The selection of wheat varieties with high arabinoxylan (AX) levels could effectively improve the daily consumption of dietary fiber. However, studies on the selection of markers for AX levels are scarce. This study analyzed AX levels in 562 wheat genotypes collected from 46 countries using a GWAS with the BLINK model in the GAPIT3. Wheat genotypes were classified into eight subpopulations that exhibited high genetic differentiation based on 31,926 SNP loci. Eight candidate genes were identified, among which those encoding F-box domain-containing proteins, disease resistance protein RPM1, and bZIP transcription factor 29 highly correlated with AX levels. The AX level was higher in the adenine allele than in the guanine alleles of these genes in the wheat collection. In addition, the AX level was approximately 10% higher in 3 adenine combinations than 2 guanine, 1 adenine, and 3 guanine combinations in genotypes of three genes (F-box domain-containing proteins, RPM1, and bZIP transcription factor 29). The adenine allele, present in 97.46% of AX-95086356 SNP, exhibited a high correlation with AX levels following classification by country. Notably, the East Asian wheat genotypes contain high adenine alleles in three genes. These results highlight the potential of these three SNPs to serve as selectable markers for high AX content.

This study aimed to identify the phytochemical compounds in purple wheat bran ‘Arriheuk’ extract, with water, 50% ethanol and 70% ethanol and determine its physiological activities. Six types of flavonoids were identified in ‘Arriheuk’ extract using a HPLC analysis. The highest total polyphenol content was observed in the 70% ethanol extract, while the highest total flavonoid content was obtained both from the water and the 70% ethanol extracts of ‘Arriheuk’. 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity was significantly high in the 48-h water extract and the 12-h 70% ethanol extract, while the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) free radical scavenging activity was not significantly different between the two extracts. Cell viability, determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, was high for cells treated with 70% ethanol extract. The apoptosis protective effect, confirmed by Hoechst staining, showed that the morphological features were significantly reduced in cells treated with the 70% ethanol extract. Oil-Red-O staining was used to confirm the protective effect of ‘Arriheuk’ extracts on the intracellular lipid accumulation in cells, and the results showed the best inhibitory effect was observed at the 70% ethanol extract. However, the effect decreased in a concentration dependent manner. These results may improve the usage of ‘Arriheuk’ as a food material in the future.

A cascade control system comprising the primary and secondary controllers suffers from a cascade-type integral windup problem in which the output saturation of the secondary control loop can considerably increase the integral part of the primary control loop. In this paper, we present a new predictive antiwindup technique that can completely eliminate the possibility of secondary controller output saturation, resulting in no cascade-type integral windup phenomenon. The proposed method does not require any type of process models; thus, its implementation is simple and straightforward, which is a very favorable advantage compared to the model-based antiwindup techniques from the practical viewpoint. Our simulation confirms that the proposed method can completely remove the primary controller's cascade-type integral windup resulting from the saturation of the secondary controller output. Further, the proposed method exhibited good control performance without needing any type of process model for the various types of processes and controllers. Our experimental study successfully demonstrated that there are no problems in applying the proposed method to real plants.

ω-5 gliadin is the major allergen that causes wheat-dependent exercise-induced anaphylaxis (WDEIA). Recently, a missing mutant wheat cultivar at 1B chromosomeSerum samples from 14 WDEIA and 7 classic wheat allergy patients were used to compare the allergenicity of ω5D and wild-type cultivars using immunoglobulin E immunoblotting, enzyme-linked immunosorbent assay (ELISA), and ImmunoCAP inhibition assays.Immunoblotting revealed that ω5D extracts had less sIgE binding to gliadins and glutenins in WDEIA sera than wild-type extracts. Immunoblot inhibition assay for gliadin sIgE reactivity also showed that ω5D gliadins had less allergenicity than wild-type gliadins. ELISA inhibition assay showed stronger allergenicity of gliadins than glutenins, although they had cross-reactivity. This assay also showed that the 50% inhibitory concentrations (IC50) of ω5D extracts against gliadin- or glutenin-sIgE reactivity were approximately 4-fold higher in WDEIA patients than those of wild-type extracts. The inhibition capacity of ω5D gliadins against recombinant ω-5 gliadin-sIgE reactivity was also lower in WDEIA patients than that of wild-type.The allergenicity of the ω5D cultivar is markedly lower for WDEIA patients in the sIgE inhibition tests. These results suggest that the ω5D cultivar may be a safe alternative for WDEIA patients.

An improved continuous-cycling method is proposed for the autotuning of the proportional–integral–derivative (PID) controller. The proposed method can identify the frequency response of the process at a preset phase angle without a modeling error. Moreover, it provides an exact frequency response even if a static disturbance is present. The proposed method is an improved version of the continuous-cycling method. The gain of the proportional controller in the continuous-cycling method is updated to obtain the continuous-cycling status automatically. To guarantee the preset phase angle of the frequency response, we place a phase shifter in the form of a time delay after the proportional controller. The results of simulation and experimental studies show that the proposed method can provide an exact frequency response even under static disturbance conditions and can be applied to real processes.

Selecting wheat with favorable spike characteristics has been a popular breeding strategy to improve sink capacity and yield potential. In the present study, quantitative trait loci (QTLs) for yield potential traits were identified using a recombinant inbred line (RIL) population derived from the cross between Taejoong and Keumkang, two Korean wheat cultivars. A linkage map encompassing a total genetic length of 6544.8 cM was constructed using 838 single nucleotide polymorphisms from the 35K Axiom Wheat Breeder&rsquo, s Array. We detected eight QTLs for four yield potential traits that are consistently identified in at least two of the three environments, that is, one for days to heading date (QDHD-1 on chromosome 7B), three for spike length (QSL-1, QSL-2, and QSL-3 on chromosomes 1D, 5A, and 6A, respectively), one for tiller number (QTN-1 on chromosome 5B), and three for length of center rachis (QLCR-1, QLCR-2, and QLCR-3 on chromosomes 1B, 5B, and 6A, respectively). Notably, Taejoong contributed the alleles for long spike at all three spike length QTLs with the additive effects of 0.6 cm, 0.6 cm, and 0.9 cm at QSL-1, QSL-2, and QSL-3, respectively. No significant two-way or three-way interaction was observed among QSL-1, QSL-2, and QSL-3, indicating that pyramiding the Taejoong alleles at the three QTLs can increase spike length additively. While the Taejoong alleles at QSL-1, QSL-2, and QSL-3 were associated with increased days to heading date, more kernels per spike, and reduced tiller number per plant, the extent of the pleiotropic effects were different among the three QTLs. Due to the limited number of molecular markers and mapping resolution, further work is required to narrow down the identified QTLs and characterize their effects more precisely. Our results would provide useful information for modulating spike characteristics and improving yield potential in wheat breeding programs.

Wheat is frequently exposed to high temperature during anthesis and ripening period, which resulted in yield loss and detrimental end-use-quality. The transcriptome analysis of wheat under high temperature stress during the early stage of the grain filling period was undertaken. Three expression patterns of differentially expressed genes (DEGs) during grain filling period were identified. The DEGs of seed storage protein and starch-branching enzyme showed continuous increases in their expressions during high temperature stress, as well as during the recovery period. The activities of the enzymes responsible for the elimination of antioxidants were significantly affected by exposure to high temperature stress. Only the WAS-2 family genes showed increased transcription levels under high temperature stress in dehulled spikelets. The relative transcription levels for sub-genome specific WAS-2 genes suggested that WAS-2 genes reacted with over-expression under high temperature stress and decreased back to normal expression during recovery. We propose the role of WAS-2 as a protective mechanism during the stage of grain development under high temperature in spikelets.

Fusarium head blight (FHB) caused by Fusarium species is a major disease of wheat and barley around the world. FHB causes yield reductions and contamination of grains with trichothecene mycotoxins including; nivalenol (NIV), deoxynivalenol (DON), 3-acetyldeoxynivalenol (3-ADON), and 15-acetylde-oxynivalenol (15-ADON). The objectives of this study were to identify strains of F. graminearum isolated in Korea from 2012-harvested wheat grain and to test the pathogenicity of these NIV- and DON-producing isolates. Three hundred and four samples of wheat grain, harvested in 2012 in Chungnam, Chungbuk, Gyeongnam, Jeonbuk, Jeonnam, and Gangwon provinces were collected. We recovered 44 isolates from the 304 samples, based on the PCR amplification of internal transcribed spacer (ITS) rRNA region and sequencing. Our findings indicate that F. asiaticum was the predominant (95% of all isolates) species in Korea. We recovered both F. asiaticum and F. graminearum from samples collected in Chungnam province. Of the 44 isolates recovered, 36 isolates had a NIV genotype while 8 isolates belonged to the DON genotype (3-ADON and 15-ADON). In order to characterize the pathogenicity of the strains collected, disease severity was assessed visually on various greenhouse-grown wheat cultivars inoculated using both NIV- and DON-producing isolates. Our results suggest that Korean F. graminearum isolates from wheat belong to F. asiaticum producing NIV, and both F. graminearum and F. asiaticum are not significantly different on virulence in wheat cultivars.

Purpose: This study assessed the quality of commercial Korean domestic wheat flours and their dry noodle-making performance. Methods: Six commercial Korean domestic wheat flours produced from different millers and a commercial all-purpose flour as a control were analyzed for moisture, ash and protein contents, solvent retention capacity (SRC), mixograph, and noodle making performance of dry noodles. Results: The moisture content of the commercial Korean domestic wheat flours varied according to the millers. The ash content of them was significantly higher than that of commercial all-purpose flour. The SRC results showed all Korean domestic flours had much lower lactic acid SRC values and gluten performance index (GPI) than the control. Mixograph data showed that one of the commercial Korean domestic flours (flour D) showed a very narrow bandwidth due to the higher damaged starch and arabinoxylan contribution based on high sodium carbonate and sucrose SRC values. Dry noodles made with commercial Korean domestic wheat flours produced significantly higher turbidity of cooked water and weight gain of cooked noodle than that with the control flour. In addition, the texture of cooked noodles made with commercial Korean domestic wheat flours was less viscoelastic, as illustrated by the less springiness, cohesiveness, gumminess and chewiness, compared to the control. Conclusion: Overall, the dry noodle-making performance of commercial Korean domestic wheat flours was inferior to that of commercial all-purpose flour due to the higher ash content and lower GPI. Lowering milling extraction and application of enzymes for reducing water absorption capacity of flour might improve the dry noodle-making performance.

Previous batch control methods, such as iterative learning control (ILC) or run-to-run (R2R) control, can significantly improve the control performance of the batch process. However, to guarantee the expected good control performance, a fairly accurate process model is required for these controllers. Also, the implementation is numerically complicated so that it is difficult to be applied to real manufacturing processes. To overcome these problems, a new batch proportional-integral-derivative (PID) control method is proposed, which borrows the concept of the conventional PID control method. Simulation studies confirm that the proposed method shows acceptable performance in tracking a setpoint trajectory, rejecting disturbances, and robustness to noises and variation of process dynamics. The application to the commercial batch process of a single crystal grower verifies that the proposed method can significantly contribute to improving the control performances of real batch processes.

This study was conducted to investigate the mixed seeding of 5 forage barley varieties (youngyang, yuyeon, dami, wooho, soman) with 1 malting barley variety (hopum). Yield of the mixed seeding was compared with each single seeding and variety combinations. Yield of mixed seeding combination was higher than each single seeding. In mixed seeding of 'Youngyang' and 'Hopum' (50% and 50% ratio), 'Yuyeon' and 'Hopum' (70% and 30% ratio), 'Dami' and 'Hopum' (50% and 50% ratio), 'Wooho' and 'Hopum' (50% and 50% ratio) were higher yield of dry weight than yield of other mixed ratios. Most of all, mixed seeding of 'Dami' and 'Hopum' were high yield mixed seeding (ratios of 50% each other). In generally, the yields of barley mixed seeding with malting barley 'Hopum' were high production. The forage quality of mixed seeding combination was higher than any each single seeding. 'Youyeon' 50% and 'Hopum' 50% ratio was more higher TDN content (69.2%) than any other mixed combination.

Pre-harvest sprouting (PHS) of wheat reduces grain yield and has deleterious effects on wheat flour quality. PHS resistance is strongly associated with grain color and seed dormancy. Resistance to PHS is a priority of wheat breeding program. We have studied a population of 122 Doubled Haploid (DH) lines from the cross between two cultivars, 'Keumkang' and 'Olgeuru', in order to detect QTLs correlated with PHS resistance. PHS was evaluated using mist spray in the humid chamber (PHS-MS), and the germination index (GI) was determined for two years. The variety 'Keumkang' has white grains and higher percentage of PHS rate (30.2%), while 'Olgeuru' has red grains and lower PHS rate (2.8%). There was significant difference in grain color according to the GI. PHS-MS was positively correlated with GI (r = 0.732, P < 0.001). Two molecular markers, Xbarc12 and Xgwm3, were found correlated with PHS resistance and GI. These markers were mapped on chromosomes 3A and 3D with LOD scores of 4.95 and 4.02, respectively. Xbarc12 explained the highest amount of phenotypic variation (R 2 ) which is 11.99% in GI and 27.25% in PHS. Xgwm3 explained 9.10% of phenotypic variation in GI. The respective value of PHS-MS and GI in Korean wheat cultivars ranged from 1.37% to 86.75% and from 0.03 to 0.73. This study provides useful information for marker-assisted wheat breeding for PHS resistance improvement in Korea.
