Superparamagnetic iron oxide (SPIO) nanoparticles have ideal properties to become a targeted magnetic drug delivery contrast probes, named theranostics. We worked with SPIO condensed colloidal nanocrystal clusters (MagAlg) prepared through a new soft biomineralization route in the presence of alginate as the polymeric shell and loaded with doxorubicin (DOX). The aim of this work was to study the in vitro cytotoxicity of these new MagAlg–DOX systems on mouse fibroblast and breast carcinoma cell lines. For proper analysis and understanding of cell behavior after administration of MagAlg–DOX compared with free DOX, a complex set of in vitro tests, including production of reactive oxygen species, comet assay, cell cycle determination, gene expression, and cellular uptake, were utilized. It was found that the cytotoxic effect of MagAlg–DOX system is delayed compared to free DOX in both cell lines. This was attributed to the different mechanism of internalization of DOX and MagAlg–DOX into the cells, together with the fact that the drug is strongly bound on the drug nanocarriers. We discovered that nanoparticles can attenuate or even inhibit the effect of DOX, particularly in the tumor MCF7 cell line. This is a first comprehensive study on the cytotoxic effect of DOX-loaded SPIO compared with free DOX on healthy and cancer cell lines, as well as on the induced changes in gene expression. The aim of the present work was to study and identify the causes and sources of defects in the glazes and decorations of archaeological ceramics containing a calcium component. As part of systematic archaeological research in the Hradcany area, several reconstructable objects and some fragmentary sherds of archaeological ceramics with a relatively high calcium content were found. Seven objects of faience were rescued from the waste pits on the premises of the third courtyard of the Prague Castle, a flower pot was found in Lumbe garden, and creamware sherds were excavated from a waste pit between the Salm and Schwarzenberg palaces on Hradcany Square. The first step of the research was the evaluation of ceramic bodies, glazes, and decorations using archaeometric methods. Subsequently, dilatometric measurements of ceramic bodies and stress state calculations of ceramic body-glaze, or glaze-decor systems were used to identify the sources of defects observed on archaeological finds. Both primary and secondary defects were identified in all studied finds. Among the primary defects, pinholes and blisters predominated. As secondary defects, crazing, blackening, corrosion, delayed peeling, and mechanical abrasion were recognised. Thanks to the combination of X-ray and optical methods, together with the determination of the stress states of the two- and three-component systems, causes of secondary defects were described. The main source of these defects were calcium residues causing volume changes in the ceramic bodies.

Postural disorders are the current problems of the present time and can be monitored in each age group and today they are not rare even among pre-schoolers. Untreated postural disorders can cause spinal deformity, pain, neurological symptoms or disorders of movement patterns. It is therefore extremely important to monitor and assess their occurrence as early as in pre-school age, when posture is still developing and taking the form and, if irregularities are found, take relevant preventive measures. This is the reason we conducted the assessment of posture among pre-schoolers. Within the project Healthy little back carried out in Ružomberok and Martin we examined 111 pre-schoolers aged 5–6. Their posture was assessed using aspexia. The aim of the examination was to determine the posture of body parts and then assess the overall posture. To assess the posture we used a modified methodology of Napoleon Wolanski. Results: 48.65% pre-schoolers with physiologic posture, 36.94% with initial postural disorder and 14.41% with serious postural disorders. Less than half of pre-schoolers have physiological posture and it is therefore necessary to implement preventive measures and programs in kindergartens to avoid the situation to worsen.

Aim. This study aims to examine, in the Bulgarian context, the relationships between vaccination attitudes and acceptance on the one hand, and use of communication channels, trust in medical authorities, and belief in misleading information on the other. Materials and methods. A cross-sectional study of Bulgarian respondents was conducted between April and May 2022 using a self-administered online survey with 1,200 participants. Logistic regression models were implemented to examine first, the association between conspiracy beliefs and COVID-19 vaccination uptake and second, between susceptibility to misinformation, sociodemographic variables, trust in medical authorities, and the use of information sources. Results. People who agreed with at least one of the conspiratorial statements were 8.56 times more likely to decline immunization. Belief in rumours and conspiracy theories was associated with a higher perceived risk of adverse events and lower perceived benefits of vaccination. In the logistic regression models, age, residence, and mistrust were associated with believing in rumours and conspiracy theories. Sourcing information from health authorities was negatively associated with belief in rumours while getting information from relatives and friends had the opposite effect. Conclusions. Misinformation had a significant impact on vaccine uptake. More efforts are needed to dispel myths and rumours surrounding vaccination. [ABSTRACT FROM AUTHOR]

Background: Racial and ethnic disparities in genomic testing could exacerbate disparities in access to precision cancer therapies and survival-particularly in the context of lung cancer where genomic testing has been recommended for the past decade. However, prior studies assessing disparities in genomic testing have yielded mixed results., Methods: We conducted a systemic review to examine racial and ethnic disparities in the use of genomic testing among lung cancer patients in the United States. Two comprehensive searches in PubMed, Embase, and Scopus were conducted (September 2022, May 2023). Original studies that assessed rates of genomic testing by race or ethnicity were included. Findings were narratively synthesized by outcome., Results: The search yielded 2739 unique records, resulting in 18 included studies. All but 1 study were limited to patients diagnosed with non-small cell lung cancer. Diagnosis years ranged from 2007 to 2022. Of the 18 studies, 11 found statistically significant differences in the likelihood of genomic testing by race or ethnicity; in 7 of these studies, testing was lower among Black patients compared with White or Asian patients. However, many studies lacked adjustment for key covariates and included patients with unclear eligibility for testing., Conclusions: A majority of studies, though not all, observed racial and ethnic disparities in the use of genomic testing among patients with lung cancer. Heterogeneity of study results throughout a period of changing clinical guidelines suggests that minoritized populations-Black patients in particular-have faced additional barriers to genomic testing, even if not universally observed at all institutions., (© The Author(s) 2024. Magnesium complexes of phthalocyanines (Pcs) and their aza-analogues have a great potential in medical applications or fluorescence detection. They are known to demetallate to metal-free ligands in acidic environments, however, detailed investigation of this process and its possible prevention is lacking. In this work, a conversion of lipophilic and water-soluble magnesium complexes of Pcs and tetrapyrazinoporphyrazines (TPyzPzs) to metal-free ligands was studied in relation to the acidity of the environment (organic solvent, water) including the investigation of the role of delivery systems (microemulsion or liposomes) in improvement in their acido-stability. The mechanism of the demetallation in organic solvents was based on an acidoprotolytic mechanism with the protonation of the azomethine nitrogen as the first step and a subsequent conversion to non-protonated metal-free ligands. In water, the mechanism seemed to be solvoprotolytic without any protonated intermediate. The water-soluble magnesium complexes were stable in a buffer with a physiological pH 7.4 while a time-dependent demetallation was observed in acidic pH. The demetallation was immediate at pH < 2 while the full conversion to metal-free ligand was done within 10 min and 45 min for TPyzPzs at pH 3 and pH 4, respectively. Incorporation of lipophilic magnesium complexes into microemulsion or liposomes substantially decreased the rate of the demetallation with the latter delivery system being much more efficient in the protection from the acidic environment. A comparison of two different macrocyclic cores revealed significantly higher kinetic inertness of magnesium TPyzPz complexes than their Pc analogues.

Male infertility refers to the inability to conceive a natural pregnancy in a fertile female, and approximately 15% of reproductive-aged couples worldwide face this problem. Several plants were used to treat fertility disorders and, among them, Lepidium meyenii, a folk medicament of Andean regions, is still used to enhance vitality and treat sterility in humans and domestic animals. The aim of the study was to evaluate the effects of L. meyenii Walpers on infertile patients by a randomized, double-blind, placebo-controlled trial. Fifty patients suffering from various reproductive-related problems were enrolled for 16 weeks to evaluate the effect of yellow maca on semen quality and serum hormone levels. Treatment with maca improved the percentage of sperm concentration by 40%, whereas the placebo improved by 76% after 8 and 16 weeks of treatment, but the results were statistically non-significant. No statistically significant change in hormone levels was reported by using maca, except a decrease in the level of free testosterone. Results are not sufficient to assess the efficacy of maca on male fertility. Further investigation and trials are required to obtain conclusive results., (© 2021 The Authors. Purpose: Determine the feasibility and utility of using a battery of tests utilized, so far, to assess neurological-cognitive functions in the typical adult population and identify the spectrum of these functions in adult SBM patients., Methods: Prospective study in which 15 participants (mean age = 28.7 ± 8.7 years, range = 19-45 years) completed the targeted battery of tests (n = 5-15/test) previously standardized to the general population. Results were compared with normative data., Results: Statistically significant differences with normative means were noted in the following tests: Montreal Cognitive Assessment (MoCA), Functional Activities Questionnaire (FAQ), and NIH Toolbox Fine Motor (Dexterity and Grip Strength) tests. Cohort means for NIH Toolbox Fluid, Crystallized, and Cognitive Composite Scores and Timed Up and GO (TUG) were not different from normative means., Conclusion: All tests were successfully completed by cohort. Whereas many aspects of cognition were normal, tests assessing visual-constructural, calculation, motor, and fluency functions did show differences from population means. Numerous tests assessing multiple domains are needed and can be used in future aging studies to appreciate the spectrum of cognitive and motor abilities in adults with SBM.

Objective: Aim of our study was to identify the number of existing posture disorders in children in primary schools and to identify the most prevalent position of body segments that cause postural disorders., Methods: The occurrence of postural disorders was analysed in the school year 2016/2017. The sample consisted of 311 pupils age 6-8 from the 16 districts of the Slovak republic. We used Klein, Thomas and Mayer method to evaluate the posture. Flat feet were evaluated according to Napoleon Wolanski scale. The results were verified by using a base rate quantity test. We determined the level of statistical significance at p = 0.05., Results: We observed statistically significant postural disorders in children. Poor posture occurs in more than 50% of the population under the study. The most critical body segments which were subjected to pathology were shoulder blades and shoulders. Pathology in these segments exceeds 80%. Flat feet occurrence was in 65% children. Adverse results were also seen in pathologic position of: pelvis anteversion (46 %), head protruding (42%) and spinal curvature deformities in the sagittal plane (30%). In the frontal plane deformities of spinae reached 13%., Conclusion: Health status of the children's locomotor system, especially postural system is poor. High prevalence of postural disorders in children requires preventive programs such as introducing corrective exercises into physical education and engaging children in sports. Knowledge of the most frequent body segments defects in children can be helpful for professionals in creating adequate corrective programs for children.

Although tau protein was long regarded as an intracellular protein with several functions inside the cell, new evidence has shown tau secretion into the extracellular space. The active secretion of tau could be a physiological response of neurons to increased intracellular amounts of tau during the progression of tau pathology. We looked for potential differences in the serum levels of toxic tau oligomers in regards to cognitive impairment of subjects. We detected tau oligomers in the serum of Alzheimer's disease (AD) patients, but they were also present to some extent in the serum of healthy older subjects where the levels positively correlated with aging (Spearman r = 0.26, p = 0.016). On the contrary, we found lower levels of tau oligomers in the serum of mild cognitive impairment (MCI) (p = 0.033) and MCI-AD (p = 0.006) patients. These results could suggest that clearance of extracellular tau proteins takes place, in part, in the periphery. In the case of MCI patients, the lower levels of tau oligomers could be the result of impaired clearance of tau protein from interstitium to blood and consequent accumulation of tau aggregates in the brain.

The latest therapeutic approaches to Alzheimer disease are using intravenous immunoglobulin (IVIG) products. Therefore, the detailed characterization of target-specific antibodies naturally occurring in IVIG products is beneficial. We have focused on characterization of antibodies isolated against tau protein, a biomarker of Alzheimer's disease, from Flebogamma IVIG product. The analysis of IgG subclass distribution indicated skewing toward IgG3 in anti-tau-enriched IgG fraction. The evaluation of their reactivity and avidity with several recombinant tau forms was performed by ELISA and blotting techniques. Truncated non-phosphorylated tau protein (amino acids 155-421) demonstrated the highest reactivity and avidity index. Background/Aims. Products of Lepidium meyenii Walp. (maca) are touted worldwide as an alimentary supplement to enhance fertility and restore hormonal balance. Enhancing properties of maca on semen parameters in animals were previously reported by various authors, but we present to the best of our knowledge the first double-blind, randomized, placebo-controlled pilot trial in men. The aim of this study was to evaluate the effects of maca on semen parameters and serum hormone levels in healthy adult men. Methods. A group of 20 volunteers aged 20-40 years was supplied by milled hypocotyl of maca or placebo (1.75 g/day) for 12 weeks. Negative controls of semen were compared to the samples after 6 and 12 weeks of maca administration; negative blood controls were compared to the samples after 12 weeks of treatment. Results. Sperm concentration and motility showed rising trends compared to placebo even though levels of hormones did not change significantly after 12 weeks of trial. Conclusion. Our results indicate that maca possesses fertility enhancing properties in men. As long as men prefer to use alimentary supplement to enhance fertility rather than prescribed medication or any medical intervention, it is worth continuing to assess its possible benefits.

Background: Despite the physiological sequestration of amyloid-β (Aβ) peptides by various carriers, interactions between peptides and protein tau appear to be pathological and involved in the development of Alzheimer's disease (AD). A recent study reported increased Aβ-tau interactions in the neurons of AD patients., Objective: We investigated the possibility that levels of Aβ-tau complexes in cerebrospinal fluid could be a prospective biomarker of AD, with greater sensitivity and specificity than Aβ1-42, tau, or phospho-tau individually., Methods: By means of ELISA, we estimated levels of the complexes in 161 people (non-demented controls, people with mild cognitive impairment (MCI), probable AD or other types of dementia)., Results: We found significant reductions in levels in people with MCI due to AD (down to 84.5%) or with AD (down to 80.5%) but not in other types of dementia. The sensitivity of the new biomarker to AD was 68.6%, the specificity 73.3% (compared to controls) or 59.1-66.1% (compared to other types of dementia). No significant correlations were observed between the complexes and the remaining biomarkers or between those and Mini-Mental State Examination score., Conclusion: We suppose that attenuated levels of complexes in cerebrospinal fluid reflect the accumulation of Aβ bound to tau in AD neurons and that changes start many years before symptom onset, analogously to those in Aβ1-42, tau, or phospho-tau. Unfortunately, these complexes are not a significantly better biomarker of AD than current biomarkers.

A new natural diketopiperazine (1) was obtained from the culture broth of Microbispora aerata strain imbas-11A, isolated from penguin excrements collected on the Antarctic Livingston Island. Compound 1 was purified consecutively by solvent extraction, silica gel column chromatography and preparative HPLC. The structure of the compound was elucidated by 1D and 2D NMR experiments and mass spectrometric investigations. The absolute configuration of compound 1 was determined by amino acid analysis and NOESY correlations. A low antiproliferative and cytotoxic effect of trans-cyclo-(D-tryptophanyl-L-tyrosyl) (1) was determined with L-929 mouse fibroblast cells, K-562 human leukemia cells and HeLa human cervix carcinoma. Trans-cyclo-(D-tryptophanyl-L-tyrosyl) (1) did not show antimicrobial activity at a concentration of 50 µg per disc against Bacillus subtilis, Staphylococcus aureus, Streptomyces viridochromogenes, Escherichia coli, Candida albicans and Mucor miehei.

Alzheimer's disease (AD) is the most common type of dementia. In connection with the global trend of prolonging human life and the increasing number of elderly in the population, the AD becomes one of the most serious health and socioeconomic problems of the present. Tau protein promotes assembly and stabilizes microtubules, which contributes to the proper function of neuron. Alterations in the amount or the structure of tau protein can affect its role as a stabilizer of microtubules as well as some of the processes in which it is implicated. The molecular mechanisms governing tau aggregation are mainly represented by several posttranslational modifications that alter its structure and conformational state. Hence, abnormal phosphorylation and truncation of tau protein have gained attention as key mechanisms that become tau protein in a pathological entity. Evidences about the clinicopathological significance of phosphorylated and truncated tau have been documented during the progression of AD as well as their capacity to exert cytotoxicity when expressed in cell and animal models. This paper describes the normal structure and function of tau protein and its major alterations during its pathological aggregation in AD.

Streptomyces sp. strain SB9 was isolated from perm frost soil samples in Spitsbergen, Arctic Ocean; it grows in a temperature range between 4 degrees C and 28 degrees C. During the survey of biologically active metabolites biosynthesized by this strain, significant amounts of alpha,alpha-trehalose (1) and glycerol (2) were detected. The compounds were isolated from the mycelium, were chromatographically separated, and the structures were elucidated on the basis of MS and NMR measurements. A possible role of trehalose in cold adaptation of the strain was examined. It was determined that the mycelium of the strain cultivated at 4 degrees C accumulated 5-fold higher amounts of trehalose in comparison with the cells cultivated at 28 degrees C. The mesofilic reference strains, Streptomyces spectabilis NRRL 2494 and Streptomyces lividans TK64, accumulated 100-fold less trahalose than the psychrotolerant Streptomyces sp. SB9. High amounts of trehalose in the cells could be a reason for adaptation of the strain to life at Arctic conditions.

The fungus, Tritirachium sp. HKI 0317, was isolated from the Antarctic lichen Neuropogon sp. Fermentation of this strain, extraction of the culture broth, and preparative separation of produced compounds furnished 4-carboxy-5,5'-dihydroxy-3,3'-dimethyldiphenylether (1), macrosphelide A (2), and macrosphelide J (3). The structures were elucidated on the basis of MS and NMR measurements, and the previously published data for this compounds.

A new sulphur-containing natural alkaloid named microbiaeratin (1a) was isolated, together with the known microbiaeratinin (2, bacillamide) from the culture filtrate of Microbispora aerata strain IMBAS-11A. The organism was isolated from penguin excrements collected on the Antarctic Livingston Island. The structure was elucidated by one- and two-dimensional NMR experiments and mass spectrometric investigations.

Sanionins A (1) and B (2) were isolated from the moss Sanionia georgico-uncinata, collected on the Antarctic Livingston Island. The compounds 1 and 2 were purified by solvent extraction, silica gel column chromatography, and preparative HPLC, consecutively. The structures of the both compounds were elucidated by 1D and 2D NMR experiments and mass spectrometric investigations. These compounds showed activity against important Gram-positive pathogens, such as mycobacteria, multiresistant staphylococci, and vancomycin resistant enterococci. This activity is combined with antiinflammatoric activity and low cytotoxicity.
