In this study, a monoclonal antibody with selectivity for clopidol was produced based on a new hapten. The 50% inhibition concentration (IC50) for the developed ic-ELISA was 0.275 ng/mL. Cross-reactivity tests showed that the monoclonal antibody had high specificity for clopidol. Using this antibody, a lateral-flow colloidal gold immunoassay (LCGA) was created for the rapid screening of clopidol in raw chicken samples. With the naked eye, the cut-off value of the strip assay was 5 μg/kg for clopidol in chicken samples. In this study, we developed a monoclonal antibody against picoxystrobin (PIC). After cell fusion, four cell lines (3C2, 4D1, 4G4 and 5D4) capable of producing monoclonal antibodies against oxystrobin were obtained. The monoclonal antibody produced by the cell line 3C2 was the most sensitive to PIC, and its IC50 value was 0.1 ng/ml. The monoclonal antibody was then used to prepare immunochromatographic strips, and its cut-off values in both phosphate-buffered saline solution and cucumber samples were 50 and 100 ng/ml, respectively. In this study, we produced a specific monoclonal antibody (mAb) against triadimefon (TDF) and triadimenol (TDN). The half-maximal inhibitory concentration for TDF and TDN was 5.25 and 9.98 ng/mL, respectively, with a linear detection range (IC20–IC80) of 1.09–25.35 and 2.29–43.61 ng/mL, respectively. Subsequently, we developed a colloidal gold immunochromatography test strip for the analysis of TDF and TDN in tomato. The results obtained from the colloidal gold immunochromatography test strip were in accordance with those obtained from the indirect competitive enzyme-linked immunosorbent assay. The medicinal properties and toxicity of aconitine in traditional Chinese medicine have caused widespread concern. In this study, we screened a highly specific and sensitive monoclonal antibody with an IC50 of 3.65 ng/mL generated by immunizing mice and cell fusion, and the limits of detection of the ic-ELISA were 1.13 and 11.76 ng/mL. We then developed a colloidal gold test strip based on the antibody. For rapid detection of aconitine in herbs, the visual limit and detection limit of the test strip are 10 and 25 ng/mL, respectively; the results are obtained within 5 min and are visible to the naked eye. Praziquantel (PZQ) is a broad-spectrum antiparasitic drug used in mammals and fish. In this study, we developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) for the detection of PZQ residues. The hapten PZQ-HS produced by introducing an amino group at a benzene ring synthesized the immunogen and the coating antigen with carrier proteins. Highly sensitive monoclonal antibodies (mAbs) were prepared following mouse immunization and cell fusion. Under optimum conditions, the half maximal inhibitory concentration of the cell line 4E9 was 7.4 ng/mL, and the working range was 2.4–22.8 ng/mL. Immunochromatographic strips (ICS) were developed for the rapid detection of PZQ residues in mackerel. The reliability of the ICS assay was verified with PZQ-negative mackerel and confirmed by HPLC/MS. The cutoff value for ICS was 50 ng/mL PZQ. A monoclonal antibody (mAb), 2C9, selective for four eugenol compounds, was developed based on the eugenol derivatives (Eug-1). The semi-inhibitory concentrations (IC50) of this antibody against eugenol, isoeugenol, methyl eugenol, and methyl isoeugenol were 0.043, 0.302, 0.016, and 0.082 μg/mL, respectively, and the detection limits (LOD) were 0.012, 0.070, 0.004 and 0.024 μg/mL. With the exception of isoeugenol, which had cross-reactivity of less than 15%, the cross-reactivity of the other compounds was good, ranging from 52.4% to 269%. In the ic-ELISA recovery test, recovery was between 80% and 94%, indicating that the method was suitable for analysis of eugenol compounds in water. The cut-off values of the immunochromatographic strips for the four compounds in water were 5, 100, 100, and 100 μg/mL, respectively, indicating that the ic-ELISA and immunochromatographic strips we developed can be used to rapidly detect eugenol compounds in water. In this study, we prepared a monoclonal antibody against pyrimethanil, for which we first derived its hapten according to the structural formula of pyrimethanil; we used indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) to screen positive mice for cell fusion; finally, we developed five cell lines (1A5, 1E7, 3A6, 3H12 and 5D4) that produced monoclonal antibodies to pyrimethanil. The monoclonal antibody produced by each cell was detected by ic-ELISA, and the monoclonal antibody produced by the 1E7 cell strain was found to be the most sensitive to pyrimethanil. The IC50 value of this antibody was 4 ng/ml. An ultrasensitive monoclonal antibody-based gold nanoparticle immunochromatographic strip assay and indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) were developed to detect florfenicol (FF) and thiamphenicol (TAP) in egg samples. The ic-ELISA, with optimized pH, methanol content and sodium chloride content, exhibited an IC50 value of 0.2 ng/mL for FF and 0.27 ng/mL for TAP, with the working range of 0.05-0.77 and 0.05-1.42 ng/mL, respectively. The optimized ic-ELISA showed negligible cross-reactivity with other phenols and broad-spectrum antibiotics. The recoveries in egg samples using the ic-ELISA ranged from 84% to 115% with a coefficient of variation of less than 5%. Based on this monoclonal antibody, a rapid and ultrasensitive immunochromatographic strip assay was developed with a cutoff value of 1 ng/mL for FF and TAP. Flunixin meglumine (FM) is a novel nonsteroidal anti-inflammatory drug for animals, which has antipyretic, analgesic, and anti-inflammatory effects. The drug, which was originally used to relieve inflammation in horses, musculoskeletal disorders, and pain, has been approved for use in endotoxemia, infectious diseases in swine, etc. A sensitive anti-FM monoclonal antibody 2H4 was prepared and used to develop an indirect competitive enzyme-linked immunosorbent assay and immunochromatographic strip assay for the detection of FM in milk. The complete antigen and coating antigen were conjugated with bovine serum albumin and ovalbumin, respectively. The monoclonal antibody 2H4, with a half inhibition concentration of 0.29 ng/mL, had a limit of detection of 0.432 ng/mL and a linear range of detection of 0.08664-0.97226 ng/mL. Vitamin B2 (riboflavin) is a water-soluble vitamin that has important roles in human health. In this study, we developed a sensitive and specific monoclonal antibody-based indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and lateral-flow immunochromatographic assay (ICA) strip for the rapid detection of vitamin B2. Following routine fusion and selection, the optimum monoclonal antibody against vitamin B2 was obtained. The 50% inhibitory concentration and limit of detection of ic-ELISA were 8.18 and 1.80 ng/mL, respectively. The cut-off value of the lateral-flow ICA strip was 50 ng/mL. A sensitive and specific anti-citrinin (anti-CIT) monoclonal antibody 1F2 was obtained following immunization and cell fusion. An indirect competitive enzyme-linked immunosorbent assay was developed with a 50% inhibitory concentration of 0.761 ng/mL and a limit of detection of 0.089 ng/mL. The recovery rates for CIT-spiked cereals (maize, wheat, and rice) ranged from 112% to 123%. A lateral-flow immunochromatographic assay was developed for both semi-quantitative and quantitative detection. With CIT-spiked cereals, the visual limit of detection was 8 ng/g and the cut-off value was 40 ng/g (semi-quantitative analysis with naked-eye detection). Using a strip scan reader, the calculated limit of detection was 1.28–1.8 ng/g for different CIT-spiked cereals. The recovery rates ranged from 110% to 127%. Red 2G (R2G), a cheap industrial colorant, cannot be added to food. An anti-R2G monoclonal antibody (mAb) was prepared by immunizing mice with the conjugate of R2G hapten and protein, which based on the 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) method. Indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and colloidal gold-based immunochromatographic assay (CG-ICA) methods were used to determine R2G in fruit drinks, red wine, and yoghurts. A standard curve of the developed ic-ELISA showed that the IC 50 of the anti-R2G mAb was 1.02 ng/mL, and limit of detection value (LOD) was 0.21 ng/mL. For the CG-ICA developed, the visual limit of detection values (vLOD) were 2 ng/mL and cut-off values of 100 ng/mL in samples. A sensitive monoclonal antibody, 3G10, derived from a mouse immunized with an immunogen of salbutamol hapten conjugated to keyhole limpet hemocyanin was developed against β-agonists, with a view to establishing a rapid gold nanoparticle immunochromatographic strip. The antibody displayed a 50% inhibitory concentration (IC50) of 1.04 ng/mL for salbutamol and simultaneously detected nine other β-agonists (clenbuterol, cimatero, brombuterol, mabuterol, terbutaline, clenpenterol, carbuterol, mapenterol, pirbuterol) with uniform cross-reactivity. The immunochromatographic strip had cut-off values of 5 ng/mL for salbutamol and clenbuterol, 20 ng/mL for cimaterol, brombuterol and mabuterol, and 50 ng/mL for terbutaline, clenpenterol, carbuterol, mapenterol and pirbuterol. In this study, we developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and an immunochromatographic strip assay based on a monoclonal antibody (mAb) against methylmercury (MeHg) to detect the presence of MeHg in tap water. Under optimum conditions (pH 8.0, 0.8% NaCl, and 0.1% Tween 20), the 50% half maximal inhibitory concentration (IC50) and limit of detection (LOD) were 16.64 and 2.03 ng/mL, respectively. The anti-MeHg mAb was specific to mercury with no cross-reactivity with other metal ions. The cut-off value of the immunochromatographic strip assay was 500 ng/mL for semi-quantitative detection, and the LOD was 11.3 ng/mL for quantitative detection. The average recovery rates of the ic-ELISA and immunochromatographic strip assay were 98.13% and 107.87%, respectively, in tap water. The misuse and illegal use of fluoroquinolones (FQs) in animal-based food products have drawn considerable attention in several countries. As a result, there has been an increased demand for efficient detection methods of FQs in food products. In this study, we developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and immunochromatographic strip device based on a monoclonal antibody against lomefloxacin (LFLX), a second generation FQ. Ic-ELISA had an IC50value of 0.19 ng/mL with a limit of detection of 0.04 ng/mL in 0.01 M phosphate-buffered saline (PBS). The intra- and inter-assay recovery rates of LFLX in bovine milk samples were 98.02–107.40% and 100.65–107.82%. The immunochromatographic assay of LFLX in PBS and spiked bovine milk samples had visual cutoff values at 1 and 5.0 ng/mL, respectively, with significant cross-reactivity with norfloxacin and enoxacin. In this study, we developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and an immunochromatographic assay to detect the furazolidone metabolite, 3-amino-2-oxazolidinone (AOZ). The detection of AOZ was based on the AOZ derivative 3-([2-nitrophenyl]methyleneamino]-2-oxazolidinone (2-NPAOZ). 3-([(3-Carboxyphenyl)methylene]-amino)-2-oxazolidinone (2-CPAOZ) was used as the immunizing hapten and a conjugate of AOZ and glyoxylic acid (AOZ-A) was used as the coating hapten. The monoclonal anti-2-NPAOZ antibody, 5G12, was generated with a half-inhibitory concentration (IC50) of 0.2 ng/mL and a line of 0.06–0.66 ng/mL. Its cross reactivity with other analogues was less than 8%. Spiked catfish samples (0.1, 0.2, and 0.5 μg/kg) were analyzed with the proposed system. The ic-ELISA showed a recovery range of 86.2–118.5% and the intra-assay and inter-assay coefficients of variation ranged from 4.3% to 9.4%. Under the optimum assay conditions, the immunochromatographic assay had a visual cut-off value of 0.5 μg/kg in catfish samples. Both methods can be used to detect AOZ in catfish samples. A sensitive monoclonal antibody, 1H2, was generated in this study that simultaneously recognizes avermectin and ivermectin. An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) was developed, with 50% inhibitory concentrations of 0.491 ng/mL for avermectin and 0.770 ng/mL for ivermectin. A lateral-flow immunochromatographic assay (ICA) strip was also developed based on this monoclonal antibody for the semiquantitative and quantitative detection of avermectin and ivermectin. The semiquantitative (with the naked eye) analysis had visual limits of detection of 10 ng/mL for avermectin and 25 ng/mL for ivermectin, with cut-off values of 25 and 50 ng/mL, respectively. Using a strip scan reader, the quantitative results had calculated limits of detection of 1.3 ng/mL for avermectin and 2.9 ng/mL for ivermectin. In an analysis of raw milk samples, the recovery rates for the ic-ELISA ranged from 94% to 112% and for the ICA strip from 110% to 125%. A highly sensitive monoclonal antibody (mAb) 3H4 against vancomycin (VAN) was prepared. Indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and lateral-flow immunochromatographic assay (ICA) were developed based on the mAb. The 50% inhibition concentration (IC50) value and limit of detection (LOD) value of ic-ELISA method for vancomycin were 0.59 and 0.06 ng/mL, and for norvancomycin were 1.51 and 0.13 ng/mL under optimized conditions as pH 7.4, 0.4% (m/v) NaCl, and 5% (v/v) acetonitrile. In lateral-flow ICA, the visual limit of detection (vLOD) value and cut-off values for vancomycin were 1 and 2.5 ng/mL, and for norvancomycin were 5 and 10 ng/mL under optimized conditions as pH 8.6 with 1 mg/mL coating antigen and 1 µg/mL gold nanoparticle-labeled mAb. In raw milk and animal feed samples, recovery rates from ic-ELISA ranged from 89.2% to 121.6%. The vLOD and cut-off value were 5–10 ng/g and 100–200 μg/kg, respectively. Sensitive and specific anti-sterigmatocystin (STG) monoclonal antibody (mAb) 4G10 was obtained by immunization and cell fusion. An indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) method and lateral-flow immunochromatographic assay (ICA) strip method were developed for the detection of STG in cereal products based on this mAb. The 50% inhibition concentration and limit of detection for the ic-ELISA method were 0.092 and 0.015 ng/mL, respectively. The visual limit of detection (vLOD) and cut-off value for the lateral-flow ICA strip method were 0.1 and 0.5 ng/mL, respectively. From the analysis of different cereal samples (wheat, maize and rice), the recovery rates ranged from 78.3% to 122.0% for the ic-ELISA method. For the lateral-flow ICA strip, the vLODs were 3, 1.2 and 3 ng/g, and the cut-off values were 12, 6 and 6 ng/g for wheat, maize and rice, respectively. Hexestrol (HES) and Diethylstilbestrol (DES) are synthetic hormones, which have been found abuse use in animal-origin food production. We developed an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and immune-chromatographic strip to detect these compounds based on monoclonal antibody. Under optimized conditions, the half-inhibition concentration (IC50) values of ic-ELISA were 0.15 µgL−1and 0.23 µgL−1for HES and DES, respectively. Spiked milk samples were analyzed. The recovery of both synthetic hormones in the milk samples was 60.48–102.19% (HES) and 89.34–100.16% (DES). A sensitive indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) method and a gold nanoparticle immunochromatographic strip were developed to detect amantadine (AM) in foods. A novel AM hapten was prepared, and a sensitive monoclonal antibody against AM was developed. The optimum ic-ELISA conditions included a pH of 7.4 and an ionic strength of 1.6% with no organic solvents. The half-inhibition concentration (IC50) value of ic-ELISA was 1.92 ng/ml, with a limit of detection of 0.62 ng/ml. The immunochromatographic test strip method had a visual cut-off value at 5 μg/kg. Chicken samples were spiked with three concentrations of AM (1, 2, and 5 μg/kg) and analysed by ic-ELISA. The use of ribavirin (RBV) as an antiviral drug for livestock has been prohibited in China, the USA, and many other countries. In this study, we developed a rapid and sensitive indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and a gold nanoparticle immunochromatographic (ICA) strip test for detecting RBV in chicken muscles. Under the optimum assay conditions, where the assay employed phosphate-buffered saline at pH 7.4, no acetonitrile, and an ionic strength of 0.8%, the quantitative working range was 1.43–26.47 ng/ml with an IC50of 6.15 ng/ml. The recovery rate for RBV in real samples ranged from 82.1% to 112.3%. The immunochromatographic test strip method had a visual cutoff value of 50 μg/kg. A novel synthesis method of methyl-3-quinoxaline-2-carboxylic acid (MQCA) and the preparation of its hapten were described. MQCA was synthesised involving in two steps with a high purity. For improving the sensitivity of detection, five different haptens were synthesised and corresponding immunogens and coating antigens were prepared. After comparing the sensitivity of immunoassay with different pairs of antibody and coating antigen, a specific immunoassay was obtained using an antibody raised against hapten (four-atom spacer arm) - BSA and a suitable coating antigen with a heterologous spacer arm group. A class-specific polyclonal antibody against fluoroquinolones was obtained by immunising rabbits with a novel designed hapten, which preserved the essential group of fluoroquinolones. With this specific polyclonal antibody, 13 fluoroquinolones antibiotics were simultaneously detected in indirect competitive enzyme-linked immunosorbent assay (ic-ELISA). The established ic-ELISA was class-specific to detect 13 fluoroquinolones such as norfloxacin, ciprofloxacin, ofloxacin etc., and the limit of detection (LOD) desirable was (0.01-10 ng/ml) and the acceptable recovery range was between 62 and 95%. In this study, a carboxyl group was introduced into the isoprocarb molecule to obtain an isoprocarb hapten, which was then coupled with a protein to obtain an artificial antigen. Three monoclonal antibody cell lines, 1D11, 6E6 and 1B5, were finally obtained by mouse immunization, cell fusion and subcloning, and the antibody produced by cell line 1B5 had the best affinity and sensitivity. The monoclonal antibody was highly sensitive and specific for isoprocarb, with an IC50 of 2.09 ng/ml and a cross‐reactivity rate of <0.21%. By optimizing the indirect competitive (ic)‐ELISA, the optimal conditions were determined to be pH 7.4, 0% methanol and 0.8% NaCl, the limit of detection value was 0.23 ng/ml, and the linear range of the ic‐ELISA was 0.46–9.62 ng/ml. The recovery rate of the isoprocarb cucumber sample was 97–99% for the ic‐ELISA method. In addition, we successfully developed an immunochromatographic test strip for the detection of isoprocarb residues. The cutoff values in phosphate‐buffered saline and cucumber extract were 10 and 25 ng/ml, respectively. Both methods met the requirements for isoprocarb residue detection in agricultural products, and can be used for semiquantitative and qualitative analysis of isoprocarb in vegetables. An anti-phenolphthalein monoclonal antibody (mAb) was prepared based on the N,N′-Carbonyldiimidazole (CDI) method through phenolphthalein conjugated with proteins. Indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) and colloidal gold-based immunochromatographic assay (ICA) methods were used to determine phenolphthalein in slimming products. A standard curve was established, and the IC 50 and limit of detection of ic-ELISA were 0.95 and 0.10 ng/mL with a linear detection range of 0.27–3.37 ng/mL. We developed a highly sensitive and specific monoclonal antibody (mAb) for ketoconazole (KCZ) detection in chicken samples. From indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) analysis, the 50% inhibitory concentration of the mAb was 0.7 ng/mL, and the cross-reactivity with fluconazole, imidazole, itraconazole, and voriconazole was <1%. Using indirect competitive enzyme-linked immunosorbent assay, spiked sample recovery was 92.5%–102.7%. Under optimal conditions, we also developed a colloidal gold immunochromatographic assay (CGIA) to detect KCZ residues in chicken samples, with a visual limit of detection of 0.5 ng/g and a cut-off value of 20 ng/g. When data were assessed using liquid chromatography with tandem mass spectrometry, no significant differences were found between assays. Also, strip results were revealed in 10 min.
