of the scale and detected two dimensions, i.e., the “Physiological dimension of fear” and the “Experiential dimension of fear”. The results of the Confirmatory Factor Analysis show adequate goodness of fit and the last version of the scale, consisting of 12 items, shows good internal consistency and convergent and discriminant validity. Positive significant correlations with the Worry Domains Questionnaire (WDQ) and the Depression Anxiety and Stress Scale (DASS-21) were also found. Moreover, results from ANOVA display significant differences between men and women, with the latter showing higher values of fear of war. Finally, t-test analyses highlight the impact of the fear of war on Italian young adults’ mental health and worry. The Italian adaptation of FOWARS has good overall psychometric properties and can be used to explore the fear of war in the Italian young adult population to highlight the psychological impact of war and its relationship with mental health.

Cellular senescence is closely linked to endothelial dysfunction, a key factor in age-related vascular diseases. Senescent endothelial cells exhibit a proinflammatory phenotype known as SASP, leading to chronic inflammation (inflammaging) and vascular impairments. Albeit in a state of permanent growth arrest, senescent cells paradoxically display a high metabolic activity. The relationship between metabolism and inflammation is complex and varies across cell types and senescence inductions. While some cell types shift towards glycolysis during senescence, others favor oxidative phosphorylation (OXPHOS). Despite the high availability of oxygen, quiescent endothelial cells (ECs) tend to rely on glycolysis for their bioenergetic needs. However, there are limited data on the metabolic behavior of senescent ECs. Here, we characterized the metabolic profiles of young and senescent human umbilical vein endothelial cells (HUVECs) to establish a possible link between the metabolic status and the proinflammatory phenotype of senescent ECs. Senescent ECs internalize a smaller amount of glucose, have a lower glycolytic rate, and produce/release less lactate than younger cells. On the other hand, an increased fatty acid oxidation activity was observed in senescent HUVECs, together with a greater intracellular content of ATP. Interestingly, blockade of glycolysis with 2-deoxy-D-glucose in young cells resulted in enhanced production of proinflammatory cytokines, while the inhibition of carnitine palmitoyltransferase 1 (CPT1), a key rate-limiting enzyme of fatty acid oxidation, ameliorated the SASP in senescent ECs. In summary, metabolic changes in senescent ECs are complex, and this research seeks to uncover potential strategies for modulating these metabolic pathways to influence the SASP.

Intertidal coastal sediments are important centers for nutrient transformation, regeneration, and storage. Sediment resuspension, due to wave action or tidal currents, can induce nutrient release to the water column and fuel primary production. Storms and extreme weather events are expected to increase due to climate change in coastal areas, but little is known about their effect on nutrient release from coastal sediments. We have conducted in-situ sediment resuspension experiments, in which erosion was simulated by a stepwise increase in current velocities, while measuring nutrient uptake or release in field flumes positioned on intertidal areas of a tidal bay (Eastern Scheldt) and an estuary (Western Scheldt). In both systems, the water column concentration of ammonium (NH4+) and nitrite (NO2−) increased predictably with greater erosion as estimated from pore water dilution and erosion depth. In contrast, the phosphate (PO43−) dynamics were different between systems, and those of nitrate (NO3−) were small and variable. Notably, sediment resuspension caused a decrease in the overlying water PO43− concentration in the tidal bay, while an increase was observed in the estuarine sediments. Our observations showed that the concentration of PO43− in the water column was more intensely affected by resuspension than that of NH4+ and NO2−. The present study highlights the differential effect of sediment resuspension on nutrient exchange in two contrasting tidal coastal environments.

Estrogen receptor α (ERα) action plays an important role in pancreatic β-cell function and survival; thus, it is considered a potential therapeutic target for the treatment of type 2 diabetes in women. However, the mechanisms underlying the protective effects of ERα remain unclear. Because ERα regulates mitochondrial metabolism in other cell types, we hypothesized that ERα may act to preserve insulin secretion and promote β-cell survival by regulating mitochondrial-endoplasmic reticulum (EndoRetic) function. We tested this hypothesis using pancreatic islet-specific ERα knockout (PERαKO) mice and Min6 β-cells in culture with Esr1 knockdown (KD). We found that Esr1-KD promoted reactive oxygen species production that associated with reduced fission/fusion dynamics and impaired mitophagy. Electron microscopy showed mitochondrial enlargement and a pro-fusion phenotype. Mitochondrial cristae and endoplasmic reticulum were dilated in Esr1-KD compared with ERα replete Min6 β-cells. Increased expression of Oma1 and Chop was paralleled by increased oxygen consumption and apoptosis susceptibility in ERα-KD cells. In contrast, ERα overexpression and ligand activation reduced both Chop and Oma1 expression, likely by ERα binding to consensus estrogen-response element sites in the Oma1 and Chop promoters. Together, our findings suggest that ERα promotes β-cell survival and insulin secretion through maintenance of mitochondrial fission/fusion-mitophagy dynamics and EndoRetic function, in part by Oma1 and Chop repression.

Coffee is considered one of the most consumed beverages in the world, while reviewed evidence regarding its capacity to modulate biomarkers of oxidative stress and inflammation remains limited. The current study made use of prominent electronic databases, including PubMed, Google Scholar, and Scopus to retrieve information from randomized controlled trials reporting on any association between coffee consumption and modulation of biomarkers of oxidative stress and inflammation in healthy individuals or those at increased risk of developing CVD. The consumption of food-derived products, including the regular intake of pepper, is increasingly evaluated for its potential benefits in protecting against diverse metabolic complications. The current study made use of prominent electronic databases including PubMed, Google Scholar, and Scopus to retrieve clinical evidence linking the intake of black and red pepper with the amelioration of metabolic complications. The findings summarize evidence supporting the beneficial effects of black pepper (Piper nigrum L.), including its active ingredient, piperine, in improving blood lipid profiles, including reducing circulating levels of total cholesterol, low-density lipoprotein cholesterol, and triglycerides in overweight and obese individuals. The intake of piperine was also linked with enhanced antioxidant and anti-inflammatory properties by increasing serum levels of superoxide dismutase while reducing those of malonaldehyde and C-reactive protein in individuals with metabolic syndrome. Evidence summarized in the current review also indicates that red pepper (Capsicum annum), together with its active ingredient, capsaicin, could promote energy expenditure, including limiting energy intake, which is likely to contribute to reduced fat mass in overweight and obese individuals. Emerging clinical evidence also indicates that pepper may be beneficial in alleviating complications linked with other chronic conditions, including osteoarthritis, oropharyngeal dysphagia, digestion, hemodialysis, and neuromuscular fatigue. Notably, the beneficial effects of pepper or its active ingredients appear to be more pronounced when used in combination with other bioactive compounds. The current review also covers essential information on the metabolism and bioavailability profiles of both pepper species and their main active ingredients, which are all necessary to understand their potential beneficial effects against metabolic diseases.

BackgroundVitamin C is one of the most consumed dietary compounds and contains abundant antioxidant properties that could be essential in improving metabolic function. Thus, the current systematic review analyzed evidence on the beneficial effects of vitamin C intake on cardiovascular disease (CVD)-related outcomes in patients with diabetes or metabolic syndrome.MethodsTo identify relevant randomized control trials (RCTs), a systematic search was run using prominent search engines like PubMed and Google Scholar, from beginning up to March 2022. The modified Black and Downs checklist was used to assess the quality of evidence.ResultsFindings summarized in the current review favor the beneficial effects of vitamin C intake on improving basic metabolic parameters and lowering total cholesterol levels to reduce CVD-risk in subjects with type 2 diabetes or related metabolic diseases. Moreover, vitamin C intake could also reduce the predominant markers of inflammation and oxidative stress like C-reactive protein, interleukin-6, and malondialdehyde. Importantly, these positive outcomes were consistent with improved endothelial function or increased blood flow in these subjects. Predominantly effective doses were 1,000 mg/daily for 4 weeks up to 12 months. The included RCTs presented with the high quality of evidence.ConclusionClinical evidence on the beneficial effects of vitamin C intake or its impact on improving prominent markers of inflammation and oxidative stress in patients with diabetes is still limited. Thus, more RCTs are required to solidify these findings, which is essential to better manage diabetic patients at increased risk of developing CVD.

Bottom trawling in shelf seas can occur more than 10 times per year for a given location. This affects the benthic metabolism, through a mortality of the macrofauna, resuspension of organic matter from the sediment, and alterations of the physical sediment structure. However, the trawling impacts on organic carbon mineralization and associated processes are not well known. Using a modelling approach, the effects of increasing trawling frequencies on early diagenesis were studied in five different sedimentary environments, simulating the effects of a deeper-penetrating gear (e.g. a tickler chain beam trawl) versus a shallower, more variable penetrating gear (e.g. an electric pulse trawl). Trawling events strongly increased oxygen and nitrate concentrations in surface sediment layers and led to significantly lower amounts of ammonium (43 %–99 % reduction) and organic carbon in the top 10 cm of the sediment (62 %–96 % reduction). As a result, total mineralization rates in the sediment were decreased by up to 28 %. The effect on different mineralization processes differed both between sediment types and between trawling frequencies. The shallow-penetrating gear had a slightly smaller effect on benthic denitrification than the deeper-penetrating gear, but there were no statistically different results between gear types for all other parameters. Denitrification was reduced by 69 % in a fine sandy sediment, whereas nitrogen removal nearly doubled in a highly eutrophic mud. This suggests that even relatively low penetration depths from bottom fishing gears generate significant biogeochemical alterations. Physical organic carbon removal through trawl-induced resuspension of sediments, exacerbated by a removal of bioturbating macrofauna, was identified as the main cause of the changes in the mineralization process.

Abstract Statins lower the risk of cardiovascular events but have been associated with mitochondrial functional changes in a tissue-dependent manner. We investigated tissue-specific modifications of mitochondrial function in liver, heart and skeletal muscle mediated by chronic statin therapy in a Göttingen Minipig model. We hypothesized that statins enhance the mitochondrial function in heart but impair skeletal muscle and liver mitochondria. Mitochondrial respiratory capacities, citrate synthase activity, coenzyme Q10 concentrations and protein carbonyl content (PCC) were analyzed in samples of liver, heart and skeletal muscle from three groups of Göttingen Minipigs: a lean control group (CON, n = 6), an obese group (HFD, n = 7) and an obese group treated with atorvastatin for 28 weeks (HFD + ATO, n = 7). Atorvastatin concentrations were analyzed in each of the three tissues and in plasma from the Göttingen Minipigs. In treated minipigs, atorvastatin was detected in the liver and in plasma. A significant reduction in complex I + II-supported mitochondrial respiratory capacity was seen in liver of HFD + ATO compared to HFD (P = 0.022). Opposite directed but insignificant modifications of mitochondrial respiratory capacity were seen in heart versus skeletal muscle in HFD + ATO compared to the HFD group. In heart muscle, the HFD + ATO had significantly higher PCC compared to the HFD group (P = 0.0323). In the HFD group relative to CON, liver mitochondrial respiration decreased whereas in skeletal muscle, respiration increased but these changes were insignificant when normalizing for mitochondrial content. Oral atorvastatin treatment in Göttingen Minipigs is associated with a reduced mitochondrial respiratory capacity in the liver that may be linked to increased content of atorvastatin in this organ.
